Overexpressed miRNA-155 dysregulates intestinal epithelial apical junctional complex in severe acute pancreatitis

AIM:To investigate whether miRNA-155(miR-155)dysregulates apical junctional complex(AJC)protein expression in experimental severe acute pancreatitis(SAP).METHODS:Twenty-four male BALB/c mice were randomly assigned to two groups:the SAP group(n=12)receiving sequential intraperitoneal injection of 50μg/kg caerulein and 10 mg/kg lipopolysaccharide over 6h,and the control group(n=12)receiving intraperitoneal injection of normal saline.Animals were sacrificed3 h following the last injection for collection of blood samples and pancreas and distal ileal segment specimens.Routine pancreas and intestine histology was used to assess SAP pathology and intestinal epithelial barrier damage.Levels of serum amylase,diamine oxidase(DAO),and tumor necrosis factor(TNF)-αwere determined using commercial kits.Total RNA samples were isolated from intestinal epithelial specimens and reversely transcribed into cDNA.miR-155 and RhoA mRNA expression profiles were determined using quantitative real-time polymerase chain reaction.Target genes for miR-155 were predicted using the miRTarBase database,RNA22 and PicTar computational methods.Western blotting was performed to quantitate the protein expression levels of the target gene RhoA,as well as zonula occludens(ZO)-1 and E-cadherin,two AJC component proteins.RESULTS:Intraperitoneal injection of caerulein and lipopolysaccharide successfully induced experimental acute pancreatic damage(SAP vs control,10.0±2.0vs 3.2±1.2,P<0.01)and intestinal epithelial barrier damage(3.2±0.7 vs 1.4±0.7,P<0.01).Levels of serum amylase(21.6±5.1 U/mL vs 14.3±4.2 U/mL,P<0.01),DAO(21.4±4.1 mg/mL vs 2.6±0.8 mg/mL,P<0.01),and TNF-α(61.0±15.1 ng/mL vs 42.9±13.9 ng/mL,P<0.01)increased significantly in SAP mice compared to those in control mice.miR-155 was significantly overexpressed in SAP intestinal epithelia(1.94±0.50 fold vs 1.03±0.23 fold,P<0.01),and RhoA gene containing three miR-155-specific binding sites in the three prime untranslated regions was one of the target genes for miR-155.RhoA(22.7±5.8 folds vs 59.6±

A photocatalytic[3+2]annulation of alkenes with vinyl azides was developed under irradiation by visible light in the presence of organic dye photocatalysts.Broad substrate scope and high functional group tolerance were demonstrated by more than 50 examples.The reaction provides a novel and efficient method for the synthesis of polyfunctionalized pyrroles under very mild metal-free conditions without other additives.

Congener-derived template to construct lithiophilic organic-inorganic layer/interphase for high volumetric capacity dendrite-free Li metal batteries

The development of lithium-metal batteries(LMBs)is seriously restricted by the out-of-control dendrites growth and infinite volume expansion.Herein,a pervasive organic-inorganic layer construction strategy is reported for the composite lithium metal anode with congener-derived organic-inorganic solid electrolyte interphase(SEI).In this strategy,the organic-inorganic Ag@polydopamine(Ag@PDA)layer is coated on the arbitrary substrates by a simple two-step method.The thin and stable congener-derived SEI is insitu formed with fewer inorganic components and more organic components during charging/discharging.The polydopamine with sufficient adhesion groups and lithiophilic Ag layer realize near-zero nucleation overpotential during lithium deposition.The low interface resistance and stable lithium deposition are achieved.Moreover,the practical areal and volumetric capacities of the composite anode with three-dimensional copper(3DCu)as the substrate are 10 mAh/cm^(2)and 1538 mAh/cm^3(vs.the mass of anode).The symmetrical cell shows very low polarization voltage(10 mV)and more than 2500 h cycles life at 1 mA/cm^(2)(1 mAh/cm^(2)).The LiNi_(0.8)Co_(0.1)Mn_(0.1)O_(2)(NCM811)-based full cells show improved capacity retention(82%)after 100 cycles at 0.5 C.The modified lithiophilic anode with congener-derived interphase provides a promising strategy to realize the next-generation dendrite-free LMBs.

