MnO_(2)-coated nanodiamond-driven photodynamic therapy for enhanced antitumor effect by addressing hypoxia and glutathione depletion

The generation of reactive oxygen species(ROS)at the tumor site to induce destruction is emerging as a novel strategy for cancer treatment,which involves photodynamic therapy(PDT).Nevertheless,tumors typically create a hypoxic environment and are equipped with an endogenous antioxidant defense system that could potentially impede the efficiency of the therapeutic *** overcome these drawbacks,herein,a tumor microenvironment-responsive the ND-PAA-CD-Ce6@MnO_(2)(NPCC@M)delivery system was fabricated by disulfide bond coupling chlorin e6(Ce6)to nanodiamond(ND)and further wrapped by MnO_(2)nanosheets to facilitate *** use of disulfide bond not only stabilizes Ce6 in the blood circulation to prevent premature leakage,but also destroys the antioxidant barrier of overexpressed glutathione(GSH)in tumor ***,the outer MnO_(2)was rapidly degraded by the endogenous hydrogen peroxide(H_(2)O_(2))in the acidic pH and GSH within the tumor cells,which leads to an abundance of O_(2)and while increases the level of 1O_(2)under laser *** results eventually broke the redox homeostasis and attenuate hypoxia,thereby inducing apoptosis and necrosis of tumor *** in vitro and in vivo biological effect has revealed a good biosafety profile and a high tumor suppression *** a novel ND-based system with tumor microenvironment-modulating capability to elevate oxygen content and promote GSH consumption in tumor cells opens new opportunities for enhanced ROS treatment paradigms.

SIRT1 inhibits apoptosis of human lens epithelial cells through suppressing endoplasmic reticulum stress in vitro and in vivo

Jurassic integrative stratigraphy,biotas,and paleogeographical evolution of the Qinghai-Tibetan Plateau and its surrounding areas

The Qinghai-Tibetan Plateau experienced a unique geological evolution during the Jurassic,driven by the termination of the Palaeotethys and the reduction of the *** Indian Plate separated from the northern margin of Gondwana and drifted northward from the Southern *** that the timing of strata serves as the basis for reconstructing geological history,the present work aimed to develop a new multiple stratigraphic and chronologic framework for the Jurassic strata of the Qinghai-Tibetan Plateau region via a synthesis of the material on lithostratigraphy,palaeontology,iso-radiometric dating,magnetostratigraphy,and other techniques with an emphasis on recent progress and *** new framework included the Jurassic System from the four major subdivisions of the plateau:the Baryan Har,Qiangtang,Lhasa-Gandise,and Southern Xizang(Himalaya).Ultimately,a more complete,refined biostratigraphic sequence was proposed,comprising the most common fossils in the plateau and those that are stratigraphically significant for the Jurassic stratigraphy,including ammonites,bivalves,brachiopods,foraminifera,radiolarians,and dinoflagellate cysts for the marine strata,and pollen and spores,and charophytes for the terrestrial *** biostratigraphic framework was correlated with the Jurassic international standard zonation of the Geological Time Scale 2020 via standard or representative species or genera of *** on this framework,we constructed a lateral correlation of the Jurassic strata between different basins of the *** palaeontologic correlation in the present work shows that the Lhasa-Gandise Block had a closer relationship with the Qiangtang Block than with the Southern Xizang Himalaya during the Jurassic ***,the Lhasa-Gandise Block and Qiangtang Block shared similar marine fauna features of the north marginal East *** contrasts the opinion suggesting that the Yarlung Zangbo Tethys was a small back-arc basin.A c

Green carbonization of waste coffee grounds into porous C/Fe hybrids for broadband and high-efficiency microwave absorption

