Objective:To investigate the anti-anaphylactic,anti-inflammatory and membrane stabilizing properties of plumerianine(compound 1)isolated from the root bark of Plumeria acuifolia ***:The anti-anaphylactic activity of compound 1(10,25 and 50 mg/kg)was studied by using models such as passive cutaneous anaphylaxis,passive paw anaphylaxis and its antiinflammatory activity against carrageenin induced paw edema and cotton pellet granuloma in albino rats was also investigated using ketotifen and indomethacin as reference ***:A dose-dependent beneficial effect was observed on leakage of evans blue dye in skin challenged with antigen and on paw anaphylaxis induced by *** compound 1 also exhibited significant(P<0.01)inhibition of rat paw edema and granuloma tissue formation,including significant protection of RBC against the haemolytic effect of hypotonic solution,an indication of membrane-stabilizing ***:Anti-anaphylactic activity of compound 1 may be possibly due to inhibition of the release of various inflammatory ***-inflammatory activity of compound may be related to the inhibition of the early phase and late phase of inflammatory events.

BACkGROUND Gamification,the use of game materials in education,has been used in several scientific fields,and studies have demonstrated its effectiveness in student *** To evaluate whether the success of 4th-and 5th-grade dentistry students increases when using the gamification applications kahoot!and *** An experimental design was used to measure the effect of gamification applications on students who were taught using the traditional *** this purpose,Mentimeter and kahoot!applications were used in this experimental *** groups of 4th-and 5th-grade students were formed:Mentimeter,kahoot!,and control *** effect of gamification applications on achievement was evaluated by administering a post-test to these groups after the instructional ***-way analysis of variance and t-tests were used for statistical *** A total of 35.5%of the students were in the Mentimeter group,29.8%in the kahoot!group,and 34.6%in the control *** the total number of students,54.8%were female and 45.2%were male,while 52.2%were 5th-grade students and 47.8%were 4th-grade *** overall grade point average was found to be 66.65(range,24-100).There was a statistically significant difference in average success scores between students of different groups(P=0.003kahoot!group(62.15).In the control group,this score was *** average success score was higher for students in the 5thgrade(68.16)and for female students(68.84).CONCLUSION Gamification has a great potential in the education of dentomaxillofacial radiology *** outperformed both the kahoot!and the traditional method in terms of average student success rates.

Traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease are three distinct neurological disorders that share common pathophysiological mechanisms involving neuroinflammation. One sequela of neuroinflammation includes the pathologic hyperphosphorylation of tau protein, an endogenous microtubule-associated protein that protects the integrity of neuronal cytoskeletons. Tau hyperphosphorylation results in protein misfolding and subsequent accumulation of tau tangles forming neurotoxic aggregates. These misfolded proteins are characteristic of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease and can lead to downstream neuroinflammatory processes, including assembly and activation of the inflammasome complex. Inflammasomes refer to a family of multimeric protein units that, upon activation, release a cascade of signaling molecules resulting in caspase-induced cell death and inflammation mediated by the release of interleukin-1β cytokine. One specific inflammasome, the NOD-like receptor protein 3, has been proposed to be a key regulator of tau phosphorylation where it has been shown that prolonged NOD-like receptor protein 3 activation acts as a causal factor in pathological tau accumulation and spreading. This review begins by describing the epidemiology and pathophysiology of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease. Next, we highlight neuroinflammation as an overriding theme and discuss the role of the NOD-like receptor protein 3 inflammasome in the formation of tau deposits and how such tauopathic entities spread throughout the brain. We then propose a novel framework linking traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease as inflammasomedependent pathologies that exist along a temporal continuum. Finally, we discuss potential therapeutic targets that may intercept this pathway and ultimately minimize long-term neurological decline.