Black hole evaporation and its remnants with the generalized uncertainty principle including a linear term

In recent years,researchers have investigated the evaporation of Schwarzschild black holes using various forms of the generalized uncertainty principle(GUP),metric quantum correction,and noncommutative geometry,respectively.However,there are differences between the GUP correction and the other two methods in terms of describing the later stages of black hole evaporation.Furthermore,some studies argue that the GUP with a negative parameter cannot effectively correct black hole evaporation,while others contend that the positivity or negativity of the GUP parameters should not affect the correction results.Taking the above into consideration,we reconsider black hole evaporation with the generalized uncertainty principle including a linear term(LGUP),and examine the case of negative parameters.The results indicate that the evaporation behavior of both Schwarzschild black holes and Reissner–Nordstr?m black holes,under LGUP correction,is consistent with the results of metric quantum correction and non-commutative geometry.Additionally,the negative parameter LGUP can also effectively correct for black hole evaporation.

Effects of dynamic ventilatory factors on ventilatorinduced lung injury in acute respiratory distress syndrome dogs

BACKGROUND:Mechanical ventilation is a double-edged sword to acute respiratory distress syndrome(ARDS) including lung injury,and systemic inflammatory response high tidal volumes are thought to increase mortality.The objective of this study is to evaluate the effects of dynamic ventilatory factors on ventilator induced lung injury in a dog model of ARDS induced by hydrochloric acid instillation under volume controlled ventilation and to investigate the relationship between the dynamic factors and ventilator-induced lung injuries(VILI) and to explore its potential mechanisms.METHODS:Thirty-six healthy dogs were randomly divided into a control group and an experimental group.Subjects in the experimental group were then further divided into four groups by different inspiratory stages of flow.Two mL of alveolar fluid was aspirated for detection of IL-8 and TNF-α.Lung tissue specimens were also extracted for total RNA,IL-8 by western blot and observed under an electronic microscope.RESULTS:IL-8 protein expression was significantly higher in group B than in groups A and D.Although the IL-8 protein expression was decreased in group C compared with group B,the difference was not statistically significant.The TNF-α ray degree of group B was significantly higher than that in the other groups(P0.05).The alveolar volume of subjects in group B was significantly smaller,and cavity infiltration and cell autolysis were marked with a significant thicker alveolar septa,disorder of interval structures,and blurring of collagenous and elastic fiber structures.A large number of necrotic debris tissue was observed in group B.CONCLUSION:Mechanical ventilation with a large tidal volume,a high inspiratory flow and a high ventilation frequency can cause significant damage to lung tissue structure.It can significantly increase the expression of TNF-α and IL-8 as well as their mRNA expression.Furthermore,the results of our study showed that small tidal ventilation significantly reduces the release of proinflamm

Ice sheet serves as a crucial indicator for assessing climate change.Mass loss in recent remote sensing-based studies indicated that the Antarctic Peninsula has rapid rates of glacier retreat and speed up of surface velocity.However,observations of seasonal variability of ice speed are limited,and glacier-area changes require multi-temporal monitoring.This study investigated the changes in area and surface velocities of∼375 glaciers on the northern Antarctic Peninsula(NAP)utilizing satellite images acquired by the Sentinel 1&2 satellites during 2018-2022.The results indicate that the glacier area reduced by approximately 166.1±44.2 km^(2)(-0.2%±0.1%per year)during the study period,with an acceleration after 2020(-0.4%±0.3%per year),and the most dramatic reduction happened on the eastern NAP.The maximum annual ice speeds on the NAP generally exceeded 3500 m per year,while the ice speeds in 2021 were the highest(exceeded 4210 m per year).The ice speed variability in austral autumn was higher than in other seasons,meanwhile the summer ice speeds showed an increasing trend.The glacier G012158E47018N,McNeile Glacier,glacier G299637E64094S and Drygalski Glacier showed the most remarkable ice speed variations represented by high daily velocities and strong fluctuations on their termini.Our results demonstrated that the variations in glacier area and seasonal ice speed on the NAP were responsive to the ice-ocean-atmosphere processes.Therefore,seasonal velocity and area variations should be considered when conducting accurate mass balance calculations,model validations and change mechanism analyses under climate warming scenarios.

Generalized Inverses and Units in a Unitary Ring

Let R be a unitary ring and a,b∈R with ab=0.We find the 2/3 property of Drazin invertibility:if any two of a,b and a+b are Drazin invertible,then so is the third one.Then,we combine the 2/3 property of Drazin invertibility to characterize the existence of generalized inverses by means of units.As applications,the need for two invertible morphisms used by You and Chen to characterize the group invertibility of a sum of morphisms is reduced to that for one invertible morphism,and the existence and expression of the inverse along a product of two regular elements are obtained,which generalizes the main result of Mary and Patricio(2016)about the group inverse of a product.

