Functional constipation(FC)is a common disorder that is characterized by diffi-cult stool passage,infrequent bowel movement,or both.FC is highly prevalent,recurs often,accompanies severe diseases,and affects quality of life;therefore,safe and effective therapy with long-term benefits is urgently needed.Microbiota treatment has potential value for FC treatment.Microbiota treatments include modulators such as probiotics,prebiotics,synbiotics,postbiotics,and fecal micro-biota transplantation(FMT).Some probiotics and prebiotics have been adopted,and the efficacy of other microbiota modulators is being explored.FMT is con-sidered an emerging field because of its curative effects;nevertheless,substantial work must be performed before clinical implementation.

In the context of the increasing scale of bridges and the increasing service life of bridges,it is very important to carry out efficient,accurate and intelligent bridge operation and maintenance.In recent years,advanced equipment,technology and intelligent algorithms have developed rapidly.It is necessary to apply advanced equipment and algorithms to bridge operation and maintenance business to facilitate the digitalization and intelligence of bridge operation and maintenance.To grasp the research progress on the bridge intelligent operation and maintenance,this paper summarizes the research progress in recent years from the aspects of intelligent detection equipment and technology,intelligent monitoring equipment and technology,intelligent data analysis,intelligent evaluation and early warning,and intelligent repair and maintenance.According to the review,more and more smart devices have been used to replace human beings to detect dangerous and hidden bridge components.At the same time,image processing,radar and other technologies have been used to analyze component damage more objectively and quantitatively.To solve the shortcomings of traditional sensors such as short life and low robustness,more non-contact measurement methods have been proposed.Scholars have proposed various intelligent algorithms to process the massive amount of bridge health monitoring data to improve the quality of the data.To achieve the rapid perception of bridge status and timely early warning of structural abnormalities,different from traditional theoretical calculations,scholars have tried to use data-driven methods to intelligently evaluate and early warning of bridge structural status.In terms of intelligent repair and maintenance,more intelligent algorithms have been used to optimize structural maintenance strategies and determine the best maintenance time by integrating multisource heterogeneous data.All these provide strong support for the automation,digitization and intelligence of bridge operation and maintenance.

This paper presents an efficient scheme for single-pixel imaging(SPI)utilizing a phase-controlled fiber laser array and an untrained deep neural network.The fiber lasers are arranged in a compact hexagonal structure and coherently combined to generate illuminating light fields.Through the utilization of high-speed electro-optic modulators in each individual fiber laser module,the randomly modulated fiber laser array enables rapid speckle projection onto the object of interest.Furthermore,the untrained deep neural network is incorporated into the image reconstructing process to enhance the quality of the reconstructed images.Through simulations and experiments,we validate the feasibility of the proposed method and successfully achieve high-quality SPI utilizing the coherent fiber laser array at a sampling ratio of 1.6%.Given its potential for high emitting power and rapid modulation,the SPI scheme based on the fiber laser array holds promise for broad applications in remote sensing and other applicable fields.

Exosomes,nanoscale extracellular vesicles(EVs)derived from the invagination of the endosomal membrane,are secreted by a majority of cell types.As carriers of DNA,mRNA,proteins,and microRNAs,exosomes are implicated in regulating biological activities under physiological and pathological conditions.Kidney-derived exosomes,which vary in origin and function,may either contribute to the pathogenesis of disease or represent a potential therapeutic resource.Membranous nephropathy(MN),an autoimmune kidney disease characterized by glomerular damage,is a predominant cause of nephrotic syndrome.Notably,MN,especially idiopathic membranous nephropathy(IMN),often results in end-stage renal disease(ESRD),affecting approximately 30%of patients and posing a considerable economic challenge to healthcare systems.Despite substantial research,therapeutic options remain ineffective at halting IMN progression,underscoring the urgent need for innovative strategies.Emerging evidence has implicated exosomes in IMN’s pathophysiology;Providing a fresh perspective for the discovery of novel biomarkers and therapeutic strategies.This review aims to scrutinize recent developments in exosome-related mechanisms in IMN and evaluate their potential as promising therapeutic targets and diagnostic biomarkers,with the hope of catalyzing further investigations into the utility of exosomes in MN,particularly IMN,ultimately contributing to improved patient outcomes in these challenging disease settings.

