AIM:To assess the clinical presentations and outcomes of idiopathic orbital inflammatory pseudotumor(IOIP)patients with orbital wall bone destruction(OWBD)and to propose an expanded classification system that includes bone ***:The study retrospectively reviewed clinical presentations,imaging findings,treatment modalities,and outcomes of six patients diagnosed histopathologically with IOIP and OWBD at the Beijing Tongren Hospital,Capital Medical University between October 2018 and June ***:Over two years,6(10%)of 60 IOIP patients at our hospital exhibited OWBD,but this may overrepresent severe *** cohort consisted of three men and three women,aged 17 to 60y(mean 35.5±16.1y).Presenting symptoms included proptosis,eyelid swelling,decreased visual acuity with pain,and palpable *** revealed multiple anatomical structures involved with the medial wall being the most common site of bone *** examination showed classic type in five patients and sclerosing type in one *** patients underwent surgical resection followed by methylprednisolone ***-up(mean 30.3±3.1mo)indicated three patients had no recurrence,while others had varying degrees of symptom persistence or ***:IOIP with bone destruction is a rare but significant subtype that mimics malignancy,leading to potential diagnostic and therapeutic *** findings suggest that complete surgical resection combined with adjunctive glucocorticoid therapy can yield favorable ***,larger-scale studies are needed to further optimize therapeutic approaches.

胫骨创伤性骨髓炎的部分患者因软组织损伤严重,胫骨与内固定外露,如不及时处理,会出现内固定失效、骨折不愈合等不良后果,严重者可因此截肢[1]。目前对于伴有软组织缺损的胫骨创伤性骨髓炎患者,临床上没有统一的治疗方法,大部分医生运用二次手术的治疗方法,但其存在治疗周期长等缺点,易出现关节功能障碍、患者满意度低、医疗费用高等一系列问题。为了寻求更佳的治疗方法,笔者所在团队以显微外科技术为主要手段,对伴有软组织缺损的胫骨创伤性骨髓炎患者进行一期手术治疗,取得了令人满意的结果。现报道如下。

RBM25 depletion suppresses the growth of colon cancer cells through regulating alternative splicing of MNK2

Increasing evidence suggests that deregulated RNA splicing factors play critical roles in tumorigenesis; however, their specific involvement in colon cancer remains largely unknown. Here we report that the splicing factor RBM25 is overexpressed in colon cancer, and this increased expression correlates with a poor prognosis of patients with colon cancer. Functionally, RBM25 ablation suppresses the growth of colon cancer cells both in vitro and in vivo. Mechanistically, our transcriptome-wide analysis of splicing events revealed that RBM25 regulates a large number of cancer-related alternative splicing events across the human genome in colon cancer. Particularly, RBM25 regulates the splicing of MNK2 by interacting with the poly G rich region in exon 14a, thereby inhibiting the selection of the proximal 3'splice site(ss), resulting in the production of the oncogenic short isoform, MNK2b. Knockdown of RBM25 leads to an increase in the MNK2a isoform and a decrease in the MNK2b isoform. Importantly, re-expression of MNK2b or blocking the 3′ ss of the alternative exon 14a with ASO partially reverses the RBM25 knockdown mediated tumor suppression. Moreover, MNK2b levels were significantly increased in colon cancer tissues, which is positively correlated with the expression level of RBM25. Collectively, our findings uncover the critical role of RBM25 as a key splicing factor in colon cancer, suggesting its potential as a prognostic marker and therapeutic target.

Protective Effects of Danmu Extract Syrup on Acute Lung Injury Induced by Lipopolysaccharide in Mice through Endothelial Barrier Repair

Objective:To investigate the effects of Danmu Extract Syrup(DMS) on lipopolysaccharide(LPS)-induced acute lung injury(ALI) in mice and explore the ***:Seventy-two male Balb/C mice were randomly divided into 6 groups according to a random number table(n=12),including control(normal saline),LPS(5 mg/kg),LPS+DMS 2.5 mL/kg,LPS+DMS 5 mL/kg,LPS+DMS 10 mL/kg,and LPS+Dexamethasone(DXM,5 mg/kg) *** pretreatment with DMS and DXM,the ALI mice model was induced by LPS,and the bronchoalveolar lavage fluid(BALF) were collected to determine protein concentration,cell counts and inflammatory *** lung tissues of mice were stained with hematoxylin-eosin,and the wet/dry weight ratio(W/D) of lung tissue was *** levels of tumor necrosis factor-α(TNF-α),interleukin(IL)-6 and IL-1βin BALF of mice were detected by enzyme linked immunosorbent *** expression levels of Claudin-5,vascular endothelial(VE)-cadherin,vascular endothelial growth factor(VEGF),phospho-protein kinase B(p-Akt) and Akt were detected by Western blot ***:DMS pre-treatment significantly ameliorated lung histopathological *** with the LPS group,the W/D ratio and protein contents in BALF were obviously reduced after DMS pretreatment(P<0.05 or P<0.01).The number of cells in BALF and myeloperoxidase(MPO) activity decreased significantly after DMS pretreatment(P<0.05 or P<0.01).DMS pre-treatment decreased the levels of TNF-α,IL-6 and IL-1β(P<0.01).Meanwhile,DMS activated the phosphoinositide 3-kinase/protein kinase B(PI3K/Akt) pathway and reversed the expressions of Claudin-5,VE-cadherin and VEGF(P<0.01).Conclusions:DMS attenuated LPS-induced ALI in mice through repairing endothelial *** might be a potential therapeutic drug for LPS-induced lung injury.

