Cytosolic HSP90 Cochaperones HOP and FKBP Interact with Freshly Synthesized Chloroplast Preproteins of Arabidopsis

Most chloroplast and mitochondrial proteins are synthesized in the cytosol of the plant cell and have to be imported into the organelles post-translationally. Molecular chaperones play an important role in preventing protein aggregation of freshly translated preproteins and assist in maintaining the preproteins in an import competent state. Pre- proteins can associate with HSP70, HSP90, and 14-3-3 proteins in the cytosol. In this study, we analyzed a large set of wheat germ-translated chloroplast preproteins with respect to their chaperone binding. Our results demonstrate that the formation of distinct 14-3-3 or HSP90 containing preprotein complexes is a common feature in post-translational protein transport in addition to preproteins that seem to interact solely with HSP70. We were able to identify a diverse and extensive class of preproteins as HSP90 substrates, thus providing a tool for the investigation of HSP90 client protein association. The analyses of chimeric HSP90 and 14-3-3 binding preproteins with exchanged transit peptides indicate an involvement of both the transit peptide and the mature part of the proteins, in HSP90 binding. We identified two partner components of the HSP90 cycle, which were present in the preprotein containing high-molecular-weight complexes, the HSP70/HSP90 organizing protein HOP, as well as the immunophilin FKBP73. The results establish chloroplast preproteins as a general class of HSP90 client proteins in plants using HOP and FKBP as novel cochaperones.

Preprotein Import into Chloroplasts via the Toc and Tic Complexes Is Regulated by Redox Signals in Pisum sativum

The import of nuclear-encoded preproteins is necessary to maintain chloroplast function. The recognition and transfer of most precursor proteins across the chloroplast envelopes are facilitated by two membrane-inserted protein complexes, the translocons of the chloroplast outer and inner envelope （Toc and Tic complexes, respectively）. Several signals have been invoked to regulate the import of preproteins. In our study, we were interested in redox-based import regulation mediated by two signals： regulation based on thiols and on the metabolic NADP＋/NADPH ratio. We sought to identify the proteins participating in the regulation of these transport pathways and to characterize the preprotein subgroups whose import is redox-dependent. Our results provide evidence that the formation and reduction of disulfide bridges in the Toc receptors and Toc translocation channel have a strong influence on import yield of all tested preproteins that depend on the Toc complex for translocation. Furthermore, the metabolic NADP＋/NADPH ratio influences not only the composition of the Tic complex, but also the import efficiency of most, but not all, preproteins tested. Thus, several Tic subcomplexes appear to participate in the translocation of different preprotein subgroups, and the redox-active compo- nents of these complexes likely play a role in regulating transport.

ATP synthases in chloroplasts （cpATPase） and mitochondria （mtATPase） are responsible for ATP production during photosynthesis and oxidative phosphorylation, respectively. Both enzymes consist of two multi- subunit complexes, the membrane-bound coupling factor O and the soluble coupling factor 1. During cpATPase biosynthesis, several accessory factors facilitate subunit production and orchestrate complex assembly. Here, we describe a new auxiliary protein in Arabidopsis thaliana, which is required for cpATPase accumulation. AtCGLD11 （CONSERVED IN THE GREEN LINEAGE AND DIATOMS 11） is a protein without any known functional domain and shows dual localization to chloroplasts and mitochondria. Loss of AtCGLDll function results in reduced levels of cpATPase and impaired photosynthetic performance with lower rates of ATP synthesis. In yeast two-hybrid experiments, AtCGLD11 interacts with the 13 subunits of the cpATPase and mtATPase. Our results suggest that AtCGLD11 functions in F1 assembly during cpATPase biogenesis, while its role in mtATPase biosynthesis may not, or not yet, be essential.

