MicroR-9c-5p and novel-mir50 co-target Akt to regulate Lasioderma serricorne reproduction

High fecundity is a common characteristic of insect pests which increases the difficulty of population ***/threonine kinase Akt is an indispensable component of the insulin signaling *** of LsAkt severely hinders reproduction in Lasioderma serricorne,a stored product insect ***,the post-transcriptional pathway of LsAkt in *** remains *** study identified 2 binding sites of miR-9c-5p and novel-mir50 in the coding sequences of *** expression profiles of 2 microRNAs(miRNAs)and LsAkt displayed an opposite pattern during the adult *** reporter assay showed that novel-mir50 and miR-9c-5p could downregulate the expression of *** of miR-9c-5p and novel-mir50 by injection of mimics inhibited the expression of LsAkt and reduced oviposition,decreased egg hatchability,and blocked ovarian *** also decreased the expression of genes involved in ovarian development(LsVg and LsVgR)and the nutritional signaling pathway(LsTOR,LsS6K,and Ls4EBP),and reduced the phosphorylation of ***,injection of miR-9c-5p and novel-mir50 inhibitors induced the expressions of LsAkt,LsVg,LsVgR,LsTOR,LsS6K,and Ls4EBP,enhanced Akt phosphorylation level,and accelerated ovarian *** of bovine insulin downregulated the expression of miR-9c-5p and novel-mir50 and upregulated the LsAkt *** also rescued the reproductive development defects associated with miR-9c-5p/novel-mir50 overexpression,forming a positive regulatory loop of insulin *** results indicate that miR-9c-5p/novel-mir50 regulates the female reproduction of *** by targeting Akt in response to insulin *** data also demonstrate the effects of the insulin/miRNA/Akt regulatory axis in insect reproduction.

Efficient and precise genomic deletion in rice using enhanced prime editing

Efficient and precise genomic deletion shows promise for investigating the function of proteins in plant research and enhancing agricultural *** this study,we tested the PRIME-Del(PDel)strategy using a pair of prime editing guide RNAs(pegRNAs)that targeted opposite DNA strands and achieved an average deletion efficiency of 55.8%for 60 bp fragment deletions at six endogenous ***,as high as 84.2%precise deletion efficiency was obtained for a 2000 bp deletion at the OsGS1 site in transgenic rice *** add the bases that were unintentionally deleted between the two nicking sequences,we used the PDel/Syn strategy,which introduced multiple synonymous base mutations in the region that had to be patched in the RT *** PDel/Syn strategy achieved an average of 58.1%deletion efficiency at six endogenous targets,which was higher than the PDel *** strategies presented in this study contribute to achieving more accurate and flexible deletions in transgenic rice plants.

Magnesium-based hydrogen storage materials represent a hydrogen storage technology with broad application prospects. As the global energy crisis and environmental pollution issues become increasingly severe, hydrogen,as a clean and efficient energy source, has garnered growing attention. Magnesium-based hydrogen storage,serving as a crucial means for storing and transporting hydrogen, is gaining prominence due to its abundant resources, low cost, low density, and high hydrogen storage density. However, challenges in terms of absorption/desorption rates, temperature, activation energy, and enthalpy during hydrogen application impede its development. To address these challenges, this paper systematically reviews current research on magnesium-based hydrogen storage materials, encompasses their types, characteristics, and hydrogen absorption ***, it delves into the impacts of nanoscale dimensions, alloying, doping, and catalysis on the performance of magnesium-based materials. The aim is to provide valuable insights for research in related fields.

