Properties of Recycled-Polyethylene Terephthalate/Polycarbonate Blend Fabricated by Vented Barrel Injection Molding

In this research, recycled-polyethylene terephthalate (PET) and polycarbonate (RPET/PC) blends fabricated by vented barrel injection molding were presented to better understand the effect of devolatilization during molding process. The effect of dried pellets, non-dried pellets, using an opened-vented hole, and using a closed-vented hole on the miscibility, morphology, thermal properties and mechanical properties of RPET/PC blends was investigated. The results indicated that no drying decreases dispersion, thermal properties, and mechanical properties of RPET/PC blends due to hydrolysis degradation of recycled-PET during the injection molding process. Using the venting system with non-dried RPET/PC blends partially improves dispersion, thermal properties and molecular weight of RPET/PC blends processed without drying, giving results that are similar to those processed with drying. Regarding the flexural properties, using the venting system without drying prevents the flexural properties from decreasing in RPET/PC blends, if the amount of RPET is less than 75 wt%. When the content of RPET is over 75 wt%, using the venting system does not eliminate the decrease in flexural properties of RPET/PC blends. When the venting system is applied to non-dried RPET, despite hydrolysis degradation of RPET not being completely eliminated, the damaging effects are nonetheless reduced compared with those samples processed without the venting system. As a result, vented barrel injection molding hardly prevents non-dried RPET/PC blends from having reduced flexural properties when the content of RPET is greater than 75 wt%.

Endogenous prostaglandin D<sub>2</sub>synthesis inhibits e-selectin generation in human umbilical vein endothelial cells

We examined the role of prostaglandin D2 (PGD2) in the formation of E-selectin following inter-leukin-1 (IL-1) stimulation in human umbilical vein endothelial cells (HUVEC) transfected with lipocaline-type PGD2 synthase (L-PGDS) genes. HUVEC were isolated from human umbilical vein and incubated with 20 U/mL IL-1 and various concentrations of authentic PGD2. The isolated HUVEC were also transfected with L-PGDS genes by electroporation. The L-PGDS-transfected HUVEC were used to investigate the role of endogenous PGD2 in IL-1-stimulated E-selectin biosynthesis. We also used an anti-PGD2 antibody to examine whether an intracrine mechanism was involved in E-selectin production. PGD2 and E-selectin levels were determined by radio-immunoassay and enzyme- immunoassay, respectively. E-selectin mRNA was assessed by real-time RT-PCR. IL-1-stimulated E-selectin production by HUVEC was dose-dependently inhibited by authentic PGD2 at concentrations greater than 10–6 mol/L. L-PGDS gene-transfected HUVEC produced more PGD2 than HUVEC transfected with the reporter gene alone. IL-1 induced increases in E-selectin production in HUVEC transfected with the reporter genes alone. However, this effect was significantly attenuated in the case of IL-1 stimulation of HUVEC trans-fected with L-PGDS genes, and accompanied by an apparent suppression of E-selectin mRNA expression. Neutralization of extracellular PGD2 by anti-PGD2- specific antibody influenced neither E-selectin mRNA expression nor E-selectin biosynthesis. HUVEC transfected with L-PGDS genes showed increased PGD2 synthesis. This increase was associated with attenuation of both E-selectin generation and E-selectin mRNA expression. The results suggest that endogenous PGD2 decreases E-selectin synthesis and E-selectin mRNA expression, probably through an intracrine mechanism.

The Determination of Interfacial Shear Strength in Short Fiber Reinforced Poly Ethylene Terephthalate by Kelly-Tyson Theory

The interfacial shear strength value measuring by the modified Kelly-Tyson equation method was studied the measurement accuracy. The measuring accuracy by using the modified Kelly-Tyson equation method is compared to the nano-indentation testing method. The results and an influential factor are described. An error in the modified Kelly-Tyson equation is verified to avoid the incorrect measurement when the interfacial shear strength was measured by the modified Kelly-Tyson equation. To study the different interfacial shear strength behavior, short fiber reinforced PET composites were fabricated. In this study, an advance fabricating technique for short fiber reinforced composite as direct fiber feeding process is conducted to fabricate GF/recycled PET for studying the interfacial shear strength. The result indicates that the modified Kelly-Tyson equation method accurately provides the accurate interfacial shear strength value, if it is conducted with the sample without a horizontally aligned fiber. So the high fiber loading content sample should be avoided to get the more accuracy result. The large horizontally aligned fiber area into specimens extremely resulted in the incorrect measurement of the interfacial shear strength value by the modified Kelly-Tyson equation method. The fiber agglomeration factor and the sensitively horizontally aligned fiber area must be considered its influence on the measuring for improving the equation effectiveness.

