Effects of Acupotomy on Immobilization-Induced Gastrocnemius Contracture and Fibrosis in Rats via Wnt/β-Catenin Signaling

Objective: To determine whether acupotomy ameliorates immobilization-induced muscle contracture and fibrosis via Wnt/β-catenin signaling pathway. Methods: Thirty Wistar rats were randomly divided into 5 groups(n=6) by a random number table, including control, immobilization, passive stretching, acupotomy, and acupotomy 3 weeks(3-w) groups. The rat model of gastrocnemius contracture was established by immobilizing the right hind limb in plantar flexion for 4 weeks. Rats in the passive stretching group received passive stretching at gastrocnemius, a daily series of 10 repetitions for 30 s each at 30-s intervals for 10 consecutive days. Rats in the acupotomy and acupotomy 3-w groups received acupotomy once and combined with passive stretching at gastrocnemius a daily series of 10 repetitions for 30 s each at 30-s intervals for 10 consecutive days. Additionally, rats in the acupotomy 3-w group were allowed to walk freely for 3 weeks after 10-day therapy. After treatment, range of motion(ROM), gait analysis [i.e., paw area, stance/swing and maximum ratio of paw area to paw area duration(Max dA/dT)], gastrocnemius wet weight and the ratio of muscle wet weight to body weight(MWW/BW) were tested. Gastrocnemius morphometric and muscle fiber cross-sectional area(CSA) were assessed by hematoxylin-eosin staining. Fibrosis-related mRNA expressions(i.e., Wnt 1, β-catenin, axin-2, α-smooth muscle actin, fibronectin, and types Ⅰ and Ⅲ collagen) were measured using real-time quantitative polymerase chain reactions. Wnt 1, β-catenin and fibronectin concentrations were measured by enzyme-linked immunosorbent assay. Types Ⅰ and Ⅲ collagen in the perimysium and endomysium were analyzed using immunofluorescence. Results: Compared with the control group, ROM, gait function, muscle weight, MWW/BW and CSA were significantly decreased in the immobilization group(all P <0.01), while protein levels of types Ⅰ and Ⅲ collagen, Wnt 1, β-catenin, fibronectin and mRNA levels of fibrosis-related genes were obviously increased(all P<0.01). Microstructure evolution and mechanical properties of brazing joint for ultra-thin-walled Inconel 718 considering grain size effect and brazing temperature

The systematic investigation of the mechanical properties and microstructure evolution process of ultra-thin-walled Inconel 718 capillary brazing joints is of great significance because of the exceptionally high demands on its application.To achieve this objective,this study investigates the impact of three distinct brazing temperatures and five typical grain sizes on the brazed joints’mechanical properties and microstructure evolution process.Microstructural evolution analysis was conducted based on Electron Back Scatter Diffraction(EBSD),Scanning Electron Microscopy(SEM),X-Ray Diffraction(XRD),High-Resolution Transmission Electron Microscopy(HRTEM),and Focused Ion Beam(FIB).Besides,the mechanical properties and fracture behavior were studied based on the uniaxial tension tests and in-situ tension tests.The findings reveal that the brazing joint’s strength is higher for the fine-grain capillary than the coarse-grain one,primarily due to the formation of a dense branch structure composed of G-phase in the brazing seam.The effects of grain size,such as pinning and splitting,are amplified at higher brazing temperatures.Additionally,micro-cracks initiate around brittle intermetallic compounds and propagate through the eutectic zone,leading to a cleavage fracture mode.The fracture stress of fine-grain specimens is higher than that of coarse-grain due to the complex micro-crack path.Therefore,this study contributes significantly to the literature by highlighting the crucial impact of grain size on the brazing properties of ultra-thin-walled Inconel 718 structures.

