RNA capping and decapping tightly coordinate with transcription,translation,and RNA decay to regulate gene expression.Proteins in the DXO/Rai1 family have been implicated in mRNA decapping and decay,and mammalian DXO was recently found to also function as a decapping enzyme for NAD+-capped RNAs(NAD-RNA).The Arabidopsis genome contains a single gene encoding a DXO/Rai1 protein,AtDXO1.Here we show that AtDXO1 possesses both NAD-RNA decapping activity and 5?-3?exonuclease activity but does not hydrolyze the m7G cap.The atdxo1 mutation increased the stability of NAD-RNAs and led to pleiotropic phenotypes,including severe growth retardation,pale color,and multiple devel-opmental defects.Transcriptome profiling analysis showed that the atdxo1 mutation resulted in upregulation@of defense-related genes but downregulation of photo-synthesis-related genes.The autoimmunity phenotype of the mutant could be suppressed by either eds1 or npr1 mutation.However,the various phenotypes associated with the atdxo1 mutant could be complemented by an enzymatically inactive AtDXO1.The atdxo1 mutation ap-parently enhances post-transcriptional gene silencing by elevating levels of siRNAs.Our study indicates that AtDXO1 regulates gene expression in various biological and physiological processes through its pleiotropic mo-lecular functions in mediating RNA processing and decay.

Serine palmitoyltransferase(SPT),a pyridoxyl-5′-phosphate-dependent enzyme,catalyzes the first and rate-limiting step in sphingolipid biosynthesis. In humans and yeast,the orosomucoid proteins(ORMs) negatively regulate SPT and thus play an important role in maintaining the levels of sphingolipids. Despite the importance of sphingoid intermediates as bioactive molecules,the regulation of sphingolipid biosynthesis through SPT is not well understood in plants. In the present study we identified and characterized the Arabidopsis thaliana ORMs,At ORM1 and At ORM2. Lossof-function of both At ORM1 and At ORM2(orm1 ami R-ORM2) stimulated de novo sphingolipid biosynthesis,leading to strong sphingolipid accumulation,especially of long-chain bases and ceramides. Yeast two-hybrid,bimolecular fluorescence complementation,and coimmunoprecipitation assays confirmed that ORM1 and ORM2 physically interact with the small subunit of SPT(ss SPT),indicating that ORMs inhibit ss SPT function. We found that orm1 ami R-ORM2 plants exhibited an early-senescence phenotype accompanied by H2O2 production at the cell wall and in mitochondria,active vesicular trafficking,and formation of cell wall appositions. Strikingly,the orm1 ami R-ORM2 plants not only showed increased the expression of genes related to ER stress and defenses,but also enhanced resistance to oxidative stress and pathogen infection. Our results indicate that At ORMs function to regulate sphingolipid biosynthesis in maintaining sphingolipid homeostasis and play a role in response to abiotic and biotic stresses.

Gut microbiota intervention attenuates thermogenesis in broilers exposed to high temperature through modulation of the hypothalamic 5‑HT pathway

Background Broilers have a robust metabolism and high body temperature,which make them less tolerant to hightemperature(HT)environments and more susceptible to challenges from elevated temperatures.Gut microbes,functioning as symbionts within the host,possess the capacity to significantly regulate the physiological functions and environmental adaptability of the host.This study aims to investigate the effects of gut microbial intervention on the body temperature and thermogenesis of broilers at different ambient temperatures,as well as the underlying mechanism involving the"gut-brain"axis.Methods Broilers were subjected to gut microbiota interference with or without antibiotics(control or ABX)starting at 1 day of age.At 21 day of age,they were divided into 4 groups and exposed to different environments for 7 d:The control and ABX groups at room temperature(RT,24±1℃,60%relative humidity(RH),24 h/d)and the control-HT and ABX-HT groups at high temperature(HT,32±1℃,60%RH,24 h/d).Results The results demonstrated that the antibiotic-induced gut microbiota intervention increased body weight and improved feed conversion in broiler chickens(Pention reduced the rectal temperature of broiler chickens(Penesisrelated genes in breast muscle and liver(Penesis capacity.Furthermore,the gut microbiota intervention blunted the hypothalamic‒pituitary‒adrenal axis and hypothalamic–pituitary–thyroid axis activation induced by HT conditions.By analyzing the cecal microbiota composition of control and ABX chickens maintained under HT conditions,we found that Alistipes was enriched in control chickens.In contrast,antibioticinduced gut microbiota intervention resulted in a decrease in the relative abundance of Alistipes(Pence was accompanied by increased hypothalamic 5-hydroxytryptamine(5-HT)content and TPH2 expression(P<0.05).Conclusions These findings underscore the critical role

