Nonfunctional alleles of long-day suppressor genes independently regulate flowering time

Due to the remarkable adaptability to various environments, rice varieties with diverse flowering times have abeen domesticated or improved from Oryza rufipogon.Detailed knowledge of the genetic factors controlling flowering time will facilitate understanding the adaptation mechanism in cultivated rice and enable breeders to design appropriate genotypes for distinct preferences. In this study,four genes（Hd1, DTH8, Ghd7 and OsPRR37） in a rice long-day suppression pathway were collected and sequenced in 154, 74,69 and 62 varieties of cultivated rice（Oryza sativa）respectively. Under long-day conditions, varieties with nonfunctional alleles flowered significantly earlier than those with functional alleles. However, the four genes have different genetic effects in the regulation of flowering time： Hd1 and Os PRR37 are major genes that generally regulate rice flowering time for all varieties, while DTH8 and Ghd7 only regulate regional rice varieties. Geographic analysis and network studies suggested that the nonfunctional alleles of these suppression loci with regional adaptability were derived recently and independently. Alleles with regional adaptability should be taken into consideration for genetic improvement. The rich genetic variations in these four genes,which adapt rice to different environments, provide the flexibility needed for breeding rice varieties with diverse flowering times.

Due to the high risk of tearing and rupture,vulnerable atherosclerotic plaques would induce serious cardiovascular and cerebrovascular diseases.Despite the available clinical methods can evaluate the vulnerability of plaques and specifically treat vulnerable plaques before a cardiovascular event,but the efficiency is still low and undesirable.Herein,we rationally design and engineer the low-intensity focused ultrasound(LIFU)-responsive FPD@CD nanomedicine for the highly efficient treatment of vulnerable plaques by facilely loading phase transition agent perfluorohexane(PFH)into biocompatible PLGA-PEG-PLGA nanoparticles(PPP NPs)and then attaching dextran sulphate(DS)onto the surface of PPP NPs for targeting delivery.DS,as a typical macrophages-targeted molecule,can achieve the precise vaporization of NPs and subsequently controllable apoptosis of RAW 264.7 macrophages as induced by acoustic droplet vaporization(ADV)effect.In addition,the introduction of DiR and Fe3O4 endows nanomedicine with near-infrared fluorescence(NIRF)and magnetic resonance(MR)imaging capabilities.The engineered FPD@CD nanomedicine that uses macrophages as therapeutic targets achieve the conspicuous therapeutic effect of shrinking vulnerable plaques based on in vivo and in vitro evaluation outcomes.A reduction of 49.4%of vascular stenosis degree in gross pathology specimens were achieved throughout the treatment period.This specific,efficient and biosafe treatment modality potentiates the biomedical application in patients with cardiovascular and cerebrovascular diseases based on the relief of the plaque rupture concerns.

Multi-dimension apportionment of clean air “parade blue”phenomenon in Beijing

The mass concentration and major chemical components of fine particulate matter were measured before,during and after Beijing＇s massive parade commemorating 70th anniversary of the Chinese Victory in World War II on September 3,2015.Regional emission inventory,positive matrix factorization（PMF）,observations from space and backward air mass trajectories were jointly applied to identify the major pollution sources and their temporal and spatial variations.The contributions of emissions variations and the meteorological conditions related to the＂parade blue＂phenomenon in Beijing and its surrounding areas were investigated in detail.The main cause of the decreased PM（2.5）mass concentration was attributed to the absolute reduction in emissions of primary air pollutants.The chemical composition of PM（2.5）varied significantly before,during and after the parade.Fugitive dust particles were well controlled,the secondary formation of PM（2.5）was reduced along with the controlled gaseous precursors＇emissions from vehicles and industrial sources during the temporary intensified control period.During the parade period,the SO2and NO2column concentrations in Beijing and the surrounding areas decreased sharply,indicating that the coordinated reduction in primary emissions from the surrounding areas of Beijing played an important role in lowering the ambient concentration of SO2and NO2and accordingly lowered PM（2.5）and improved the regional air quality.A comparison of the temperature,humidity,and wind speed and direction during the same periods in 2014 and 2015 showed that the meteorological conditions positively influenced the achievement of＂parade blue＂.

Establishment of an optimized CTC detection model consisting of EpCAM,MUC1 and WT1 in epithelial ovarian cancer and its correlation with clinical characteristics

Objective:Emerging studies have demonstrated the promising clinical value of circulating tumor cells(CTCs)for diagnosis,disease assessment,treatment monitoring and prognosis in epithelial ovarian cancer.However,the clinical application of CTC remains restricted due to diverse detection techniques with variable sensitivity and specificity and a lack of common standards.Methods:We enrolled 160 patients with epithelial ovarian cancer as the experimental group,and 90 patients including 50 patients with benign ovarian tumor and 40 healthy females as the control group.We enriched CTCs with immunomagnetic beads targeting two epithelial cell surface antigens(EpCAM and MUC1),and used multiple reverse transcription-polymerase chain reaction(RT-PCR)detecting three markers(EpCAM,MUC1 and WT1)for quantification.And then we used a binary logistic regression analysis and focused on EpCAM,MUC1 and WT1 to establish an optimized CTC detection model.Results:The sensitivity and specificity of the optimized model is 79.4%and 92.2%,respectively.The specificity of the CTC detection model is significantly higher than CA125(92.2%vs.82.2%,P=0.044),and the detection rate of CTCs was higher than the positive rate of CA125(74.5%vs.58.2%,P=0.069)in early-stage patients(stage I and II).The detection rate of CTCs was significantly higher in patients with ascitic volume≥500 mL,suboptimal cytoreductive surgery and elevated serum CA125 level after 2 courses of chemotherapy(P<0.05).The detection rate of CTC;and CTC;was significantly higher in chemo-resistant patients(26.3%vs.11.9%;26.4%vs.13.4%,P<0.05).The median progression-free survival time for CTC;patients trended to be longer than CTC;patients,and overall survival was shorter in CTC;patients(P=0.043).Conclusions:Our study presents an optimized detection model for CTCs,which consists of the expression levels of three markers(EpCAM,MUC1 and WT1).In comparison with CA125,our model has high specificity and demonstrates better diagnostic values,especially for early-stage ovarian cancer.Detection of

