Second-generation direct-acting-antiviral hepatitis C virus treatment: Efficacy, safety, and predictors of SVR12

AIm To gather data on the antiviral efficacy and safety of second generation direct acting antiviral(DAA) treatment with respect to sustained virological response(SVR) 12 wk after conclusion of treatment, and to determine predictors of SVR12 in this *** Two hundred and sixty patients treated with SOF combination partners PR(n = 51), R(n = 10), SmV(n = 30), DCV(n = 81), LDV(n = 73), or 3D(n = 15).144/260 were pre-treated, 89/260 had liver cirrhosis, 56/260 had portal hypertension with platelets mELD score ≥ 10 and 17/260 were postliver transplantation patients. 194/260 had HCV GT1, 44/260 HCV *** Two hundred and forty/256(93.7%) patients achieved SVR12(m ITT); 4/260 were lost to follow-up. SVR12 rates for subgroups were: 92% for SOF/DCV, 93% for each SOF/SmV, SOF/PR, 94% for SOF/LDV, 100% for 3D, 94% for pretreated, 87% for liver cirrhosis, 82% for patients with platelets m relapse, 6 prematurely discontinued treatment, of which 4 died. Negative predictors of SVR12 were a platelet count mELD score ≥ 10(P ment GT3 had significantly lower SVR rates than GT1(P = 0.016). Side effects were mild. CONCLUSION Excellent SVR12 rates and the favorable side-effect profile of DAA-combination therapy can be well translated into "real-world". Patients with advanced liver disease, signs of portal hypertension, especially with platelets me comparatively higher rates of virological failure.

AIm:To search for further immunodominant peptides of the pyruvate dehydrogenase complex E2-component (PDC-E2) recognized by antimitochondrial antibodies (AmA) in primary biliary cirrhosis (PBC). mETHODS:Sera from 95 patients with PBC were tested by enzyme-linked immunosorbent assay against 33 synthetic overlapping peptides (25 amino acids; aa) covering the entire length of the E2-subunit of PDC-E2. Furthermore,the inner lipoyl peptide 167-184 was used in an unlip oylated and a lipoylated form as well as coupled to ovalbumin. Sera from 11 AmA negative/ANA posit ive PBC patients,63 patients with other liver disorders and 22 healthy blood donors served as ***:Of the 95 PBC-sera,74% reacted with the peptide 475-499 and 58% with the pept ide 407-431 located within the catalytic domain of PDC-E2. Patients with other disorders or healthy controls were positive in only up to 18%. Antibodies to the unlipoylatedand lip oylated pept ide 167-184 within the inner lipoyl domain were found in only 5% and 11% of the PBC sera,respectively; using ovalbumin-coupled peptides,the incidence increased up to 57% (unlipoylated form). CONCLUSION:Peptides within the catalytic site of PDC-E2 rather than the previously reported lipoyl binding peptide 167-184 may represent major immunodomin ant epitopes recognized by AmA in PBC.

Background:Nitrogen(N)deposition alters litter decomposition and soil carbon(C)sequestration by influencing the microbial community and its enzyme *** atmospheric N deposition comprises of inorganic N(IN)and organic N(ON)***,most studies have focused on IN and its effect on soil C cycling,whereas the effect of ON on microbial enzyme activity is poorly *** we studied the effects of different forms of externally supplied N on soil enzyme activities related to decomposition in a temperate *** nitrate was chosen as IN source,whereas urea and glycine were chosen as ON *** ratios of IN to ON(Control,10:0,7:3,5:5,3:7,and 0:10)were mixed with equal total amounts of N and then used to fertilize the grassland soils for 6 ***:Our results show that IN deposition inhibited lignin-degrading enzyme activity,such as phenol oxidase(POX)and peroxidase(PER),which may restrain decomposition and thus induce accumulation of recalcitrant organic C in grassland *** contrast,deposition of ON and mixed ON and IN enhanced most of the C-degrading enzyme activities,which may promote the organic matter decomposition in grassland *** addition,theβ-N-acetyl-glucosaminidase(NAG)activity was remarkably stimulated by fertilization with both IN and ON,maybe because of the elevated N availability and the lack of N limitation after long-term N fertilization at the grassland ***,differences in soil pH,soil dissolved organic carbon(DOC),and microbial biomass partially explained the differential effects on soil enzyme activity under different forms of N ***:Our results emphasize the importance of organic N deposition in controlling soil processes,which are regulated by microbial enzyme activities,and may consequently change the ecological effect of N ***,more ON deposition may promote the decomposition of soil organic matter thus converting C sequestration in grassland soils into a

Periodic noble metal nanoparticles offer a wide spectrum of applications including chemical and biological sensors,optical devices,and model catalysts due to their extraordinary *** sensing purposes and catalytic studies,substrates made of glass or fused-silica are normally required as supports,without the use of metallic adhesion ***,precise patterning of such uniform arrays of silica-supported noble metal nanoparticles,especially at sub-100 nm in diameter,is challenging without adhesion *** this paper,we report a robust method to large-scale fabricate highly ordered sub-20 nm noble metal nanoparticles,i.e.,gold and platinum,supported on silica substrates without adhesion layers,combining displacement Talbot lithography(DTL)with dry-etching *** photoresist nanocolumns at diameters of~110 nm are patterned on metal-coated oxidized silicon wafers using DTL,and subsequently transferred at a 1:1 ratio into anti-reflection layer coating(BARC)nanocolumns with the formation of nano-sharp tips,using nitrogen plasma *** BARC nanocolumns are then used as a mask for etching the deposited metal layer using inclined argon ion-beam *** find that increasing the etching time results in coneshaped silica features with metal nanoparticles on the tips at diameters ranging from 100 nm to sub-30 nm,over large areas of 3×3 cm^(2).moreover,subsequent annealing these sub-30 nm metal nanoparticle arrays at high-temperature results in sub-20 nm metal nanoparticle arrays with~10^(10) uniform particles.

We report a robust fabrication method for patterning freestanding Pt nanowires for use as thermal anemometry probes for small-scale turbulence *** e-beam lithography,high aspect ratio Pt nanowires(~300 nm width,~70µm length,~100 nm thickness)were patterned on the surface of oxidized silicon(Si)*** wet etching processes with dry etching processes,these Pt nanowires were successfully released,rendering them freestanding between two silicon dioxide(SiO2)beams supported on Si ***,the unique design of the bridge holding the device allowed gentle release of the device without damaging the Pt *** total fabrication time was minimized by restricting the use of e-beam lithography to the patterning of the Pt nanowires,while standard photolithography was employed for other parts of the *** demonstrate that the fabricated sensors are suitable for turbulence measurements when operated in constant-current mode.A robust calibration between the output voltage and the fluid velocity was established over the velocity range from 0.5 to 5 m s−1 in a SF6 atmosphere at a pressure of 2 bar and a temperature of 21°*** sensing signal from the nanowires showed negligible drift over a period of several ***,we confirmed that the nanowires can withstand high dynamic pressures by testing them in air at room temperature for velocities up to 55 m s−1.

Seed induces and promotes the crystallization of calcium phosphate, and acts as carrier of the recovered phosphorus （P）. In order to select suitable seed for P recovery from wastewater, three seeds including Apatite （AP）, Juraperle （JP） and phosphate-modified Juraperle （m-JP） were tested and compared. Batch and fixed-bed column experiments of seeded crystallization of calcium phosphate were undertaken by using synthetic wastewater with 10 mg/L P phosphate. It shows that AP has bad enduring property in the crystallization process, while JP has better performance for multiple uses, and m-JP is a hopeful seed for P recovery by crystallization of calcium phosphate.