A new class of backward particle systems with sequential interaction is proposed to approximate the mean-field backward stochastic differential equations.It is proven that the weighted empirical measure of this particle system converges to the law of the McKean-Vlasov system as the number of particles grows.Based on the Wasserstein met-ric,quantitative propagation of chaos results are obtained for both linear and quadratic growth conditions.Finally,numerical experiments are conducted to validate our theoretical results.

Corrosion behavior of Gd_(2)Zr_(2)O_(7)thermal barrier coatings under Fe-containing environmental sediment attack

environmental sediments mainly consisting of CaO–MgO–Al_(2)O_(3)–SiO_(2)(CMAS)corrosion are a serious threat to thermal barrier coatings(TBCs),in which Fe element is usually ignored.Gd_(2)Zr_(2)O_(7)TBCs are famous for their excellent CMAS resistance.In this study,the characteristics of Fe-containing environmental sediments(CMAS-Fe)and their corrosiveness to Gd_(2)Zr_(2)O_(7)coatings were investigated.Four types of CMAS-Fe glass with different Fe contents were fabricated.Their melting points were measured to be 1322–1344℃,and the high-temperature viscosity showed a decreasing trend with increasing Fe contents.The corrosion behavior of four types of CMAS-Fe to Gd_(2)Zr_(2)O_(7)coatings at 1350℃was investigated.At the initial corrosion stage(0.1 h),anorthite was precipitated in CMAS-Fe with a high Ca:Si ratio,while Fe-garnet was formed in the melt with the highest Fe content.Prolonging the corrosion time resulted in the formation of a reaction layer,which exhibited an interpenetrating network composed of Gd-oxyapatite,ZrO_(2),and residual CMAS-Fe.Some spinel was precipitated within the reaction layer.After 1 h or even longer time,the reaction layers tended to be stable and compact,which had comparable hardness and fracture toughness to those of Gd_(2)Zr_(2)O_(7)coatings.Under the cyclic CMAS-Fe attack,the residual CMAS-Fe in the interpenetrating network provided a pathway for the redeposited CMAS-Fe infiltration,resulting in the continuous growth of the reaction layer.As a result,the Gd_(2)Zr_(2)O_(7)coatings had a large consumption in the thickness,degrading the coating performance.Therefore,the Gd_(2)Zr_(2)O_(7)coatings exhibit unsatisfactory corrosion resistance to CMAS-Fe attack.

Dear Editor,An adaptive consensus control algorithm for uncertain multi-agent systems(MAS),capable of guaranteeing unified prescribed performance,is presented in this letter.Unlike many existing prescribed performance related works,the developed control exhibits some features.Firstly,a distributed prescribed time observer is introduced so that not only each follower is able to estimate the leader’s signal within a predetermined time,but also the control design for each agent is independent with its neighbors.

Immediate implantation of ultrafine fiber slow-release system based on cell electrospinning to induce osteogenesis of mesenchymal stem cells