Recently great effort s have been focused on designing high-performance microwave absorbers using sustainable biomass resources,but there remains a lack of green and efficient fabrication ***,inspired by natural porous character of biomass waste,we demonstrated a green one-step route to convert waste coffee grounds into porous C/Fe hybrids,and further explored their potential applications for broadband and high-efficiency microwave *** this design,the WCG-20-750(incorporated 20 wt%Fe(C_(5)H_(7)O_(2))_(3)catalyst and carbonized at 750℃)exhibited porous microstructure with the highest char yield of 55.45 wt%.Furthermore,the as-prepared C/Fe hybrids from WCG-20-750 displayed excellent microwave absorption ***,the minimum reflection loss(RL_(min))reached to-52.86 dB and the widest effective absorption bandwidth(EAB)was 6.40 GHz at the thickness of 3.0 *** work provides an economically viable and environmentally friendly strategy to convert biomass wastes into value-added microwave absorbers,ultimately making contributions to the upcycling of renewable biomass resources and the fostering of sustainable environment.

The BTB/TAZ domain-containing protein CmBT1-mediated CmANR1 ubiquitination negatively regulates root development in chrysanthemum

Roots are fundamental for plants to adapt to variable environmental *** development of a robust root system is orchestrated by numerous genetic determinants and,among them,the MADS-box gene ANR1 has garnered substantial *** research has demonstrated that,in chrysanthemum,CmANR1positively regulates root system ***,the upstream regulators involved in the CmANR1-mediated regulation of root development remain *** this study,we successfully identified bric-a-brac,tramtrack and broad(BTB)and transcription adapter putative zinc finger(TAZ)domain protein CmBT1 as the interacting partner of CmANR1 through a yeasttwo-hybrid(Y2H)screening ***,we validated this physical interaction through bimolecular fluorescence complementation and pull-down *** assays revealed that CmBT1 exerted a negative influence on root development in *** both in vitro and in vivo assays,it was evident that CmBT1mediated the ubiquitination of CmANR1 through the ubiquitin/26S proteasome *** ubiquitination subsequently led to the degradation of the CmANR1 protein and a reduction in the transcription of CmANR1-targeted gene CmPIN2,which was crucial for root development in *** analysis suggested that CmBT1 modulated root development,at least in part,by regulating the level of CmANR1 ***,these findings shed new light on the regulatory role of CmBT1 in degrading CmANR1 through ubiquitination,thereby repressing the expression of its targeted gene and inhibiting root development in chrysanthemum.

Efficient intracellular delivery of protein drugs is critical for protein *** combination of protein drugs with chemotherapeutics represents a promising strategy in enhancing anti-cancer ***,co-delivery systems for efficient delivery of these two kinds of drugs are still lacking because of their different ***,we show a well-designed delivery system based on dynamic covalent bond for efficient intracellular co-delivery of ribonuclease A(RNase A)and doxorubicin(DOX).Two polymers,PEG-b-P(Asp-co-AspDA)and PAE-b-P(Asp-co-AspPBA),and two 2-acetylphenylboronic acid(2-APBA)-functionalized drugs,2-APBA-RNase A and 2-APBA-DOX,self-assemble into mixed-shell nanoparticles(RNase A/DOX@MNPs)via dynamic phenylboronic acid(PBA)-catechol bond between PBA and dopamine(DA)*** PBA-catechol bond endows the nanoparticles with high stability and excellent stimulus-responsive drug release *** the slight acidic environment at tumor tissue,RNase A/DOX@MNPs are positively charged,promoting their *** cellular uptake into endosome,further protonation of PAE chains leads to the rupture of endosomes because of the proton sponge effect and the cleavage of PBA-catechol bond promotes the release of two *** cytoplasm,the high level of GSH removed the modification of 2-APBA on *** restored RNase A and DOX show a synergistic and enhanced antic-cancer *** system may be a promising platform for intracellular co-delivery of protein drugs and chemotherapeutics.

