Two-dimensional polyacrylamide gel electrophoresis analysis of indomethacin-treated human colon cancer cells

AIM: To establish the two-dimensional gel electrophoresis (2-DE) profiles of indomethacin (IN)-treated human colon cancer cell line HCT116, and to provide a new way to study its anti-tumor molecular mechanism through analyzing a variety of protein ***: Two-DE profiles of HCT116 were established in IN-treated and untreated groups. Total proteins were separated by immobilized pH gradient-based 2-DE. The gels were stained by silver, scanned by ImageScanner,and analyzed with Image Master ***: Clear background, well-resolved and reproducible 2-DE patterns of HCT116 cells were acquired in IN-treated and untreated group. The average deviation of spot position was 0.896±0.177 mm in IEF direction and 1.106±0.289 mm in SDS-PAGE direction respectively. In IN-treated group,1 169±36 spots were detected and 1 061±32 spots were matched, the average matching rate was 90.6% in three gels. In untreated group, 1 256±50 spots were detected and 1 168±46 spots were matched, the average matching rate was 93.0% in three gels. Forty-five differential protein spots were displayed between IN-treated and untreated groups. Of which, 34 protein spots decreased and 9showed higher expression in IN-treated group, and only two protein spots showed an expression in untreated ***: Two-DE profiles of IN-treated and untreated HCT116 cells were established. Apparent 45 different protein spots were detected in IN-treated and untreated HCT116 cells. The analysis on differential protein spots may serve as a new way to study the molecule mechanism of IN-treated colon cancer.

A First-principles Calculation of Structures and Stability of Al_(13)I Cluster

Using first-principles pseudo-potential plane wave method, the energetics, geometrical and electronic structures of three Al13I cluster isomers were calculated. The calculation results of the binding energy indicate Al13I cluster is more stable than Al13 cluster although its electrons are not a magic number as in Alia cluster, and among Al13I cluster isomers the ＂Bridge＂ structure is the most stable, the second is the ＂Ontop＂ structure, and the worst is the ＂Hollow＂ structure. By analyzing the geometrical structures of Al13I cluster isomers, it is found that after I atom and Al13 cluster combine the geometrical structures of Al13 moieties are changed besides Al13I Hollow cluster, in which the Alia moiety is still a regular icosahedron. For Al13I Ontop cluster, the Al13 moiety has a shrinking trend to I, whereas in Al13I Bridge cluster it is distorted. Mulliken population analysis shows for the interaction of electrons between Al-I atoms in Al13I cluster not only there exists an ionic bonding but there is a covalent bonding. Part of electrons in the Alia cluster transfer to I as Al13 cluster and I atom combine. The order of the strength of covalent bonding between Al13 moiety and I in Al13I cluster isomers is Al13IBridge〉Al13IHollow〉Al13I Ontop. Further analysis of electric structures of Al13 and Al13I clusters indicates a higher stability of Al13I cluster than Al13 cluster can be attributed to the s-p hybridization of 3s and 3p electrons of Al in Al13 moiety induced by 1 doped, which leads to fewer electrons N（EF） at EF in Al13I and a larger energy gap △EH-L between HOMO and LUMO levels in Al13I cluster. The distinguish of structural stability of Al13I cluster isomers mainly originates from their different magnitudes .in decrease of N（EF） and increase of △EH-L relative to Al13 cluster. The fewest N（EF） and the largest △EH-L are responsible for the high stability of Al13I Bridge cluster.

Composite coating on GH4169 alloy is prepared by laser cladding yttria-stabilized zirconia(YSZ)@Ni core-shell powdersmixed with NiCoCrAlY alloy *** cyclic oxidation behavior of the coatings,especially the growth process of theoxide layer,is investigated based on experimental research and first-principle *** results indicate that theoxidation resistance of coated GH4169 alloy is beer than that of uncoated GH4169 *** coating has three layers:acellular dendrite outer layer,a planar YSZ interlayer,and an inner layer composed of Cr2O3 formed during laser *** oxidation at 1000 and 1050℃,as the oxidation time increases,the cellular dendrite outer layer becomes thicker,andthe planar yttria-stabilized zirconia interlayer becomes *** the planar interlayer and Cr2O3 inner layer,and Al2O3 layer ***,cracks formed in the intcrfa.e of Al2O3/Cr2O3 owing to thcir weak interfa.e strength,whichled to the fa.lure of the composite coating.

Discrete vortices on anisotropic lattices

We consider the effects of anisotropy on two types of localized states with topological charges equal to 1 in two-dimensional nonlinear lattices, nsing the discrete nonlinear SchrSdinger equation as a paradigm model. We find that on-site-centered vortices with different propagation constants are not globally stable, and that upper and lower boundaries of the propagation constant exist. The region between these two boundaries is the domain outside of which the on-site-centered vortices are unstable. This region decreases in size as the anisotropy parameter is gradually increased. We also consider off-site-centered vortices on anisotropic lattices, which are unstable on this lattice type and either transform into stable quadrupoles or collapse. We find that the transformation of off-site- centered vortices into quadrupoles, which occurs on anisotropic lattices, cannot occur on isotropic lattices. In the quadrupole case, a propagation-constant region also exists, outside of which the localized states cannot stably exist. The influence of anisotropy on this region is almost identical to its effects on the on-site-centered vortex case.

