An UPLc-MS/MS method for simultaneous determination of multiple constituents in Guizhi Fuling capsule with ultrafast positive/negative ionization switching

Guizhi Fuling capsule(GFc), a traditional chinese medicine(TcM) with effects of promoting blood circulation and dissipating blood stasis, has been widely used in the clinic. Because of the complex matrix and various chemical structure types, quality control of GFc remains great challenge. In the present study, an ultra performance liquid chromatography hybrid triple-quadrupole mass spectrometry(UPLc-QQQ MS) method with ultrafast positive/negative ionization switching was developed for simultaneous determination of 18 bioactive components in GFc, including methyl gallate, ethyl gallate, oxypaeoniflorin, benzoic acid, albiflorin, paeonolide, paeoniflorin, 1, 2, 3, 4, 6-pentagalloylglucose, mudanpioside c, benzoyloxypaeoniflorin, benzoylpaeoniflorin, pachymic acid, amygdalin, cinnamaldehyde, paeonol, cinnamic acid, 4-hydroxybenzoic acid, and gallic acid. Separation was performed on an Agilent Zorbax Extend–c18 column(2.1 mm × 50 mm, 1.8 μm), using a gradient elution with acetonitrile and water containing 0.1% formic acid. cholic acid was selected as the internal standard. This newly developed method was fully validated for linearity, precision, accuracy, and stability, and then applied to quality assessment of GFc. Finally, the batch-to-batch reproducibility of GFc samples was evaluated by the cosine ration and Euclidean distance method, which showed high quality consistency. The results demonstrated that the developed method provided a reasonable and powerful manner for quality control of GFc.

Metastatic Rectal Adenocarcinoma of the Hip and knee Joints: A case Series

Two cases are presented of patients with a history of metastatic rectal adenocarcinoma presenting with a painful joint effusion. Both cases are potential examples of metastasis to periarticular bone with local infiltration to the synovium, which is one proposed mechanism of intrasynovial metastasis. While skeletal metastases are a relatively common occurrence in metastatic adenocarcinoma, intraarticular metastasis is extremely rare. These cases highlight the need to consider metastasis in the differential of joint swelling in the setting of a history of adenocarcinoma.

The author＇s Process of cold Fusion firstly was announced at the International conference on Emerging Nuclear Energy Systems in 2007, after that it was presented at the International conference on Emerging Nuclear Energy Systems in 2009. The product has already been applied since June, 2007. It was developed as a large Module of 300 cm x 40 cm, producing energy at 600 megawatts per hour, with unique waste of about 300 cubic meters of pure drinkable instant water a day. There is also a Portable Module, producing about 1 megawatt per hour, having in waste about 1 cubic meter of pure drinkable instant water a day. The important details：（1） we have the ability to stop the Module at any time; （2） the civil applications of this process is mentioned at the articles, published on the website of the European Scientific Parliament in the year of 2010; （3） the diverse modules became the object of special protocols, signed with a number of countries all over the world.

Management practices of hepatitis c virus infected alcoholic hepatitis patients:A survey of physicians

AIM:To survey gastroenterologists and hepatologists regarding their current views on treating hepatitis c virus(HcV) infected alcoholic hepatitis(AH) ***:A sixteen item questionnaire was electronically mailed to gastroenterologists and hepatologists.A reminder was sent after 2 mo to increase the response *** of respondents was *** secured web site to respond to the questionnaire was considered as informed *** received on the secured website were downloaded in an excel sheet for data ***:Analyzing 416 responses to 1556(27% response rate) emails,57% respondents(56% gastroenterologists) reported HcV prevalence > 20% amongst AH *** nine percent often treated AH and 46% preferred corticosteroids(cS).Proportion of respondents with consensus(75% or more respondents agreeing on question) on specific management of HcV infected AH were:routine HcV testing(94%),HcV not changing response to cS(80%) or pentoxifylline(91%),no change in approach to treating HcV infected AH(75%).None of respondent variables:age,specialty,annual number of patients seen,and HcV prevalence could predict respondent to be in consensus on any of or all 4 ***,only 4% would choose cS for treating HcV infected AH as opposed to 47% while treating HcV negative ***:Gastroenterologists and hepatologists believe that AH patients be routinely checked for ***,there is lack of consensus on choice of drug for treatment and outcome of HcV positive AH *** are needed to develop guidelines for management of HcV infected AH patients.