Nucleus-Encoded Light-Harvesting Chlorophyll a/b Proteins are Imported Normally into Chlorophyll b-Free Chloroplasts of Arabidopsis

Chloroplast-located proteins which are encoded by the nuclear genome have to be imported from the cytosol into the organelle in a posttranslational manner. Among these nuclear-encoded chloroplast proteins are the light- harvesting chlorophyll a/b-binding proteins （LHCPs）. After translation in the cytosol, precursor proteins of LHCPs are imported via the TOC/TIC translocase, processed to their mature size to insert into thylakoid membranes where they recruit chlorophylls a and b to form pigment-protein complexes. The translocation of proteins is a highly regulated process which employs several regulators. To analyze whether CAO （chlorophyll a oxigenase） which converts chlorophyll a to chlorophyll b at the inner chloroplast membrane, is one of these regulators, we performed import reactions utilizing a homozygous loss-of-function mutant （cao-1）. We imported in vitro translated and 35S-labeled precursor proteins of light- harvesting proteins of photosystem II LHCB1, LHCB4, and LHCB5 into chloroplasts isolated from cao-1 and show that import of precursor proteins and their processing to mature forms are not impaired in the mutant. Therefore, regulation of the import machinery cannot be responsible for the decreased steady-state levels of light-harvesting complex （LHC） proteins. Regulation does not take place at the transcriptional level either, because Lhcb mRNAs are not down-regulated. Additionally, reduced steady-state levels of LHCPs also do not occur due to posttranslational turnover of non-functional LHCPs in chloroplasts. Taken together, our data show that plants in the absence of CAO and therefore devoid of chlorophyll b are not influenced in their import behavior of LHC proteins.

Decreasing nutrient concentrations in soils and trees with increasing elevation across a treelineecotone in Rolwaling Himal, Nepal

At a global scale, tree growth in alpine treeline ecotones is limited by low temperatures. At a local scale, however, tree growth at its upper limit depends on multiple interactions of influencing factors and mechanisms. The aim of our research was to understand local scale effects of soil properties and nutrient cycling on tree growth limitation, and their interactions with other abiotic and biotic factors in a near-natural Himalayan treeline ecotone. soil samples of different soil horizons, litter, decomposition layers, and foliage samples of standing biomass were collected in four altitudinal zones along three slopes, and were analysed for exchangeable cations and nutrient concentrations, respectively. Additionally, soil and air temperature, soil moisture, precipitation, and tree physiognomy patterns were evaluated. Both soil nutrients and foliar macronutrient concentrations of nitrogen(N), magnesium(Mg), potassium(K), and foliar phosphorus(P) decrease significantly with elevation. Foliar manganese(Mn) concentrations, bycontrast, are extraordinarily high at high elevation sites. Potential constraining factors on tree growth were identified using multivariate statistical approaches. We propose that tree growth, treeline position and vegetation composition are affected by nutrient limitation, which in turn, is governed by low soil temperatures and influenced by soil moisture conditions.

Repeated and random components in Oklahoma's monthly precipitation record

Precipitation across Oklahoma exhibits a high degree of spatial and temporal variability and creates numerous water resources management challenges. Risk-based decision making in agricultural and water resources management may find value in knowing the extent to which historical precipitation can provide guidance for inferring expectations of future monthly precipitation. A 123-year long monthly precipitation record from the central Oklahoma climate division was evaluated, in a proof-of-concept analysis, to establish whether a simple monthly precipitation decomposition into repeatable (expected) and random (unexpected) components could identify the odds of encountering repeatable or random monthly precipitation in the future. The metric for identifying precipitation kind was based on it falling within or outside of a predetermined range, defined by the relative difference between random and a percentage of repeatable precipitation. For climate conditions in central Oklahoma, the odds of future precipitation being repeatable were between 18% and 33%, depending on the month of the year. The corresponding odds of precipitation being random were 67–82%. Thus, management decisions that rely solely on historical precipitation records to anticipate future monthly precipitation have a low probability of success in central Oklahoma.

