OsWRKY65 enhances immunity against fungal and bacterial pathogens in rice

Diverse bacterial and fungal pathogens attack plants,causing biotic stress and severe yield losses globally.These losses are expected to become more serious as climate change improves conditions for many pathogens.Therefore,identifying genes conferring broad-spectrum disease resistance and elucidating their underlying mechanisms provides important resources for plant breeding.WRKY transcription factors affect plant growth and stress responses.However,the functions of many WRKY proteins remain to be elucidated.Here,we demonstrated the role of rice(Oryza sativa)WRKY groupⅢtranscription factor OsWRKY65 in immunity.OsWRKY65 localized to the nucleus and acted as transcriptional repressor.Genetic and molecular functional analyses showed that OsWRKY65 increases resistance to the fungal pathogen Fusarium fujikuroi through downregulation of GA signaling and upregulation of JA signaling.Moreover,OsWRKY65 modulated the expression of the key genes that confer susceptibility or resistance to Xanthomonas oryzae pv.oryzae to enhance immunity against the pathogen.In particular,OsWRKY65directly bound to the promoter region of OsSWEET13 and repressed its expression.Taken together,our findings demonstrate that the OsWRKY65 enhances resistance to fungal and bacterial pathogens in rice.

Background Genome editing has been considered as powerful tool in agricultural fields.However,genome editing progress in cattle has not been fast as in other mammal species,for some disadvantages including long gestational periods,single pregnancy,and high raising cost.Furthermore,technically demanding methods such as microinjection and somatic cell nuclear transfer(SCNT)are needed for gene editing in cattle.In this point of view,electroporation in embryos has been risen as an alternative.Results First,editing efficiency of our electroporation methods were tested for embryos.Presence of mutation on embryo was confirmed by T7E1 assay.With first combination,mutation rates for MSTN and PRNP were 57.6%±13.7%and 54.6%±13.5%,respectively.In case of MSTN/BLG,mutation rates were 83.9%±23.6%for MSTN,84.5%±18.0%for BLG.Afterwards,the double-KO embryos were transferred to surrogates and mutation rate was identified in resultant calves by targeted deep sequencing.Thirteen recipients were transferred for MSTN/PRNP,4 calves were delivered,and one calf underwent an induction for double KO.Ten surrogates were given double-KO embryos for MSTN/BLG,and four of the six calves that were born had mutations in both genes.Conclusions These data demonstrated that production of genome edited cattle via electroporation of RNP could be effectively applied.Finally,MSTN and PRNP from beef cattle and MSTN and BLG from dairy cattle have been born and they will be valuable resources for future precision breeding.

近年来,经自然腔道取标本手术(NOSES)治疗结直肠癌已引起了广泛关注。NOSES应用于结直肠手术的潜在优势包括减轻术后疼痛,减少伤口并发症,减少对术后镇痛的需求,加快肠道功能的恢复,缩短住院时间,更好的美容效果,以及给患者带来了更佳的心理状态。尽管NOSES的手术创伤显著减轻,然而该技术也存在一些潜在问题。尤其随着这项新技术的兴起,人们对其无菌的把控、肿瘤学结果和病例选择等问题表示出关切。因此,迫切需要一部操作指南来规范NOSES手术在结直肠癌中的开展。经过国际NOSES联盟全体成员的三轮讨论,最终形成了该部共识。这一共识有助于NOSES在全球范围内的长远发展。

Effect of organic mineral supplementation in reducing oxidative stress in Holstein calves during short‑term heat stress and recovery conditions

Background This study investigated the effects of inorganic and organic minerals on physiological responses,oxidative stress reduction,and rumen microbiota in Holstein bull calves(123.81±9.76 kg;5 months old)during short-term heat stress(HS)and recovery periods.Eight Holstein calves were randomly assigned to four treatment groups:no mineral supplementation(Con),inorganic minerals(IM),organic minerals(OM),and high-concentration organic minerals(HOM)and two thermal environments(HS and recovery)using 4×2 factorial arrangement in a crossover design of four periods of 35 d.Calves were maintained in a temperature-controlled barn.The experimental period consisted of 14 d of HS,14 d of recovery condititon,and a 7-d washing period.Results Body temperature and respiration rate were higher in HS than in the recovery conditions(P0.05).The principal coordinate analysis represented that the overall rumen microbiota was not influenced by mineral supplementation;however,temperature-induced microbial structure shifts were indicated(PERMANOVA:P<0.05).At the phylum level,Firmicutes and Actinobacteria decreased,whereas Fibrobacteres,Spirochaetes,and Tenericutes increased(P<0.05),under HS conditions.The genus Treponema increased under HS conditions,while Christensenella was higher in recovery conditions(P<0.05).Conclusion HOM supplementation during HS reduced cor

