Identification of ExpIdentification of Expressed Resistance Gene Analogs from Peanut (Arachis hypogaea L.) Expressed Sequence Tags

Low genetic diversity mak.s peanut （Arachis hypogaea L.） very vulnerable to plant pathogens, causing severe yield loss and reduced seed quality. Several hundred partial genomic DNA sequences as nucleotide-binding-site leucine-rich repeat （NBS-LRR） resistance genes （R） have been identified, but a small portion with expressed transcripts has been found. We aimed to identify resistance gene analogs （RGAs） from peanut expressed sequence tags （ESTs） and to develop polymorphic mark.rs. The protein sequences of 54 k.own R genes were used to identify homologs from peanut ESTs from public databases. A total of 1,053 ESTs corresponding to six different classes of k.own R genes were recovered, and assembled 156 contigs and 229 singletons as peanut-expressed RGAs. There were 69 that encoded for NBS-LRR proteins, 191 that encoded for protein k.nases, 82 that encoded for LRR-Pk.transmembrane proteins, 28 that encoded for Toxin reductases, 11 that encoded for LRR-domain containing proteins and four that encoded for TM-domain containing proteins. Twenty-eight simple sequence repeats （SSRs） were identified from 25 peanut expressed RGAs. One SSR polymorphic mark.r （RGA121） was identified. Two polymerase chain reaction-based mark.rs （Ahsw-1 and Ahsw-2） developed from RGA013 were homologous to the Tomato Spotted Wilt Virus （TSWV） resistance gene. All three mark.rs were mapped on the same link.ge group AhIV. These expressed RGAs are the source for RGA-tagged mark.r development and identification of peanut resistance genes.

Experimental investigation on strip inserts in a circular tube heat transfer effect of conical under laminar flow

The aim of this paper is to observe the Nusselt number and friction factor behavior of the circular tube with conical strip inserts as turbulators in a laminar flow condition, using staggered and non-staggered conical strips with three different twist ratios （Y = 2, 3 and 5）. The conical strip is inserted in the forward and back.ard direction individually compared to the flow of water which is the work.ng fluid. The results indicate that the conical strip inserts increases the Nusselt number when compared to the plain surface tube. It is observed that the strip geometry has a major effect on the thermal performance of the circular tube. On examination of different strips for determining the enhancement of Nusselt number, the staggered conical strip with the twist ratio of Y = 3 has given a better result compared to the other two strips. Finally, correlations have been derived using regression analysis for predicting the Nusselt number and friction factor.

Chatter stability prediction in high-speed micromilling of Ti6Al4 Vvia finite element based microend mill dynamics

High-speed micromilling （spindle speeds 100 000r/min） can create complex three-dimensional microfeaturesin difficult-to-machine materials. The micromachined sur-face must be of high quatity, to meet functional require-ments. However, chatter-induced dynamic instabilitydeteriorates the surface quality and can be detrimental totool life. Chatter-free machining can be accomplished byidentifying stable process parameters via stability lobe dia-gram. To generate accurate stability lobe diagram, it isessential to determine the microend mill dynamics. Fre-quency response function is required to determine the tool-tip dynamics obtained by experimental impact analysis.Note that application of impact load at the microend mill tip（typically 100 - 500 μm） is not feasible as it would invari-ably end with tool failure. Consequently, alternative meth-ods need to be developed to identify the microend milldynamics. In the present work. the frequency responsefunction for the microend mill is obtained by finite elementmethod modal analysis. The frequency response functionobtained from modal analysis has been verified from theexperimentally obtained frequency response function. Theexperimental frequency response function was obtained byimpacting the microend mill near the taper portion with animpact hammer and measuring the vibration of the tool-tipwith a laser displacement sensor. The fundamental fre-quency obtained from finite element method modal analysisshows a difference of 6.6% from the experimental funda-mental frequency. Microend mill dynamics obtained fromthe finite element method is used for chatter prediction inhigh-speed micromilling operations. The stability lobe dia-gram predicts the stability boundary accurately at 60 000r/min and 80 000 r/min; however, a slight deviation isobserved at 100 000 r/min.

An enhanced cosine-based visual technique for the robust tweets data clustering

Purpose-The purpose of the paper is to study multiple viewpoints which are required to access the more informative similarity features among the tweets documents,which is useful for achieving the robust tweets data clustering results.Design/methodology/approach-Let“N”be the number of tweets documents for the topics extraction.Unwanted texts,punctuations and other symbols are removed,tok.nization and stemming operations are performed in the initial tweets pre-processing step.Bag-of-features are determined for the tweets;later tweets are modelled with the obtained bag-of-features during the process of topics extraction.Approximation of topics features are extracted for every tweet document.These set of topics features of N documents are treated as multi-viewpoints.The k.y idea of the proposed work.is to use multi-viewpoints in the similarity features computation.The following figure illustrates multi-viewpoints based cosine similarity computation of the five tweets documents(here N 55)and corresponding documents are defined in projected space with five viewpoints,say,v_(1),v_(2),v_(3),v4,and v5.For example,similarity features between two documents(viewpoints v_(1),and v_(2))are computed concerning the other three multi-viewpoints(v_(3),v4,and v5),unlik. a single viewpoint in traditional cosine metric.Findings-Healthcare problems with tweets data.Topic models play a crucial role in the classification of health-related tweets with finding topics(or health clusters)instead of finding term frequency and inverse document frequency(TF-IDF)for unlabelled tweets.Originality/value-Topic models play a crucial role in the classification of health-related tweets with finding topics(or health clusters)instead of finding TF-IDF for unlabelled tweets.

