Crohn's disease genotypes of patients in remission vs relapses after infliximab discontinuation

AIM： To investigate genetic differences between Crohn＇s disease （CD） patients with a sustained remission vs relapsers after discontinuing infliximab while in cortico- steroid-free remission. METHODs： Forty-eight CD patients received infliximab and were in full corticosteroid-free clinical remission but then discontinued infliximab for reasons other than a loss of response, were identified by review of an electronic database and charts. Infliximab-associated remis- sion was defined as corticosteroid-free plus normaliza- tion of clinical disease activity [CD activity index （CDAI）〈 150] during follow-up visits based on physician global assessments. A CD relapse （loss of infliximab-induced remission） was clinically defined as a physician visit for symptoms of disease activity （CDAI 〉 220） and a thera- peutic intervention with CD medication（s）, or a hospital- ization with complications related to active CD. Genetic analyses were performed on samples from 14 patients （n = 6 who had a sustained long term remission after stopping infliximab, n -- 8 who rapidly relapsed after stopping infliximab）. Nucleotide-binding oligomerization domain 2 （NOD2）/caspase activation recruitment do- main 15 （CARD15） polymorphisms （R702W, G908R and L1007fs） and the inflammatory bowel disease 5 （IBDs） polymorphisms （IGR2060a1 and IGR3081a1） were ana- lyzed in each group. REsULTs： Five single nucleotide polymorphisms of IBD5 and NOD2/CARD15 genes were successfully analyzed for all 14 subjects. There was no signifcant increase in frequency of the NOD2/CARD15 polymor- phisms （R702W, G908R and L1007fs） and the IBD5 polymorphisms （IGR2060al and IGR3081a1） in either group of patients; those whose disease relapsed rap- idly or those who remained in sustained long term remission following the discontinuation of infliximab. We analyzed the biogenic silica(Bsi)content and produced a diatom-based summer sea-surface temperature(ssT)reconstruction for sediment core GC4 from the Holsteinsborg Dyb,West *** aim was to reconstruct marine productivity and climatic fluctuations during the last *** Bsi content and diatom abundance suggest relatively high marine productively during the interval of AD 1000–1400,corresponding in time to the Medieval Warm Period(MWP).The summer ssT reconstruction indicates relatively warm conditions during AD 900–1100,followed by cooling after AD *** extended cooling period during AD 1400–1900 is characterized by prolonged low in reconstructed ssT and high sea-ice *** Bsi values fluctuated during this period,suggesting varying marine productivity during the Little Ice Age(LIA).There is no significant correlation between the Bsi content and ssT during the last millennium,suggesting that the summer ssT has little influence on marine productively in the Holsteinsborg Dyb.A good correspondence between the Bsi content and the element Ti counts in core GC4 suggests that silicate-rich meltwater from the Greenland ice sheet was likely responsible for changes in marine productively in the Holsteinsborg Dyb.

Melanoma of the vulva: A retrospective study of 46 cases from Queensland

Objective: To review the records of cases of vulvar melanoma referred to the Queensland Centre for Gynaecological Cancer (QCGC) between 1986 and 2009 recognising the high incidence of cutaneous melanoma in Queensland. Methods: Vulvar melanoma case data from QCGC were reviewed and analysed using the computer software statistical Package for the social sciences (sPss) 11.0. Results: Of the 46 cases 22 died of their disease, nine died of other diseases, ten are still alive and five lost to follow up. The mean age at diagnosis was 65.5 years for nodular melanoma and 69 years for superficial spreading melanoma. All cases were Caucasian. Time from on-set of symptoms to diagnosis averaged 3.2 months. In all 46 cases diagnosis was confirmed histologically. Presenting symptoms included lumps, itch, stinging, bleeding and pain. The most common presenting complaint was a lump. The initial treatment was surgical without a trend to a less radical approach to management. Conclusions: Vulvar melanoma prognosis remains guarded. Increased tumor size, depth of invasion, mitotic rate, groin lymph node positivity and status of disease at the margins of surgical specimens reliably equate to long term outcome. The incidence of vulvar melanoma was not increased in Queensland.

Mercury bioaccumulation in aquatic biota along a salinity gradient in the saint John River estuary

Although estuaries are critical habitats for many aquatic species, the spatial trends of toxic methylmercury（MeHg） in biota from fresh to marine waters are poorly understood. Our objective was to determine if MeHg concentrations in biota changed along a salinity gradient in an estuary. Fourspine stickleback（Apeltes quadracus）, invertebrates（snails,amphipods, and chironomids）, sediments, and water were collected from ten sites along the saint John River estuary, New Brunswick, Canada in 2015 and 2016, with salinities ranging from 0.06 to 6.96. Total mercury（proxy for MeHg） was measured in whole fish and MeHg was measured in a subset of fish, pooled invertebrates, sediments, and water. stable sulfur（δ34s）, carbon（δ13 C）, and nitrogen（δ15N） isotope values were measured to assess energy sources（s, C） and relative trophic level（N）. There were increases in biotic δ13C and δ34s from fresh to more saline sites and these measures were correlated with *** aqueous MeHg was higher at the freshwater than more saline sites, only chironomid MeHg increased significantly with salinity. In the saint John River estuary, there was little evidence that MeHg and its associated risks increased along a salinity gradient.

