A renaissance in cell-free protein synthesis(CFPS)is underway,enabled by the acceleration and adoption of synthetic biology *** has emerged as a powerful platform technology for synthetic gene network design,biosensing and on-demand *** primarily of bacterial origin,cell-free extracts derived from a variety of host organisms have been explored,aiming to capitalise on cellular diversity and the advantageous properties associated with those ***,cell-free extracts produced from eukaryotes are often overlooked due to their relatively low yields,despite the potential for improved protein folding and posttranslational *** we describe further development of a Pichia pastoris cell-free platform,a widely used expression host in both academia and the biopharmaceutical *** a minimised Design of Experiments(DOE)approach,we were able to increase the productivity of the system by improving the composition of the complex reaction *** was achieved in a minimal number of experimental runs,within the constraints of the design and without the need for liquid-handling *** doing so,we were able to estimate the main effects impacting productivity in the system and increased the protein synthesis of firefly luciferase and the biopharmaceutical HSA by 4.8-fold and 3.5-fold,*** study highlights the ***-based cell-free system as a highly productive eukaryotic platform and displays the value of minimised DOE designs.

Abstract We investigated the social relationships among adult females in two multimale-multifemale groups of black howler monkeys Alouatta pigra during a 14-month study in Palenque National Park, mexico. Based on over 900 focal hours and over 5400 scan samples recording neighboring group members, we found that females very rarely engaged in agonistic interactions and no dominance hierarchy could be discerned. Relationships among resident females were primarily affiliative, but females of one study group spent a higher proportion of time in close proximity and engaged in affiliative interactions with one another at higher rates than females in the other study group. The strength of female relationships increased with the birth of an infant. Al- though no females immigrated during the study period, the temporary association of three extragroup females with our study groups implies that the social system of black howler monkeys is more dynamic than previously suggested. These findings sug- gest that female black howler monkeys behave more similarly to female red howler monkeys A. seniculus than to female mantled howler monkeys A. palliata

In urban or suburban areas with a large number of access points, four-lane undivided highways are prone to crashes due to left-turning and through movements in a single lane. many studies recommended expensive countermeasures like conversion from undivided to divided road with physical separation. One inexpensive alternative is reconfiguring the existing roadways by either increasing or decreasing the number of lanes. This study investigated the safety impact of converting four lane undivided roadways （4U） to five lane undivided roadways （ST） with a two way left turn lane （TIkrLTL）. This study used Empirical Bayes method to determine the safety impact of this inexpensive countermeasure. In this study, data from eight sites from Louisiana were collected for investigation, and site- specific crash modification factor （CmF） values were calculated. Although ST is usually not preferable due to its exposure of higher number of crashes in the existing literature, the findings of the current study indicated a positive safety impact. The benefit-cost ratio of this conversion ranges from 97 to 379. The current findings indicate that conversion of 4U to 5T is a feasible inexpensive solution for urban roadways with lower volume and a limited number of driveways.

Previous studies have revealed the significance of cytokine interleukin 1 （IL-1） in the onset and progression of rheumatoid arthritis （RA）. The precise molecular mechanisms related to IL-1 underlying RA is still elusive. We conducted a whole genome-wide tran- scriptomal comparison of wild-type （WT） and arthritis-prone IL-1 receptor antagonist （IL-lrn） deficient BALB/c mice to address this issue. To refine our search efforts, gene expression profiling was also performed on paired wild-type and arthritis-resistant IL-lrn deficient DBA/1 mice as internal controls when identifying causative arthritis candidate genes. Two hundred and fifteen transcripts were found to be dysregulated greater than or equal to 2-fold in the diseased mice. The altered transcriptome in BALB/c mice revealed increased myeloid cell activities and impaired lymphocyte functionality, suggesting dual regulatory effects of IL-1 hyperactivity on immunological changes associated with arthritis development. Phase-specific gene expression changes were identified, such as early increase and late decrease of heat shock protein coding genes. moreover, common gene expression changes were also observed, especially the upregulation of paired Ig-like receptor A （Pira） in both early and late phases of arthritis. Real-time PCR was performed to validate the expression of Pira and an intervention experiment with a major histocompatibility complex （mHC） class I inhibitor （brefeldin A） was carded out to investigate the role of suppressing Pira activity. We conclude that global pattern changes of common and distinct gene expressions may represent novel opportunities for better control of RA through early diagnosis and development of alternative therapeutic strategies.

