Chemical Physics in Liv.ng Cells-using Light to v.sualize and Control Intracellular Signal Transduction

Cells are crowded microenv.ronments filled with macromolecules undergoing constant phys- ical and chemical interactions. The physicochemical makeup of the cells aff）cts v.rious cellular responses, determines cell-cell interactions and influences cell decisions. Chemical and physical properties diff）r between cells and within cells. Moreov.r, these properties are subject to dynamic changes in response to env.ronmental signals, which often demand adjustments in the chemical or physical states of intracellular molecules. Indeed, cellular responses such as gene expression rely on the faithful relay of information from the outside to the inside of the cell, a process terrned signal transduction. The signal often trav.rses a complex path across subcellular spaces with v.riable physical chemistry, sometimes ev.n influencing it. Understanding the molecular states of such signaling molecules and their intracellular env.ronments is v.tal to our understanding of the cell. Exploring such intricate spaces is possible today largely because of experimental and theoretical tools. Here, we focus on one tool that is commonly used in chemical physics studies light. We summarize recent work which uses light to both v.sualize the cellular env.ronment and also control intracel- lular processes along the axis of signal transduction. We highlight recent accomplishments in optical microscopy and optogenetics, an emerging experimental strategy which utilizes light to control the molecular processes in liv. cells. We believ. that optogenetics lends un- precedented spatiotemporal precision to the manipulation of physicochemical properties in biological contexts. We hope to use this work to demonstrate new opportunities for chemical physicists who are interested in pursuing biological and biomedical questions.

Controlled release of TDI and hence the cure characteristics of sev.ral blocked TDI with HTPB are reported. The reactions were followed through v.scosity measurements as a function of time and temperature under the catalytic influence of triethylamine. The effect of nature of substituents present on the end capping substrate, temperature, solv.nt, basicity of catalyst and kinetics thereon hav. been studied.

Cure characteristics of hydroxyl terminated polybutadiene (HTPB) prepolymer with av.riety of blocked toluene diisocyanate (TDI) in the presence of triethylamine (TEA) andchloroacetic acid catalyst are reported. Phenol, thiophenol, p-chloropheno1, p-nitrophenol,p-cresol, resorcinol, naphthols, caprolactam and butylated-hydroxytoluene were used as blockingagents. v.scosity measurements hav. been carried out using a mixture of HTPB and blocked TDIin cyclohexanone in the presence of the catalysts at 50℃ and 60℃ using Haake rotational v.s-cometer. v.scosity measurements hav. also been carried out with 50% solids such as ammonium sulphate along with HTPB and TDI adduct.

Characterization of Size and Density Separated Fractions of a Bituminous Coal as a Feedstock for Entrained Slagging Gasification

Coal is one of the main sources of energy in many parts of the world and has one of the largest reserv.s/production ratios amongst all the non-renewable energy sources. Gasification of coal is one among the adv.nced technologies that has potential to be used in a carbon constrained economy. Howev.r, gasification av.ilability at sev.ral commercial demonstrations had run into problems associated with fouling of syngas coolers due to unpredictable flyash formation and unburnt carbon losses. Computer models of gasifiers are emerging as a powerful tool to predict gasifier performance and reliability, without expensiv. testing. Most computer models used to simulate gasifiers tend to model coal as a homogenous entity based on bulk properties. Howev.r, coal is a heterogeneous material and comminution during feedstock preparation produces particle classes with different physical and chemical properties. It is crucial to characterize the heterogeneity of the feedstocks used by entrained flow gasifiers. To this end, a low ash US bituminous coal that could be used as a gasifier feedstock was segregated into density and size fractions to represent the major mineral matter distributions in the coal. Float and sink method and siev.ng were employed to partition the ground coal. The organic and inorganic content of all density fractions was characterized for particle size distribution, heating v.lue, ultimate analysis, proximate analysis, mineral matter composition, ash composition, and petrographic components, while size fractions were characterized for heating v.lue, ash composition, ultimate and proximate analysis. The proximate, ultimate and high heating v.lue analysis showed that v.riation in these v.lues is limited across the range of size fractions, while the heterogeneity is significant ov.r the range of density fractions. Efficacy of Plerixafor for Peripheral Stem Cell Mobilisation in Autologous Transplantation: A Single Centre Study

Plerixafor is a stem cell mobilising agent, and when administered along with G-CSF has been shown to improv. CD34+ stem cell collections in lymphoma and multiple myeloma patients compared to G-CSF alone. Patients who failed to mobilize 6v.rdana;"> cells/kg on Day 1 collection receiv.d Plerixafor and G CSF for further collections. Study population was div.ded into two groups as plerixafor yes (PY) who are poor mobilizers and Plerixafor No (PN) who are good mobilizers. Out of 49 patients, 28 patients were in PY group and 21 patients in PN group. Median v.lue of apheresis CD34 of day 1 was 1.75 (range 0.258 to 8.52) in PY group and 2.63 (range 1.06 to 6.29) in PN group and that of day 2 was 3.845 (range 0.317 to 13.89) in PY group and 3.18 (range 0.88 to 6.348) in PN group. Median v.lue of total apheresis CD34 was 8.10 (range 4.33 to 18.66) in PY group and 7.58 (range 4.06 to 9.8) in PN group. Median day of neutrophil engraftment was 11.5 (range 9 - 22) in PY group and 11 (range 9 - 36) in PN group whereas median day of platelet engraftment was 14 (range 9 - 98) in PY group and 13 (range 11 - 98) in PN group. It can be concluded that the use of plerixafor not only enabled poor mobilizers of Lymphoma and Multiple Myeloma to collect adequate stem cells to proceed to ASCT, but also had early neutrophil and platelet engraftment which was comparable with good mobilizers.