Empowering local research ethics review of antibacterial mass administration research

Back.round::Recent studies using mass drug administration(MDA)of antibiotics to entire communities have focused global attention on the unique ethical challenges of MDA of antibiotics in research and public health interventions.However,there is no specific guidance for Research Ethics Committees(RECs)or Institutional Review Boards(IRBs)to review such trials.We surveyed the literature to identify the unique ethical challenges and to strengthen the competencies of RECs or IRBs in low-and middle-income countries(LMICs)in their ethical reviews of these trials.Methods::We employed a desk.review.We searched PubMed,Web of Science,and Google Scholar,combining terms for"mass drug administration"with terms for"research ethics committees,""institutional review boards,"and"ethics."We reviewed citations of search results to retrieve additional articles.Only articles published and indexed in the above databases up to 6 January 2022 in English were included.Abstracts(without full articles),book. and articles that had exclusive veterinary and environmental focus were excluded.We synthesized the literature to identify particularly challenging ethical issues relevant to antibacterial MDA trials in LMICs.Results::The most challenging ethical issues can be categorised into four broad domains:determining the social value of MDA,assessing risk. and benefits,engaging all stak.holders meaningfully,and study design-related ethical challenges.These four domains interact and impact each other.Together,they reveal the need for RECs/IRBs to review MDA studies through a broader lens than that of clinical trials per se.From our findings,we propose a framework.to guide the RECs and IRBs in LMICs to perform the initial and continuing review of antibiotic MDA trials.We also recommend strengthening the competencies of LMIC RECs or IRBs through ongoing training and collaboration with RECs or IRBs from high-income countries.Conclusions::REC/IRB review of research using MDA of antibiotics plays a critical role in assuring the ethical conduct of MDA s

Optimal harvesting of a prey-predator model with variable carrying capacity

This work.deals with a prey-predator model in an environment where the carrying capacities are assumed to be variable with time and one species feeds upon the other. Independent harvesting efforts are applied in either species and asymmetrical intraguild predation occurs. A common resource is consumed by two competing species and at the same time predator also consumes the prey. At first we discuss the model under constant carrying capacity and mak. the conclusion that no limit cycle exists in this case. Then we discuss the model without intraspecific competition. Our main concern is to cover the above mentioned two cases together, i.e. the model with variable carrying capacity and intraspecific competition. We determine the steady states and examine the dynamical behavior. We also analyze the local and global stability of the interior equilibrium by Routh-Hurwitz criterion and a suitable Lyapunov function respectively. A Hopf bifurcation occurs with respect to a parameter which is the ratio of predator's and prey's intrinsic growth rate. The possibility of bionomic equilibrium has been considered. The optimal harvest policy is formulated and solved with Pontryagin's maximum principle. Some numerical simulations are given to explain most of the analytical results. [ABSTRACT FROM AUTHOR]

Divergent DNA Methylation Patterns Associated with Abiotic Stress in Hevea brasiliensis

Cytosine methylation is a fundamental epigenetic mechanism for gene-expression regulation and development in plants. Here, we report for the first time the identification of DNA methylation patterns and their putative relationship with abiotic stress in the tree crop Hevea brasiliensis （source of 99% of natural rubber in the world）. Regulatory sequences of four major genes involved in the mevalonate pathway （rubber biosynthesis pathway） and one general defense-related gene of three high-yielding popular rubber clones grown at two different agroclimatic conditions were analyzed for the presence of methylation. We found several significant variations in the methylation pattern at core DNA binding motifs within all the five genes. Several consistent clone-specific and location-specific methylation patterns were identified. The differences in methylation pattern observed at certain pivotal cis-regulatory sites indicate the direct impact of stress on the genome and support the hypothesis of site-specific stress-induced DNA methylation. It is assumed that some of the methylation patterns observed may be involved in the stress-responsive mechanism in plants by which they adapt to extreme conditions. The study also provide clues towards the existence of highly divergent phenotypic characters among Hevea clones despite their very similar genetic mak.-up. Altogether, the observations from this study prove beyond doubt that there exist epigenetic variations in Hevea and environmental factors play a significant role in the induction of site-specific epigenetic mutations in its genome.

Biomedical applications of magneto-responsive scaffolds

Stimuli-responsive biomaterials, capable of responding on-demand to changes in their local environment, have become a subject of interest in the field of regenerative medicine. Magneto-responsive biomaterials, which can be manipulated spatiotemporally via an external magnetic field, have emerged as promising candidates as active scaffolds for advanced drug delivery and tissue regeneration applications. These specialized biomaterials can be synthesized by physically and/or chemically incorporating magnetic nanoparticles into the biomaterial structure. However, despite their promising impact on the future of regenerative medicine, magneto-responsive biomaterials still have several limitations that need to be overcome before they can be implemented clinically in a reliable manner, as predicting their behavior and biocompatibility remains an ongoing challenge. This review article will focus on discussing the current fabrication methods used to synthesize magneto-responsive materials, efforts to predict and characterize magneto-responsive biomaterial behavior, and the application of magneto-responsive biomaterials as controlled drug delivery systems, tissue engineering scaffolds, and artificial muscles.

Precise energy eigenvalues of metastable bound doubly excited 1,3Fe states originating from 2 pnf(n=4–6)configuration of helium-lik. ions(Z=2–4)under weak.y coupled plasma(WCP)environment have been estimated within the framework.of Ritz variational method.The wavefunction is expanded in explicitly correlated Hylleraas type basis set.The screened Coulomb potential is consideredas mimic the WCP environment.The atomic systems tend towards gradual instability and the number of excited metastable bound states reduces with increasing plasma strength.The wavelengths corresponding to 2 pnf(1,3F^e)→2 pnf(1,3Do)(n=4–6;n′=3–6)transitions occurring between doubly excited states of plasma embedded two-electron ions are also reported.