Interworking between Modbus and internet of things platform for industrial services

In the era of rapid development of Internet of Things(IoT),numerous machine-to-machine technologies have been applied to the industrial domain.Due to the divergence of IoT solutions,the industry is faced with a need to apply various technologies for automation and control.This fact leads to a demand for an establishing interworking mechanism which would allow smooth interoperability between heterogeneous devices.One of the major protocols widely used today in industrial electronic devices is Modbus.However,data generated by Modbus devices cannot be understood by IoT applications using different protocols,so it should be applied in a couple with an IoT service layer platform.oneM2M,a global IoT standard,can play the role of interconnecting various protocols,as it provides flexible tools suitable for building an interworking framework for industrial services.Therefore,in this paper,we propose an interworking architecture between devices working on the Modbus protocol and an IoT platform implemented based on oneM2M standards.In the proposed architecture,we introduce the way to model Modbus data as oneM2M resources,rules to map them to each other,procedures required to establish interoperable communication,and optimization methods for this architecture.We analyze our solution and provide an evaluation by implementing it based on a solar power management use case.The results demonstrate that our model is feasible and can be applied to real case scenarios.

A comparative study on kinetics and dynamics of two dump truck lifting mechanisms using MATLAB simscape

In this paper,two lifting mechanism models with opposing placements,which use the same hydraulic hoist model and have the same angle of 50°,have been developed.The mechanical and hydraulic simulation models are established using MATLAB Simscape to analyze their kinetics and dynamics in the lifting and holding stages.The simulation findings are compared to the analytical calculation results in the steady state,and both methods show good agreement.In the early lifting stage,Model 1 produces greater force and discharges goods in the container faster than Model 2.Meanwhile,Model 2 reaches a higher force and ejects goods from the container cleaner than its counterpart at the end lifting stage.The established simulation models can consider the effects of dynamic loads due to inertial moments and forces generated during the system operation.It is crucial in studying,designing,and optimizing the structure of hydraulic-mechanical systems.

Integrative multi-omics approaches reveal that Asian cultivated rice domestication influences its symbiotic relationship with arbuscular mycorrhizal fungi

Potential changes in the symbiotic relationship between rice(Oryza sativa)and microorganisms have occurred during the domestication of Asian cultivated rice(O.sativa)from common wild rice(Oryza rufipogon)and in response to global climate change,along with evolving adaptations to the environment.The potential genes may express differently or dominate the symbiotic relationships between arbuscular mycorrhizal fungi(AMF)and plants,which may be beneficial to rice breeding.To date,research on this important topic has been limited.In this study,we aimed to examine the symbiotic relationships of Asian common wild and cultivated rice species with AMF.By conducting a comparative metagenomic analysis of the rhizospheres of wild and cultivated rice species,we identified differences in Rhizophagus intraradices-related genes associated with wild and cultivated rice,as well as functional genes of AMF.Furthermore,we obtained root-related genes associated with AMF from transcriptome data of rice roots.Our results collectively suggest that R.intraradices-related genes in the rhizosphere of wild rice may be more conducive to its colonization.Additionally,bacteria from the Nitrosomonadaceae and Nitrospiraceae families identified in the rhizosphere of wild rice exhibited positive correlations with R.intraradices-related genes with protein identifiers 1480749 and1871253,which may indicate that nitrobacteria can enhance the functions of R.intraradices in association with wild rice.Next,in a case study using comparative transcriptome analysis of root samples obtained from R.intraradices-inoculated wild and cultivated rice plants,we found significantly higher expression levels of the strigolactone pathway-related genes DWARF3(D3)and DWARF14(D14)in R.intraradices-inoculated common wild rice than in R.intraradices-inoculated cultivated rice.This study provides a theoretical basis for identifying the effects of domestication on mycorrhizal symbiosis-related genes,which could be promoted in wild rice in the future.

