Induction of Oral Tolerance in Neonatal Mice by Transfer of Food Allergens as IgA-Immune Complexes in Breast Milk

Various opinions have been presented on the merits and demerits that breastfeeding gives for the allergic onset of the babies. In this report, we focused on whether food proteins eaten by mother mice and secreted into breast milk as IgA-immune complexes contribute to the allergy prevention through oral tolerance in infants who ingest the milk. BALB/c mice were divided into two groups;E-group fed only egg white proteins and M-group fed only cow’s milk proteins as a dietary protein source. After immunizing M-group infants fed their own mother’s milk with ovalbumin/alum, diarrhea associated with experimental Th2 intestinal inflammation was induced by oral administration of ovalbumin. The diarrhea was dramatically suppressed in E-group infants. Concomitantly, low level of serum anti-ovalbumin- and ovomucoid-IgG1 and IgE, suppression of IL-4 synthesis by spleen cells, and low incidence of anaphylactic death after intravenous injection of ovalbumin were observed preferentially in E-infants. Immune complexes of respective dietary proteins and IgA were found in the breast milk obtained from each group of mother. Oral administration of pseudo immune complex chemically synthesized with ovalbumin and monoclonal mouse IgA in advance effectively suppressed anti-ovalbumin-IgG1 synthesis in adult mice after immunization with ovalbumin. The tolerance induced by the pseudo immune complex of ovalbumin diminished spontaneously while mice did not take egg white proteins. Thus, immune tolerance and then prevention of allergic disorder against dietary proteins were acquired via breastfeeding by mothers feeding the relevant proteins, probably through the immune complexes of dietary proteins and sIgAs secreted into breast milk.

Iterative risk management and risk-sensitive public investment planning are increasingly seen as essential elements of natural disaster resilience. This article assesses the disaster risk facing the hazard-prone Southeast Asian country of Cambodia and discusses its fiscal preparedness and need for proactive disaster risk *** study provides a bottom-up assessment of flood and cyclone risks to public and private buildings including educational structures, health facilities, and housing and estimates the total direct economic damage to range from approximately USD 304 million for a 5-year return period event to USD 2.26 billion for a 1000-year return period event. These estimates were further analyzed using the fiscal risk due to disasters, which indicates that Cambodia will likely face a resource gap whenever a hazard as large as that of a 28-year return period event strikes. Given the frequent occurrence of disasters and rapid accumulation of capital assets taking place, proactive risk reduction is highly advisable. But interviews with national policymakers also revealed that there are a number of barriers to effective risk reduction and management in Cambodia. The general lack of awareness regarding risk-based concepts and the limited availability of local risk information necessitate a continued and sustained effort to build iterative risk management in Cambodia.

Erythronium japonicum is a representative species of spring ephemeral, forest floor plants in substitution forests in Japan. Seed germination tests were conducted to observe the effects of temperature, light, and moisture content on the germination of E. japonicum. E. japonicum seeds did not germinate at constant temperatures of 5°C, 10°C, 15°C, 20°C, or 25°C. Seeds germinated at 5°C and 10°C after storage at 20°C for 1 or 2 months. Seed germinability was affected by desiccation to only 40% moisture content. Seeds germinated well in both light and dark conditions. Conditions under the leaf litter layer showed unique characteristics of temperature, light, and moisture content. The temperature was around 20°C under the leaf litter layer, even though it was 30°C above. The moisture content under the leaf litter layer was higher than that above. The leaf litter blocked out the sunlight. The temperature of 20°C necessary for E. japonicum germination occurred below, but not above, the leaf litter layer. The avoidance of decreasing in germinability from reduced seed moisture content in a short time is consistent with the high moisture conditions under the leaf litter layer. The leaf litter layer plays an important role in providing optimal conditions of temperature, light, and moisture content for the germination of E. japonicum seeds.

＜正＞ We have previously shown that amlodipine, an L - type voltage - dependent Ca2+ channel blocker, inhibited the cell growth of human epidermoid carcinoma A431 cells that lack relevant Ca2+ channels. In this study, we examined the effect of amlodipine on cell cycle distribution and cell cycle - specific protein expression in A431 cells by flow cytometric analysis and Western blotting, respec- tively. Treatment with amlodipine （20-30 uM for 24 hrs） induced Gl phase accumulation, which was associated with decreases in the phosphorylated form of retinoblastoma protein （pRB）, a regulator of Gl - S - phase transition and in protein levels of cyclin Dl and cyclindependent kinase 4 （CDK4）, Gl - specific cell- cycle proteins. On the other hand, the expression of p21Wafl/Cipl, an inhibitor protein of CDK/cyclin complexes, was increased by amlodipine. These data suggest that amlodipine induced the expression of p21Wafl/Cipl and con-comitantly inhibited the CDK4 and CDK4 - mediated phosphorylation of pRB, which resulted in Gl cell - cycle arrest and growth inhibition.