Experimental and Theoretical Study of Reactions between Manganese Oxide Cluster Cations and Hydrogen Sulfide

Manganese oxide cluster cations Mnm180n＋ were prepared by laser ablation and reacted with hydrogen sulfide （H2S） in a fa.t flow reactor under thermal collision conditions. A time-of-flight mass spectrometer was used to detect the cluster distributions before and after the interactions with H2S. The experiments suggest that oxygen-for-sulfur （O/S） ex- change reaction to release water took place in the reactor for most of the manganese oxide cluster cations： MnmlSOn＋＋H2S→Mnm18On-1S＋＋H218O. Density functional theory cal- culations were performed for reaction mechanisms of Mn202＋＋H2S, Mn203＋＋H2S, and Mn204＋＋H2S. The computational results indicate these O/S exchange reactions are both thermodynamically and kinetically fa.orable, thus in good agreement with the experimental observations. The O/S exchange reactions identified in this gas-phase cluster study parallel similar behavior of related condensed phase reaction systems.

Seasonal waterbird population changes in Lashihai Lake in northwest Yunnan, China

We studied the waterbird population at Lashihai Lake, Yunnan Province, China, which is a Ramsar Site(Wetland of International Importance), to determine seasonal variation in the species composition and size of the waterbird population. The study was conducted at five selected spots along Lashihai Lake at the same time each week from August 2011 to September 2013. In total, 62 waterbird species were recorded, of which 38.71%, 35.48%, 16.13%, and 9.68% were winter migrants, passage visitors, residents, and summer migrants, respectively. We found important seasonal changes in waterbird species composition and population size. Waterbird species richness was highest from September to the following April, with the total species numbers peaking in December. Total individual numbers peaked twice from late November to early December and mid-to-late February. However, waterbird species and individual numbers were comparatively lower from May to August. The change in species composition was determined by the arrival and departure dynamics of winter migrants and passage visitors. Winter migrants primarily caused the periodic changes in population size. Of concern,species and overall waterbird numbers seemed to be lower than the numbers in historical records. The decline of waterbird numbers implies that environmental changes caused by the implementation of the dam upstream of Lashihai Lake may have had adverse effects on this waterbird population. This study confirms the existence of major seasonal changes in species composition and size of the waterbird population at Lashihai Lake. Furthermore, the findings demonstrate that this wetland is of high conservation importance for waterbirds using the Central Asian–Indian and Asian–Pacific migratory routes.

Consideration of stable isotopes in precipitation is valuable for investigating hydrological ***,correcting the measured isotopic composition of precipitation under below-cloud evaporation is *** accurate description of the underlying processes affecting stable isotopic composition of precipitation could help improve our understanding of the water *** transitivity between monsoonal and arid climates was reflected by the evaporation rate of fa.ling raindrops in precipitation in the Qilian Mountains,a typical transition zone between Tibetan Plateau and arid region of *** 1310 precipitation event-scale samples,based on stable isotope analysis method,the mean below-cloud evaporation rate(f)in the study area was measured as 12.00%during the summer half-year(May-October).The evaporation rate on the northern slopes(12.70%)of the Qilian Mountains in China was significantly higher than that on the southern slopes(9.98%).The transition between monsoonal and arid climates was reflected in the evaporation rate of fa.ling raindrops during precipitation in the Qilian Mountains of ***-cloud evaporation contributed to a noticeable enrichment of stable isotopes in the precipitation in the study *** monthly precipitationδ^(18)O enrichment rate in the Qilian Mountains of China from May to October was 29.18%,23.35%,25.60%,22.99%,31.64%,and 14.72%,*** every 1.00%increase in the evaporation rate of raindrops in Qilian Mountains of China,the changes in the concentration of oxygen isotopes from the bottom of the clouds to the ground increased by 0.92‰;however,with an evaporation rate offa.tor affecting below-cloud evaporation in the study area.

Effect of droplet characteristics on liquid-phase distribution in spray zone of internal mixing air-mist nozzle

In continuous casting production,droplet characteristics are important parameters for evaluating the nozzle atomization quality,and have a significant impact on the secondary cooling effect and the slab *** order to study the behavior of atomized droplets after reaching the slab surfa.e and to optimize the spray cooling effect,the influence of droplet diameter and droplet velocity on the migration behavior of droplets in the secondary cooling zone was analyzed by FLUENT *** show that the droplets in the spray zone and on the slab surfa.e are mainly concentrated in the center,thus,the liquid volume fraction in the center is higher than that of either *** the droplet diameter increases,the region of high liquid volume fraction on the slab surfa.e becomes wider,and the liquid phase distribution in the slab width direction becomes *** increasing the droplet velocity at the nozzle exit has little effect on droplet diffusion in the spray zone,the distribution becomes more uneven due to more liquid reaches the slab surfa.e per unit time.A prediction formula of the maximum water flow rate on the slab surfa.e for specific droplet characteristics was proposed based on dimensionless analysis and validated by simulated data.A nozzle spacing of 210 mm was recommended under the working conditions in this study,which ensures effective coverage of the spray water over the slab surfa.e and enhances the distribution uniformity of water flow rate in the transverse direction.
