Prediction of performance and evaluation of flexible pavement rehabilitation strategies

Five test sections with different additives and strategies were established to rehabilitate a State-maintained highway more effectively in Rhode Island （RI）： control, calcium chloride, asphalt emulsion, Portland cement and geogrid. Resilient moduli of subgrade soils and subbase materials before and after full depth rehabilitation were employed as input pa- rameters to predict the performance of pavement structures using AASHTOWare Pave- ment ME Design （Pavement ME） software in terms of rutting, cracking and roughness. It was attempted to use Level i input （which includes traffic full spectrum data, climate data and structural layer properties） for Pavement ME. Traffic data was obtained from a Weigh- in-Motion （WIM} instrument and Providence station was used for collecting climatic data. Volumetric properties, dynamic modulus and creep compliance were used as input pa- rameters for 19 mm （0.75 in.} warm mix asphalt （WMA） base and 12.S mm （0.5 in.） WMA surface layer. The results indicated that all test sections observed AC top-down （longitu- dinal） cracking except Portland cement section which passed for all criteria. The order in terms of performance （best to worst） for all test sections by Pavement ME was Portland cement, calcium chloride, control, geogrid, and asphalt emulsion. It was also observed that all test sections passed for both bottom up and top down fatigue cracking by increasing thickness of either of the two top asphalt layers. Test sections with five different base/ subbase materials were evaluated in last two years through visual condition survey and measurements of deflection and roughness to confirm the prediction, but there was no serious distress and roughness. Thus these experiments allowed selecting the best reha- bilitation/reconstruction techniques for the particular and/or similar highway, and a framework was formulated to select an optimal technique and/or strategy for future rehabilitation/reconstruction projects. Cold In-Place Recycling as a Sustainable Pavement Practice

Pavement rehabilitation and reconstruction methods with CIR （cold in-place recycling） are alternatives that can effectively reduce the high stresses and waste produced by conventional pavement strategies. An attempt was made to predict the performance, particularly low-temperature cracking resistance characteristics of CIR mixtures. These were prepared with the mix design procedure developed at the URI （University of Rhode Island） for the FHWA （Federal Highway Administration） to reduce wide variations in the application of CIR mixtures production. This standard was applied to RAP （reclaimed asphalt pavement） to produce CIR mixtures with CSS-Ih asphalt emulsion as the additive. By adjusting the number of gyrations of the SGC （Superpave gyratory compactor） for compaction, the field density of 130 pcf was represented accurately. To secure a base line, HMA （hot mix asphalt） samples were produced according to the Superpave volumetric mix design procedure. The specimens were tested using the IDT （indirect tensile） tester according to the procedure of AASHTO T 322 procedure at temperatures of-20, -10 and 0 ℃ （-4, 14, and 32°F, respectively）. The obtained results for the creep compliance and tensile strength were used as input data for the MEPDG （mechanistic empirical pavement design guide）. The analysis results indicated that no thermal or low-temperature cracking is expected over the entire analysis period of 20 years for both HMA and CIR mixtures. Thus, it appears that CIR is a sustainable rehabilitation technique which is also suitable for colder climates, and it is recommended to conduct further investigation of load-related distresses such as rutting and fatigue cracking.

Exploration of Mutation and DNA Methylation of Polo-Like Kinase 1 (PLK1) in Colorectal Cancer