Waterfowl industry is an essential part of modern agricultural industry system.Enterprises and breeding farmers in waterfowl industry are new agricultural business entities,backbones for development of waterfowl industry,and main undertakers of business risks and major promoters of industry development.From the survey and analysis of several enterprises,farmers and business organizational form of waterfowl industry,it concluded that the business organizational form and the relationship between interested parties exert great influence on the development of waterfowl industry.According to these,it came up recommendations including optimizing business organizational form and reasonably selecting logistics mode according to profits and risks of business entities in the industry chain and main points of input and technological progress,in the hope of helping enterprises to resist risks,conduct scientific management,so as to promote stable development of waterfowl industry.

细胞外囊泡(extracellular vesicles,EV)是由脂质双分子层包裹着蛋白质、核酸等生物分子组成的天然纳米结构颗粒。EV作为细胞间无细胞通讯的一种方式,通过传递包括遗传信息在内的大量生物分子来影响细胞间的通讯。此外,EV还参与多种生物学功能的调控,如免疫调节、细胞间竞争、水平基因转移和致病性等。革兰氏阳性细菌分泌的EV携带多种化合物,这些化合物在细菌竞争、生存、入侵、抗生素耐药和感染方面发挥多样化作用。目前对于细菌EV的研究主要集中在革兰氏阴性细菌中,对革兰氏阳性细菌EV的研究报道较少,其中对葡萄球菌EV的报道主要是金黄色葡萄球菌和表皮葡萄球菌。这篇综述介绍了葡萄球菌EV的化学成分组成、功能和分泌的影响因素及临床应用。

针对加载预训练模型的传统SSD(Single Shot MultiBox Detector)模型不能更改网络结构,设备内存资源有限时便无法使用的问题,该研究提出一种不使用预训练模型也能达到较高检测精度的灵武长枣图像轻量化目标检测方法。首先,建立灵武长枣目标检测数据集。其次,以提出的改进DenseNet网络为主干网络,并将Inception模块替换SSD模型中的前3个额外层,同时结合多级融合结构,得到改进SSD模型。然后,通过对比试验证明改进DenseNet网络和改进SSD模型的有效性。在灵武长枣数据集上的试验结果表明,不加载预训练模型的情况下,改进SSD模型的平均准确率(mAP,mean Average Precision)为96.60%,检测速度为28.05帧/s,参数量为1.99×10^(6),比SSD模型和SSD模型(预训练)的mAP分别高出2.02个百分点和0.05个百分点,网络结构参数量比SSD模型少11.14×10^(6),满足轻量化网络的要求。即使在不加载预训练模型的情况下,改进SSD模型也能够很好地完成灵武长枣图像的目标检测任务,研究结果也可为其他无法加载预训练模型的目标检测任务提供新方法和新思路。

China is the largest rice-producing country,but the genomic landscape of rice diversity has not yet been clarified.In this study,we re-sequence 1070 rice varieties collected from China(400)and other regions in Asia(670).Among the six major rice groups(aus,indica-I,indica-II,aromatic,temperate japonica,and tropical japonica),almost all Chinese varieties belong to the indica-II or temperate japonica group.Most Chinese indica varieties belong to indica-II,which consists of two subgroups developed during different phases of rice breeding.The genomic segments underlying the differences between these subgroups span36.32 Mb.The Chinese japonica rice varieties fall into the temperate japonica group,consisting of two subgroups based on their geographical distribution.The genomic segments underlying the differences between these subgroups span 27.69 Mb.These differentiated segments in the Chinese indica varieties span 45 genes with nonsynonymous mutations that are closely related to variations in plant height and grain width.Fifty-four genes with nonsynonymous mutations are associated with the differences in heading date between the two Chinese japonica subgroups.These findings provide new insights into rice diversity in China that will facilitate the molecular breeding.

The overuse of nitrogen(N)fertilizer in fields has increased production costs and raised environmental concerns.Increasing the N use efficiency(NUE)of rice varieties is crucial for sustainable agriculture.Here we report the cloning and characterization of OsNPF3.1,a gene that controls rice NUE.An amino acid mutation in the OsNPF3.1 coding region caused different NUEs in wild and cultivated rice.OsNPF3.1,which is expressed mainly in the aerial parts of rice,also affects rice plant height,heading date,and thousand-grain weight.The OsNPF3.1 protein is located in the plasma membrane.When OsNPF3.1 was subjected to artificial selection,two naturally varying loci were associated with NUE,of which OsNPF3.1Chr6_8741040differed between indica and japonica rice.OsNPF3.1 can be used as a new target gene for breeding rice varieties with high NUE.