A method for detecting two-dimensional plane stress distribution in basin-type insulator based on critically refracted longitudinal wave

Basin-type insulator often has small cracks due to stress *** current method cannot accurately reflect the stress condition of the insulator to find the stress concentration *** solve these problems,a method for detecting two-dimensional plane stress(δ_(1) andδ_(2))within different depth ranges in a basin-type insulator is proposed based on critically refracted longitudinal(LCR)***,the acoustoelastic equation characterising the relationship between the variation of LCR wave propagation time and the plane stress was ***,the propagation characteristics of LCR wave in epoxy resin samples were ***,the stress distribution within different depth ranges of the insulator subjected to hydraulic load was measured using the proposed method,including direction(θ),δ_(1) andδ_(2).The results show that the magnitude of the stress alone cannot accurately characterise the stress *** with equal distances to the centre have similar stress magnitudes,but their directions are not the *** increasing depth,θremains essentially unchanged at the same location,whileδ1 andδ2 decrease,and the rate of decrease varies at different *** the measured and simulated data,the results showed that they were in good agreement,and the maximum errors of stress value andθwere 0.69 MPa and 2.97°,respectively,which confirmed the feasibility and accuracy of the stress detection in the proposed method.

Isotopic insights on quantitative assessments of interaction of eco-hydrological processes in multi-scale karst watersheds

The dynamics of hydrological processes and the storage mechanisms of karst water resources are the most important issues in karst *** impact of environmental changes on water quantity,and the evaluation and quantification of eco-hydrological processes remain poorly *** this study,high-frequency continuous monitoring in multi-scale karst watersheds in Southwest China combined the approaches of water isotopes and the hybrid single-particle lagrangian integrated trajectory(HYSPLIT)model to identify the recharge mechanisms between atmospheric vapor,rainfall,surface water,and groundwater,and to reveal the interaction of eco-hydrological *** dominant moisture sources in Puding(PD)County were the Indian Ocean(43-69%)and local moisture(24-33%).Theδ^(18)O and deuterium excess(d-excess)values showed a positive correlation indicating that secondary or sub-cloud evaporation was prominent in the wet *** water line-conditioned excess(lc-excess)indicated that karst water interacted with recent precipitation,groundwater,and evaporation across *** to its specific hydrogeological structure,surface water and rainwater have a higher contribution rate to groundwater *** Chenqi stream replenished the Houzhai River mainly in the form of groundwater,with percentages ranging from 38.1 to 93.5%in the wet season,and 47.8-80.1%in the dry *** the Houzhai outlet,surface water and groundwater interconverted frequently with a percentage of 45.6-49.1%.We believe this is the first systematic study to quantify the supply relationship between water vapor transport,rainfall,surface water and groundwater in the Chinese karst zone,making a significant move forward in the field of karst hydrological processes and improving the efficiency of water resource evaluation and management.

Aerodynamic characteristics of a pitching airfoil with leading-edge morphing

This paper focuses on the effect of the phase offset of Leading-Edge(LE)morphing on the aerodynamic characteristics of a pitching NACA0012 *** an unstretched camber and using polynomial interpolation,an explicit expression for LE nonlinear morphing is proposed and implemented for the large pitching motion of the *** field results and aerodynamic forces are obtained by solving the unsteady Reynolds-averaged Navier-Stokes equations for both the airfoil’s pitching motion and LE ***,the index of instantaneous aerodynamic power is used to quantify the work done by the airflow in a dynamic *** to the instantaneous aerodynamic power and energy map,which denotes the energy transfer between the airfoil’s oscillation and flow field,the airfoil is subject to stall *** results show that LE morphing with an optimal phase offset of 315°reduces the energy extraction from the flow field,suppressing the stall flutter *** optimal phase offset is effective at different pitching axis positions of the *** results signify that LE morphing can suppress stall flutter by advancing the occurrence of the first LE vortex and increasing the nose-down moment during the upstroke period.

