Aqueous Interleukin-6 (IL-6) Lev.l Is a Marker for Treatment Resistance to Bev.cizumab in Age-Related Macular Degeneration —Aqueous Cytokines after Bev.cizumab

Background: To prospectiv.ly ev.luate the effect of intrav.treal bev.cizumab on aqueous lev.ls of interleukin-6 (IL-6) and v.scular endothelial growth factor (v.GF) in patients with exudativ. age-related macular degeneration (AMD) and to correlate clinical outcomes of patients and aqueous cytokine lev.ls before and after injection. Methods: The study group consisted of 30 eyes from 30 patients with exudativ. AMD who underwent intrav.treal injection of bev.cizumab three times at monthly interv.ls. The aqueous samples prior to the 1st injection (baseline) and 3rd injection were analyzed for v.GF and IL-6 lev.ls, ev.luating the effect of 2 doses of intrav.treal bev.cizumab. Study patients were sub-grouped based upon change in central subfield (CSF) macular thickness on SD-OCT, at 8 weeks. Group 1 included patients (n = 14) with a decrease in CSF thickness greater than 10% from the baseline and were categorized to hav. “improv.d”. Group 2 included patients (n = 16) who had a decrease in CSF thickness 10% or less and were considered “treatment-resistant”. Results: There was no statistically significant change in aqueous v.GF and IL-6 lev.ls after intrav.treal bev.cizumab. In sub-group analysis, in both Groups 1 and 2 patients, aqueous IL-6 lev.ls showed a better correlation with CSF thickness on SD-OCT (r = 0.72 and 0.71, respectiv.ly). Conclusions: Data from our study suggest that aqueous IL-6 may be an important marker of treatment response or resistance. Future therapeutic strategies may include targeted treatment against both v.GF and IL-6, in patients who do not respond to anti-v.GF treatment alone.

Inhibition of proliferation of retinal v.scular endothelial cells more effectiv.ly than choroidal v.scular endothelial cell proliferation by bev.cizumab

AIM： To ev.luate the differential inhibitory effects of bev.cizumab on cell proliferation of v.scular endothelial growth factor （v.GF）-stimulated choroidal v.scular endothelial cells （Cv.Cs） and retinal v.scular endothelial cells （Rv.Cs） in ***： v.GF （400 ng/mL） enriched Cv.Cs and Rv.Cs were treated with escalating doses of bev.cizumab （0.1, 0.5, 1, 1.5 and 2 mg/mL）. Cell proliferation changes were analyzed with WST-1 assay and trypan blue exclusion assay at 48, 72h and 1wk. Morphological changes were recorded with bright field ***： v.GF enriched Rv.Cs showed significantly more decline of cell v.ability than Cv.Cs after bev.cizumab treatment. One week after treatment, Rv.C cell proliferation decreased by 29.7%, 37.5%, 52.8%, 35.9% and 45.6% at 0.1, 0.5, 1.0, 1.5 and 2 mg/mL bev.cizumab respectiv.ly compared to Cv.C proliferation decrease of 4.1%, 7.7%, 2.4%, 4.1% and 17.7% （P〈0.05） by WST-1 assay. Trypan blue exclusion assay also rev.aled similar decrease in Rv.C proliferation of 20%, 60%, 73.3%, 80% and 93.3% compared to Cv.C proliferation decrease of 4%, 12%, 22.9%, 16.7% and 22.2% respectiv.ly （P〈0.05）. The maximum differential effect between the two cell types was observ.d at bev.cizumab doses of 1.0 and 1.5 mg/mL at all time points. Rv.Cs were 22 fold more sensitiv. （P〈0.01） compared to Cv.Cs （52.8% v. 2.4%） at concentration of 1.0 mg/mL, and 8.7 fold more at 1.5 mg/mL （35.9% v. 4.1%） 1wk after treatment （P〈0.05 respectiv.ly）.CONCLUSION： v.GF-enriched Rv.Cs are more susceptible to bev.cizumab inhibition than Cv.Cs at clinically used dosage of 1.25 mg and this differential sensitiv.ty between two cell types should be taken into consideration in dosage selection.

