Investigation of h.AR2 antagonists as a potential strategy to modulate bovine leuk.cytes

Back.round Dairy cows experiencing k.tosis after calving suffer greater disease incidence and are at greater risk.of leaving th. h.rd. In vitro administration of beta-h.droxybutyric acid(Bh.A;th. primary blood k.tone) h.s inh.bitory effects on th. function of bovine leuk.cytes. Bh.A is a ligand of h.AR2 and th. activation of th.se receptors promotes an anti-inflammatory response wh.ch.may be related with.immunosuppression observed in transition dairy cattle. Th. objective of th.s study was to identify and test antagonists for h.AR2 in bovine immune cells cultured with.*** We observed expression of h.AR2 at th. protein level with.n lymph.cytes, monocytes, and granulocytes. Th. proportion of cells expressing h.AR2 tended to be greater in mid-lactation compared to early lactation cows;th. increase was a result of increased proportion of T and B cells expressing h.AR2. Stimulation of h.AR2 with.niacin or Bh.A promoted Ca^(2+) mobilization in neutroph.ls and mononuclear cells. Mononuclear cells treated with.Bh.A h.d diminish.d intracellular Ca^(2+) responses wh.n h.AR2 was k.ock.d down by si RNA silencing, indicating Ca^(2+) mobilization was mediated by h.AR2 signaling. Two candidate antagonists for h.AR2, synth.sized from niacin(NA-1 and NA-5), were tested;monocytes and neutroph.ls pre-treated with.NA-1 and NA-5 h.d reduced Ca^(2+) mobilization after incubation with.Bh.A. Furth.rmore, NA-5 but not NA-1 prevented Bh.A-associated reductions in cyclic *** We demonstrated th.t h.AR2 is present on bovine leuk.cytes and h.s greater expression later in lactation. We confirmed th.t Bh.A and niacin derived h.AR2 antagonists alter bovine leuk.cyte activity. Our results demonstrate th.t both.Bh.A and niacin affect bovine leuk.cyte Ca^(2+) mobilization in a h.AR2-dependent manner.

using Portable Gamma-Ray Spectrometry for Testing uranium Migration: A Case Study from th. Wadi El k.reim Alk.line Volcanics,Central Eastern Desert, Egypt

Th. 300±20 Ma anomalously radioactive trach.tes of Wadi El k.reim, central Eastern Desert, are a significant example of u-mineralization related to th. alk.line volcanics in Egypt. Extensive portable gamma-ray spectrometric data h.s been utilized to identify geological factors controlling uranium mobility in th. geological units along th. th.ee detailed study locations of k.b Al-Abyad, South.Wadi(W) Al-Tarafawy and W. Al-Fark.ah. th.ir eT h.eu ratios averaging around 4.1, 3.7 and 5.6 respectively. Quantitative analysis with.th. integration of mobility maps and geological studies suggest two systems controlling u-migration with.n th. geological units(confined system and unconfined system). In th. confined system, th. syngenetically formed u h.ve experienced mobility after leach.ng and are redistributed in th. presence of an incorporation carrier during transportation(probably as carbonate complexes). Th.n th. retardant for uranium is ach.eved by sorption or by coprecipitation with.th. aid of Fe oxy-h.droxide, and finally th. formation of immobile secondary u-bearing minerals tak.s place along a lith.geoch.mical trap. In contrast to th. confined system, th. unconfined one is basically lack.ng th. lith.geoch.mical trap wh.ch.influences th. final accumulation of u-bearing minerals. Th. radioactivity of th. trach.te rock. arises from th. radioactive minerals uranoph.ne and betauranoph.ne with.u-and/or Th.bearing minerals samarsk.te, Th.rich.REE silicates, monazite and allanite.

Th. auth.rs consider systems of th. form wh.re th. matrix A(u) is assumed to be strictly h.perbolic and with.th. property th.t th. integral curves of th. eigenvector fields are straigh. lines. For th.s class of systems one can define a natural Riemann solver, and h.nce a Godunov sch.me, wh.ch.generalize th. standard Riemann solver and Godunov sch.me for conservative systems. Th.s paper sh.ws convergence and L1 stability for th.s sch.me wh.n applied to data with.small total variation. Th. main step in th. proof is to estimate th. increase in th. total variation produced by th. sch.me due to quadratic coupling terms. using Duh.mel’s principle, th. problem is reduced to th. estimate of th. product of two Green k.rnels, representing probability densities of discrete random walk.. Th. total amount of coupling is th.n determined by th. expected number of crossings between two random walk. with.strictly different average speeds. Th.s provides a discrete analogue of th. arguments developed in [3,9] in connection with.continuous random processes.

Th. edge transport code SOLPS5.0 is used to model edge plasmas in th. experi- mental sh.ts on JT-60u and th. profiles of th. transverse particle and h.at transport coefficients D, Xe and Xi along th. outer midplane are obtained by fitting th. simulational results to th. experimental data in L-mode sh.t 39090 and h.mode sh.ts 37851, 37856. Th. modelling and fitting results sh.w th.t with.n th. pedestal region in h.mode sh.ts 37851 and 37856 th. radial particle transport coefficient D exh.bits a significant drop, but, for L-mode sh.t 39090, th. obvious drop in both.D and Xe was not found.

A multi-relaxation-time discrete Boltzmann model(DBM) with.split collision is proposed for both.subsonic and supersonic compressible reacting flows, wh.re ch.mical reactions tak. place among various components. Th. ph.sical model is based on a unified set of discrete Boltzmann equations th.t describes th. evolution of each.ch.mical species with.adjustable acceleration, specific h.at ratio, and Prandtl number. On th. righ.-h.nd side of discrete Boltzmann equations, th. collision,force, and reaction terms denote th. ch.nge rates of distribution functions due to self-and crosscollisions, external forces, and ch.mical reactions, respectively. Th. source terms can be calculated in th.ee ways, among wh.ch.th. matrix inversion meth.d possesses th. h.gh.st ph.sical accuracy and computational efficiency. Th.ough.Ch.pman-Ensk.g analysis, it is proved th.t th. DBM is consistent with.th. reactive Navier-Stok.s equations, Fick.s law and th. Stefan-Maxwell diffusion equation in th. h.drodynamic limit. Compared with.th. one-step-relaxation model, th. split collision model offers a detailed and precise description of h.drodynamic, th.rmodynamic, and ch.mical nonequilibrium effects. Finally, th. model is validated by six bench.ark., including multicomponent diffusion, mixture in th. force field, k.lvin-h.lmh.ltz instability, flame at constant pressure, opposing ch.mical reaction, and steady detonation.
