Objective: To investigate matrix metalloproteinases(mmP)-2 and mmP-9 inhibitory effect of Salsola komarovii Iljin, an edible halophyte with health beneficial ***: Salsola komarovii crude extracts(SKI), and solvent(n-hexane, 85% aq. meOH, n-BuOH, and H2O) fractionated extracts of SKI were prepared. Gelatin zymography was carried out to observe mmP enzymatic activity. The release of the mmP enzymes was measured by enzyme-linked imm.nosorbent assay. Expression of mmPs in m RnA and protein level were investigated by polymerase chain reaction analysis and imm.noblotting, ***: SKI and SKI fractions inhibited active mmP-2 and mmP-9 amount in the treated cell culture medium. Also, SKI suppressed the release of mmP-2 and mmP-9 from stimulated HT1080 hum.n fibrosarcoma cells. Furthermore, SKI suppressed the m RnA and protein expression of mmP-2 and mmP-9. SKI fractions showed parallel effects except for H2O fraction which did not yield any significant mmP inhibitory effect. Am.ng fractions, 85% aq. me OH was the most active fraction to inhibit both the enzymatic effect and expression of mmP-2 and ***: SKI may contain potential mmP release inhibitory compounds. Salsola komarovii is a promising source of compounds against mmP and could be utilized in the developm.nt of antitumor agents.

Objective: To exam.ne whether Caulerpa okamurae ethanolic extract(COE) could inhibit obesitymediated inflammation, improve glucose metabolism and increase insulin sensitivity, using in vitro cell models of RAW 264.7 macrophages and 3 T3-L1 ***: We cocultured 3 T3-L1 adipocytes in direct contact with lipopolysaccharide-stimulated RAW264.7 macrophages and induced insulin resistance in 3 T3-L1 adipocytes with tumor necrosis factor-a(TnF-a) in the presence or absence of 250 mg/m L of COE. We investigated various markers of inflammation, glucose regulation and insulin sensitivity in these models using Griess reagent to measure nitric oxide(nO) production, 2-[n-(7-nitrobenz-2-oxa-1,3-diazol-4-yl) am.no]-2-deoxyglucose to measure glucose uptake, Western blot analysis to quantify protein expression and reverse transcriptasepolymerase chain reaction to evaluate m RnA ***: We found that COE(250 mg/m L) significantly inhibited the lipopolysaccharide-induced inflammatory response in RAW 264.7 macrophages by downregulating nO production, nitric oxide synthase 2 expression and nuclear translocation of nuclear factor-j B. COE also showed similar anti-inflammatory activity in coculture, along with decreased TnF-a, interleukin-6 and m.nocyte chemoattractant protein m RnA expression. In addition, COE also improved glucose uptake in coculture by upregulating glucose transporter-4(GLUT-4) and adiponectin and reducing serine phosphorylation of insulin receptor substrate-1(IRS1). In the TnF-a-induced insulin resistance model of 3 T3-L1 adipocytes, COE significantly improved both basal and insulin-stimulated glucose uptake, accompanied by phosphorylation of IRS1 at tyrosine 632, phospho-50 adenosine m.nophosphate-activated protein kinase a and glycogen synthase kinase-3 b(Ser9) as well as upregulation of ***: Together, these findings suggest that COE has potential to treat or prevent obesity-induced metabolic disorders.

AIm: To investigate the anti-inflammatory activities of the sem.n extract of Cuscuta chinensis Lam.(Cuscutae Sem.n; CS) on the production of inflammatory mediators, nitric oxide(nO), prostaglandin 2(PGE2), and proinflammatory cytokines in lipopolysaccharide(LPS)-stimulated BV-2 microglia. mETHOD: BV-2 cells were treated with CS extract for 30 m.n, and then stimulated with LPS or without for 24 h. The levels of nO, PGE2 and proinflammatory cytokines were measured by Griess assay and ELISA. The expression of inducible nitric oxide synthase(inOS), and cyclooxygenase(COX)-2 m.nA and protein was determ.ned by RT-PCR and Western blot, respectively. The phosphorylation of extracellular signal-regulated kinase 1/2(ERK1/2), Jun n-term.nal kinase(JnK), and p38 mitogen-activated protein kinase(mAPK), and the nuclear expression of nuclear factor(nF)-κB p65 were investigated by Western blot analysis. RESULTS: CS extract significantly decreased the production of nO and PGE2 by suppressing the expression of inOS and COX-2 in activated microglia. CS extract decreased the production of TnF-α, IL-1β, and IL-6 by down-regulating their transcription levels. In addition, CS extract suppressed the phosphorylation of ERK1/2, JnK, and p38 mAPK, and the nuclear translocation of nF-κB p65 in activated microglia. COnCLUSIOn: These results indicate that CS extract is capable of suppressing the inflammatory response by microglia activation, suggesting that CS extract has potential in the treatm.nt of brain inflammation.