Implications of tree species–environment relationships for the responsiveness of Himalayan krummholz treelines to climate change

Climate warming is expected to advance treelines to higher elevations. However, empirical studies in diverse mountain ranges give evidence of both advancing alpine treelines as well as rather insignificant responses. In this context, we aim at investigating the sensitivity and responsiveness of the near-natural treeline ecotone in Rolwaling Himal, Nepal, to climate warming. We analysed population densities of tree species along the treeline ecotone from closed forest stands via the krummholz belt to alpine dwarf shrub heaths （3700-4200 m） at 50 plots in 2013 and 2014. We quantified species - environment relationships, i.e. the change of environmental conditions （e.g., nutrient and thermal deficits, plant interactions） across the ecotone by means of redundancy analyses, variation partitioning and distance-based Moran＇s eigenvector maps. In particular, we focus on explaining the high competitiveness of Rhododendron campanulatum forming a dense krummholz belt and on the implications for the responsiveness of Himalayan krummholz treelines to climate change. Results indicate that treeline trees in the ecotone show species-specific responses to the influence of environmental parameters, and that juvenile and adult tree responses are modulated by environmental constraints in differing intensity. Moreover, the species - environment relationships suggest that the investigated krummholz belt will largely prevent the upward migration of other tree constrain the future response species and thus of Himalayan krummholz treelines to climate warming.

Tryptophan-derived, indolic metabolites possess diverse functions in Arabidopsis innate immunity to microbial pathogen infection. Hers, we investigate the functional role and regulatory characteristics of indolic metabolism in Arabidopsis systemic acquired resistance （SAR） triggered by the bacterial pathogen Pseudomonas syringae. Indolic metabolism is broadly activated in both P. syringae-inoculated and distant, non-inoculated leaves. At inoculation sites, camalexin, indol-3-ylmethylamine （13A）, and indole-3-carboxylic acid （ICA） are the major accumulating compounds. Camalexin accumulation is positively affected by MYB122, and the cytochrome P450 genes CYP81F1 and CYP81F2. Local 13A production, by contrast, occurs via indole glucosinolate breakdown by PEN2- dependent and independent pathways. Moreover, exogenous application of the defense hormone salicylic acid stimulates 13A generation at the expense of its precursor indol-3-ylmethylglucosinolate （13M）, and the SAR regulator pipecolic acid primes plants for enhanced P. syringae-induced activation of distinct branches of indolic metabolism. In uninfected systemic tissue, the metabolic response is more specific and associated with enhanced levels of the indolics 13A, ICA, and indole-3-carbaldehyde （ICC）. Systemic indole accumulation fully depends on functional CYP79B2/3, PEN2, and MYB34/51/122, and requires functional SAR signaling. Genetic analyses suggest that systemi- cally elevated indoles are dispensable for SAR and associated systemic increases of salicylic acid. However, soil-grown but not hydroponically -cultivated cyp79b2/3 and pen2 plants, both defective in indolic secondary metabolism, exhibit pre-induced immunity, which abrogates their intrinsic ability to induce SAR.

Groundwater is the main source of water in the semi-arid Calera watershed, located in the State of Zacatecas, Mexico. Due to increasing population, rapid industrial growth, and increased irrigation to meet growing food demand, groundwater extraction in the Calera watershed are exceeding recharge rates. Therefore, development and evaluation of alter-native water management strategies are needed for sustainable development of the region. The soil and Water Assessment Tool (SWAT) model was selected for this purpose as it has been used to simulate a wide range of agricultural production, the extensive testing and application in diverse watersheds worldwide, and the potential for future linkage of this model to groundwater models. However, crucial flow data which are commonly used for calibrating hydrologic models are not available in this watershed. This paper describes a novel calibration methodology that uses biomass and water balance approach which has potential for calibration of hydrologic models in ungauged or data-scarce watersheds, which are prevalent in many parts of the world. Estimated long-term annual average actual evapotranspiration (AET), and deep aquifer recharge rates and plant biomass values based on the expert knowledge of researchers and managers in the watershed were used as targets for calibration. The model performance was assessed using the Nash-Sutcliffe effi-ciency coefficient (NSE), coefficient of determination (R2), and percent bias (PBIAS, %) statistics. On average, the calibrated SWAT model yielded annual Nash-Sutcliffe efficiency coefficient values of 0.95, 0.99, and 0.85 for AET, recharge, and biomass, respectively. The coefficient of determination, values for AET, recharge, and biomass were 0.95, 0.94, and 0.99 respectively. The percent bias values of ±2.21%, ±0.18%, and ±0.96% for AET, recharge, and biomass, respectively, indicated that the model reproduced the calibration target values of the three water budget variables within an acce