Effects of UV radiation on the RNA/DNA ratio of Copepods from Antarctica and Chile

The effect of ultraviolet （UV） radiation on marine organisms has been an important focus of recent research, with depletion of the ozone layer resulting in increased UV radiation at high latitudes. several studies have identified negative impacts of UV radiation on the biology of zooplanktonic organisms. This study used the RNA/DNA ratio as a measure of stress in copepod assemblages from Fildes Bay in Antarctica and Quintay Bay on the central coast of Chile, two areas with high UV radiation but different photobiologic histories. Controlled time-light experiments were performed with copepods from the two locations, exposing them to white light, UV light, or darkness. The results showed different responses to UV radiation. Copepods from Fildes Bay showed a slow metabolic response to UV radiation after 4 and 8 h of exposure. Copepods from Quintay Bay showed a fast metabolic response after 4 h of exposure （4 orders of magnitude higher than that for Fildes Bay copepods） followed by a rapid return toward baseline after 8 h of exposure. These different responses probably reflect the time the eopepod assemblages have been exposed to increased UV radiation and the extent of adaptive stress responses to cope with that increased UV radiation. The results of this study show that the RNA/DNA ratio is a useful indicator of the physiologic status of marine organisms and is a useful tool to measure the effects of changing environmental conditions on marine ecosystems, such as those associated with global climate change.

Dual Effects of IL-1 Overactivity on the Immune system in a Mouse Model of Arthritis due to Deficiency of IL-1 Receptor Antagonist

Previous studies have revealed the significance of cytokine interleukin 1 （IL-1） in the onset and progression of rheumatoid arthritis （RA）. The precise molecular mechanisms related to IL-1 underlying RA is still elusive. We conducted a whole genome-wide tran- scriptomal comparison of wild-type （WT） and arthritis-prone IL-1 receptor antagonist （IL-lrn） deficient BALB/c mice to address this issue. To refine our search efforts, gene expression profiling was also performed on paired wild-type and arthritis-resistant IL-lrn deficient DBA/1 mice as internal controls when identifying causative arthritis candidate genes. Two hundred and fifteen transcripts were found to be dysregulated greater than or equal to 2-fold in the diseased mice. The altered transcriptome in BALB/c mice revealed increased myeloid cell activities and impaired lymphocyte functionality, suggesting dual regulatory effects of IL-1 hyperactivity on immunological changes associated with arthritis development. Phase-specific gene expression changes were identified, such as early increase and late decrease of heat shock protein coding genes. Moreover, common gene expression changes were also observed, especially the upregulation of paired Ig-like receptor A （Pira） in both early and late phases of arthritis. Real-time PCR was performed to validate the expression of Pira and an intervention experiment with a major histocompatibility complex （MHC） class I inhibitor （brefeldin A） was carded out to investigate the role of suppressing Pira activity. We conclude that global pattern changes of common and distinct gene expressions may represent novel opportunities for better control of RA through early diagnosis and development of alternative therapeutic strategies.

Highly active and low-cost catalysts for electrochemical reactions such as the hydrogen evolution reaction （HER） are crucial for the development of efficient energy conversion and storage technologies. Theoretical simulations have been instrumental in revealing the correlations between the electronic structure of materials and their catalytic activity, and guide the prediction and development of improved catalysts. However, difficulties in accurately engineering the desired atomic sites lead to challenges in making direct comparisons between experi- mental and theoretical results. In Mos2, the Mo-edge has been demonstrated to be active for HER whereas the s-edge is inert. Using a computational descriptor- based approach, we predict that by incorporating transition metal atoms （Fe, Co, Ni, or Cu） the s-edge site should also become HER active. Vertically standing, edge-terminated Mos2 nanofilms provide a well-defined model system for verifying these predictions. The transition metal doped Mos2 nanofilms show an increase in exchange current densities by at least two-fold, in agreement with the theoretical calculations. This work opens up further opportunities for improving electrochemical catalysts by incorporating promoters into particular atomic sites, and for using well-defined systems in order to understand the origin of the promotion effects.

Influence of sugarcane Bagasse Fiber size on Biodegradable Composites of Thermoplastic starch

Although thermoplastic starch(TPs)is biodegradable,its low mechanical resistance limits its wide *** bagasse(sB)fibers can be used as reinforcement in TPs matrix composites,but the influence of fiber size on the properties of the composite is still *** this study,TPs composites reinforced with sB short fibers of four sizes were processed and characterized in order to analyze the influence of fiber size on the mechanical properties of the TPs/sB *** was observed that the interaction between fiber and matrix was good and optimized when the fibers are sifted in sieves between 30 and 50 mesh,obtaining fibers with average length of 1569±640μm and average diameter of 646±166μ*** these composites,increases of more than 660%in the modulus and more than 100%in the maximum tension were verified when compared to the pure TPs.

The microbiomes associated with bee nests influence colony health through various mechanisms,although it is not yet clear how honeybee congeners differ in microbiome assembly processes,in particular the degrees to which floral visitations and the environment contribute to different aspects of *** used DNA metabarcoding to sequence bacterial 16s rRNA from honey and stored pollen from nests of 4 honeybee species(Apis cerana,***,***,and ***)sampled throughout Yunnan,China,a global biodiversity *** developed a computational pipeline integrating multiple databases for quantifying key facets of diversity,including compositional,taxonomic,phylogenetic,and functional ***,we assessed candidate drivers of observed microbiome dissimilarity,particularly differences in floral visitations,habitat disturbance,and other key environmental *** revealed that microbiome alpha diversity was broadly equivalent across the study sites and between bee species,apart from functional diversity which was very low in nests of the reclusive *** in microbiome composition across Yunnan was driven predominantly by pollen *** disturbance negatively impacted both compositional and phylogenetic alpha diversity of nest microbiomes,but did not correlate with microbial *** herein make progress in understanding microbiome diversity associated with key pollinators in a biodiversity hotspot,and provide a model for the use of a comprehensive informatics framework in assessing pattern and drivers of diversity,which enables the inclusion of explanatory variables both subtly and fundamentally different and enables elucidation of emergent or unexpected drivers.