Thermal stability of the Cr-coated zirconium alloy microstructure prepared by pulsed laser deposition

Cr-coated zirconium alloy was prepared by pulsed laser deposition(PLD)for the application of accident-tolerant fuel cladding in light water *** microstructural characteristics of the Cr coating and its evolution with temperature were investigated using grazing incidence X-ray diff raction and in situ heating transmission electron microscopy(TEM).Results show that the microstructure of the laser-deposited Cr coatings consists mainly of fine and non-specific shaped nano-crystals in the inner layer and columnar crystals in the outer *** recrystallization of the Cr-coating layer starts at 300–400℃ to release the high strain introduced by PLD,and the grain coalescence starts at temperatures>400°*** annealing,the(110)-texture gradually intensifi es because of its high reticular density and low close-packed ***,in situ heating TEM observation shows the presence of cavities on the Cr–Zr interface,which may result from the interdiff usion and/or the transformation from amorphous to crystalline.

Changes of the peripapillary vascular parameters inpremature infants without retinopathy of prematurityusing U-net segmentation

AIM:To quantitatively assess the changes in mean vascular tortuosity(mVT)and mean vascular width(mVW)around the optic disc and their correlation with gestational age(GA)and birth weight(BW)in premature infants without retinopathy of prematurity(ROP).METHODS:A single-center retrospective study included a total of 133(133 eyes)premature infants[mean corrected gestational age(CGA)43.6wk]without ROP as the premature group and 130(130 eyes)CGA-matched fullterm infants as the control *** peripapillary mVT and mVW were quantitatively measured using computerassisted ***:Premature infants had significantly higher mVT(P=0.0032)and lower mVW(P=0.0086)by 2.68(10^(4) cm^(-3))and 1.85μm,*** analysis with GA showed significant differences(P=0.0244)in mVT between the early preterm and middle to late preterm groups,but the differences between mVW were not significant(P=0.6652).The results of the multiple linear regression model showed a significant negative correlation between GA and BW with mVT after adjusting sex and CGA(P=0.0211 and P=0.0006,respectively).For each day increase in GA at birth,mVT decreased by 0.1281(10^(4) cm^(-3))and for each 1 g increase in BW,mVT decreased by 0.006(10^(4) cm^(-3)).However,GA(P=0.9402)and BW(P=0.7275)were not significantly correlated with ***:Preterm birth significantly affects the peripapillary vascular parameters that indicate higher mVT and narrower mVW in premature infants without *** in these parameters may provide new insights into the pathogenesis of ocular vascular disease.

Glycidol is a common lipid-derived foodborne toxicant mainly presents in refined oils and related *** endothelial cells may be potential targets of the deleterious effects associated with glycidol *** human umbilical vein endothelial cells(HUVECs),we found that glycidol treatment promoted endothelialto-mesenchymal transition(EndMT)at a lower concentration(0.5 mmol/L),while induced apoptosis and inflammation at a higher concentration(1 mmol/L).These harmful effects were achieved by the activation of NF-κB/MAPK signaling pathway and were mediated by reactive oxygen species(ROS).In addition,the protective potential of 6-C-(E-2-fluorostyryl)naringenin(6-CEFN)against glycidol was evaluated and compared with *** pre-treated with 6-CEFN,but not naringenin,displayed resistance to endothelial dysfunction caused by glycidol.

Damage Characteristics Analysis and Fractal Study of Shale With Prefabricated Fractures under Thermal-mechanical Coupling

To study the damage and failure of shale with different fracture inclination angles under uniaxial compression loading,in this work,RFPA2D-Thermal,a two-dimensional real failure process analysis software,was used for numerical *** simulation results show that quartz in shale mainly affects the tensile and compressive strength of shale by increasing rock *** coupling of temperature and pressure will cause lateral and volume destruction of shale,which enables the shale body to be more easily *** inclination is the key factor affecting shale damage *** failure mode of shale with low-and high-angle fractures is mainly shear failure,and the compressive strength does not vary with crack *** damage mode of obliquely intersecting fractured shale is slip damage along the fracture face,the compressive strength decreases and then increases with the fracture inclination,and a minimum value *** acoustic emission simulation results of the damage process effectively reflect the accumulated internal damage and macroscopic crack appearance until fracture instability when the prefabricated fractured shale is subjected to uniaxial compressive *** crack inclinations of 0°and 120℃ corresponds to the most complex"N"shape damage *** crack inclinations of 30°and 60°,and the damage mode is an inverted"λ"shape.