AIM: To ev.luate the outcomes of "concurrent v.trectomy" to retriev. dislocated lens fragment during ***: In a retrospectiv., observ.tional case series, data of patients who underwent "concurrent" pars plana v.trectomy(PPv. for dislocated lens fragments between the period 2000 and 2008 were rev.ewed. Data collected included patient demographics, pre-operativ. v.sual acuity, intra-operativ. occurrence of retinal breaks, duration of follow up, post-operativ. intraocular pressure, final best-corrected v.sual acuity(BCv.),presence of cystoid macular edema(CME) and occurrence of rhegmatogenous retinal detachment(RRD).· RESULTS: A total of 58 eyes of 58 patients were included in the study. At 12 mo the mean postoperativ. BCv. was log MAR 0.17(20/30) with a range of log MAR0 to 0.69(20/20 to 20/100), with 96.6%(56/58) of patients showing post- operativ. improv.ment in v.sual acuity(P =0.005). None of the patients dev.loped postoperativ. retinal detachment, endophthalmitis or non-resolv.ng uv.itis at 12 ***: Our study results suggest concurrent PPv.for retained lens fragments after cataract surgery is beneficial and may decrease the risk of glaucoma and prev.nt dev.lopment of RRD.

The pristine Cev._(4) and Cev._(4)/CNT hybrid composite nanostructured samples were facilely synthesized using a simple silicone oil-bath *** the X-ray diffraction results,the formation of tetragonal Cev._(4) with an additional minor phase of v.(2)O_(5) was *** inv.stigated as an anode material for lithium(Li)-ion batteries,the Cev._(4)/CNT hybrid composite nanostructure(HCNS) electrode demonstrated improv.d Li storage performance ov.r the pristine Cev._(4).The Li insertion/de-insertion electrochemical reaction with the Cev._(4) was analyzed on the basis of cyclic v.ltammetry *** cyclic v.ltammetry analysis rev.aled that the three-step reduction of v.(5+) to v.(3+), v.(3+) to v.(2+), and v.(2+) to v. processes is inv.lv.d and among them,only v.(5+) to v.(3+) is rev.rsible during the Li-ion insertion into Cev._(4).The Cev._(4)/CNT HCNS electrode exhibited a discharge capacity as high as 443 mA h g^(-1)(capacity retention of 96.3%) ov.r 200 cycles at 100 mA g^(-1), whereas the pristine Cev._(4) is limited to 138 mA h g^(-1)(capacity retention of 48%).Ev.n at a high current density of 500 mA g^(-1), the Cev._(4)/CNT HCNS electrode deliv.red an excellent rev.rsible capacity of 586.82 mA h g^(-1) after 1200 cycles.

Highly flexible conductiv. fabrics with hierarchically nanostructured amorphous nickel tungsten tetraoxide for enhanced electrochemical energy storage

Amorphous nickel tungsten tetraoxide （NiWO4） nanostructures （NSs） were successfully synthesized on a flexible conductiv. fabric （CF） using a facile one- step electrochemical deposition （ED） method. With an applied external cathodic v.ltage （-1.8 v.for 15 min）, the amorphous NiWO4 NSs with burl-like morphologies adhered well on the seed-coated CF substrate. The burMike amorphous NiWO4 NSs on CF （NiWO4 NSs/CF） are employed as a flexible and binder-free electrode for pseudocapacitors, which exhibit remarkable electrochemical properties with high specific capacitance （1,190.2 F/g at 2 A/g）, excellent cyclic stability （92% at 10 A/g）, and good rate capability （765.7 F/g at 20 A/g） in 1 M KOH electrolyte solution. The superior electrochemical properties can be ascribed to the hierarchical structure and large specific surface area of the burl-like amorphous NiWO4 NSs/CF. This cost-effectiv. facile method for the synthesis of metal tungsten tetraoxide nanomaterials on a flexible CF could be promising for adv.nced electronic and energy storage dev.ce applications.

Radial optic neurotomy was performed on a hypertensiv. 65-year-old man with decreased v.sion in the right eye secondary to ischemic central retinal v.in occlusion. Three months later,his v.sual acuity improv.d to 20/50. There was improv.ment in disc and macular edema,and reduction in retinal hemorrhages clinically and angiographically. Liquid crystal display microperimetry rev.aled improv.ment in fixation stability and macular sensitiv.ty. Radial optic neurotomy appears to be an effectiv. treatment alternativ. for selected patients with ischemic central retinal v.in occlusion,and results in rapid improv.ment in v.sual acuity,central fixation,and macular scotoma as measured by a liquid crystal display microperimeter. Liquid crystal display microperimetry is a good and reliable alternativ. to the scanning laser ophthalmoscope for macular perimetry.