Vomiting-induced pharyngeal perforation during bowel preparation for colonoscopy:A case report

BACKGROUND Effective bowel cleansing is essential for a successful colonoscopy.Laxatives,such as polyethylene glycol,are commonly used for bowel preparation.Vomiting is a frequent complication during bowel preparation,and forceful vomiting can potentially lead to esophageal perforation,as reported in several previous cases.However,pharyngeal perforation during bowel preparation has not been previously documented.Here,we present a case of pharyngeal perforation induced by forceful vomiting during bowel preparation.CASE SUMMARY A 38-year-old man with a history of hypertension,dyslipidemia,diabetes mellitus,and end-stage renal disease on hemodialysis was admitted for evaluation of recurrent abdominal pain.The patient complained of sudden pain in the neck,throat,and anterior chest following forceful vomiting during bowel preparation.Physical examination revealed crepitus under the skin of the neck and anterior chest on palpation,and upper gastrointestinal endoscopy revealed pharyngeal perforation.The perforation site was located above the upper esophageal sphincter,which distinguished it from Boerhaave’s syndrome.Conservative medical management was chosen after consultation with a thoracic surgeon and an otolaryngologist,considering the patient's mild symptoms,stable vital signs,and the small size of the lesion;the perforation resolved without endoscopic or surgical intervention.The patient was discharged from hospital two weeks after the perforation.CONCLUSION Despite its rarity,pharyngeal perforation should be considered a potential complication of bowel preparation for colonoscopy.

Velocity equation for grenades while impacting on dry sand media

This paper deals with the collision of sphere shape grenades with sand media.The central issue of the article is the establishing of an empirical velocity equation of the grenade while impacting on sand that is used to solve motion equations of the mechanical mechanism inside the impact grenade fuze.The paper focuses on impact velocities that are lower than 5 m s^(-1).An experiment was conducted to study the velocity of the grenade while impacting on dry sand.A high-speed camera video was used to capture the grenade positions.The grenade velocity in the impact process was generated from these video data.Some types of fitting curves are used to regress the velocity equation of the grenade while interacting with the sand media and the best-fitting model is chosen.The result shows the regression curve has a high correlation with the experiment data for grenade velocities below 5 m s^(-1).The received regression equation is useful for analyzing the working ability of the inertial mechanism inside the impact grenade or analyzing and choosing the appropriate parameters of each part in the inertial mechanism to meet the required characteristics of the mechanism.

Rare-earth silicates are promising environmental barrier coatings(EBCs)that can protect SiC_(f)/Si C_(m)substrates in next-genera tion gas turbine blades.Notably,RE_(2)Si_(2)O_(7)(RE=Yb and Ho)shows potential as an EBC due to its coefficient of thermal expansion(CTE)compatible with substrates and high resistance to water vapor corrosion.The target operating temperature for next-generation tur bine blades is 1400°C.Corrosion is inevitable during adhesion to molten volcanic ash,and thus,understanding the corrosion behavior o the material is crucial to its reliability.This study investigates the high-temperature corrosion behavior of sintered RE_(2)Si_(2)O_(7)(RE=Yb and Ho).Samples were prepared using a solid-state reaction and hot-press method.They were then exposed to volcanic ash at 1400°C for 224,and 48 h.After 48 h of exposure,volcanic ash did not react with Yb_(2)Si_(2)O_(7)but penetrated its interior,causing damage.Meanwhile Ho_(2)Si_(2)O_(7)was partially dissolved in the molten volcanic ash,forming a reaction zone that prevented volcanic ash melts from penetrating the interior.With increasing heat treatment time,the reaction zone expanded,and the thickness of the acicular apatite grains increased The Ca:Si ratios in the residual volcanic ash were mostly unchanged for Yb_(2)Si_(2)O_(7)but decreased considerably over time for Ho_(2)Si_(2)O_(7).Th Ca in volcanic ash was consumed and formed apatite,indicating that RE^(3+)ions with large ionic radii(Ho>Yb)easily precipitated apatit from the volcanic ash.
