A rice tubulin tyrosine ligase-like 12 protein affects the dynamic and orientation of microtubules

The detyrosination/retyrosination cycle is the most common post-translational modification of α-*** of the conserved C-terminal tyrosine of α-tubulin by a still elusive tubulin tyrosine carboxypeptidase, and religation of this tyrosine by a tubulin tyrosine ligase(TTL), are probably common to all eukaryotes. Interestingly, for plants, the only candidates qualifying as potential TTL homologs are the tubulin tyrosine ligase-like 12 proteins. To get insight into the biological functions of these potential TTL homologs, we cloned the rice TTL-like 12 protein(Os TTLL12)andgeneratedoverexpression Os TTLL12-RFP lines in both rice and tobacco BY-2 cells. We found, unexpectedly, that overexpression of this Os TTLL12-RFP increased the relative abundance of detyrosinated α-tubulin in both coleoptile and seminal root, correlated with more stable microtubules. This was independent of the respective orientation of cortical microtubule, and followed by correspondingly changing growth of coleoptiles and seminal roots. A perturbed organization of phragmoplast microtubules and disoriented cell walls were further characteristics of this phenotype. Thus, the elevated tubulin detyrosination in consequence of Os TTLL12 overexpression affects structural and dynamic features of microtubules, followed by changes in the axiality of cell plate deposition and, consequently, plant growth.

A rational regulation of the solar water splitting reaction pathway by adjusting the surface composition and phase structure of catalysts is a substantial approach to ameliorate the sluggish reaction kinetics and improve the energy conversion *** this study,we demonstrate a nanocrystalline iron pyrophosphate(Fe_(4)(P_(2)O_(7))_(3),FePy)-regulated hybrid overlayer with amorphous iron phosphate(FePO_(4),FePi)on the surface of metal oxide nanostructure with boosted photoelectrochemical(PEC)water *** manipulating the facile electrochemical surface treatment followed by the phosphating process,nanocrystalline FePy is localized in the FePi amorphous overlayer to form a heterogeneous hybrid *** FePy-regulated hybrid overlayer(FePy@FePi)results in significantly enhanced PEC performance with long-term *** with the homogeneous FePi amorphous overlayer,FePy@FePi can improve the charge transfer efficiency more significantly,from 60% of FePi to 79%of FePy@*** density-functional theory calculations reveal that the coexistence of FePi and FePy phases on the surface of metal oxide results in much better oxygen evolution reaction kinetics,where the FePi was found to have a typical down-hill reaction for the conversion from OH*to O_(2),while FePy has a low free energy for the formation of OH*.

Background:Germ cell mitotic arrest is conserved in many vertebrates,including birds,although the time of entry or exit into quiescence phase *** arrest is essential for the normal differentiation of male germ cells into spermatogonia and accompanies epigenetic reprogramming and meiosis inhibition from embryonic development to ***,mitotic arrest was not well studied in chickens because of the difficulty in obtaining pure germ cells from relevant developmental ***:We performed single-cell RNA sequencing to investigate transcriptional dynamics of male germ cells during mitotic arrest in DAZL::GFP *** differentially expressed gene analysis and K-means clustering to analyze cells at different developmental stages(E12,E16,and hatch),we found that metabolic and signaling pathways were regulated,and that the epigenome was reprogrammed during mitotic *** particular,we found that histone H3K9 and H3K14 acetylation(by HDAC2)and DNA demethylation(by DNMT3B and HELLS)led to a transcriptionally permissive chromatin ***,we found that global DNA demethylation occurred gradually after the onset of mitotic arrest,indicating that the epigenetic-reprogramming schedule of the chicken genome differs from that of the mammalian *** hypomethylation persisted after hatching,and methylation was slowly re-established 3 weeks ***:We found a unique epigenetic-reprogramming schedule of mitotic-arrested chicken prospermatogonia and prolonged hypomethylation after *** will provide a foundation for understanding the process of germ-cell epigenetic regulation in several species for which this process is not clearly *** findings on the biological processes related to sex-specific differentiation of prospermatogonia could help studying germline development in vitro more elaborately.

AIM： To investigate the effect of selective Cyclooxygenase-2 （COX-2） inhibitor 4-[5-（4-Chloro-phenyl）-3- （trifluoromethyl）- 1H-pyrazol- 1-yl] benzenesulfonamide （SC-236）, on the cholecystokinin （CCK）-octapeptideinduced acute pancreatitis （AP） in rats. METHODS： Wistar rat weighing 240 g to 260 g were divided into three groups. （1） Normal DMSO treated group, （2） SC-236 at 4 mg/kg treated group; SC-236 systemically administered via the intravenous （i.v.） catheter, followed by 75 μg/kg CCK octapeptide subcutaneously three times, after 1, 3 and 5 h. This whole procedure was repeated for 5 d. （3） Dimethyl sulfoxide （DMSO） treated group： an identical protocol was used in this group as in the SC-236 cohort （see 2. above）. Repeated CCK octapeptide treatment resulted in a typical experimentally induced pancreatitis in the Wistar rats. RESULTS： SC-236 improved the severity of CCK- octapeptide-induced AP as measured by laboratory criteria [the pancreatic weight/body weight （p.w/ b.w） ratio, the level of serum amylase and lipase]. The SC-236 treated group showed minimal histologic evidence of pancreatitis and a significant reduction in myeloperoxidase activity. SC-236 also increased heat shock protein （HSP）-60 and HSP72 compared with the DMSO-treated group in the CCK-octapeptide-induced AP and also reduced the pancreatic levels of COX-2. Furthermore, SC-236 reduced proinflammatory cytokine synthesis and inhibited NF-KB activation compared with the DMSO-treated group in the CCK-octapeptide-induced AP. CONCLUSION： Our results suggested that COX-2 plays pivotal role in the development of AP and COX-2 inhibitors may play a beneficial role in preventing AP.

