Diabetic retinopathy(DR)is a form of *** oxidative stress in the mitochondria and cell membranes decreases ischemic injury and end-organ damage to the *** approaches are needed,which reduce the risk and improve the outcomes of DR while complementing current therapeutic ***(Hcy)elevation and oxidative stress are potential therapeutic targets in *** genetic polymorphisms such as those of methylenetetrahydrofolate reductase(MTHFR),increase Hcy and DR risk and *** with DR have high incidences of deficiencies of crucial vitamins,minerals,and related compounds,which also lead to elevation of Hcy and oxidative *** the effects of the MTHFR polymorphism and addressing comorbid deficiencies and insufficiencies reduce the impact and severity of the *** approach provides safe and simple strategies that support conventional care and improve *** vitamin co-factor availability also impairs the release of neurotrophic and neuroprotective growth ***,this accounts for variability in presentation and response of DR to conventional ***,there are straightforward recommendations for addressing these issues and supporting traditional treatment *** have reviewed the literature for nutritional interventions that support conventional therapies to reduce disease risk and *** combinations of vitamins b1,b2,b6,L-methylfolate,methylcobalamin(b12),C,D,natural vitamin E complex,lutein,zeaxanthin,alpha-lipoic acid,and n-acetylcysteine are identified for protecting the retina and *** medical foods have been successfully used as therapy for *** based on this review and our clinical experience are developed for clinicians to use to support conventional therapy for *** from both type 1 diabetes mellitus(T1DM)and type 2 diabetes mellitus(T2DM)have similar retinal findings and responses to nutritional therapies.

Nitrogen Constrains the Growth of Late Summer Cyanobacterial blooms in Lake Erie

Phosphorus (P) is generally considered to be the main limiting nutrient to freshwater phytoplankton productivity. However, recent research is drawing attention to the importance of nitrogen (N) in freshwater eutrophication and N often constrains growth of cyanobacteria in small lakes. In this study we determined phytoplankton nutrient limitation in a large lake, Lake Erie during two growing seasons. During 2010 and 2011, nutrient enrichment bioassays (+P, +N and, +P and N) were conducted monthly from June through September with water collected in Maumee bay (site Mb18) and in the center of the western basin (site WbC). Nutrient concentrations were monitored every other week. At Mb18, total P concentration was often >3 mmol/L and nitrate concentration decreased from >250 mmol/L in early summer to mmol/L in late summer. Nitrogen and P levels were about five-fold less at WbC. bioassays indicated that phytoplankton nutrient limitation varied in summer, spatially, and even among phytoplankton groups. For site Mb18, +P increased chlorophyll concentration in one of the eight bioassays, indicating that P did not typically limit production. For site WbC, +P increased chlorophyll concentration in six of the eight bioassays. As a result of very low ambient nitrate concentration (mmol/L) in late summer, +N (without P) increased chlorophyll concentration, suggesting symptoms of N-limitation. The N-fixing cyanobacterium Anabaena became dominant following N-limitation. This study highlights the need to reduce P loading to restore water quality. Furthermore, due to low nitrate concentration, the severity of the cyanobacterial blooms could be worse if not for N-limitation in western Lake Erie.

Development of gemcitabine-resistant patient-derived xenograft models of pancreatic ductal adenocarcinoma

Intrinsic determinants of optic nerve regeneration

Objective To review the functions of these intracellular signals in their regulation of retinal ganglion cell （RGC） axon regeneration. Data sources Relevant articles published in English or Chinese from 1970 to present were selected from PubMed. Searches were made using the terms ＂intrinsic determinants, axon regeneration, RGC, optic nerve regeneration, and central nervous system axon regeneration.＂ Study selection Articles studying the mechanisms controlling RGC and central nervous system （CNS） axon regeneration were reviewed. Articles focusing on the intrinsic determinants of axon regeneration were selected. Results Like other CNS neurons of mammals, RGCs undergo a developmental loss in their ability to grow axons as they mature, which is a critical contributing factor to the failure of nerve regeneration and repair after injury. This growth failure can be attributed, at least in part, by the induction of molecular programs preventing cellular overgrowth and termination of axonal growth upon maturation. Key intracellular signals and transcription factors, including b cell lymphoma/leukemia 2, cyclic adenine monophosphate, mammalian target of rapamycin, and Kr^Jppel-like transcription factors, have been identified to play central roles in this process. Conclusions Intense effort and substantial progress have been made to identify the various intrinsic growth pathways that regulate RGC axon regeneration. More work is needed to elucidate the mechanisms of and the interrelationship between the actions of these factors and to successfully achieve regeneration and repair of the severed RGC axons.

Predictive Value of Clinical and Laboratory Parameters for Overt bone Marrow Metastasis in Patients with Solid Malignancy

bone marrow (bM) metastases are known to worsen the outcome in patients with malignancies and alter the choice of the treatment regimen. In an attempt to identify routine parameters for the prediction of bM metastasis we evaluated the clinical and laboratory data of 103 patients with metastatic solid tumors. We also assessed which of these factors appeared to influence survival. Three laboratory abnormalities were found to be predictive of metastasis on bM biopsy: thrombocytopenia 9/L OR 14.4;p = 0.02;hypocellular or dry tap aspirate OR 7.0;p = 0.02 and high serum alkaline phosphatase OR 6.5;p = 0.03. Three additional factors were found to be associated with deceased survival. Thrombocytopenia was associated with decreased survival from a median of 0.8 months to 4.1 months, p by bone marrow examination was associated with reduced survival from 1.8 months vs. 4.0 months, p = 0.03 in individuals where bM metastases were not the presenting feature of malignancies. Use of these simple clinical parameters may help in the early diagnosis of bone marrow metastases and improve clinical approach.

Taxonomic and metabolic shifts in the Coorong bacterial metagenome driven by salinity and external inputs

The Coorong estuary lies at the terminus of Australia's largest river system, the Murray-Darling; both are strongly influenced by human activities; including farming and extensive flow modification. Metagenomic approaches were used to determine the planktonic bacterial community composition and potential metabolic function at two extremes in the Coorong, the river mouth which exhibits marine-like salinity, and the hypersaline upper-reaches of the estuary. Significant shifts in taxa and metabolic function were seen between the two sites. The river mouth exhibited an increase in abundance of R hodobacteriaceae and Alteromonadaceae; families readily able to adapt to change in nutrient conditions; and the potentially pathogenic families b rucellaceae, Enterobacteriaceae and Vibrionaceae. Metabolisms over-represented include motility and chemotaxis, RNA metabolism and membrane transport, all of which are involved in actively searching for and obtaining nutrients. Also over-represented were metabolisms involved in population succession and stress response. An over-representation of taxa and metabolisms indicative of environmental change is reflective of anthropogenically af fected riverine input. In the hypersaline upper reaches of the estuary, the halophilic family Ectothiorhodospiraceae was over-represented, as were the families Flavobacteriaceae, Cytophagaceae and Nocardioidaceae, members of which are able to survive over a wide salinity range. Metabolisms over-represented here were reflective of increased bacterial growth, characteristic of hypersaline environments, and included DNA metabolism, nucleotide and nucleoside synthesis and cell cycle. Coorong metagenomes clustered taxonomically and metabolically with other planktonic metagenomes, but remained an outlier of this group with only 71% and 84% similarity, respectively.