Explainable-AI-based two-stage solution for WSN object localization using zero-touch mobile transceivers

Artificial intelligence technology is widely used in the field of wireless sensor networks(WSN).Due to its inexplicability, the interference factors in the process of WSN object localization cannot be effectively eliminated. In this paper, an explainable-AI-based two-stage solution is proposed for WSN object localization. In this solution, mobile transceivers are used to enlarge the positioning range and eliminate the blind area for object localization. The motion parameters of transceivers are considered to be unavailable,and the localization problem is highly nonlinear with respect to the unknown parameters. To address this,an explainable AI model is proposed to solve the localization problem. Since the relationship among the variables is difficult to fully include in the first-stage traditional model, we develop a two-stage explainable AI solution for this localization problem. The two-stage solution is actually a comprehensive consideration of the relationship between variables. The solution can continue to use the constraints unused in the firststage during the second-stage, thereby improving the performance of the solution. Therefore, the two-stage solution has stronger robustness compared to the closed-form solution. Experimental results show that the performance of both the two-stage solution and the traditional solution will be affected by numerical changes in unknown parameters. However, the two-stage solution performs better than the traditional solution, especially with a small number of mobile transceivers and sensors or in the presence of high noise. Furthermore,we have also verified the feasibility of the proposed explainable-AI-based two-stage solution.

Insufficient TRPM5 Mediates Lipotoxicity-induced Pancreaticβ-cell Dysfunction

Objective:While the reduction of transient receptor potential channel subfamily M member 5(TRPM5)has been reported in islet cells from type 2 diabetic(T2D)mouse models,its role in lipotoxicity-induced pancreaticβ-cell dysfunction remains unclear.This study aims to study its role.Methods:Pancreas slices were prepared from mice subjected to a high-fat-diet(HFD)at different time points,and TRPM5 expression in the pancreaticβcells was examined using immunofluorescence staining.Glucose-stimulated insulin secretion(GSIS)defects caused by lipotoxicity were mimicked by saturated fatty acid palmitate(Palm).Primary mouse islets and mouse insulinoma MIN6 cells were treated with Palm,and the TRPM5 expression was detected using qRT-PCR and Western blotting.Palm-induced GSIS defects were measured following siRNA-based Trpm5 knockdown.The detrimental effects of Palm on primary mouse islets were also assessed after overexpressing Trpm5 via an adenovirus-derived Trpm5(Ad-Trpm5).Results:HFD feeding decreased the mRNA levels and protein expression of TRPM5 in mouse pancreatic islets.Palm reduced TRPM5 protein expression in a time-and dose-dependent manner in MIN6 cells.Palm also inhibited TRPM5 expression in primary mouse islets.Knockdown of Trpm5 inhibited insulin secretion upon high glucose stimulation but had little effect on insulin biosynthesis.Overexpression of Trpm5 reversed Palm-induced GSIS defects and the production of functional maturation molecules unique toβcells.Conclusion:Our findings suggest that lipotoxicity inhibits TRPM5 expression in pancreaticβcells both in vivo and in vitro and,in turn,drivesβ-cell dysfunction.

Detoxification strategy of Microcystis aeruginosa to the toxicity of Cd(Ⅱ):role of EPS in alleviating toxicity

Although many studies have found that cadmium(Cd)can be toxic to microalgae,only a few reports focused on the role of extracellular polymeric substances(EPS)in Cd(Ⅱ)detoxification.The biochemical and physiological endpoints of Microcystis aeruginosa,including the composition and functional groups of soluble EPS(SL-EPS),loosely bound EPS(LB-EPS),and tightly bound EPS(TB-EPS),were detected to elucidate the toxicity and detoxification mechanisms of Cd(Ⅱ)for cyanobacteria.Toxicological and physiological assays on M.aeruginosa showed that the 0.25-mg/L Cd(Ⅱ)resulted in a larger inhibition on growth and F_(v)/F_(m).Nevertheless,Cd(Ⅱ)significantly induced much higher contents of superoxide dismutase(SOD),intracellular microcystin LR(MC-LR),extracellular MC-LR,and EPS.Scanning electron microscopy with energy dispersive X-ray spectroscopy confirmed that Cd(Ⅱ)was absorbed into the EPS layer.Fourier transform infrared spectrum analysis revealed that the functional groups bound with Cd(Ⅱ)of algae biomass,SL-EPS,LB-EPS,and TB-EPS were somewhat different.The C=O/C=N groups ofδ-lactam or protein were their prominent functional groups,suggesting that amide or proteins in the EPS played a key role in the adsorption in Cd(Ⅱ).The concentration of 0.25 mg/L of Cd(Ⅱ)may change the chemical structure of EPS by altering the production of protein-like substances containing tryptophan.This study indicated that M.aeruginosa could detoxify Cd(Ⅱ)stress via induction of antioxidant capacity(higher SOD activity and MC synthesis),EPS production,and modification in chemical structure of EPS.

