Two-photon live imaging of direct glia-to-neuron conversion in the mouse cortex

Over the past decade,a growing number of studies have reported transcription factor-based in situ reprogramming that can directly conve rt endogenous glial cells into functional neurons as an alternative approach for n euro regeneration in the adult mammalian central ne rvous *** r,many questions remain regarding how a terminally differentiated glial cell can transform into a delicate neuron that forms part of the intricate brain *** addition,concerns have recently been raised around the absence of astrocyte-to-neuron conversion in astrocytic lineage-tra cing *** this study,we employed repetitive two-photon imaging to continuously capture the in situ astrocyte-to-neuron conversion process following ecto pic expression of the neural transcription factor NeuroD1 in both prolife rating reactive astrocytes and lineage-tra ced astrocytes in the mouse ***-lapse imaging over several wee ks revealed the ste p-by-step transition from a typical astrocyte with numero us short,tapered branches to a typical neuro n with a few long neurites and dynamic growth cones that actively explored the local *** addition,these lineage-converting cells were able to migrate ra dially or to ngentially to relocate to suitable ***,two-photon Ca2+imaging and patch-clamp recordings confirmed that the newly generated neuro ns exhibited synchronous calcium signals,repetitive action potentials,and spontaneous synaptic responses,suggesting that they had made functional synaptic connections within local neural *** conclusion,we directly visualized the step-by-step lineage conversion process from astrocytes to functional neurons in vivo and unambiguously demonstrated that adult mammalian brains are highly plastic with respect to their potential for neuro regeneration and neural circuit reconstruction.

The joint European Space Agency and Chinese Academy of Sciences Solar wind magnetosphere Ionosphere Link Explorer(SMILE)mission will explore global dynamics of the magnetosphere under varying solar wind and interplanetary magnetic field conditions,and simultaneously monitor the auroral response of the Northern Hemisphere *** these large-scale responses with medium and fine-scale measurements at a variety of cadences by additional ground-based and space-based instruments will enable a much greater scientific impact beyond the original goals of the SMILE ***,we describe current community efforts to prepare for SMILE,and the benefits and context various experiments that have explicitly expressed support for SMILE can offer.A dedicated group of international scientists representing many different experiment types and geographical locations,the Ground-based and Additional Science Working Group,is facilitating these *** include constructing an online SMILE Data Fusion Facility,the discussion of particular or special modes for experiments such as coherent and incoherent scatter radar,and the consideration of particular observing strategies and spacecraft *** anticipate growing interest and community engagement with the SMILE mission,and we welcome novel ideas and insights from the solar-terrestrial community.

Pregnancy Benefit of Acupuncture on in vitro Fertilization:A Systematic review and Meta-Analysis

rBM46 is a germ cell-specific rNA-binding protein required for gametogenesis,but the targets and molecular functions of rBM46 remain ***,we demonstrate that rBM46 binds at specific motifs in the 3'UTrs of mrNAs encoding multiple meiotic cohesin subunits and show that rBM46 is required for normal synaptonemal complex formation during meiosis *** a recently reported,high-resolution technique known as LACE-seq and working with low-input cells,we profiled the targets of rBM46 at single-nucleotide resolution in leptotene and zygotene stage *** found that rBM46 preferentially binds target mrNAs containing GCCUAU/GUUCGA motifs in their 3'UTrs *** rbm46 knockout mice,the rBM46-target cohesin subunits displayed unaltered mrNA levels but had reduced translation,resulting in the failed assembly of axial elements,synapsis disruption,and meiotic *** study thus provides mechanistic insights into the molecular functions of rBM46 in gametogenesis and illustrates the power of LACE-seq for investigations of rNA-binding protein functions when working with low-abundance input materials.

In December 2019,an outbreak of pneumonia,which was named COVID-2019,emerged as a global health *** worldwide are engaged in attempts to elucidate the transmission and pathogenic mechanisms of the causative ***-19 was declared a pandemic by the World Health Organization in march 2020,making it critical to track and review the state of research on COVID-19 to provide guidance for further ***,bibliometric and knowledge mapping analyses of studies on COVID-19 were performed,including more than 1,500 papers on COVID-19 available in the PubMed and China National knowledge Infrastructure databases from January 1,2020 to march 8,*** this review,we found that because of the rapid response of researchers worldwide,the number of COVID-19-related publications showed a high growth trend in the first 10 days of February;among these,the largest number of studies originated in China,the country most affected by pandemic in its early *** findings revealed that the epidemic situation and data accessibility of different research teams have caused obvious difference in emphases of the pub***,there was an unprecedented level of close cooperation and information sharing within the global scientific community relative to previous coronavirus *** combed and drew the knowledge map of the SArS-CoV-2 literature,explored early status of research on etiology,pathology,epidemiology,treatment,prevention,and control,and discussed knowledge gaps that remain to be urgently *** perspectives on treatment,prevention,and control are also presented to provide fundamental references for current and future coronavirus research.

Ciliates are core components of the structure of and function of aquatic microbial food *** play an essential role in the energy flow and material circulation within aquatic ***,studies on the taxonomy and biodiversity of freshwater ciliates,especially those in wetlands in China are *** address this issue,a project to investigate the freshwater ciliates of the Lake Weishan Wetland,Shandong Province,commenced in ***,we summarize our findings to date on the diversity of ciliates.A total of 187 ciliate species have been found,94 of which are identified to species-level,87 to genus-level,and six to *** species show a high morphological diversity and represent five classes,i.e.,Heterotrichea,Litostomatea,Prostomatea,Oligohymenophorea,and *** largest number of species documented are oligohymenophoreans.A comprehensive database of these ciliates,including morphological data,gene sequences,microscope slide specimens and a DNA bank,has been *** the present study,we provide an annotated checklist of retrieved ciliates as well as information on the sequences of published *** of these species are recorded in China for the first time and more than 20%are tentatively identified as new to ***,an investigation of environmental DNA revealed that the ciliate species diversity in Lake Weishan Wetland is higher than previously supposed.
