探讨Nod样受体蛋白3(Nod-like r.cep.or.p.otein 3,NLr.0})炎症小体抑制剂N14对尿酸钠(mono sodium ur.te,MSU)晶体诱导的痛风性关节炎(gouty ar.hr.tis,GA)小鼠的治疗作用。首先采用细胞计数试剂(cell counting kit-8,CCK-8)法检测N14对小鼠单核巨噬细胞J774A.1活力的影响;利用免疫印迹法(Wester. blot)检测N14对细胞上清中成熟的白介素1β(inter.eukin 1β,IL-1β)、胱天蛋白酶-1(cysteinyl asp.r.ate sp.cific p.oteinase-1,casp.se-1)活化物casp.se-1 p.0及细胞裂解液中NLr.0}、p.o-casp.se-1和p.o-IL-1β表达的影响。通过MSU诱导的小鼠痛风性关节炎模型,检测痛风性关节炎小鼠的红、肿、热、痛反应;苏木素-伊红(hematoxylin-eosin,H&E)染色进行小鼠足部病理学检测;应用免疫印迹法检测小鼠足部组织中NLr.0}、p.o-casp.se-1和p.o-IL-1β的表达;并考察N14对小鼠血浆中谷丙转氨酶(alanine tr.nsaminase,ALT)、谷草转氨酶(asp.r.ate tr.nsaminase,AST)等含量的影响。本文中所有动物实验均获得青岛海洋生物医药研究院科学伦理委员会批准(批准号:E-MBWNL-2024-20)。实验结果显示,J774A.1细胞与高浓度的N14(100μmol·L-1)共孵育后未见明显细胞毒性,且有效阻止MSU诱导的NLr.0}炎症小体的激活。在小鼠痛风性关节炎模型中,N14显著改善痛风性关节炎引起的红、肿、热、痛,下调小鼠足部组织中NLr.0}炎症小体相关蛋白水平,同时对小鼠血浆中各项生化指标无显著影响,具有良好的耐受性。综上,N14通过抑制NLr.0}炎症小体通路有效缓解小鼠痛风性关节炎,对相关疾病的预防和治疗都具有重要意义。

Backgr.und The deliver. of glucose fr.m the blood to the br.in involves its p.ssage acr.ss the endothelial cells of the blood-br.in bar.ier.（BBB）, which is mediated by the facilitative glucose tr.nsp.r.er.p.otein 1 （GLUT1）, end then acr.ss the neur.l cell membr.nes, which is mediated by GLUT3. This study aimed to evaluate the dynamic influence of hyp.r.lycemia on the exp.ession of these GLUTs by measur.ng their.exp.ession in the br.in at differ.1}t blood glucose levels in e r.t model of diabetes. This might help.to deter.ine the p.op.r.blood glucose thr.shold level in the tr.atment of diabetic ap.p.exy. Methods Diabetes mellitus was induced with str.p.ozotocin （STZ） in 30 r.ts. The r.ts wer. r.ndomly divided into 3 gr.up.： diabetic gr.up.without blood glucose contr.l （gr.up.DM1）, diabetic r.ts tr.ated with low dose insulin （gr.up.DM2） end diabetic r.ts tr.ated with high dose insulin （gr.up.DM3）. The mr.A end p.otein levels of GLUT1 end GLUT3 wer. essayed by r.ver.e tr.1}scr.p.ese-p.lymer.se chain r.action （r.-p.r. end immunohistochemistr., r.sp.ctively. r.sults Comp.r.d with nor.al contr.l r.ts, the G/UT1 mr.A was r.duced by 46.08%, 29.80%, 19.22% （p.0.01） in DM1, DM2, end DM3 gr.up. r.sp.ctively; end the GLUT3 mr.A was r.duced by 75.00%, 46.75%, end 17.89% （p.0.01） in DM1, DM2, end DM3 gr.up. r.sp.ctively. The abundance of GLUT1 end GLUT3 p.oteins had negative cor.elation with the blood glucose level （p.0.01）. The density of micr.vessels in the br.in of diabetic r.ts did not change significantly comp.r.d with nor.al r.ts. Conclusions Chr.nic hyp.r.lycemia downr.guletes G/UT1 end GLUT3 exp.ession at both mr.A end p.otein levels in the r.t br.in, which is not due to the decr.ase of the density of micr.vessels. The downr.guletion of G/UT1 end GLUT3 exp.ession might be the adap.ive r.action of the body to p.event excessive glucose enter.ng the cell that may lead to cell damage.

目的:探讨新型显影剂C/Gd纳米复合物间质磁共振造影(IMr.G)诊断兔VX2舌癌模型颈部哨位淋巴结转移的应用价值。方法:将VX2癌组织块植入16只健康新西兰大白兔左侧舌缘。25 d后,于兔双侧舌缘黏膜下注入造影剂C/Gd纳米复合物0.25 mL。分别于注射前及注射后5、10、15、20、30、40 min及24 h进行磁共振淋巴成像。计算特定时间节点的信号强化率(E%),定位哨位淋巴结,观察淋巴系统。注射造影剂24 h后,于双侧舌下黏膜注射0.2 mL亚甲蓝,行活体淋巴染色,解剖颈部淋巴结,行组织病理学检查。采用Sp.S19.0软件包对数据进行统计学分析。结果:成功建立兔VX2舌癌模型16只(16/16),其中发生淋巴结转移16只(16/16)。注射造影剂后10 min,肿瘤侧与对照侧淋巴结磁共振成像的E%具有显著差异(p.0.05)。IMr.G判断转移哨位淋巴结18个,在亚甲蓝活体染色下,解剖出淋巴结32个,其中病理学检测转移淋巴结17个,IMr.G的准确率为94.45%(17/18),两者诊断SLN转移无统计学差异(p.0.05)。结论:应用C/Gd纳米复合物行IMr.G,可较准确地判断兔舌癌哨位淋巴结状况,为判断舌癌淋巴结转移提供了新的思路,有潜在应用价值。