Bone morphogenetic protein 4(BMP4)can induce the formation of chicken primordial germ cells(PGCs)in vitro;however,its regulatory mechanism in poultry remains unknown.This study aimed to use RNA-seq to analyze PGCs in chicken embryos and iPGCs induced by BMP4 in vitro,clarify the internal regulatory factors of PGCs,analyze the mechanism of the formation of PGCs,and lay a theoretical foundation for the further optimization of PGCs induction systems.Embryonic stem cells(ESCs),PGCs and iPGCs induced by BMP4 in vitro were collected.The transcriptional maps of the three cell types were studied using RNA-seq.The results showed 6,142 genes differentially expressed between PGCs and iPGCs,of which 2,728 were upregulated in iPGCs and 3,414 were downregulated in iPGCs.Compared to that in ESCs,BMP4 was significantly upregulated in PGCs and iPGCs.KEGG results showed that both the TGF-βand Wnt signaling pathways were activated during the formation of PGCs in vitro and in vivo,and the activation was more significant during iPGCs induced by BMP4.The expression of Nodal,an inhibitory factor of TGF-βsignaling,was significantly decreased in PGCs and iPGCs,but was not expressed in iPGCs,which further supports our conclusion.Additionally,the Lysosome and PI3K-AKT signaling pathways were significantly enriched in PGCs and iPGCs,respectively.Further,transmission electron microscopy(TEM)results showed that the number of autolysosomes was significantly higher after the addition of BMP4,which is consistent with the KEGG results.Furthermore,the number of PGCs was significantly reduced after ATG14 was interfered in vivo and in vitro.In conclusion,this study screened out the key signaling pathways during the formation of PGCs,aiming to provide help for enriching the mechanism network regulating PGCs formation in chicken and laying a theoretical foundation for further improving the efficiency of inducing PGCs in vitro.

Study on the interannual variability of the Kerama Gap transport and its relation to the Kuroshio/Ryukyu Current system

An analysis of a 68-year monthly hindcast output from an eddy-resolving ocean general circulation model reveals the relationship between the interannual variability of the Kerama Gap transport(KGT)and the Kuroshio/Ryukyu Current system.The study found a significant difference in the interannual variability of the upstream and downstream transports of the East China Sea-(ECS-)Kuroshio and the Ryukyu Current.The interannual variability of the KGT was found to be of paramount importance in causing the differences between the upstream and downstream ECS-Kuroshio.Additionally,it contributed approximately 37%to the variability of the Ryukyu Current.The interannual variability of the KGT was well described by a two-layer rotating hydraulic theory.It was dominated by its subsurface-intensified flow core,and the upper layer transport made a weaker negative contribution to the total KGT.The subsurface flow core was found to be mainly driven by the subsurface pressure head across the Kerama Gap,and the pressure head was further dominated by the subsurface density anomalies on the Pacific side.These density anomalies could be traced back to the eastern open ocean,and their propagation speed was estimated to be about 7.4 km/d,which is consistent with the speed of the local first-order baroclinic Rossby wave.When the negative(positive)density anomaly signal reached the southern region of the Kerama Gap,it triggered the increase(decrease)of the KGT towards the Pacific side and the formation of an anticyclonic(cyclonic)vortex by baroclinic adjustment.Meanwhile,there is an increase(decrease)in the upstream transport of the entire Kuroshio/Ryukyu Current system and an offshore flow that decreases(increases)the downstream Ryukyu Current.
