Troponin based studies in search of a biomarker for cardiac arrest

Cardiac arrest is shown to be a cause of a large number of deaths not only in Pakistan but around the globe. The prevalence of this dis-ease demands identification of its etiology. The science of proteomics can be used to identify cardiac specific proteins. The subsequent over expression or under expression of these pro-teins can be utilized as targets not only for therapeutical interventions but also for identi-fying molecular signatures for Cardiac diseases. In context of a number of studies which have shown that the specificity of serum biomarkers like troponin (cTnI and cTnT) are questionable as they may also appear in serum in pathologi-cal conditions other than cardiac dysfunction, the search of a specific marker for cardiac arrest becomes imperative. In this study protein pro-filing of cardiac arrest patients was performed after its quantification through Bradford assay. SDS-PAGE and 2 DE techniques were used as to characterize proteins. The samples of the pa-tients prior to characterizing of proteins were subjected to lipid and cardiac enzymes profiling. The results of these investigations have shown an increase in almost all of these parameters by many folds from that of normal values. In addi-tion to this the samples were found out to be positive for troponin T which strongly confirms the incidence of the cardiac arrest. The results of SDS-PAGE exhibited the induction of three proteins of 100 kDa, 97 kDa and of 66 kDa with 100 kDa as the most highly expressed protein. In addition to that SDS-PAGE gels have shown the down regulation of 45 kDa protein, again indi-cating the changes as a result of cardiac arrest. 2DE gel patterns of cardiac arrest samples demonstrated higher number of protein spots as compare to control in the alkaline range, which might suggest their role in cardiac dysfunction. Prospects of Using Platelets as Peripheral Marker to Study the Role of GABA in Autism

Literature indicated that platelets could be used as a model for neuronal receptors such as γ-amino butyric acid (GABA) and serotonin. Research work exhibited the presence of low levels of GABA and high levels of serotonin concentration in the platelets of autistic children as compare to their healthy counter parts. There are also other evidences pointing out to the significant role of GABA in autism such as association of g-band frequency with the cortical concentration of GABA and gabapentin (GABA analogue) specifically inhibits the cytosolic branched chain amino transferase (BCATc);an enzyme responsible to modulate glutamate availability for the synthesis of GABA.

Effects of Inoculum Density on Plant Growth and Hydrocarbon Degradation

The combined use of plants and bacteria is a promising approach for the remediation of soil contaminated with organic pollutants. Different biotic and abiotie factors can affect the survival and activity of the applied bacteria and consequently plant growth and phy- toremediation efficiency. The effect of inoculum density on the abundance and expression of alkune-degrading genes in the rhizosphere of plant vegetated in hydrocarbon-contaminated soil has been rarely observed. In this study, an alkane-degrading bacterium （Pantoea sp. strain BTRH79）, at different inoculum densities （10^5 to 10^8 cells cm^-3 soil）, was inoculated to ryegrass （Lolium perenne） vegetated in diesel-contaminated soil to find the optimum inoculum density needed for its efficient colonization and hydrocarbon degradation activity. Bacterial inoculation improved plant growth and hydrocarbon degradation. Maximum plant growth and hydrocarbon degra- dation were observed with the inoculum having the highest cell density （10^8 cells cm^-3 soil）. Moreover, the inoculum with higher cell density exhibited more abundance and expression of alkane hydroxylase gene, CYP153. This study suggests that the inoculum density is one of the main factors that can affect bacterial colonization and activity during phytoremediation.

Chronic active hepatitis(CAH) is acknowledged as an imperative risk factor for the development of liver injury and hepatocellular *** histological end points of CAH are chronic inflammation,fibrosis and cirrhosis which are coupled with increased DNA synthesis in cirrhotic vs healthy normal *** potential mechanism involved in CAH includes a combination of processes leading to liver cell necrosis,inflammation and cytokine production and liver scaring(fibrosis).The severity of liver damage is regulated by Hepatitis B virus genotypes and viral *** viral and cellular factors that contribute to liver injury are discussed in this *** injury caused by the viral infection affects many cellular processes such as cell signaling,apoptosis,transcription,DNA repair which in turn induce radical effects on cell survival,growth,transformation and *** consequence of such perturbations is resulted in the alteration of bile secretion,gluconeogenesis,glycolysis,detoxification and metabolism of carbohydrates,proteins,fat and balance of *** identification and elucidation of the molecular pathways perturbed by the viral proteins are important in order to design effective strategy to minimize and/or restore the hepatocytes injury.

Hepatocellular carcinoma(HCC)is one of the leading causes of death induced by cancer in the modern world and majority of the cases are related to chronic hepatitis B virus(HBV)***-encoded X protein(HBx)is known to play a pivotal role in the pathogenesis of viral induced *** is a multifunctional protein of17 kDa which modulates several cellular processes by direct or indirect interaction with a repertoire of host factors resulting in *** might interfere with several cellular processes such as oxidative stress,DNA repair,signal transduction,transcription,protein degradation,cell cycle progression and apoptosis.A number of reports have indicated that HBx is one of the most common viral ORFs that is often integrated into the host genome and its sequence variants play a crucial role in *** mutational or deletion analysis it was shown that carboxy terminal of HBx has a likely role in protein-protein interactions,transcriptional transactivation,DNA repair,cell,signaling and pathogenesis of *** accumulated evidence thus far suggests that it is difficult to understand the mechanistic nature of HBx associated HCC,and HBx mediated transcriptional transactivation and signaling pathways may be a major *** article addresses the role of HBx in the development of HCC with particular emphasis on HBx mutants and their putative targets.