The sterilization conditions between experiment and dairy industrial level are different and concern about influence on product’s qualities. In this study, the fermentation properties of Streptococcus thermophilus, alone and in combination with Lactobacillus delbrueckii subsp. bulgaricus, were evaluated in skim milk that had been subjected to distinct sterilization conditions. Growth, organic acid generation, and EPS production were determined using pasteurized or autoclaved milk. When S. thermophilus was cultivated in pasteurized skim milk, the growth was strain-dependent. On the other hand, growth of S. thermophilus was accelerated in autoclaved milk. Exocellular polysaccharide (EPS) production by L. bulgaricus was not affected by the combination of S. thermophilus strains. Thus, we observed that yogurt fermented by L. bulgaricus was minimally affected by the combination of S. thermophilus strains;growth of L. bulgaricus was maintained under the constant environment. These results should facilitate the development of fermented milk produced from L. bulgaricus in the dairy industry.

Collagen-Based Fuel Cell and Its Proton Transfer

We have fabricated the fuel cell based on the tissue derived biomaterial “collagen” and investigated its proton transfer. It was found that “collagen” becomes the electrolyte of fuel cell in the humidified condition. The power density of the fuel cell becomes typically 8.6 W/m2 in the 80% humidity. Further, these results indicate that collagen exhibits proton conductivity in the humidified condition. Both of proton conductivity and dielectric constant increase by the increase of humidity. From the analyses of the frequency dependence of AC conductivity, it was found that proton conductivity and the dielectric dispersion observed in the humidified condition are caused by the formation of the water bridge, which is bonded with the collagen peptide chain. Considering that hydration induces the formation of the water bridge and that increases proton conductivity and dielectric constant, it is deduced that proton transfer in the fuel cell based on collagen is caused by the breaking and rearrangement of hydrogen bond in the water bridge.

Floods affect more people globally than any other type of natural hazard. Great potential exists for new technologies to support flood disaster risk reduction. In addition to existing expert-based data collection and analysis, direct input from communities and citizens across the globe may also be used to monitor, validate, and reduce flood risk. New technologies have already been proven to effectively aid in humanitarian response and recovery. However, while ex-ante technologies are increasingly utilized to collect information on exposure, efforts directed towards assessing and monitoring hazards and vulnerability remain limited. Hazard model validation and social vulnerability assessment deserve particular attention. New technologies offer great potential for engaging people and facilitating the coproduction of knowledge.

目的调查护士对有喂养困难的唇腭裂患儿所采用的临床奶瓶喂养技术现状。方法采用质性描述性研究设计。于2021年12月至2022年1月,向日本1109所设有产科、新生儿科或小儿牙科病房的医院分别发放匿名问卷。参与者是具有5年以上唇腭裂患儿护理工作经验的护士。问卷包括喂养技术4个方面的开放式问题,包括:奶瓶喂养前的准备、乳头插入方法、吸吮帮助和停止奶瓶喂养的标准。根据意义相似性对资料进行分类和分析。结果共获得410份有效问卷。资料分析结果显示,奶瓶喂养前的准备工作分为7个类别(如,改善患儿的口腔运动、保持患儿平稳呼吸),27个子类别;在乳头插入方法中,分为4个类别(如,用乳头在口腔内形成负压封闭裂口、插入乳头时不接触裂口),11个子类别;在吸吮帮助中,分为5个类别(如,促进觉醒、口腔产生负压),13个子类别;在停止奶瓶喂养的标准中,分为5个类别(如,觉醒水平降低、生命体征变差),16个子类别。96.0%护士希望学习唇腭裂患儿的奶瓶喂养技术。结论本研究确定了多项用于唇腭裂喂养困难患儿的奶瓶喂养技术。然而,这些技术之间存在相互矛盾,方法的有效性也未得到评估。未来需要进行干预研究,以确定各项技术的益处或潜在的危害。
