Prolonged high-fat-diet feeding promotes non-alcoholic fatty liver disease and alters gut microbiota in mice

BACKGROUND Non-alcoholic fatty liver disease (NAFLD) has become an epidemic largely due to the worldwide increase in obesity. While lifestyle modifications and pharmacotherapies have been used to alleviate NAFLD, successful treatment options are limited. One of the main barriers to finding safe and effective drugs for long-term use in NAFLD is the fast initiation and progression of disease in the available preclinical models. therefore, we are in need of preclinical models that (1) mimic the human manifestation of NAFLD and (2) have a longer progression time to allow for the design of superior treatments. AIM to characterize a model of prolonged high-fat diet (HFD) feeding for investigation of the long-term progression of NAFLD. MEtHODS In this study, we utilized prolonged HFD feeding to examine NAFLD features in C57BL/6 male mice. We fed mice with a HFD (60% fat, 20% protein, and 20% carbohydrate) for 80 wk to promote obesity (Old-HFD group, n = 18). A low-fat diet (LFD)(14% fat, 32% protein, and 54% carbohydrate) was administered for the same duration to age-matched mice (Old-LFD group, n = 15). An additional group of mice was maintained on the LFD (Young-LFD, n = 20) for a shorter duration (6 wk) to distinguish between age-dependent and age-independent effects. Liver, colon, adipose tissue, and feces were collected for histological and molecular *** Prolonged HFD feeding led to obesity and insulin resistance. Histological analysis in the liver of HFD mice demonstrated steatosis, cell injury, portal and lobular inflammation and fibrosis. In addition, molecular analysis for markers of endoplasmic reticulum stress established that the liver tissue of HFD mice have increased phosphorylated Jnk and CHOP. Lastly, we evaluated the gut microbial composition of Old-LFD and Old-HFD. We observed that prolonged HFD feeding in mice increased the relative abundance of the Firmicutes phylum. Acetaminophen and zinc phosphide for lethal management of invasive lizards Ctenosaura similis

Reducing populations of invasive lizards through trapping and shooting is feasible in many cases but effective integrated management relies on a variety of tools, including toxicants. In Florida, using wild-caught non-native black spiny-tailed iguanasCtenosaura similis, we screened acetaminophen and zinc phosphide to determine their suitability for effective population management of this prolific invasive species. Of the animals that received acetaminophen, none died except at the highest test dose, 240 mg per lizard, which is not practical for field use. Zinc phosphide produced 100% mortality at dose levels as little as 25 mg per lizard, equivalent to about 0.5% in bait which is lower than currently used in commercial baits for commensal rodent control. We conclude that zinc phosphide has potential as a useful tool for reducing populations of invasive lizards such as the black spiny-tailed iguana provided target-selective delivery methods are developed.

Based on the normal pulsed laser ablation method,femtosecond pulsed laser deposition(fs-PLD)is adopted in vacuum for the production of tiO2nanoparticle-assembled *** study the morphology and electronic characteristics of tiO2nanoparticle-assembled films deposited at different oxygen background gas pressures from high vacuum(~10-4Pa)to 100 Pa and different deposition *** results show that tiO2nanoparticle-assembled films obtained in high vacuum present both a mixture with rutile phase and anatase phase and a pure rutile *** the same time,there are more mesoporous structures in the film after annealing,which is beneficial for the enhancement of photocatalytic *** water splitting experiment,part of the tiO2nanoparticle-assembled films embedded with a small mass fraction of CdS nanoparticles(~5%)present an interesting photocurrent enhancement with a maximum value of^0.2mA/cm2under a solar simulator.

Clinical profile of mechanically ventilated COVID-19 patients:A retrospective observational study from Dubai

Background:We did a retrospective analysis of critical coronavirus disease 2019(COVID-19)patients admitted to our intensive care unit(ICU).the objective was to evaluate the outcome,risk factors and effect of prone position in critically ill patients requiring invasive mechanical ventilation(IMV).Patients and methods:the data were collected regarding demographics,comorbidities,laboratory parameters and *** regression was used for analysis of the association of risk factors to the ***:From 15 March to 30 May 2020,35(59.3%)out of 59 critical COVID-19 requiring IMV were admitted to a tertiary care hospital in *** day-28 ICU mortality was 28.8% and 48.6% in patients requiring *** position(PP)was used in 17(48.6%)patients for median duration of 19(5-20)hours with significant PaO_(2)/FiO_(2) *** kidney injury was common(30.5%),and half of the patients required renal replacement therapy(RRt)with higher mortality(77.8%).Lactate dehydrogenase(LDH)odd ratio(OR)-1.006[95%CI-1.00-1.01],D-dimer(OR-1.003[1.000-1.000]),low total leucocyte count(OR-1.135[1.01-1.28]),and lymphopenia(OR-0.909[0.84-0.98])were independently associated with increased risk of ***:IMV requirement in patients with COVID-19 is associated with higher *** markers like LDH,D-dimer,and lymphopenia can be used to predict the *** patients with COVID-19 on IMV respond significantly with prone position,and it should be considered early with a longer duration.
