Discovery and structure-activity relationship studies of novel tetrahydro-β-carboline derivatives as apoptosis initiators for treating bacterial infections

Developing and excavating new agrochemicals with highly active and safe is an important tactic for protecting crop health and food safety.In this paper,to discover the new bactericide candidates,we designed,prepared a new type of1,2,3,4-tetrahydro-β-carboline(THC)derivatives and evaluated the in vitro and in vivo bioactivities against the Xanthomonas oryzae pv.oryzae(Xoo),Xanthomonas axonopodis pv.citri(Xac),and Pseudomonas syringae pv.actinidiae(Psa).The in vitro bioassay results exhibited that most title molecules possessed good activity toward the three plant pathogenic bacteria,the compound A17 showed the most active against Xoo and Xac with EC50 values of 7.27 and 4.89 mg mL^(-1)respectively,and compound A8 exhibited the best inhibitory activity against Psa with EC50value of 4.87 mg mL^(-1).Pot experiments showed that compound A17 exhibited excellent in vivo antibacterial activities to manage rice bacterial leaf blight and citrus bacterial canker,with protective efficiencies of 52.67 and 79.79%at 200 mgmL^(-1),respectively.Meanwhile,compound A8 showed good control efficiency(84.31%)against kiwifruit bacterial canker at 200 mg mL^(-1).Antibacterial mechanism suggested that these compounds could interfere with the balance of the redox system,damage the cell membrane,and induce the apoptosis of Xoo cells.Taken together,our study revealed that tetrahydro-β-carboline derivatives could be a promising candidate model for novel broadspectrum bactericides.

Yuzaoea gen. nov., a new biraphid diatom(Bacillariophyceae)genus and its phylogenetic significance

The flexed frustules in pennate diatoms are usually associated with monoraphid diatoms. Interestingly, we found a biraphid diatom species with flexed frustules in an offshore intertidal beach environment on Weizhou Island,Beihai City, Guangxi Zhuang Autonomous Region, China. Therefore, based on morphological characteristics, we described a new genus of diatoms Yuzaoea sinensis gen. et sp. nov. CH Li, HH liu, YH Gao & CP Chen. The frustule of this genus is characterized by heterogeneous frustule with one concave valve and one convex valve,complete raphe on both valves, straight and moderately eccentric raphe, uniseriate striae and girdle bands with a single row of areolae. The most identifying feature of this genus was the flexed frustule, which is rare in biraphid diatoms and common in monoraphid diatoms. We compared the morphometric characteristics of genus Yuzaoea with genus Rhoikoneis and several genera within the family Rhoicospheniaceae, including Rhoicosphenia,Campylopyxis, and Cuneolus. Phylogenetic analyses based on SSU rRNA and rbc L showed that the genus Yuzaoea was the sister group to the clade of Rhoicosphenia with a high support value(bootstrap values = 100%), and the clade “Yuzaoea+Rhoicosphenia” was sister to the clade of monoraphid diatoms, in which the genera Achnanthidium, Planothidium and some Cocconeis with high support values(bootstrap = 100%).Morphologically, the genus Yuzaoea shares many morphological features with monoraphid diatoms like genera Achnanthidium and Planothidium and the members within the Rhoicospheniaceae. Therefore, based on a combined morphological studies and phylogenetic results we suggested that this branch may represented the evolution of one kind monoraphid diatoms, from biraphid diatoms(e.g. genus Yuzaoea), to incompleted biraphid diatoms(e.g. genera Rhoicosphenia, Campylopyxis), to monoraphid diatoms(e.g. genera Achnanthidium and Planothidium).

PUMD:a PU learning-based malicious domain detection framework

Domain name system(DNS),as one of the most critical internet infrastructure,has been abused by various cyber attacks.Current malicious domain detection capabilities are limited by insufficient credible label information,severe class imbalance,and incompact distribution of domain samples in different malicious activities.This paper proposes a malicious domain detection framework named PUMD,which innovatively introduces Positive and Unlabeled(PU)learning solution to solve the problem of insuffcient label information,adopts customized sample weight to improve the impact of class imbalance,and effectively constructs evidence features based on resource overlapping to reduce the intra-class distance of malicious samples.Besides,a feature selection strategy based on permutation importance and binning is proposed to screen the most informative detection features.Finally,we conduct experiments on the open source real DNS traffic dataset provided by QI-ANXIN Technology Group to evaluate the PUMD framework's abil-ity to capture potential command and control(C&C)domains for malicious activities.The experimental results prove that PUMD can achieve the best detection performance under different label frequencies and class imbalance ratios.