The recent trend in tourism marketing is focus on customer relationship management. Tourism industry today is one of the highest revenue generating industry and strategic approach for sustainable development of this industry hosts benefits not only for the tourism related stakeholders but also the community and economy on the whole. Oman is one of the most preferred destinations for the tourists especially after the declaration of Muscat, Arab tourism Capital for 2012. Thus to materialize this honor and position, the scientific study and analysis of tourist behavior will help to predict the future trend of tourism and will give direction for effort investment. This work presents a novel strategy to identify, analyze and highlights the main tourist behavioral factors that could increase tourists＇ loyalty to a specific destination or agency. The analytical TSS （tourism support system） will also classify customers into two categories--first category will be classified as ＂LT （loyal tourists）＂ and second category as ＂NLT （non-loyal tourists）＂. Dataset is collected from a tourism business organization. Twenty-four attributes and 545 instances were collected and were analyzed by algorithms like logistics, forest of random trees, naive Bayes, J48 and Id3. The explanatory variables were defined, and some transformations were done to identify the response variable. Entropy was used and adapted in order to find the response variable from the explanatory variables. The results obtained from this work confirm that the generated rules can be used for future prediction and tourism business can be improved and efforts can be directed in right place for the right consumer resulting in high return on investment.

The Role of Trace Metals and Environmental Factors in the Onset and Progression of Schizophrenia in Pakistani Population

Exposure to toxic chemicals appears to be one of the major factors in the onset of Schizophrenia. Present study was designed to find out the association of socio-economic, clinical and heavy metals such as chromium (Cr), lead (Pb) copper (Cu), zinc (Zn), iron (Fe), manganese (Mn) selenium (Se) and arsenic (As) as the principle environmental factors that appear to impact schizophrenic condition in Pakistani population. In this study we have tried to expose some factors such as drug abuse, marital status, education, monthly income that could be related to the disease. These parameters were not investigated before in Pakistani subjects. A self-made questionnaire was developed to collect and record the history of patient’s social and economical status. The level of transition metals in the whole blood was also measured by using Inductive couple plasma optical emission spectroscopy (ICP-OES). Present study has found higher levels of Cr, Pb, Zn, Se, As and Cu and lower levels of Fe and Mn in the blood of newly diagnosed (ND) patients when compared to the controls. While comparing ND with the patients who were on medication (Old Diagnosed) we, found lower level of these metals except for copper in ND. However no significant differences were observed between any trace metal levels between the studied groups. The survey concludes that economic status, marital status and illegal drugs are significantly associated with schizophrenia. Pakistani men who use cannabis are significantly higher in numbers when compared to women in acquiring the psychiatric symptoms.

Host nucleotide polymorphism in hepatitis B virusassociated hepatocellular carcinoma

Hepatocellular carcinoma(HCC) is etiologically linked with hepatitis B virus(HBV) and is the leading cause of death amongst 80% of HBV patients. Among HBV affected patients, genetic factors are also involved in modifying the risk factors of HCC. However, the genetic factors that regulate progression to HCC still remain to be determined. In this review, we discuss several single nucleotide polymorphisms(SNPs) which were reportedly associated with increased or reduced risk of HCC occurrence in patients with chronic HBV infection such as cyclooxygenase(COX)-2 expression specifically at COX-2-1195G/A in Chinese, Turkish and Egyptian populations, tumor necrosis factor α and the three most commonly studied SNPs: PAT-/+, Lys939Gln(A33512C, rs2228001) and Ala499Val(C21151T, rs2228000). In genome-wide association studies, strong associations have also been found at loci 1p36.22, 11q22.3, 6p21(rs1419881, rs3997872, rs7453920 and rs7768538), 8p12(rs2275959 and rs37821974) and 22q11.21. The genes implicated in these studies include HLA-DQB2, HLA-DQA1, TCF19, HLA-C, UBE2L3, LTL, FDX1, MICA, UBE4 B and PG. The SNPs found to be associated with the above-mentioned genes still require validation in association studies in order to be considered good prognostic candidates for HCC. Screening of these polymorphisms is very beneficial in clinical experiments to stratify the higher or lower risk for HCC and may help in designing effective and efficient HCC surveillance programs for chronic HBV-infected patients if further genetic vulnerabilities are detected.

Chronic hepatitis C virus(HCV) infection exhibits a wide range of extrahepatic complications, affecting various organs in the human body. Numerous HCV patients suffer neurological manifestations, ranging from cognitive impairment to peripheral neuropathy. Overexpression of the host immune response leads to the production of immune complexes, cryoglobulins, as well as autoantibodies, which is a major pathogenic mechanism responsible for nervous system dysfunction. Alternatively circulating inflammatory cytokines and chemokines and HCV replication in neurons is another factor that severely affects the nervous system. Furthermore, HCV infection causes both sensory and motor peripheral neuropathy in the mixed cryoglobulinemia as well as known as an important risk aspect for stroke. These extrahepatic manifestations are the reason behind underlying hepatic encephalopathy and chronic liver disease. The brain is an apt location for HCV replication, where the HCV virus may directly wield neurotoxicity. Other mechanisms that takes place by chronic HCV infection due the pathogenesis of neuropsychiatric disorders includes derangement of metabolic pathways of infected cells, autoimmune disorders, systemic or cerebral inflammation and alterations in neurotransmitter circuits. HCV and its pathogenic role is suggested by enhancement of psychiatric and neurological symptoms in patients attaining a sustained virologic response followed by treatment with interferon; however, further studies are required to fully assess the impact of HCV infection and its specific antiviral targets associated with neuropsychiatric disorders.
