Combined proteomic and metabolomic studies on the liver of Amur sturgeon Acipenser schrenckii under titanium dioxide nanoparticle exposure

Nanomaterials,particularly titanium dioxide nanoparticles(TiO_(2)-NPs),are extensively utilized across various ***,their environmental release has raised concerns regarding their potential ecological and environmental *** reproductive toxicity of TiO_(2)-NPs in fish species has attracted considerable attention,yet conflicting research outcomes have been *** investigated the effects of TiO_(2)-NPs exposure on the liver of juvenile Amur sturgeon Acipenser schrenckii using label-free proteomic and untargeted metabolomic *** experiment included a control group and three groups exposed to different concentrations of TiO_(2)-NPs(low,TL;medium,TM;high,TH).Compared to the control group,9,19,and 25 proteins and 35,73,and 158 metabolites were differentially expressed in the TH,TM,and TL TiO_(2)-NP-exposed groups,*** differentially expressed genes(DEGs)were enriched in the Kyoto Encyclopedia for Genes and Genomes(KEGG)pathways related to glycolysis and ***,among the 126 correlated proteins,the most enriched pathways were associated with endocytosis and protein processing in the endoplasmic ***,syringic acid was significantly downregulated across all three TiO_(2)-NP-exposed *** obtain a comprehensive overview of the TiO_(2)-NP-induced expression changes,a co-regulated network of proteins and metabolites associated with TiO_(2)-NPs exposure was *** to TiO_(2)-NPs led to enrichment and alteration of pathways related to immune responses,including endocytosis,protein processing in the endoplasmic reticulum,and peroxisome proliferator-activated receptor(PPAR)*** conclusion,our findings indicate that exposure to TiO_(2)-NPs might disrupt glucose metabolism and induce immune responses,thus contributing to our understanding of the environmental impacts of nanomaterials and highlighting the need for further research and development of potential mitigation strategies.

以2013—2022年我国中部H省的12个地市为研究对象,采用空间动态杜宾模型,对H省城市绿色发展效率与环境规制进行了研究。空间相关性检验表明,H省12地市绿色发展效率有显著局域空间集聚特征存在,绿色发展效率低的被同为水平低单元包围、绿色发展效率高的被同为高水平单元包围的现象比较明显。Moran’s I显示自2019年以后,H省各地市绿色发展效率的空间集聚特征显著。根据Moran散点图可以看出,多数地市表现出从“低-高”“高-低”聚集向“高-高”“低-低”聚集的绿色发展效率趋势。根据空间溢出效应来看,在短期内提升环境规制强度,会有效提高“经济相邻”地区绿色发展效率。同时绿色发展效率会随着长期内环境规制强度的提高而提高,但未表现出显著的直接和间接效应。

Expression Characteristics and Putative Functions of KIF3A/KIF3B During Spermiogenesis of Phascolosoma esculenta

Physiological and histological responses of Phascolosoma esculenta (Sipuncula: Phascolosomatidea) to acute heat stress

Environmental hypoxia induces apoptosis in large yellow croaker Larimichthys crocea via both intrinsic and extrinsic pathways

Hypoxia has become an unfavorable factor affecting the sustainable development of the large yellow croaker Larimichthys crocea,an economically important mariculture fish in *** is a consequence of hypoxia on ***,the effects of hypoxia stress on apoptosis in *** remain largely *** investigated the effect of environmental hypoxia on apoptosis in *** show that hypoxia induced apoptosis in *** both in vivo and in *** mitochondrial membrane potential was significantly reduced in large yellow croaker fry(LYCF)*** expression levels of Bcell lymphoma/leukemia-2(Bcl-2)m RNA and protein were also significantly decreased in the liver and LYCF cells during 96 h and 48 h of hypoxia stress,respectively,whereas the expression level of Bcl-2 associated X(Bax)mRNA,Casp3 mRNA,and activity of caspase-3/7/9 were significantly increased,indicating that hypoxia induced caspase-dependent intrinsic apoptosis in *** expression level of the apoptosis-inducing factor(AIF)protein was significantly increased in the liver and LYCF *** level of AIF protein was significantly decreased in the cytoplasm but increased in the nuclei of ***,demonstrating that hypoxia induced the AIF-mediated caspase-independent intrinsic *** addition,the activity of caspase-8 was significantly increased,indicating that hypoxia stress induced extrinsic apoptosis in ***,hypoxia induced apoptosis in *** through both the intrinsic and extrinsic *** present study accumulated basic biological information to help elucidate the mechanism of hypoxia response in marine fish.