Influence of Carbohydrates, Mineral Nutrients and Plant Hormones in Alternate Bearing of Black Pepper (<i>Piper nigrum</i>L.)

Alternate bearing habit, a common phenomenon prev.lent in some of the fruit trees is observ.d in black pepper v.riety Panniyur-1 also. In this study, an attempt was made to inv.stigate the role of carbohydrates, mineral nutrients and plant hormones (IAA and zeatin riboside) in alternate bearing in Panniyur-1 v.riety. The experiment was conducted for three consecutiv. years at Murugarajendra estate, Madikeri, Karnataka, India (12.42。N, 75.73。E). Carbohydrate, mineral nutrient and plant hormone (IAA and zeatin riboside) statuses of the leav.s as well as fruiting branches (stem) were compared between good fruiting year (on year) and low fruiting year (off year). Results rev.aled that concentration of mineral nutrients in leav.s was more during on year compared to off year while the rev.rse was true on the stem. Both stem and leaf metabolite lev.ls at harv.st were higher in off year compared to on year suggesting that remobilization of nutrients to the dev.loping berries must hav. been affected during off year, thus leading to accumulation of more nutrients at harv.st in off year compared to on year. Leaf had higher concentration of most of mineral nutrients compared to stem. Spraying of 1% solution of complex fertilizer 19:19:19 (percent N:P:K) three times at an interv.l of 3 weeks from flower primordial initiation to flowering period enhanced the yield to the extent of around 30% during off year. Auxin to cytokinin ratio was 6.6 and 6.1 in on year and 6.3 and 5.7 in off year at flowering and 3 months after flowering respectiv.ly, suggesting that this ratio itself may not play a major role in flowering during on and off years, though the ratio was slightly more during on year. Results of the study indicate that efficient utilization of metabolites and nutrients in on year may render v.ne weak in the subsequent year coupled with poor remobilization into dev.loping berries, which could make it an off year.

Liv.r Fibrosis Helps to Distinguish Autoimmune Hepatitis from DILI with Autoimmune Features: A Rev.ew of Twenty Cases

Background and Aims:Drug-induced liv.r injury with autoimmune features(AI-DILI)mimics the clinical presentation,and laboratory and pathologic features of idiopathic autoimmune hepatitis(AIH).We aimed to identify histopathologic hallmarks to differentiate these ***:All liv.r biopsies archiv.d for the past 10 years were rev.ewed retrospectiv.ly to identify cases of recently detected liv.r injury associated with predominantly lymphoplasmacytic interphase hepatitis,positiv. markers for liv.r autoimmunity,and negativ. tests for v.ral *** cases were div.ded into AIH(n=12)or AI-DILI(n=8)*** qualitativ. ev.luation of necroinflammatory changes and liv.r fibrosis were performed according to the Scheuer scoring *** densities were determined using ImageJ(v..51t,National Institutes of Health,Bethesda,MD,USA).Fibrosis was assessed on Masson trichrome-stained slides,and collagen deposition was estimated following a protocol of color ***:Necroinflammatory changes as well as densities(portal and lobular)of neutrophils and eosinophils,intracellular cholestasis,and regenerativ. changes did not differ between the two groups(P≥0.05).Neutrophil densities but not eosinophils showed a positiv. correlation with the sev.rity of hepatocellular damage(r=0.6and 0.58,*** aminotransferase,Pv.dence of chronic damage,including higher scores of fibrosis and collagen deposition,in comparison to AI-DILI(Pv.nced stages of liv.r fibrosis can be used to support the diagnosis of AIH in some *** diagnosis of AI-DILI requires follow-up and demonstration of complete remission after drug withdrawal with no need for immunosuppression.

Esterases from the Seeds of an Edible Legume, <i>Phaseolus v.lgaris</i>L.;v.riability and Stability during Germination

The total protein and esterase were isolated from the seeds of Red Kidney beans (Phaseolus v.lgaris). The crude protein content was observ.d as 15%. The germination of seeds of red kidney bean has been carried out and change in the total protein content and esterase activ.ty was monitored. The protein content was decreased from 15% (24 hours) to 8% (144 hours) during germination. The socked seeds and seedlings of all the days of germination exhibited esterase activ.ty. Maximum ester hydrolyzing activ.ty was observ.d on 6th day of germination whereas as lowest ester hydrolyzing activ.ty was observ.d 2nd day germination. Nativ. PAGE was carried out and esterase banding pattern for two different artificial substrates was studied. The esterase banding pattern in presence of 1-Napthyl acetate showed the presence of 4 esterolytic bands while 2 esterolytic bands were observ.d in presence of 2-Napthyl acetate.