Entacapone,a catechol-O-methyltransferase inhibitor,can strengthen the therapeutic effects of levodopa on the treatment of Parkinson’s ***,few studies are reported on whether entacapone can affect hippocampal neurogenesis in *** investigate the effects of entacapone,a modulator of dopamine,on proliferating cells and immature neurons in the mouse hippocampal dentate gyrus,60 mice(7 weeks old)were randomly divided into a vehicle-treated group and the groups treated with 10,50,or 200 mg/kg *** results showed that 50 and 200 mg/kg entacapone increased the exploration time for novel object *** staining results revealed that after entacapone treatment,the numbers of Ki67-positive proliferating cells,doublecortin-positive immature neurons,and phosphorylated cAMP response element-binding protein(pCREB)-positive cells were significantly *** blot analysis results revealed that treatment with tyrosine kinase receptor B(TrkB)receptor antagonist significantly decreased the exploration time for novel object recognition and inhibited the expression of phosphorylated TrkB and brain-derived neurotrophic factor(BDNF).Entacapone treatment antagonized the effects of TrkB receptor *** results suggest that entacapone treatment promoted hippocampal neurogenesis and improved memory function through activating the BDNF-TrkB-pCREB *** study was approved by the Institutional Animal Care and Use Committee of Seoul National University(approval ***-130730-1)on February 24,2014.

Fabrication of miniature helical gears by powder extrusion using gas atomized Zn-22%Al powder

Powder extrusion, which is based on the superplastic behavior of Zn-22%A1 eutectoid alloy, was proposed to reduce the forming load and promises to provide fine microstructures in the manufacture of miniature helical gears. The specifications of the helical gears were as follows： module, 0.3; number of teeth, 12; and helix angle, 15°. Compacted powders were consolidated by sintering and solution heat treatment. The consolidated billets consisted of lamellar and fine-grained microstructures. Extrusion experiments were carried out under the following conditions： temperature, 250 ℃; strain rates, 2.36× 10-3 s^-1 and 1.18× 10^-1 s-1. The mechanical properties of the extruded helical gears were investigated by measurement of the Vickers hardness and extrusion load, and by scanning electron microscopy.

Among many biomaterials,gelatin methacrylate(GelMA),a photocurable protein,has been widely used in 3D bioprinting process owing to its excellent cellular responses,biocompatibility and ***,GelMA still shows a low processability due to the severe temperature dependence of *** overcome this obstacle,we propose a two-stage temperature control system to effectively control the viscosity of *** optimize the process conditions,we evaluated the temperature of the cooling system(jacket and stage).Using the established system,three GelMA scaffolds were fabricated in which different concentrations(0,3 and 10 wt%)of silanated silica particles were *** evaluate the performances of the prepared scaffolds suitable for hard tissue regeneration,we analyzed the physical(viscoelasticity,surface roughness,compressive modulus and wettability)and biological(human mesenchymal stem cells growth,western blotting and osteogenic differentiation)***,the composite scaffold with greater silica contents(10 wt%)showed enhanced physical and biological performances including mechanical strength,cell initial attachment,cell proliferation and osteogenic differentiation compared with those of the *** results indicate that the GelMA/silanated silica composite scaffold can be potentially used for hard tissue regeneration.

Influence of tannin-rich pine bark supplementation in the grain mixes for meat goats:Growth performance,blood metabolites,and carcass characteristics

The objective was to evaluate the use of condensed tannin(CT)-rich ground pine bark(PB)in grain mixed diets on meat goat growth performance,blood metabolites,and carcass *** four Kiko crossbred(Capra aegugrus hircus)growing male kids(BW=36.9±2,5 kg)at approximately 8 months of age were assigned randomly to 2 treatments with 3 replicates per treatment and 4 goats per *** goats were fed grain mixed diets composed of either 30%bermudagrass hay(BGH)plus concentrate(control)or 30%PB plus *** were fed at 1.2%of *** addition,all goats grazed a crabgrass/bermudagrass(CB)-based *** feeding trial lasted for 55 *** ground PB as a supplement did not negatively affect BW,average daily gain(ADG),carcass characteristics,meat pH,and meat color compared to the control *** gamma-glutamyl transferase(P=0.03),glucose(P<0.01)and Ca concentrations(P=0.04)were higher for PB than for BGH,*** 30%PB supplementation does not negatively affect animal performance,blood metabolites,and carcass parameters.

Emamectin benzoate 9.7%SL as a new formulation for a trunkinjections against pine wood nematode,Bursaphelenchus xylophilus

In this study,we investigated the preventive effects of emamectin benzoate 9.7%SL,which was newly developed to reduce the injection volume and number of injection holes required to protect against pine wood *** of the Pinus thunbergii trees injected with emamectin benzoate 9.7%SL at 0.3 mL/cm diameter at breast height(DBH)died within 2 years of inoculation with pine wood *** benzoate 9.7%SL injected at 0.6 mL/cm DBH resulted in no tree mortality for 3 *** residue of emamectin benzoate 9.7%SL in pine twigs injected with 0.3 mL/cm DBH was 0.490μg/g at 1 year after injection and 0.303μg/g after 2 *** residues values are greater than 0.031μg/g,previously determined IC95 value for emamectin benzoate against the pine wood *** field experiment and residue analysis showed that emamectin benzoate 9.7%SL could be a substitute agent for emamectin benzoate 2.15%EC,which is widely used to prevent pine wood nematode in the field and that injection volume and number of injection holes can be greatly educed using this new formulation,which will reduce injury to the cambium,interruption of water movement,and infection of inoculation wounds by wood-decay or blue stain fungi.