In this study,we developed a simple screening procedure for the determ.nation of 18 anthelm.ntics(including benzimidazoles,macrocyclic lactones,salicylanilides,substituted phenols,tetrahydropyrimidines,and imidazothiazoles)in five animal-derived food matrices(chicken muscle,pork,beef,milk,and egg)using liquid chromatography-tandem mass *** were extracted using acetonitrile/1% acetic acid(milk and egg)and acetonitrile/1% acetic acid with 0.5 mL of distilled water(chicken muscle,pork,and beef),and purified using saturated n-hexane/acetonitrile.A reversed-phase analytical column and a mobile phase consisting of(A)10 mm amm.nium formate in distilled water and(B)methanol were used to achieve optimal chromatographic ***-matched standard calibration curves(R^(2)≥0.9752)were obtained for concentration equivalent to ×1/2,×1,×2,×3,×4,and×5 fold the maximum residue limit(mRL)stipulated by the Korean m.nistry of Food and Drug *** of 61.2e118.4%,with relative standard deviations(RSDs)of ≤19.9%(intraday and interday),were obtained for each sample at three spiking concentrations(×1/2,×1,and ×2 the mRL values).Limits of detection,limits of quantification,and matrix effects were 0.02e5.5 mg/kg,0.06e10 mg/kg,and -98.8 to 13.9%(at 20 μg/kg),*** five samples of each food matrix(chicken muscle,pork,beef,milk,and egg)purchased from large retailers in Seoul that were tested,none of the target analytes were *** has therefore been shown that this protocol is adaptable,accurate,and precise for the quantification of anthelm.ntic residues in foods of animal origin.

Objective： To investigate whether the dried root of Phellodendron amurense Ruprecht（Phellodendri cortex; PC） extract improves arthritic symptoms through anti-inflammatory and imm.ne-modulatory effects in collagen-induced arthritis in mice. methods： Rheumatoid arthritis（RA） was induced in male DBA/1 mice by imm.nization with type Ⅱ collagen（ColⅡ）. CIA mice were divided into 5 groups（n=10 per a group） with normal, CIA control, PC extract（50 mg/kg and 100 mg/kg）-treated, and meloxicam（50 mg/kg）-treated as the reference drug. The PC extract or meloxicam were adm.nistered orally in CIA mice once a day for 14 days after arthritis induction. Arthritic score, levels of anti-ColⅡ IgG2a antibody, prostaglandin E2（PGE2）, tumor necrosis factor（TnF）-α, and interleukin（IL）-17 in the sera of CIA mice were measured. Histopathological changes in the ankle joints of CIA mice were also analyzed by staining with hematoxylin and eosin（H and E）, safranin-O and imm.nohistochemistry using anti-TnF-α and anti-IL-17 antibodies. Results： The arthritic score was increased in CIA mice in a time-dependent m.nner, as were the serum levels of anti-ColⅡ IgG2a antibody, PGE2, TnF-α, and IL-17. However, the oral adm.nistration of PC extract at 50 and 100 mg/kg in CIA mice significantly decreased the arthritic scores, and the serum levels of anti-ColⅡ IgG2a, PGE2, TnF-α, and IL-17 compared with those in the CIA group（P〈0.05 or P〈0.01）. Furthermore, histopathological improvem.nt of the joint architecture in CIA mice was observed after adm.nistration of PC extract. PC extract also significantly inhibited the expression of TnF-α and IL-17 in the joints of CIA mice by suppressing the expression of their m RnA and proteins. Conclusion： PC extract may improve the pathological progression of RA through the inhibition of joint destruction by synovial inflammation and imm.ne-stimulation, therefore, it would be a potential anti-arthritic agent in RA.

Impact of UV-A Radiation on Erythemal UV and UV-index Estimation over Korea

Because total UV （TUV） in the UV-A region is 100 times higher than in the UV-B region, UV-A is a considerable compo- nent when calculating erythemal UV （EUV） and UV-index. The ratio of EUV to TUV in the UV-A value [EUV（A）/TUV（A）] is investigated to convert the EUV（A） from TUV（A） for broadband observation. The representative value of EUV（A）/TUV（A）, from the simulation study, is 6.9 × 10^- 4 changing from 6.1 ×10^-4 to 7.0 × 10^ -4 as aerosol optical depth, total ozone and solar zenith angle change. By adopting the observational data of EUV（B） and TUV（A） from UV-biometer measurem.nts at Yonsei University [（37.57°n, 126.95°E）, 84 m above sea level], the EUV irradiance increases to 15% of EUV（B） due to the consideration of EUV（A） from the data of TUV（A） observation. Compared to the total EUV observed from the Brewer spec- trophotometer at the same site, the EUV（B） from the UV-biometer observes only 95% of total EUV, and its underestimation is caused by neglecting the effect of UV-A. However, the sum of EUV（B） and EUV（A） [EUV（A＋B）] from two UV-biometers is 10% larger than the EUV from the Brewer spectrophotometer because of the spectral overlap effect in the range 320-340 nm. The correction factor for the overlap effect adjusts 8% of total EUV.