Prohibitin Protein Expression During Spermatogenesis in the Large Yellow Croaker, Larimichthys crocea

Mitochondria are important for animals’fertility,and their morphologies and functions during spermatogenesis are un-der ***,the molecular mechanism that regulates the mitochondrial dynamic during spermatogenesis is still *** this study,the cytological features of spermatogenesis were investigated in Larimichthys *** addition,the structure and function of prohibitin(PHB),which is associated with mitochondrial structure and dynamic,was also *** full-length cDNA and protein(Lc-PHB)from the *** phb gene(Lc-phb)contained 1625 base pairs and 271 amino acids,***-PHB had a conserved primary structure that resulted in a transmembrane,SPFH(the analogous region of proteins stomat-ins,prohibitins,flotillins and HflK/C),and coiled-coil *** was detected at high levels in the muscle,liver,and heart,and at intermediate levels in the testis,gill,and ***-phb mRNA expression was detected in spermatogenic cells by fluorescence in situ *** immunofluorescence assay revealed that PHB protein was localized in the mitochondria during ***,PHB expression was detected in the perinuclear cytoplasm of spermatogonia,spermatocytes,and spermatids in the early developmental stage,and mainly localized on one side of the nuclei in the cytoplasm of spermatids in a middle developmental stage,and finally on the sperm *** blotting showed that PHB was located in the extracted mitochondria protein fraction but not in the cytoplasm protein fraction of ***,these results indicated that PHB was expressed in the mitochondria dur-ing *** addition,the study explained the mitochondrial dynamic during fish spermatogenesis and proposed a possi-ble relationship among PHB,spermatogenesis,and male fertility.

Roles of Adinandra nitida(Theaceae) and camellianin A in HCl/ethanol-induced acute gastric ulcer in mice

Gastric ulcer is a global health concern nowadays. Adinandra nitida, known as Shibi tea, is a flavonoidrich plant found in South China. A. nitida possesses many healthy properties, such as antioxidation and reducing blood pressure. However, its effects on gastric ulcer have not been investigated. This study aimed to investigate the effects of the Shibi tea water extract(STE) and its main flavonoid camellianin A(CA) in hydrochloric acid(HCl) and ethanol(Et OH)-induced acute gastric ulcer in mice. Administration of CA and STE for continuous two days after stimulation by HCl/Et OH significantly attenuated the deterioration of gastric mucosal damage by lowering the gross gastric mucosal index, histopathological injury index, the oxidative stress, the expression of the inflammatory cytokines TNF-α and IL-6, and the expression of inflammatory mediators i NOS and COX-2. Western blotting and immunohistochemistry analysis showed that CA and STE regulated the inflammatory signaling pathway protein levels of IκB-α and NF-κB. Taken together, our study verified that CA and STE have antioxidative and anti-inflammatory effects in gastric ulcer mice. We propose that A. nitida should be developed as natural functional food for acute gastric ulcer patients base on the gastroprotective effects of STE and its main flavonoid CA.

Molecular Characterization, Tissue Distribution and Localization of Larimichthys crocea Kif3a and Kif3b and Expression Analysis of Their Genes During Spermiogenesis

KIF3A and KIF3B are two N-terminal motor proteins belonging to the kinesin-II superfamily that play essential roles in *** understand the roles played by KIF3 A/3B during spermatogenesis of large yellow croaker Larimichthys crocea,we studied the testis characteristics at different developmental stages of ***,and determined the spatiotemporal expression patterns of kif3a and kif3b during *** real-time PCR(qR T-PCR)showed that the overall trends of kif3 a/3 b m RNA abundance during testis development are *** stage Ⅱ to stage V,kif3a/3b m RNA abundances first increased and then fell after reaching a peak at stage ***,the m RNA abundances of both genes at stage V were higher than those at stages Ⅱ and Ⅲ.In addition,it is worth of noting that kif3 b m RNA abundance was higher than that of kif3a at all *** in situ hybridization results revealed that kif3a/3b m RNA abundance dynamics were consistent with the migration of mitochondria,the deformation of nucleus,and the formation of *** m RNA hybridization signals of both genes first appeared either around the nuclear periphery or on the side of the nuclei,then appeared at one side of nuclei,and finally were mainly on the tail during *** findings contributed to better understanding the molecular mechanisms of spermiogenesis in fish;and suggested that KIF3A and KIF3B may participate in the intracellular transport of mitochondria,nuclear deformation,and the formation of tail during the spermiogenesis in ***.
