Chlamydomonas reinhardtii strains lacking phytoene synthase, the first enzyme of carotenoid biosynthesis, are white. They lack carotenoid pigments, have very low levels of chlorophyll, and can grow only heterotrophically in the dark. Our electron and fluorescence microscopic studies showed that such a mutant strain （Itsl-204） had a proliferated plastid envelope membrane but no stacks of thylakoid membranes within the plastid. It accumulated cytoplasmic compartments that appeared to be autophagous vacuoles filled with membranous material. The Its1 mutants apparently lacked pyrenoid bodies, which normally house ribulose bisphosphate carboxylase-oxygenase （Rubisco）, and accumulated many starch granules. Although these mutant strains cannot synthesize the carotenoid and carotenoid-derived pigments present in the phototactic organelle （eyespot）, the mutant we examined made a vestigial eyespot that was disorganized and often mislocalized to the posterior end of the cell. The absence of a pyrenoid body, the accumulation of starch, and the disorganization of the eyespot may all result from the absence of thylakoids. The ultrastructure of Its1 mutant strains is similar to but distinct from that of previously described white and yellow mutant strains of C. reinhardtii and is similar to that of naturally colorless algae of the Polytoma group.

Strategy for treatment of nonerosive reflux disease in Asia

The paper is to review the clinical and pathophysiologic differences between of nonerosive reflux disease (NERD) and reflux esophagitis (RE), and to propose a treatment strategy for NERD, especially for patients in Asia. A Medline search was performed regarding the clinical and pathophysiologic differences between NERD and RE, and treatment of NERD and RE. The characteristics of NERD patients in Asia are as follows: (1) high proportion of female patients, (2) low frequency of hiatal hernia, (3) high frequency of H pylori infection, (4) severe glandular atrophy of the gastric mucosa, and (5) frequent resistance to proton pump inhibitor (PPI) therapy. In Asian NERD patients, exposure of the esophagus to acid is not increased, and esophageal motility is normal. These characteristics are similar to those of patients in Western countries. Our recommended first-choice treatment is administration of PPI in combination with a prokinetic agent. However, at present, because there is limited evidence regarding effective treatments for NERD, it is best to try several different treatment strategies to find the best treatment for each patient.

The major Rhesus （Rh） protein of the green alga Chlamydomonas reinhardtii, Rhl, is homologous to Rh proteins of humans. It is an integral membrane protein involved in transport of carbon dioxide. To localize a fusion of intact Rhl to the green fluorescent protein （GFP）, we used as host a white （Its1） mutant strain of C. reinhardtii, which is blocked at the first step of carotenoid biosynthesis. The Its1 mutant strain accumulated normal amounts of Rhl heterotrophically in the dark and Rhl-GFP was at the periphery of the cell co-localized with the cytoplasmic membrane dye FM4-64. Although Rhl carries a potential chloroplast targeting sequence at its N-terminus, Rhl-GFP was clearly not associated with the chloroplast envelope membrane. Moreover, the N-terminal half of the protein was not imported into chloroplasts in vitro and N-terminal regions of Rhl did not direct import of the small subunit of ribulose bisphosphate carboxylase （SSU）. Despite caveats to this interpretation, which we discuss, current evidence indicates that Rhl is a cytoplasmic membrane protein and that Rhl-GFP is among the first cytoplasmic membrane protein fusions to be obtained in C. reinhardtii. Although Its1 （white） mutant strains cannot be used to localize proteins within sub-compartments of the chloroplast because they lack thylakoid membranes, they should nonetheless be valuable for localizing many GFP fusions in Chlamydomonas.

This study was aimed at the preparation of an electrode for Zinc–air battery, which had excellent catalytic activity by use of electroplating of alloys made of abundant metal, such as Fe and Ni. The oxygen overvoltage of the Fe-Ni-W alloy plated electrode was the smallest through the measurement. The elemental composition and the enlargement of the surface area were confirmed by SEM and EDX analysis. Involvement of Fe and W of Fe-Ni-W alloy plated electrode will be one factor for its high catalytic activity. Thus plated Fe-Ni-W alloy electrodes were compared with other Fe alloy plated elec-trodes considering to their cathode performance as Zinc-air battery. The catalytic activity of Fe-Ni-W plated electrode showed the best performance comparing to Fe-Ni alloy plated electrodes as cathode for Zinc-air battery. Also comparing to the platinum electrode which had been widely used as cathode in the field of Zinc-air battery, the Fe-based alloy plated electrode showed better performance as the electrodes considering to its oxygen evolution reaction.