Polo-like kinase 1 (PLK1) is a serine/threonine kinase that plays critical roles in cell cycling and DNA damage response. Overexpression of PLK1 is associated with the poorer prognosis of cancers, including colorectal cancer (CRC). Although the downstream pathways of the overexpression that lead to oncogenesis have been extensively studied, little is known about the factors that cause the overexpression of PLK1 in CRC. DNA methylation was reported to be affecting the expression of PLK1 in some cancers. The study aims to investigate the contribution of genetic mutation and DNA methylation of the PLK1 gene to the overexpression of PLK1 in CRC. The study involves data mining from Catalogue of Somatic Mutations in Cancer (COSMIC) and UniProtKB, Sanger sequencing on DNA from the cell lines HCT116, SW48, Colo320DM and T84 to analyse the possible mutation of PLK1. Other than that methylation status of the PLK1 promoter in CRC are also analysed by using mass spectrometry (MS) and pyrosequencing. Data from the COSMIC show the low incidences of PLK1 mutation for CRC (3.02%) with 46 mutations identified. One of the mutation p.R337Q (c.1010G > A) is located in the D-box which is an important motif for protein ubiquitination and eventually proteasomal degradation. Hence this mutation may potentially result in stabilisation of the PLK1 protein. Mutations are detected at the upstream silencer region, the promoter region and Exon1 in HCT116 but are not located at the protein binding or functioning site. Similarly, the same mutation at promoter region is detected in SW48. Differential trends of changes in methylation status of PLK1 in the IR treated CRC cell lines detected by MS reveal the possible association between the methylation and the radiosensitivity. Furthermore, pyrosequencing shows that PLK1 methylation status in tumour tissues with high expression of PLK1 is not significantly different from those with no PLK1 expression. Evaluating the Technical and Economic Feasibility of Adding a Power Recovery System to the Steam Condenser of a Lignite Coal-Fired Power Plant

Steam is the typical working fluid to drive turbo-generators in coal-fired power plants. It is an effective working fluid, but some of its energy is extracted in an unusable form when condensed. A Power Recovery Cycle (PRC) using a more volatile Secondary Working Fluid (SWF) added to the steam cycle could improve energy efficiency. PRCs have been applied to the flue gas and for combined cycle systems but not to traditional plant steam cycles. This paper details an analysis of adding a steam cycle PRC to a 500 MW lignite coal-fired power plant. A validated model of the plant was developed and PRCs using the three most attractive SWFs, benzene, methanol and hydrazine, were then added to the model. Adding a benzene, methanol, or hydrazine steam cycle PRC will produce an additional 59, 34, and 49 MW, respectively. An AACE Class 4 factored broad capital cost estimate and comparable operating costs and revenue estimates were developed to evaluate PRC feasibility. The benzene, methanol, and hydrazine processes had 2019 Net Present Values (NPVs) @12% of -$32, -$59, and +$35 million ± 40%, respectively. Thus, a PRC may be profitable at current or modest increases to U.S. Upper Midwest electricity prices of around $0.0667/kWh.

Allometric relationships between primary size measures and sapwood area for six common tree species in snow-dependent ecosystems in the Southwest United States

High-elevation,snow-dependent,semiarid ecosystems across southwestern United States are expected to be vulnerable to climate change,including drought and fi re,with implications for various aspects of the water *** that end,much less is known about the dynamics of transpiration,an important component of the water cycle across this *** the individual-tree scale,transpiration is estimated by scaling mean sap fl ux density by the hydroactive sapwood area(SA).SA also remains a key factor in eff ectively scaling individual tree water-use to stand *** across large spatial scales is normally established by relating SA of a few trees to primary size measures,e.g.,diameter at breast height(DBH),tree height(H),or canopy diameter(CD).Considering the importance of SA in scaling transpiration,the primary objective of this study was therefore to establish six species-specifi c(aspen,maple,white fi r,ponderosa pine,Douglas fi r,Englemann spruce)allometric relationships between SA and three primary size measures(DBH,CD,or H)across two high-elevation,snow-dependent,semiarid ecosystems in New Mexico and *** on multiple statistical criteria(coeffi cient of determination,index of agreement,Nash–Sutcliff e effi-ciency)and ease of measurement in the forest,we identifi ed DBH as the primary independent variable for estimating SA across all *** on group regression analysis,we found allometric relationships to be signifi cantly(p<0.05)diff erent for the same species(ponderosa pine,Douglas-fi r)across diff erent ***,our allometric relationships provide a valuable database for estimating transpiration at diff erent spatial scales from sap fl ow data in some of our most vulnerable ecosystems.