The relationship between the protective performance of flexible polymer material and material parameters(elasticmodulus,viscosity coefficient)is explored,an impact collision motion equation between two bodies is establishedfrom the viscoelastic material constitutive,and the relationship between the kinematic response and the materialparameters is obtained.Based on the Kelvin constitutive model,a theoretical model for impact between the pro-tective body and the protected body is established,then the dynamic response is obtained.The feasibility of themodel was verified by drop hammer experiment,and the material parameters(elastic modulus,viscosity coeffi-cient)were obtained by formula.The model is discretized and the relationship between local impact response andmaterial parameters is analyzed.The discussion results on the relationship between the impact response and theprotective material performance indicate that adjusting the elastic modulus,viscosity coefficient,and thicknessof the protective material can effectively improve protective effect.

Effects of long-term fencing on soil microbial community structure and function in the desert steppe,China

One of the goals of grazing management in the desert steppe is to improve its ecosystem.However,relatively little is known about soil microbe communities in the desert steppe ecosystem under grazing management.In this study,we investigated the diversity and aboveground biomass of Caragana korshinskii Kom.shrub communities in long-term fencing and grazing areas,combined with an analysis of soil physical-chemical properties and genomics,with the aim of understanding how fence management affects plant-soil-microbial inter-relationships in the desert steppe,China.The results showed that fence management(exclosure)increased plant diversity and aboveground biomass in C.korshinskii shrub area and effectively enhanced soil organic carbon(233.94%),available nitrogen(87.77%),and available phosphorus(53.67%)contents.As well,the Shannon indices of soil bacteria and fungi were greater in the fenced plot.Plant-soil changes profoundly affected the alpha-and beta-diversity of soil bacteria.Fence management also altered the soil microbial community structure,significantly increasing the relative abundances of Acidobacteriota(5.31%-8.99%),Chloroflexi(3.99%-5.58%),and Glomeromycota(1.37%-3.28%).The soil bacterial-fungal co-occurrence networks under fence management had higher complexity and connectivity.Based on functional predictions,fence management significantly increased the relative abundance of bacteria with nitrification and nitrate reduction functions and decreased the relative abundance of bacteria with nitrate and nitrite respiration functions.The relative abundances of ecologically functional fungi with arbuscular mycorrhizal fungi,ectomycorrhizal fungi,and saprotrophs also significantly increased under fence management.In addition,the differential functional groups of bacteria and fungi were closely related to plant-soil changes.The results of this study have significant positive implications for the ecological restoration and reconstruction of dry desert steppe and similar areas.

Esophageal cancer is one of the leading causes of cancer death in the world,with approximately half of the new cases occurring in China every year.^(1)Esophageal squamous cell carcinoma(ESCC)is the main subtype,accounting for more than 90%,and the five-year survival rate is less than 10%.Using large-scale genome analysis,many driver mutations and key pathways associated with ESCC have been identified.However,these genomic signatures have not improved the clinical management of EscC patients,or established effective targeted therapy.^(2)Esophageal cancer still lacks representative molecular markers.

Metal oxides with oxygen vacancies have a significant impact on catalytic activity for the transformation of organic pollutants in waste-to-energy(WtE)incineration processes.This study aims to investigate the influence of hematite surface oxygen point defects on the formation of environmentally persistent free radicals(EPFRs)from phenolic compounds based on the first-principles calculations.Two oxygen-deficient conditions were considered:oxygen vacancies at the top surface and on the subsurface.Our simulations indicate that the adsorption strength of phenol on theα-Fe_(2)O_(3)(0001)surface is enhanced by the presence of oxygen vacancies.However,the presence of oxygen vacancies has a negative impact on the dissociation of the phenol molecule,particularly for the surface with a defective point at the top layer.Thermo-kinetic parameters were established over a temperature range of300-1000 K,and lower reaction rate constants were observed for the scission of phenolic O-H bonds over the oxygen-deficient surfaces compared to the pristine surface.The negative effects caused by the oxygen-deficient conditions could be attributed to the local reduction of FeⅢto FeⅡ,which lower the oxidizing ability of surface reaction sites.The findings of this study provide us a promising approach to regulate the formation of EPFRs.

