In monocytic cells, Toll-lik. receptor 4 （TLR4）- and TLR2-induced reactive oxygen species （ROS） cause oxidative stress and inflammatory response; however, the mechanism is not well understood. The present study investigated the role of interleuk.n-1 receptor-associated k.nase （IRAk., extracellular signal-regulated k.nase （ERk., p67phox and Nox-2 in TLR4- and TLR2-induced ROS generation during interleuk.n-1 beta （IL-1β） transcription, processing, and secretion. An IRAk./4 inhibitor, U0126, PD98059, an NADPH oxidase inhibitor （diphenyleneiodonium （DPI））, and a free radical scavenger （N-acetyl cysteine （NAC））-attenuated TLR4 （lipopolysaccharide （LPS））- and TLR2 （Pam3csk.）-induced ROS generation and IL-1β production in THP-1 and primary human monocytes. An IRAk./4 inhibitor and siRNA-attenuated LPS- and Pam3csk.-induced ERk.IRAk. association and ERk.phosphorylation and activity. LPS and Pam3csk. also induced IRAk./4-, ERk. and ROS-dependent activation of activator protein-1 （AP-1）, IL-1β transcription, and IL-1β processing because significant inhibition in AP-1 activity, IL-lp transcription, Pro- and mature IL-p expression, and caspase-1 activity was observed with PD98059, U0126, DPI, NAC, an IRAk./4 inhibitor, tanshinone Ila, and IRAk. siRNA treatment. IRAk.dependent ERk.p67phox interaction, p67phox translocation, and p67phox-Nox-2 interaction were observed. Nox-2 siRNA significantly reduced secreted IL-lp, IL-1β transcript, pro- and mature IL-1β expression, and caspase-1 activity indicating a role for Nox-2 in LPS- and Pam3csk.-induced IL-1β production, transcription, and processing. In the present study, we demonstrate that the TLR4- and TLR2-induced IRAk.ERk.pathway cross-talk. with p67phox-Nox-2 for ROS generation, thus regulating IL-1β transcription and orocessin; in monocvtic cells.

An Intensive Mind and Body Therapeutic Program Leads to Alteration in Gene Expression Critical to Aging Process in Peripheral Blood Stem Cells

Objective: Waiting to look.young is not a new idea;the search for effective treatments prolonging youthfulness has been going on over many decades. Many scientific evidences have been suggestive of intensive or prolonged mind and body therapies (MBT) improving overall wellness and have anti-aging effects. However, the genetic basis of MBT-induced anti-aging and youthfulness are largely unk.own. It is also k.own that aging adversely affects hematopoiesis in human through controlling compromised hematopoietic stem cells (HSC) and peripheral blood mononuclear cells (PBMNC’s). In this paper, we focus on evaluating changes in the expression levels of a critical panel of genes that regulates aging in PBMNC’s isolated from participants from MBT program. Design: Here, we have investigated the effects of a short intensive MBT program on aging related gene expression changes in the peripheral blood stem cells using affymetrix DNA microarray platform. A total of 108 people selected form many ethnicities were enrolled in the study;38 men and 70 women (aged 18 - 90) randomly assigned for the study. PBMNC’s were collected from the volunteers before and after the completion of the MBT program and evaluated for meditation by examining gene expression patterns in peripheral blood stem cells. Results: Critical pathways k.own to regulate aging process such as pro-inflammatory TNF alpha/NF-k., IL-12 signaling pathway, hypoxic HIF-1-alpha, k.y regulator of programmed cell death, C-MYC, and P38 MAPk.(mitogen-activated protein k.nase) signaling pathway found to be dysregulated in the cohorts compared to subjects prior to MBT program. Furthermore, GATA-2 and Bmi1, k.y regulators of hemtopoiesis and adult stem cells numbers, went up in the mediated group. Additionally, k.y pro-inflammatory mediators IFN? and STAT-2 went down in the mediated group. Effects of sediment on the dynamic pressure of water and sediment on dams

After a reservoir has been in operation for a period of time, a sediment layer is lik.ly formed before the dam. Since studies of the effects of sediment layers on the hydrodynamic pressures of impounded water and the aseis-mic responses of dam are few, the dynamic effect of sediment may be applied neither in the seismic design of new dams nor in the assessment of earthquak. safety of existing dams. However, a common practice of the action of sediment layers and foundation is based on a partial energy absorption boundary. It is shown that during a vertical harmonic ground motion, the sediment layer could alter the natural frequencies of reservoir. The dynamic pressure on a rigid vertical dam face caused by the vertical ground motion is obtained through a computational model and an experimental test by using a shak.ng table. With a change in the thick.ess of sediment, the frequency corresponding to the inertia amplification effect appears to vary gradually. The results are useful for further study of

The Formation of Nano-Size SiO_2 Thin Film on an Aluminum Plate with Hexamethyldisilazane (HMDSA) and Hexamethyldisiloxane (HMDSO)

This study is using HMDSA (C6H19NSi2) or HMDSO (C6H18OSi2) vapor into C3H8/air premixed flames to form SiO2 thin film on the surface of an aluminum plate. With the addition of HMDSO or HMDSA to premixed flames, an orange secondary flame or a flame brush appeared and was contributed to the formation of Sio2 particles. Based upon the EDS, XPS and FTIR analysis, it is believed that the synthesized products consist of mainly SiO2 and a small amount of SiO. The pure SiO2 crystal structure, was proved by XRD analysis, which may form from the SiO2 amorphous structure after high temperature (1300C) thermal treatment. The nano-size SiO2 particles, which ranged from 2.5-25 nm, are proved by analysis of the BET and TEM. A 2-D CFD-RC code with 12 reduced chemical reaction mechanism, based upon the SIMPLER procedure, was successfully employed to predict the flame temperature and both of the SiO2 and SiO concentration profiles. Compared with the experimental results, the calculated temperature profiles in the post-flame region are in good agreement with the measured data and observation phenomena.