Numerical study of the influence of the atmospheric pressure on the thermal environment in the passenger cabin

The cabin air pressure remains lower than the horizontal atmospheric pressure when the airplane is in flight.Air pressure is one of the parameters that must be taken into consideration while studying the thermal environment of an airplane cabin.There are still no reference values for aircraft cabins despite the fact that numerous studies on low pressure heat transfer have demonstrated the connection between convective heat transfer coefficient(CHTC)and air pressure.In this paper,a correction method for CHTC under low pressure conditions was established by using the dummy heat dissipation in the low-pressure cabin experiment.On this basis,a thermal environment simulation model was developed,then was applied to the simulation of a seven-row aircraft cabin containing 42 passengers,and the CHTC and heat loss of dummy surface in the cabin were obtained.Finally,the results of PMV calculated by using heat dissipation and air parameters at sampling points were compared.The results show that the modified CHTC can accurately reflect the cabin thermal environment under low pressure conditions,and the correction of CHTC can be realized by adjusting the turbulent Prandtl number,which is nonlinear correlated with the pressure.The simulation results of the thermal environment in the seven-row cabin show that the CHTC changes by about 42%before and after modification.The air pressure decreases during take-off,which reduces the average CHTC of the crew surface from 5.09 W/(m^(2)·K)to 4.56 W/(m^(2)·K),but the air temperature rises by about 0.2°C as a whole.The deviation of PMV results calculated by using simulated heat loss data and using air parameters of measuring points in space is up to 0.5,but the latter is representative for calculating the thermal comfort level of the whole cabin.

Proteomics Study of Benzene Metabolite Hydroquinone Induced Hematotoxicity in K562 Cells

Objective Hydroquinone(HQ),one of the phenolic metabolites of benzene,is widely recognized as an important participant in benzene-induced hematotoxicity.However,there are few relevant proteomics in HQ-induced hematotoxicity and the mechanism hasn’t been fully understood yet.Methods In this study,we treated K562 cells with 40μmol/L HQ for 72 h,examined and validated protein expression changes by Label-free proteomic analysis and Parallel reaction monitoring(PRM),and performed bioinformatics analysis to identify interaction networks.Results One hundred and eighty-seven upregulated differentially expressed proteins(DEPs)and 279 downregulated DEPs were identified in HQ-exposed K562 cells,which were involved in neutrophilmediated immunity,blood microparticle,and other GO terms,as well as the lysosome,metabolic,cell cycle,and cellular senescence-related pathways.Focusing on the 23 DEGs and 5 DEPs in erythroid differentiation-related pathways,we constructed the network of protein interactions and determined 6 DEPs(STAT1,STAT3,CASP3,KIT,STAT5B,and VEGFA)as main hub proteins with the most interactions,among which STATs made a central impact and may be potential biomarkers of HQ-induced hematotoxicity.Conclusion Our work reinforced the use of proteomics and bioinformatic approaches to advance knowledge on molecular mechanisms of HQ-induced hematotoxicity at the protein level and provide a valuable basis for further clarification.

Study of the multi-physics field-coupled model of the two-stage electrostatic precipitator

The two-stage electrostatic precipitator is widely used to purify oil mist particles.However,there is limited research on the influences of relative humidity,particle deposition characteristics,and the generation of gaseous pollutants.Therefore,this paper established a numerical model of the electrostatic oil mist purifier and applied it to a two-stage electrostatic precipitator.Then the model was used to investigate the corona discharge characteristics under different relative humidity conditions in the charged zone,the particle deposition characteristics,the purification efficiency,the ozone concentration distribution,and the oil vapor concentration distribution in the collection zone.The results indicate that,with a constant temperature,the corona current decreases as relative humidity increase,and there is a quadratic relationship between relative humidity and current.The variation in relative humidity has little impact on the purification efficiency.The maximum ozone concentration occurs near the electrode line,and its concentration is influenced by the discharge current and inlet airflow velocity.The oil vapor concentration reaches its maximum value at the side plates,with a value of 19 ppb,while it reaches the minimum value at the collecting zone electrode plate,with a value of 2 ppb.The temperature is the main factor affecting the volatilization of the oil film,with higher temperatures resulting in higher oil vapor.