Ascophyllum nodosum and Fucus vesiculosus ameliorate restenosis via improving inflammation and regulating the PTEN/PI3K/AKT signaling pathway

Restenosis is a common complication following coronary angioplasty.The traditional use of seaweeds for health benefits has increasingly been explored,however few studies exist reporting its protective effects on the development of restenosis and gut dysbiosis.The aim of this study was to investigate the potential of seaweed extracts(SE) of Ascophyllum nodosum and Fucus vesiculosus in inhibiting intimal hyperplasia in a rat model of restenosis and its underlying mechanisms in macrophages and vascular smooth muscle cells(vSMCs).16S rRNA sequencing was done to investigate the regulatory effect of SE on the gut microbiome of injured rats.As indicated by the results,SE significantly inhibited the progression of intimal hyperplasia in vivo,attenuated inflammation in macrophages and could inhibit the proliferation,dedifferentiation and migration of vSMCs.It was observed through immunoblotting assays that treatment with SE significantly upregulated PTEN expression in macrophages and inhibited the upregulation of PI3K and AKT expression in vSMCs.Meanwhile,according to the 16S rRNA gene sequencing analysis,supplementation with SE modulated gut microbiota composition in injured rats.In conclusion,SE could ameliorate intimal hyperplasia by inhibiting inflammation and vSMCs proliferation through the regulation of the PTEN/PI3K/AKT pathway and modulating the gut microbiome.

Multi-scale data joint inversion of minerals and porosity in altered igneous reservoirs—A case study in the South China Sea

There are abundant igneous gas reservoirs in the South China Sea with significant value of research,and lithology classification,mineral analysis and porosity inversion are important links in reservoir evaluation.However,affected by the diverse lithology,complicated mineral and widespread alteration,conventional logging lithology classification and mineral inversion become considerably difficult.At the same time,owing to the limitation of the wireline log response equation,the quantity and accuracy of minerals can hardly meet the exploration requirements of igneous formations.To overcome those issues,this study takes the South China Sea as an example,and combines multi-scale data such as micro rock slices,petrophysical experiments,wireline log and element cutting log to establish a set of joint inversion methods for minerals and porosity of altered igneous rocks.Specifically,we define the lithology and mineral characteristics through core slices and mineral data,and establish an igneous multi-mineral volumetric model.Then we determine element cutting log correction method based on core element data,and combine wireline log and corrected element cutting log to perform the lithology classification and joint inversion of minerals and porosity.However,it is always difficult to determine the elemental eigenvalues of different minerals in inversion.This paper uses multiple linear regression methods to solve this problem.Finally,an integrated inversion technique for altered igneous formations was developed.The results show that the corrected element cutting log are in good agreement with the core element data,and the mineral and porosity results obtained from the joint inversion based on the wireline log and corrected element cutting log are also in good agreement with the core data from X-ray diffraction.The results demonstrate that the inversion technique is applicable and this study provides a new direction for the mineral inversion research of altered igneous formations.

Preparation and properties of high-energy-density aluminum/boroncontaining gelled fuels

Energetic nanofluid fuel has caught the attention of the field of aerospace liquid propellant for its high energy density(HED), but it suffers from the inevitable solid-liquid phase separation problem. To resolve this problem, herein we synthesized the high-Al-/B-containing(up to 30%(mass)) HED gelled fuels, with low-molecular-mass organic gellant Z, which show high net heat of combustion(NHOC), density, storage stability, and thixotropic properties. The characterizations indicate that the application of energetic particles to the gelled fuels obviously destroys their fibrous network structures but can provide the new particle-gellant gelation microstructures, resulting in the comparable stability between 1.0%(mass) Z/JP-10 + 30%(mass) Al or B and pure JP-10 gelled fuel. Moreover, the gelled fuels with high-content Al or B exhibit high shear-thinning property, recovery capability, and mechanical strength, which are favorable for their storage and utilization. Importantly, the prepared 1.0%(mass) Z/JP-10 + 30%(mass) B(or 1.0%(mass) Z/JP-10 + 30%(mass) Al) shows the density and NHOC 1.27 times(1.30) and 1.43 times(1.21)higher than pure JP-10, respectively. This work provides a facile and valid approach to the manufacturing of HED gelled fuels with high content of energetic particles for gel propellants.