Effect of BaO and MgO on structure and properties of aluminate slag with various CaO/Al_(2)0_(3)ratios

To provide theoretical guidance for performance stability control of low-reactive mold fluxes,the effects of BaO and MgO on the structure and properties of aluminate slag with various CaO/Al_(2)0_(3)(C/A)ratios were investigated using the Fourier transform infrared spectrometer,Raman spectroscope,hemispherical melting point instrument,rotational viscometer and X-ray *** results indicated that with BaO and MgO addition,the structure polymerization was first weakened and then enhanced at C/A of 1.1,and the transition contents corresponded to 8 wt.%BaO and 2 wt.%MgO,respectively,while the structure polymerization decreased continuously at C/A of *** the viscosity change was well consistent with the structure evolution,the polymerization degree played a more prominent role in the slag viscosity than superheat degree when the melting temperature difference was within 40℃.The break temperature decreased initially and then increased with augment of BaO and MgO at C/A of 1.1,while it manifested a decrease trend with BaO addition,and it decreased obviously but then turned to increase with MgO increment at C/A of *** crystallization phase and crystallization ratio kept stable with BaO increment,while the crystallization ratio rose greatly with MgO promoting LiAlO_(2)precipitation at C/A of *** crystal types of all experimental slags were mainly Ca_(12)Al_(14)O_(33)and CaF_(2)at C/A of 1.3,and the precipitation of crystalline phase BaAl2O4 demonstrated a rising trend,while that of Ca_(12)Al_(14)O_(33)gradually declined with BaO augment.

Direct methanol fuel cell technology recently becomes the focus of both academic and engineering circles,which stimulates the exploitation and utilization of advanced electrode catalysts with high activity and long ***,we demonstrate a robust bottom-up approach to the spatial construction of three-dimensional(3D) spinel manganese-cobalt oxide-modified N-doped graphene nanoarchitectures decorated with ultrasmall Pt nanoparticles(Pt/MnCo_(2)O_(4)-NG) via a controllable selfassembly *** incorporation of MnCo_(2)O_(4)nanocrystals provides abundant hydroxyl sources to promote the oxidative removal of CO-like byproducts on Pt sites,while the existence of 3D porous N-doped graphene networks facilitates the transportation of both ions and electrons in the hybrid system,thus giving rise to remarkable synergetic coupling effects during the methanol oxidation ***,the optimized Pt/MnCo_(2)O_(4)-NG nano architecture expres ses exceptional electrocatalytic properties with a large electrochemically active surface area of 99.5 m^(2)·g^(-1),a high mass activity of1508.3 mA·mg^(-1),strong toxicity resistance and reliable long-term durability,which have obvious competitive advantages over those of conventional Pt/carbon black,Pt/carbon nano tube,Pt/graphene,and Pt/N-doped graphene catalysts with the same Pt usage.

Rapid report of source parameters of 2023 M6.2 Jishishan,Gansu earthquake sequence

The M6.2 earthquake in Jishishan,Gansu Province,on December 18,2023,caused extraordinary earthquake *** was located in the northern part of the north−south seismic zone,which is a key area for earthquake monitoring in *** newly built dense strong motion stations in this area provide unprecedented conditions for high-precision earthquake relocation,especially the earthquake focal *** paper uses the newly built strong motion and traditional broadband seismic networks to relocate the source locations of the M3.0 and above aftershocks and to invert their focal *** horizontal error of earthquake location is estimated to be 0.5−1 km,and the vertical error is 1−2 *** focal depth range of aftershocks is 9.6−14.6 km,distributed in a 12-km-long strip with SSE *** in the south are more concentrated horizontally and vertically,while aftershocks in the north are more *** focal mechanisms of the main shock and aftershocks are relatively consistent,and the P-axis orientation is consistent with the regional strain *** is a seismic blank area of M3.0 and above,about 3−5 km between the main shock and *** is suggested that the energy released by the main shock rupture is concentrated in this *** on the earthquake location and focal mechanism of the main shock,it is inferred that the Northern Lajishan fault zone is the seismogenic structure of the main shock,and the main shock did not occur on the main fault,but on a secondary *** initial rupture depth and centroid depth of the main shock were 12.8 and 14.0 km,*** source rupture depth may not be the main reason for the severe earthquake disaster.