GLP-1 and GIP promote insulin secretion from pancreatic β-cells by inducing intracellular signals such as Ca2+ and cAMP. Metformin primarily acts by inhibiting glucogenesis in the liver and promoting glucose metabolism in the muscle. It is used as a concomitant drug with the incretin in the treatment of T2D. We focused on intracellular signals under various glucose concentrations and assessed the effects of metformin on incretin signaling in MIN6 β-cells. Metformin inhibited incretin-induced [Ca2+]i in the presence of 5.5 mM glucose but not 16.7 mM glucose. In accordance with low [Ca2+]i, insulin secretion from MIN6 cells declined, despite enhanced incretin-induced cAMP production. Abundant expressions of Adcy 6 and 9, which are negatively controlled by Ca2+ signals, were detected in MIN6 cells. Thus, increasing cAMP production was associated with the inhibition of Ca2+ mobilization by metformin. However, we show that metformin controls insulin secretion by inhibiting incretin-mediated [Ca2+]i under normoglycemic conditions.

Computer-based simulations are essential for clarifying the hemodynamics of brain aneurysms. Since cerebrovascular disease is often fatal, it is strongly desirable to predict its progression. While previous studies have clarified the initiation mechanism of aneurysms, their growth mechanism remains unclear. Consequently, it is difficult to develop a diagnostic system for predicting aneurysm rupture. This study seeks to clarify the mechanism of aneurysm growth by identifying significant hydrodynamic factors. We focus on a single ruptured aneurysm that was followed up for five years. Computer simulations and fluid dynamic experiments with silicone vessel models were performed. To confirm the reliability of data in the computer simulations, we conducted particle image velocimetry measurements in steady flow. We then performed computer simulations for pulsatile conditions to determine an effective index for aneurysm growth. We obtained good agreement between the trends in the obtained computer simulation and experimental data. Numerical simulations for pulsatile flow in three models revealed that aneurysms grew in regions having a low wall shear stress, a low aneurysm formation indicator, and a high oscillatory shear index.

Short Hydration in Chemotherapy with Cisplatin plus S-1 for Advanced or Recurrent Gastric Cancer: A Retrospective Study

Background: Despite there are a few reports that assessed the S-1 + CDDP regimen with short hydration regimen for unresectable or metastatic gastric cancer, there is no consensus on the best regimen for short hydration. The aim of study was to evaluate the safety and the efficacy of S-1 plus cisplatin doublet chemotherapy with short hydration. Methods: S-1 was administered orally (p.o.) twice daily for the first 3 weeks of a 5-week cycle. Dose of S-1 administered was calculated according to the body surface area. CDDP was given as an intravenous (i.v.) infusion of 60 mg/m2 on day 8 of each cycle. Patients received the total of 1900 ml infusion containing 1000 ml of acetate Ringer’s solution as pre- and post-hydraion. 300 ml of 20% mannitol was administered as a diuretic. Results: 35 patients with unresectable or recurrent gastric cancer were enrolled. The reasons for termination of S-1 + CDDP were as follows: 21 (63.6%) by progressive disease;12 (31.4%) by toxicity. Even though 12 of 35 patients (34.2%) were discontinued S-1 + CDDP chemotherapy, only one patient was discontinued by Grade 2 of increased creatinine. TTF (time to progression) was 174 days (3 - 586 days), and the median of the total number of treatment cycles of S-1 + CDDP was 3.31. Median overall survival, as secondary endpoint, was 518 days. Conclusions: Our study suggested that the short hydration regimen is as safe and efficient as the continuous hydration regimen.

Objective. Population-specific differences in the genetic susceptibility to inflammatory bowel disease (IBD) are indicated by the fact that Crohn’s disease (CD) in Japanese patients does not have any of the common CARD15 variants that are associated with CD in Caucasians. Recently, the disease-causing mutation in the IBD5 haplotype was identified. The TC haplotype, composed of L503F in SLC22A4 and -207G/C in SLC22A5 promoters, was reported to alter the function of the organic cation transporter and to be associated with CD in Caucasians. To determine whether the TC haplotype is also associated with IBD in a Japanese population, we genotyped L503F and -207G/C variants in Japanese subjects. Furthermore, we also performed a case-control association study with all representative single nucleotide polymorphisms (SNPs) in IBD5 using previous information of linkage disequilibrium extension reported in Japanese patients to determine whether there were variants in IBD5 specifically associated with IBD in Japanese patients. Material and methods. A total of 758 Japanese individuals, 241 patients with CD, 247 patients with ulcerative colitis (UC) and 270 healthy controls, were analyzed in this study. Genotyping for SNPs was determined by polymerase chain reaction-restriction fragment length polymorphism analysis. Results. We found L503F and -207G/C to be very rare (< 1%frequency) in CD, UC and HC in the Japanese population. Furthermore,we also found that none of the representative SNPs in IBDS was associated with CD or UC in the Japanese subjects. Conclusions. In contrast to Caucasians, IBD5 is not a major component of the susceptibility to IBD in the Japanese population.
