Androgens and chronic inflammation,which play essential roles in the development of benign prostatic hyperplasia(BPH),are considered to be important factors in disorders of prostate homeostasis.These two factors may lead to pathological hyperplasia in the prostate transition zone of patients with BPH.However,few studies have examined the mechanism of how dihydrotestosterone(DHT)affects chronic inflammation in prostate tissue during the progression of BPH.This study examined the performance of DHT in lipopolysaccharide-treated M1 macrophages and the subsequent effects on the proliferation of prostate stromal and epithelial cells.We found that DHT increased secretion of the pro-inflammatory factor tumor necrosis factor(TNF)-αfrom M1 macrophages differentiated from THP-1 cells.The supernatant of M1 macrophages promoted the proliferation of WPMY-1 prostate stromal cells by upregulating B-cell lymphoma-extra large(Bcl-xL)and cellular Myc(c-Myc)levels by activating TNF-α-mediated nuclear factor-kappa B(NF-κB)and mitogen-activated protein kinase(MAPK)pathways.Moreover,this supernatant increased the expression of androgen receptor in WPMY-1 cells,which was TNF-α-independent.Additionally,TNF-αprotein expression was significantly higher in patients with BPH and a large prostate volume than that in those with a small prostate volume.Further analysis showed that higher serum testosterone combined with prostate-specific androgen concentrations was related to TNF-αexpression.This study suggests that DHT modulates the inflammatory environment of BPH by increasing TNF-αexpression from lipopolysaccharide-treated M1 macrophages and promotes the proliferation of prostate stromal cells.Targeting TNF-α,but not DHT,may be a promising strategy for patients with BPH.

Human adipose-derived stem cells(hASCs)are a promising cell type for bone tissue regeneration.Circular RNAs(circRNAs)have been shown to play a critical role in regulating various cell differentiation and involve in mesenchymal stem cell osteogenesis.However,how circRNAs regulate hASCs in osteogenesis is still unclear.Herein,we found circ_0003204 was significantly downregulated during osteogenic differentiation of hASCs.Knockdown of circ_0003204 by si RNA or overexpression by lentivirus confirmed circ_0003204 could negatively regulate the osteogenic differentiation of hASCs.We performed dual-luciferase reporting assay and rescue experiments to verify circ_0003204 regulated osteogenic differentiation via sponging miR-370-3p.We predicted and confirmed that miR-370-3p had targets in the 3′-UTR of HDAC4 m RNA.The following rescue experiments indicated that circ_0003204 regulated the osteogenic differentiation of hASCs via miR-370-3p/HDAC4 axis.Subsequent in vivo experiments showed the silencing of circ_0003204 increased the bone formation and promoted the expression of osteogenic-related proteins in a mouse bone defect model,while overexpression of circ_0003204 inhibited bone defect repair.Our findings indicated that circ_0003204 might be a promising target to promote the efficacy of hASCs in repairing bone defects.

Electrochemical nitrogen reduction(NRR)is deemed as a consummate answer for the traditional Haber–Bosch technology.Breaking the linear correlations between adsorption and transition-state energies of intermediates is vital to improve the kinetics of ammonia synthesis and obtain a less energy-intensive process.Herein,carbon-encapsulated mixed-valence Fe_(7)(PO_(4))_(6) was prepared and applied as an electrocatalyst for high-efficiency NRR.A dramatic faradaic efficiency(FE)of 36.93%and an NH_(3) production rate of 13.1μg h^(-1) mg_(cat)^(-1) were obtained at-0.3 V versus RHE,superior to nearly all Fe-based catalysts.Experiments and DFT calculations revealed that the superior performance was ascribed to the synergistic effect of mixed-valence iron pair,which braked the linear correlations to improve the kinetics of ammonia from collaborative hydrogenation and*NH_(3) separation.This work proves the feasibility of mixedvalence catalysts for nitrogen reduction and thus opening a new avenue towards artificial nitrogenfixation catalysts.

Repairing glans dehiscence after failed hypospadias repair is challenging for pediatric surgeons.Here,we introduced and evaluated a newly modified Mathieu technique,Mathieu combined tunnel(MCT),which involves multiple custom-designed flaps for the shortage of flap source material after repeated operations;we also constructed a tunnel to avoid the glans incision that may carry new risks of dehiscence.This retrospective study included 26 patients who were consecutively admitted to the First Affiliated Hospital of Sun Yat-Sen University(Guangzhou,China)for glans dehiscence repair after failed hypospadias repair from October 2014 to October 2020;sixteen patients underwent surgery using the MCT(MCT group)and ten patients underwent surgery using the tubularized incised plate(TIP)technique(TIP group).The operative time,blood loss,postoperative complications,normal urethral meatus rate,success rate,and Hypospadias Objective Penile Evaluation(HOPE)score were compared between the two groups.The MCT group achieved an overall satisfactory penile appearance and voiding function,with a higher rate of normal urethral meatus(15/16,93.8%)and a lower rate of glans dehiscence(1/16,6.2%),compared with the TIP group(70.0%and 30.0%,respectively).However,these differences were not statistically significant,possibly because of the limited number of patients(all P>0.05).Mean postoperative HOPE scores were similar in the MCT group(mean±standard deviation:8.83±0.89)and TIP group(8.94±0.57)(P>0.05).No significant differences were found between the two groups in terms of blood loss and success rate,nor in the rates of various complications(e.g.,fistula,urethral stricture,and glans dehiscence).In conclusion,the MCT technique appears to be feasible and reliable for repairing glans dehiscence after failed hypospadias repair.