We analyzed the amino acid residues present in the water-soluble and transmembrane proteins of 6 thermophilic and 6 mesophilic species of the domains Archaea and Eubacteria, and characterized them as favorable or unfavorable. The characterization was performed by comparing the observed number of each amino acid residue to the expected number calculated from the percentage of nucleotides present in each gene. Amino acids that were more or less abundant than expected were considered as favorable or unfavorable, respectively. Comparisons of amino acid compositions indicated that the water-soluble proteins were rich in charged residues such as Glu, Asp, Lys, and His, whereas hydrophobic residues such as Trp, Phe, and Leu were abundant in transmembrane proteins. Interestingly, our results found that although the Trp residue was abundant in transmembrane proteins, it was not defined as favorable by our calculations, indicating that increased numbers of a particular amino acid does not necessary indicate it is a favorable residue. Amino acids with high G + C content such as Ala, Gly, and Pro were frequently observed as favorable in species with low G + C content. Comparatively, amino acids with low G + C content such as Phe, Tyr, Lys, Ile, and Met were frequently observed as favorable in species with high G + C content. These are the examples to increase the supply of amino acids than expected. Amino acids with neutral G + C content, i.e., Glu and Asp were favorable in water-soluble proteins from all species analyzed, and Cys was unfavorable both in water-soluble and transmembrane proteins. These results indicate that amino acid compositions are essentially determined by the nucleotide sequence of the genes, and the amino acid content is altered by a deviation from expectation.

A Study on Relationship among Blade Camber Direction and Pitch Angle and Performance of a Small Straight-Blade Darrieus Wind Turbine by Using Scale Test Model and Gurney Flap

Straight-blade Darrieus vertical axis wind turbines are used as medium and small size wind turbine because of higher power output in vertical axis wind turbine (VAWT). In our previous study, the relationship between the performance and Reynolds number based on airfoil chord length had been investigated by using small-scale test models of lift-type VAWT, and the results showed that the performance of tested wind turbine models with small diameter was clearly lower than that of the large-scale field test machine, and its performance also varies significantly with the blade pitch angle. In this study, we focused on the performance of a small-scale straight-blade Darrieus VAWT, the relationship among the blade airfoil camber direction and the pitch angle, and the performance of the small-scale VAWT was examined experimentally by using a small-scale VAWT test model with Gurney flap which was a small flat plate. Gurney flaps with its height h, as a ratio to the blade chord length c, h/c = 0.036 to 0.055, were attached to the blades of the VAWT test model, in addition, the attaching direction of the Gurney flap on the blade was examined for both inward and outward of the rotor, and the pitch angle was also examined for a range of −5 to 10 degrees. These results are discussed comparing with the result of the VAWT without Gurney flap and considering the numerical results for the single blade with/without the Gurney flap. The results showed that the performance of the tested VAWT was reversed between the inward and outward Gurney flaps around a pitch angle of 10 degrees. That is, the inward Gurney flap was superior at a pitch angle of less than 10 degrees, while the outward Gurney flap was effective at a pitch angle of more than 10 degrees. Periodontal disease is recognized as a risk factor for cardiovascular disease (CVD). Some studies have suggested that serum lipids might play a role in the association between periodontal disease and CVD. However, this hypothesis remains unproven. Present study evaluated the association between periodontal disease and CVD-related parameters including electrocardiographic (ECG) parameters and serum lipid levels in the elderly. This study was a prospective cohort study. A total of 107 subjects (57 males and 50 females) from Niigata City’s community-dwelling elderly (all aged 75 years old) who possessed at least 20 teeth and did not exhibit ECG abnormalities at the baseline underwent annual examinations for 5 years. Logistic regression analysis was used to assess the relationship between periodontal condition and the presence of ECG abnormalities, after adjusting for confounding factors including serum lipid levels. Logistic regression analysis showed that the subjects who exhibited a poor periodontal condition at the baseline had a 2.97 times greater risk of suffering from ECG abnormalities than the subjects who displayed a good periodontal condition at the baseline (p = 0.019). Furthermore, the subjects who displayed a poor periodontal condition and an unfavorable serum lipid profile at the baseline showed a significantly higher frequency of ECG abnormalities (63.6%) than those who exhibited a healthy periodontal condition and a favorable serum lipid profile (21.6%, p = 0.008) as well as those who displayed a healthy periodontal condition and an unfavorable serum lipid profile (21.4%, p = 0.010). As a conclusion, periodontal condition could be a marker of CVD, even in the healthy elderly.

Medical malpractice litigation related to gastrointestinal endoscopy in Japan:A two-decade review of civil court cases

AIM: To examine the allegations in malpractice litigations related to gastrointestinal endoscopy in Japan. METHODS: A retrospective review of cases tried in the civil court system during the 21-year period from 1985 to 2005, identified in a computerized legal database, was undertaken. RESULTS: Eighteen malpractice litigations and a total of 30 allegations were identified. Of the 18 (44%) malpractice litigations, 8 (44%) were related to eso- phagogastroduodenoscopy, 4 (22%) to colonoscopy, 4 (22%) to endoscopic sphincterotomy, and 2 (11%) to endoscopic retrograde cholangiopancreatography. Seventeen (94%) cases pertained to complications, and the remaining (6%) case pertained to misdiagnosis. In 10 cases, the patient died of the complications. Allegations were categorized as: (1) performance error during the endoscopic procedure (n = 12, 40%); (2) lack of informed consent (n = 9, 30%); (3) performance error during the treatment after the endoscopic procedure (n = 4, 13%); (4) premedication error (n = 3, 10%); (5) diagnostic error (n = 1, 3%); and (6) indication error for the endoscopic procedure (n = 1, 3%). CONCLUSION: These data may aid in the design of risk prevention strategies to be used by gastrointestinal endoscopists.