This study presents the development and evaluation of a poly(3-hydroxybutyrate-co-4-hydroxybutyrate)(P34HB)ultrafine fiber slow-release system for in vivo osteogenic induction of human umbilical cord mesenchymal stem cells(HUCMSCs).Utilizing dual-nozzle and cell electrospinning techniques,the system encapsulates L-ascorbic acid-2-phosphate magnesium(AsP),βB-glycerophosphate sodium and dexamethasone(DEX)within the'fbers,ensuring sustained osteogenic differentiation.The scaffold's morphology,characterization,hydrophilicity,mechanical properties and cellular behavior were examined.Immediate subcutaneous implantation in rabbits was con-ducted to observe its ectopic osteogenic induction effect.Successfully fabricated P34HB ultrafine fiber slow-release sys-tem.Charact ization med.theuniform distribution of HUCMSCs and inducing components within the scaffold,with no chemical reactions affecting the active components.In vitro tests showcased a prolonged release of DEX and ASP,while biocompatibility assays highlighted the scaffold's suitability for cellular growth.Alizarin Red,type I collagen,and osteopontin(OPN)staining verifed the scaffold's potent osteogenic induction effect on HUCMsCs.Notably,immediate implantation into New Zealand White rabbits led to significant new bone formation within 8 weeks.These findings underscore the system's potential for immediate in vivo implantation without prior in vitro induction,marking a promising advancement in bone tissue engineering.

Molecular mechanism of Xuebijing in treating pyogenic liver abscess complicated with sepsis

BACKGROUND:Xuebijing(XBJ)can alleviate the inflammatory response,improve organ function,and shorten the intensive care unit(ICU)stay in patients with pyogenic liver abscess(PLA)complicated with sepsis,but the molecular mechanisms have not been elucidated.This study aimed to explore the molecular mechanism of XBJ in treating PLA complicated with sepsis using a network pharmacology approach.METHODS:The active ingredients and targets of XBJ were retrieved from the ETCM database.Potential targets related to PLA and sepsis were retrieved from the GeneCards,PharmGKB,DisGenet,Online Mendelian Inheritance in Man(OMIM),Therapeutic Targets Database(TTD),and DrugBank databases.The targets of PLA complicated with sepsis were mapped to the targets of XBJ to identify potential treatment targets.Protein-protein interaction networks were analyzed using the STRING database.Potential treatment targets were imported into the Metascape platform for Gene Ontology(GO)functional enrichment and Kyoto encyclopedia of Genes and Genomes(KEGG)pathway enrichment analyses.Molecular docking was performed to validate the interactions between active ingredients and core targets.RESULTS:XBJ was found to have 54 potential treatment targets for PLA complicated with sepsis.Interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor(TNF)were identifi ed as core targets.KEGG enrichment analysis revealed important pathways,including the interleukin-17(IL-17)signaling pathway,the TNF signaling pathway,the nuclear factor-kappa B(NF-κB)signaling pathway,and the Toll-like receptor(TLR)signaling pathway.Molecular docking experiments indicated stable binding between XBJ active ingredients and core targets.CONCLUSION:XBJ may exert therapeutic eff ects on PLA complicated with sepsis by modulating signaling pathways,such as the IL-17,TNF,NF-κB,and TLR pathways,and targeting IL-1β,IL-6,and TNF.

A DNA tetrahedron-based ferroptosis-suppressing nanoparticle: superior delivery of curcumin and alleviation of diabetic osteoporosis

Diabetic osteoporosis(DOP)is a significant complication that poses continuous threat to the bone health of patients with diabetes;however,currently,there are no effective treatment strategies.In patients with diabetes,the increased levels of ferroptosis affect the osteogenic commitment and differentiation of bone mesenchymal stem cells(BMSCs),leading to significant skeletal changes.To address this issue,we aimed to target ferroptosis and propose a novel therapeutic approach for the treatment of DOP.We synthesized ferroptosis-suppressing nanoparticles,which could deliver curcumin,a natural compound,to the bone marrow using tetrahedral framework nucleic acid(tFNA).This delivery system demonstrated excellent curcumin bioavailability and stability,as well as synergistic properties with tFNA.Both in vitro and in vivo experiments revealed that nanoparticles could enhance mitochondrial function by activating the nuclear factor E2-related factor 2(NRF2)/glutathione peroxidase 4(GPX4)pathway,inhibiting ferroptosis,promoting the osteogenic differentiation of BMSCs in the diabetic microenvironment,reducing trabecular loss,and increasing bone formation.These findings suggest that curcumin-containing DNA tetrahedron-based ferroptosissuppressing nanoparticles have a promising potential for the treatment of DOP and other ferroptosis-related diseases.