Objective:Chronic fatigue syndrome(CFS)is a prevalent symptom of post-coronavirus disease 2019(COVID-19)and is associated with unclear disease mechanisms.The herbal medicine Qingjin Yiqi granules(QJYQ)constitute a clinically approved formula for treating post-COVID-19;however,its potential as a drug target for treating CFS remains largely unknown.This study aimed to identify novel causal factors for CFS and elucidate the potential targets and pharmacological mechanisms of action of QJYQ in treating CFS.Methods:This prospective cohort analysis included 4,212 adults aged≥65 years who were followed up for 7 years with 435 incident CFS cases.Causal modeling and multivariate logistic regression analysis were performed to identify the potential causal determinants of CFS.A proteome-wide,two-sample Mendelian randomization(MR)analysis was employed to explore the proteins associated with the identified causal factors of CFS,which may serve as potential drug targets.Furthermore,we performed a virtual screening analysis to assess the binding affinity between the bioactive compounds in QJYQ and CFS-associated proteins.Results:Among 4,212 participants(47.5%men)with a median age of 69 years(interquartile range:69–70 years)enrolled in 2004,435 developed CFS by 2011.Causal graph analysis with multivariate logistic regression identified frequent cough(odds ratio:1.74,95%confidence interval[CI]:1.15–2.63)and insomnia(odds ratio:2.59,95%CI:1.77–3.79)as novel causal factors of CFS.Proteome-wide MR analysis revealed that the upregulation of endothelial cell-selective adhesion molecule(ESAM)was causally linked to both chronic cough(odds ratio:1.019,95%CI:1.012–1.026,P=2.75 e^(−05))and insomnia(odds ratio:1.015,95%CI:1.008–1.022,P=4.40 e^(−08))in CFS.The major bioactive compounds of QJYQ,ginsenoside Rb2(docking score:−6.03)and RG4(docking score:−6.15),bound to ESAM with high affinity based on virtual screening.Conclusions:Our integrated analytical framework combining epidemiological,genetic,and in silico data provides a novel strategy

以提高我国养殖鱼类产品品质的动物福利策略为目标，对国内外从动物福利到鱼福利的发展历程进行了归纳分析并提出展望。首先，基于对陆生脊椎动物福利的五项自由原则和三大范畴的系统回顾，对比动物福利在家畜家禽业的研究发展历程，对鱼类福利的发展历程与现状进行了脉络梳理；然后，针对鱼类福利的量化评估问题，从生理应激、行为应激以及心理应激三大应激响应机制，对鱼类福利评价体系相关的研究进展进行了回顾，重点针对评价指标及评价方法进行了归纳总结；接下来，针对养殖鱼类产业，重点对养殖过程、离水捕捞以及致死处置三个主要环节的鱼类福利特点及研究利用现状进行了分析讨论。最后，分析指出我国渔后动物福利存在盲区，提出了品质易逝期动物福利方案的设计思路，和基于品质易逝期动物福利原则以实现锁鲜调控的展望。

Brittle culm 25, which encodes an UDP-xylose synthase, affects cell wall properties in rice

Because plant mechanical strength influences plant growth and development,the regulatory mechanisms underlying cell-wall synthesis deserve investigation.Rice mutants are useful for such research.We have identified a novel brittle culm 25(bc25)mutant with reduced growth and partial sterility.BC25 encodes an UDP-glucuronic acid decarboxylase involved in cellulose synthesis and belongs to the UXS family.A single-nucleotide mutation in BC25 accounts for its altered cell morphology and cellwall composition.Transmission electron microscopy analysis showed that the thickness of the secondary cell wall was reduced in bc25.Monosaccharide analysis revealed significant increases in content of rhamnose and arabinose but not of other monosaccharides,indicating that BC25 was involved in xylose synthesis with some level of functional redundancy.Enzymatic assays suggested that BC25 functions with high activity to interconvert UDP-glucuronic acid(UDP-Glc A)and UDP-xylose.GUS staining showed that BC25 was ubiquitously expressed with higher expression in culm,root and sheath,in agreement with that shown by quantitative real-time(q RT)-PCR.RNA-seq further suggested that BC25 is involved in sugar metabolism.We conclude that BC25 strongly influences rice cell wall formation.

专科护士在临床中发挥越来越重要的作用,同时他们在独立面对患者时(尤其是造口治疗师/伤口造口专科护士)也常常为伦理难题所困扰。在前期构建的“临床护理伦理结构化分析表格”基础上,采用案例陈述法,以1例产后脂肪液化合并线结反应的慢性伤口患者的护理为例,分析造口治疗师在解决治疗冲突时的伦理思考。进而建议专科护士在解决患者疑难复杂问题时,需要充分考虑患者的意愿与背景特征,可采用“伦理分析四盒子模型”,以达到分析及解决伦理冲突问题的目的。

目的:探讨磁共振(MRI)和高频超声诊断儿童髋关节滑膜炎(SHC)的价值。方法:107例小儿髋部疼痛患者作为研究对象,均给予MRI和高频超声检查。分析两种检查方法对SHC的诊断价值及对不同类型SHC的鉴别价值。结果:高频超声检查与临床诊断结果的一致性高于MRI(Kappa=0.717 vs 0.586);联合检查诊断SHC的AUC为0.844,大于MRI检查的0.812(P0.05)。结论:MRI和高频超声联合检查对SHC具有较高的诊断价值。

有研究提出,严重烧伤会导致长时间的高代谢应激反应^([1])。这种高代谢应激反应,也被称为代谢亢进的现象,是严重烧伤患者需要克服的一个经典的、高度复杂的、多因素的挑战。研究表明,代谢亢进中会出现例如高血糖、高胰岛素血症,全身脂质水平升高以及长期静息能量消耗(Resting energy expenditure,REE)提升^([2])等情况。此外,这种显著且持久的代谢亢进,还会伴随着皮质醇、儿茶酚胺、细胞因子和急性期蛋白的显著升高,提示烧伤患者在烧伤后处于一种高炎症状态^([3])。上述一系列的情况,会对患者烧伤后的身体造成极大的负担。
