We investigated the role of the Na/H exchanger 3(NHE3)regulatory factor 1(NHERFl)on intestinal salt and water absorption,brush border membrane morphology and on the NHE3 mRNA expression,protein abundance,and transport activity in the murine ***1-deficient mice displayed reduced jejunal fluid absorption in vivo,as well as a attenuated Na absorption in isolated mucosa of jejunal and colonic tissue but not of ileal mucosa in ***,cAMP-mediated inhibition of both parameters remained *** basal NHE3 transport rate was decreased in surface colonocytes,while inhibition by cAMP and cGMP was normal. Immunodetection of NHE3 revealed normal NHE3 localization in the brush border membrane(BBM)of NHERF1 null mice,but NHE3 abundance,as measured by Western blot,was significantly reduced in isolated membranes from the small and large ***,the microvilli in the proximal colon,but not in the small intestine, were significantly shorter in NHERF1 null *** knockout of PDZK1(NHERF3),another member of the NHERF family of adapter proteins,which binds to both NHE3 and NHERF1,further reduced basal NHE3 activity and caused complete loss of cAMP-mediated NHE3 *** activator of the exchange protein activated by cAMP(EPAC)had no effect on jejunal fluid absorption in vivo,but slightly inhibited NHE3 activity in surface colonocytes in *** conclusion,NHERF1 has segment-specific effects on intestinal salt absorption,NHE3 transport rates and NHE3 membrane abundance without affecting mRNA ***,unlike PDZK1,NHERF1 is not required for NHE3 regulation by cyclic nucleotides.

An Experimental Simulation of Addressing Auto-Configuration Issues for Wireless Sensor Networks

Applications of Wireless Sensor devices are widely used byvarious monitoring sections such as environmental monitoring, industrialsensing, habitat modeling, healthcare and enemy movement detection *** were found that 16 bytes packet size (payload) requires MediaAccess Control (MAC) and globally unique network addresses overheads asmore as the payload itself which is not reasonable in most situations. Theapproach of using a unique address isn’t preferable for most Wireless SensorNetworks (WSNs) applications as well. Based on the mentioned drawbacks,the current work aims to fill the existing gap in the field area by providingtwo strategies. First, name/address solutions that assign unique addresseslocally to clustered topology-based sensor devices, reutilized in a spatialmanner, and reduce name/address size by a noticeable amount of 2.9 basedon conducted simulation test. Second, name/address solutions that assignreutilizing of names/addresses to location-unaware spanning-tree topologyin an event-driven WSNs case (that is providing minimal low latenciesand delivering addressing packet in an efficient manner). Also, to declinethe approach of needing both addresses (MAC and network) separately, itdiscloses how in a spatial manner to reutilize locally unique sensor devicename approach and could be utilized in both contexts and providing anenergy-efficient protocol for location unawareness clustered based *** comparison, an experimental simulation test performed and given theaddresses solution with less overhead in the header and 62 percent fairpayload efficiency that outperforms 34 percent less effective globally uniqueaddresses. Furthermore, the proposed work provides addresses uniquenessfor network-level without using network-wide Duplicate Address Detection(DAD) algorithm. Water and Heavy Metal Transport in Roadside Soils

Roads with very high traffic loads in regions where soils are low in both pH and sorption capacity might be a source of percolation water loaded with heavy metals. Looking at some "worst case" scenarios, this study focused on the input of traffic related pollutants and on Pb, Cd, Cu, Zn, Ni and Cr concentrations in the soil matrix and soil solution, respectively. The analysis also included pH and electrical conductivity and at some sites DoC. The investigations were carried out on sandy soils with more or less low pH values at four motorway sites in Germany. The average of daily traffic was about 50 000 up to 90 000 vehicles. Soil pore water was collected in two soil depths and at four distances from the road. The pH in general decreased with increasing distance from the roadside. The elevated pH near the roadside was presumably caused by deposition of dust and weathering residues of the road asphalt, as well as by infiltration of salt that was used during winter time. At these road sites, increased heavy metal concentrations in the soil matrix as well as in the soil solution were found. However, the concentrations seldom exceeded reference values of the German Soil Protection Act. The soil solution concentrations tended to increase from the road edge to 10 m distance, whereas the concentration in the soil matrix decreased. Elevated DoC concentrations corresponded with elevated Cu concentrations but did not substantially change this tendency. High soil water percolation rates were found near the roads. Thus, even low metal concentrations of percolation water could yield high metal loads in a narrow area beside the road.

Sex chromosome turnover and biodiversity in fishes

The impact of sex chromosomes and their turnover in speciation remains a subject of ongoing debate in the field of evolutionary biology. Fishes are the largest group of vertebrates, and they exhibit unparalleled sexual plasticity, as well as diverse sex-determining (SD) genes, sex chromosomes, and sex-determination mechanisms. This diversity is hypothesized to be associated with the frequent turnover of sex chromosomes in fishes. Although it is evident that amh and amhr2 are repeatedly and independently recruited as SD genes, their relationship with the rapid turnover of sex chromosomes and the biodiversity of fishes remains unknown. We summarize the canonical models of sex chromosome turnover and highlight the vital roles of gene mutation and hybridization with empirical evidence. We revisit Haldane’s rule and the large Xeffect and propose the hypothesis that sex chromosomes accelerate speciation by multiplying genotypes via hybridization. By integrating recent findings on the turnover of SD genes, sex chromosomes, and sexdetermination systems in fish species, this review provides insights into the relationship between sex chromosome evolution and biodiversity in fishes.

Dear Editor,Abnormal DNA methylation,a process whereby tumor suppressors tend to be hypermethylated and silenced,is a hallmark of cancer cells[1].Removing the methylation by demethylation agents such as azacitidine and decitabine is one of the strategies to treat cancers and has been successfully used to treat certain hematological and solid tumors[2].However,there are no established DNA methylation markers or signatures that can accurately predict patients’response to treatment[2];thus,the identification of reliable predictive biomarkers for effective therapy remains a critical need in clinical practice.

Risk-adjustment in hepatobiliarypancreatic surgery

AIM: The present study evaluates the performance of the PoSSUM, the American Society of Anesthetists (ASA), APACHE and Childs classification in predicting mortality and morbidity in hepatopancreaticobiliary (HPB) surgery. We describe especially the limitations and advantages of risk in stratifying the patients. METHoDS: We investigated 177 randomly chosen patients undergoing elective complex HPB surgery in a single institution with a total of 71 pre-operative and intra-operative risk factors. Primary endpoint was in-hospital mortality and morbidity. ordered logistic regression analysis was used to identify individual predictors of operative morbidity and mortality. RESULTS: The operative mortality in the series was 3.95%. This compared well with the p-PoSSUM and APACHE predicted mortality of 4.31% and 4.29% respectively. Postoperative complications amounted to 45% with 24 (13.6%) patients having a major adverse event. on multrvariate analysis the pre-operative PoSSUM physiological score (oR = 1.18, P = 0.009) was superior in predicting complications compared to the ASA (P= 0.108), APACHE (P= 0.117) or Childs classification (P= 0.136). In addition, serum sodium, creatinine, international normalized ratio (INR), pulse rate, and intra-operative blood loss were independent risk factors. A combination of the PoSSUM variables and INR offered the optimal combination of risk factors for risk prognostication in HPB surgery. CoNCLUSIoN: Morbidity for elective HPB surgery can be accurately predicted and applied in everyday surgical practice as an adjunct in the process of informed consent and for effective allocation of resources for intensive and high-dependency care facilities.