In recent years,solution-processible semiconductors with perovskite or perovskite-inspired structures have been extensively investigated for optoelectronic applications.In particular,silver-bismuth-halides have been identified as especially promising because of their bulk properties and lack of heavily toxic elements.This study investigates the potential of Ag2BiI5 for near-infrared(NIR)-blind visible light photodetection,which is critical to emerging applications(e.g.,wearable optoelectronics and the Internet of Things).Self-powered photodetectors were realized and provided a near-constant≈100 mA W−1 responsivity through the visible,a NIR rejection ratio of>250,a long-wavelength responsivity onset matching standard colorimetric functions,and a linear photoresponse of>5 orders of magnitude.The optoelectronic characterization of Ag2BiI5 photodetectors additionally revealed consistency with one-center models and the role of the carrier collection distance in self-powered mode.This study provides a positive outlook of Ag2BiI5 toward emerging applications on low-cost and low-power NIR-blind visible light photodetector.

Mannose,a different isomer of the hydroxyl group at the C-2 position of glucose,shares the same transport carrier protein with glucose to enter cells and participate in the regulation of glucose metabolism.It affects cell growth,differentiation,and function and plays an active role in tumor immunity and inflammatory processes.This paper provides theoretical support for expanding the clinical applications of mannose by exploring its constitution,metabolic pathways,and role in regulating immune cell function and treating immunology-related diseases.

In this article, we propose a novel, multilevel, dynamic factor model, to determine endogenously clustered regions for the investigation of regional clustering and synchronization of provincial business fluctuations in China. The parameter identification and model estimation was conducted using the Markov Chain Monte Carlo method. We then conducted an empirical study of the provincial business fluctuations in China(31 Chinese provinces are considered except Hong Kong, Macao, and Taiwan due to the data unavailability), which were sampled from January 2000 to December 2015. Our results indicated that these provinces could be clustered into four regions: leading, coincident, lagging, and overshooting. In comparison with traditional geographical divisions, this novel clustering into four regions enabled the regional business cycle synchronization to be more accurately captured. Within the four regional clusters it was possible to identify substantial heterogeneities among regional business cycle fluctuations, especially during the periods of the 2008 financial crisis and the ‘four-trillion economic stimulus plan'.

Development of defoliating insects and their preferences for host plants under varying temperatures in a subtropical evergreen forest in eastern China

Abstract The aim of this work was to understand the development of defoliating insects and their preferences for host plants under varying temperatures in a subtropical evergreen broad-leaved forest in China. We measured the main developmental parameters of three typical defoliating insects （i.e., Ourapteryx ebuleata szechuana, Biston marginata, and Euproctis angulata） and their preferences for five host plants at temperatures from 16~C to 31 ~C at 3~C intervals in the Tiantong National Forest Research station in eastern China. The results showed the following. 1） An appropriate rise in temperature increases the survival rate with an increase in the number of offspring. The developmental durations for these three insects were shortened, and pupal weight increased with an increase in temperature. 2） A shift in the preference for host plants for these three insects was observedat elevated tempera- tures. They all preferred to feed on Schima superba and Castanopsis sclerophylla at elevated temperatures, show- ing an opposite response to the other three plants. The daily leaf consumption of the three insects was positively correlated with their feeding preference, with more leaves being consumed from the plants they preferred. 3） For O. ebuleata szechuana larvae, daily leaf consumption initially increased and then decreased with increasing temperatures. In contrast, Biston marginata and Euproctis angulata larvae consumed more leaves at elevated temperatures. The feeding preferences of O. ebuleata szechuana and Biston marginata were more sensitive to changing temperatures than that of Euproctis angulata laevae. We concluded that increased numbers of offspring and generations, pupal weights, and a shift in preference to two plants for these three defoliating insects might lead to severe damage to these two plants which would enhance the fragmentation and decrease the stability of the forest communities under changing temperatures. Insufficient TRPM5 Mediates Lipotoxicity-induced Pancreatic β-cell Dysfunction

Objective:While the reduction of transient receptor potential channel subfamily M member 5(TRPM5)has been reported in islet cells from type 2 diabetic(T2D)mouse models,its role in lipotoxicity-induced pancreatic β-cell dysfunction remains unclear.This study aims to study its role.Methods:Pancreas slices were prepared from mice subjected to a high-fat-diet(HFD)at different time points,and TRPM5 expression in the pancreatic β cells was examined using immunofluorescence staining.Glucose-stimulated insulin secretion(GSIS)defects caused by lipotoxicity were mimicked by saturated fatty acid palmitate(Palm).Primary mouse islets and mouse insulinoma MIN6 cells were treated with Palm,and the TRPM5 expression was detected using qRT-PCR and Western blotting.Palm-induced GSIS defects were mimicked following siRNA-based Trpm5 knockdown.The detrimental effects of Palm on primary mouse islets were also assessed after overexpressing Trpm5 via an adenovirus-derived Trpm5(Ad-Trpm5).Results:HFD feeding decreased the mRNA levels and protein expression of TRPM5 in mouse pancreatic islets.Palm reduced TRPM5 protein expression in a time-and dose-dependent manner in MIN6 cells.Palm also inhibited TRPM5 expression in primary mouse islets.Knockdown of Trpm5 inhibited insulin secretion upon high glucose stimulation but had little effect on insulin biosynthesis.Overexpression of Trpm5 reversed Palm-induced GSIS defects and the production of functional maturation molecules unique to β cells.Conclusion:Our findings suggest that lipotoxicity inhibits TRPM5 expression in pancreatic β cells both in vivo and in vitro and,in turn,drives β-cell dysfunction.