Aging is a contributor to liver ***,the concept of liver aging has become prominent and has attracted considerable interest,but its underlying mechanism remains poorly *** our study,the internal mechanism of liver aging was explored via multi-omics analysis and molecular experiments to support future targeted *** aged rat liver model was established with D-galactose,and two other senescent hepatocyte models were established by treating HepG2 cells with D-galactose and *** then performed transcriptomic and metabolomic assays of the aged liver model and transcriptome analyses of the senescent hepatocyte *** livers,genes related to peroxisomes,fatty acid elongation,and fatty acid degradation exhibited down-regulated expression with aging,and the hepatokine Fgf21 expression was positively correlated with the down-regulation of these *** senescent hepatocytes,similar to the results found in aged livers,FGF21 expression was also ***,the expressions of cell cycle-related genes were significantly down-regulated,and the down-regulated gene E2F8 was the key cell cycle-regulating transcription *** then validated that FGF21 overexpression can protect against liver aging and that FGF21 can attenuate the declines in the antioxidant and regenerative capacities in the aging *** successfully validated the results from cellular and animal experiments using human liver and blood *** study indicated that FGF21 is an important target for inhibiting liver aging and suggested that pharmacological prevention of the reduction in FGF21 expression due to aging may be used to treat liver aging-related diseases.

生成对抗网络(GAN)在图像生成、蛋白质设计领域有着广泛应用,但是对于噬菌体尾部蛋白的生成少有研究。提出一种基于GAN的噬菌体尾部蛋白序列生成方法。首先使用Wasserstein距离作为模型的目标函数。其次采用多层感知机(MLP)作为模型的基本结构。然后将MLP扩展为多路径结构。实验结果表明,上述方法取得了0.9241的质量得分、0.8498的多样性得分和1.7739的总得分,优于其它常用的生成方法。相较于单路径MLP,多路径MLP提高了序列的生成效果。所提方法能够生成高质量噬菌体尾部蛋白序列,同时保证生成序列的多样性。

Different concentrations of hyaluronic acid eye drops for dry eye syndrome: a systematic review and Meta-analysis

AIM:To compare high or low concentration of hyaluronic acid eye drops(HY)for dry eye syndromes(DES).METHODS:Randomized controlled trials(RCTs)comparing various concentrations of HY were searched in PubMed,Embase,Web of Science,Cochrane,SinoMed,CNKI,Wanfang Database,CQVIP,and Chinese journals databases between inception and July *** standardized mean differences(SMD)or weighted mean difference(WMD)with 95%confidence intervals(CI)from RCTs evaluating Schirmer’s I test(SIT),corneal fluorescein staining score(CFS),tear breakup time(TBUT),DES score(DESS),and Ocular Surface Disease Index(OSDI)were *** analysis,Egger’s test and Meta-regression analysis were performed for all ***:We conducted a Meta-analysis of 10 RCTs that met the inclusion criteria,involving 1796 ***-concentrations group significantly improved the outcome of CFS according to random effects modelling(SMD,-3.37;95%CI,-5.25 to-1.48;P=0.0005).The rest of the results were not statistically significant,including indicators such as SIT,TBUT,DESS and ***:For dry eyes with positive corneal staining,a high concentration of HY is recommended,whereas in other cases,a high concentration of HY does not offer a more pronounced advantage over a low concentration of HY in the treatment of dry eyes.

The relationship between uric acid and blood pressure in patients with diabetes:results from NHANES database

Objective This study aimed to explore the association between uric acid(UA)and blood pressure(BP)in patients with diabetes receiving anti-diabetic *** A cross-sectional study with 2878 individuals from the National Health and Nutrition Examination Survey(NHANES)was *** linear regression analysis was applied to explore the relationship between UA and BP in the antihypertensive and anti-diabetic group(n=1966)and the anti-diabetic group(n=912).Results Between the group with antihypertensive and anti-diabetic therapy and the group with anti-diabetic therapy alone,no significant association was found with systolic blood pressure(SBP).However,the significantly negative association was present in diastolic blood pressure(DBP),and the size of the effect was(β,-0.35(95%CI:-0.68~-0.02))and(β,-0.60(95%CI:-1.13~-0.08)),*** trend test was not significant between UA and SBP in two groups,but the trend test was significant between UA and DBP in the antihypertensive and anti-diabetic group(P=0.005).Conclusion In our study,we found that the association between UA and DBP in patients with diabetes receiving anti-diabetic therapy was negative.