Salt Stress Induced Modulations in Growth, Compatible Solutes and Antioxidant Enzymes Response in Two Cultivars of Safflower (Carthamus tinctorius L. Cultivar TSF1 and Cultivar SM) Differing in Salt Tolerance

Safflower is an important, traditional, multipurpose oil crop. This was to investigate the effect of different salinity levels on morphological, physiological, biochemical and antioxidant response of two safflower cultivars (Carthamus tinctorius L. cultivar TSF1 and cultivar SM) differing in salt tolerance. Salinity stress (0.0%, 1.0%, 1.5% and 2.0% of NaCl) was induced to safflower plants after 19 days of vegetative growth. After 12 days of stress impositions, plants were harvested and analysed for various parameters. The results revealed that cultivar TSF1 showed maximum growth, dry weight, cell membrane stability and more water content in both root and leaf tissues at higher salinity levels than cultivar SM. Salt stress resulted an accumulation of more soluble sugars, amino acids, proline and glycine betaine at high salinity level confers the tolerance potential of cultivar TSF1 over cultivar SM. Salt stress induces more increase in the enzyme activity of superoxide dismutase, ascorbate peroxidase, guaiacol peroxidase and catalase in tolerant cultivar than sensitive one. The results indicate that each cultivar adopt specific strategy at distinct salinity level for resistance against salinity. The possible conclusion is that improved tolerance in cultivar TSF1 to salinity may be accomplished by better management of growth, physiological attributes and antioxidative defence mechanisms.

Introduction: This study investigates the Experimental and Theoretical Investigation of Steel and Glass Fiber Reinforced Polymer (GFRP) Reinforced Slab Incorporating Alccofine and M-sand. Objective: Specific objectives include evaluating the mechanical properties and structural behaviour of steel and GFRP-reinforced one-way slabs and comparing experimental and theoretical predictions. Methods: Four different mix proportions were arrived at, comprising both conventional concrete and Alccofine-based concrete. In each formulation, a combination of normal river sand and M-sand was utilized. Results: Concrete with Alccofine exhibits superior mechanical properties, while M-sand incorporation minimally affects strength but reduces reliance on natural sand. GFRP-reinforced slabs display distinct brittle behaviour with significant deflections post-cracking, contrasting steel-reinforced slabs’ gradual, ductile failure. Discrepancies between experimental data and design recommendations underscore the need for guideline refinement. Conclusion: Alccofine and M-sand enhance concrete properties, but reinforcement type significantly influences slab behaviour. GFRP-reinforced slabs, though exhibiting lower values than steel, offer advantages in harsh environments, warranting further optimization.

The effect of Monascus purpureus extract (MPE) on probiotic lactic acid bacteria (LAB) was investigated to ascertain its application in fermented foods. Viable count of LAB was not affected after 24 hours of incubation in Man Rogosa Sharpe (MRS) broth containing MPE. The agar well-diffusion assay did not show any inhibition zone. The biotransformation of isoflavone glycosides by LAB in culture medium supplemented with MPE increased antioxidant activities. These data suggest that, nutritive and biological functionality of fermented foods can be improved by the use of MPE.

Human secretin is responsible for carrying a number of physiological functions including energy and water homeostasis, thus making secretin receptor a promising target for drug development. For GPCRs （G protein-coupled receptors）, radioactive ligands are usually used in conventional binding assays to characterize the binding affinities of the ligands. An alternative non-hazardous fluorescence based binding assay is lucrative over the radio-ligand assays. Here, we have developed a FRET （fluorescence resonance energy transfer） competitive binding assay for human secretin receptor. The receptor gene sequence is cloned in the SNAP （single nucleotide amplified polymorphisms） tag-plasmid and expressed in CHO （chinese hamster ovary）-k1 cells. Its expression and function is confirmed with immunofluorescence localization and receptor activation. The receptor and the ligand are labeled with fluorescent donor （Tb） and acceptor （Alexa488）. FRET signals are produced when the labeled ligand is bound to the receptor and the same drop when it is displaced by the test compounds. The saturation concentration of the receptor labeling is 100 nM, and the ligand kd value is 500 nM. At these concentrations, the IC50 of unlabeled secretin is 1.63 4- 3.55 nM. Additionally, few class-B ligands are screened and hold good correlation with traditional radio-ligand assay. Henceforth, this FRET binding assay can be efficiently used as a primary screening tool for peptide analogs.