Steroid hormones, alkylphenol ethoxylates（APEOs） and phthalic acid esters（PAEs） are emerging endocrine disrupting chemicals（EDCs） that can interfere with the endocrine function in organisms at low concentrations. The occurrence, distribution behavior, removal rate and the fate of 31 target EDCs in sewage treatment plants, which consist of various treatment facilities and receiving water in Hong Kong, were investigated. Estrone,nonylphenol-di-ethoxylate and diethyl phthalate were found to be dominant in each group of influent samples with concentrations ranging from 11–33, 747–3945 and 445–4635 ng/L,respectively. Conversely, progesterone, nonylphenol-mono-ethoxylate and bis（2-ethylhexyl）phthalate were the most abundant in dewatered sludge, with concentrations ranging from 0.9–237, 75–19,743 and 4310–37,016 ng/g（dry weight）. The removal rates of primary sedimentation and disinfection approaches were lower than 30% for most of the chemicals,while those of activated sludge and reverse osmosis were greater than 80% for more than two-thirds of the compounds, noticeably decreasing the estrogenic risk of sewage discharged into the environment. Steroid hormones were removed via biological degradation, while some APEOs and PAEs adsorbed to the sludge. Victoria Harbor poses a low to medium estrogenic risk mainly contributed by estrone and estradiol and deserves attention.

Purpose： This study aimed to investigate whether workload intensity modulates exercise-induced effect on reaction time （RT） performances, and more specifically to clarify whether cognitive control that plays a crucial role in rapid decision making is altered. methods： Fourteen participants performed a Simon Task while cycling 20 min at a light （first ventilatory threshold, VT~ - 20%）, moderate （VTI）, or very hard （VTj ＋ 20%） level of exercise. Results： After 15 min of cycling, RTs are faster than during the first 5 min of exercise. This benefit does not fluctuate with the intensity of exercise and enlarges as RT lengthens. Despite a numerical difference suggesting a greater facilitation during moderate exercise （-16 ms） than during a light exercise （-10 ms）, the benefit is not statistically different. Interestingly, we did not observe any signs of worsening on RT or on accuracy during very hard exercise. Conclusion： Cognitive control is extremely robust and appears not to be affected by the intensity of exercise. The selective inhibition and the between-trials adjustments are effective from the beginning to the end of exercise, regardless of the workload output.

many experimental and clinical observations suggest that intestinal microflora plays a potential role in the pathogenesis of inflammatory bowel disease （IBD）. manipulation of the luminal content using antibiotics or probiotics represents a potentially effective therapeutic option. The available studies do not support the use of antibiotics in ulcerative colitis （UC）. Antibiotics are effective in treating septic complications of Crohn＇s disease （CD） but their use as a primary therapy is more controversial, although this approach is frequently and successfully adopted in clinical *** is evidence that probiotic therapy may be effective in the prevention and treatment of mild to moderate UC. In contrast, a lack of successful study data at present precludes the widespread use of probiotics in the treatment of CD. Both antibiotics and probiotics appear to play a beneficial role in the treatment and prevention of pouchitis and further trials are warranted to fully quantify their clinical efficacy.

Retinal disorders are a group of ocular diseases whose onset is associated with a number of aberrant molecular and cellular processes or physical damages that affect retinal structure and function resulting in neural and vascular degeneration in the *** research has primarily focused on delaying retinal disease with minimal success in preventing or reversing neuronal *** this review,we explore a relatively new field of research involving circular RNAs,whose potential roles as biomarkers and mediators of retinal disease pathogenesis have only just *** knowledge of circular RNAs function is limited given its novelty,current evidence has highlighted their roles as modulators of microRNAs,regulators of gene transcription,and biomarkers of disease development and ***,we summarize how circular RNAs may be implicated in the pathogenesis of common retinal diseases including diabetic retinopathy,glaucoma,proliferative vitreoretinopathy,and age-related macular ***,we explore the potential of circular RNAs as novel biomarkers and therapeutic targets for the diagnosis and treatment of retinal diseases.