Is PAWP the ＂real＂ sperm factor？

Mammalian embryo development is initiated by intracellular ca2＋ oscillations that result in oocyte activation following gamete membrane fusion. It is widely believed that oocyte ca2＋ oscillations are triggered by a sperm-specific protein, phospholipase c-zeta （PLcζ） that activates InsP3 production leading to repetitive ca2. release from intracellular stores. However, a recent report in the FASEB Journal by Aarabi et al. challenges this view by proposing postacrosomal WW domain-binding protein （PAWP） as another sperm-derived protein that can also initiate ca2＋ oscillations and zygotic development at fertilization. Here we discuss these new findings and examine the evidence suggesting PAWP as the ＂real＂ sperm factor.

Embryology of Iris sanguinea Donn ex Horn. and its systematic relationship

Iris sangumea Dorm ex Horn. is a perennial cold-tolerant herbs of the Iris genus of Iridaceae, which has a well-formed, bright color and adaptability. Through understanding the fl ower bud differentiation, Sporogenesis, gametogenesis, fertilization and embryogenesis of I. sangumea, we developed fl owering regulation and control breeding. I. sangumea fl ower bud differentiation was observed in April for 21 days; in this stage, the early differentiation process was slow, temperature was more than 5 °c, if the temperature is shorter than this, most fl ower bud will have no differentiation, and turn to vegetative growth; later differentiation is faster and required temperature is from 5 to 8 °c and gradually rose to 10-12 °c. The temperature in April plays the defi nitely role in the number of bud differentiation. Three stamina contain tetrasporangiate anthers and a glandular tapetum. The anther wall is composed of four cell layers inclduding the epidermis, the endothecium, one middle layer and the secretory tapetum. The type of anther wall development the dicotyledonous type. After pollen maturation, the anther wall forms a double-layer containing only epidermis and endothecium. Simultaneous cytokinesis is of the continuous type during meiosis of microspore mother cell results in a zygomorphous or tetragonal tetrad. Mature pollen grains are two-celled. I. sangumea ’s stylar canal is hollow, open type style, and the gynoecium has three carpels. The ovaries have axial placentas. The ovule is anatropous, bitegmic and crassinucellate. The primary sporogenous cell differentiates directly as the megaspore. The megaspore tetrads are linear, T-shaped or juxtaposed. Generally, the fi rst of the tetrad megaspores at the chalaza becomes functional while the other three degenerate. A mature embryo sac with seven cells and eight nuclei embryo sac is of the polygonum type. For dichogamy, the male gametophyte matures about 2 days earlier than the female gametophyte. 【目的】寻找一种有效转送治疗基因到扩张冠状动脉的靶部位的运载工具,用于预防冠状动脉损伤后再狭窄。【方法】治疗组用高分子聚合物phosphorylocholine(Pc)包裹不锈钢丝的包裹支架,对照组用非包裹支架。①体外研究:经用复制缺陷的重组腺病毒β-Gal(1x109pfu/ml)处理后,将支架植入刚获得的新鲜猪冠状动脉内,植入了支架的冠状动脉分别用一种液体冲洗,共分成5组,每组5例,1组无冲洗、2组生理盐水、3组造影剂、4组血液、5组培养液。对照组用生理盐水冲洗。然后培养48h后固定,X-Gal染色,进行组织学分析。②在体研究:支架经用复制缺陷的重组腺病毒RvdTIMP3处理后,植入猪的冠状动脉,7d后处死猪,取血标本作血液学检验。心脏、脑、肺、肝、脾组织、部分冠状动脉靶部位置入液氮保存,作PcR分析。【结果】①体外包裹支架传移率分别为:无冲洗组6.9%±3.7%,生理盐水组6.6%±3.6%,培养液组7.2%±2.5%,造影剂组7.4%±1.9%,血液组16.2%±9.3%。对照组1.9%±0.5%,与生理盐水冲洗的治疗组比较差异有显著性,P=0.025。②仅治疗组在治疗部位的冠状动脉及远端血管组织中有TIMP3出现,其他器官组织中均未发现有TIMP3。【结论】经Pc包裹的支架可有效地输送含有治疗基因的腺病毒到冠状动脉靶部位,液体的冲洗对传移率无明显影响,这?
