Anti-scarring effects of butaprost on human subconjunctival Tenon's fibroblasts

AIM： To investigate the toxicity of the E-prostanoid 2（EP2） receptor agonist, butaprost against human subconjunctival（Tenon＇s capsule） fibroblasts, and to determine the underlying mechanism. METhoDS： We isolated Tenon＇s fibroblasts from the subconjunctival area of healthy subjects and evaluated the types of EP receptors expressed using quantitative realtime reverse transcription polymerase chain reaction（RTPCR）. The toxicity of butaprost against the fibroblasts was evaluated using methyl thiazolyl tetrazolium and lactic dehydrogenase assays. The inhibition of conjunctival fibroblast proliferation by butaprost was assessed by measuring α-actin levels. The underlying mechanism was assessed by measuring intracellular cyclic adenosine monophosphate（c AMP） levels. Intergroup differences were statistically analyzed using an independent t-test. Densitometry of the Western blot bands was performed using the Image J software. RESULTS： Quantitative real-time RT-PCR revealed that the fibroblast EP2 receptor levels were higher than those of the other EP receptors. Butaprost did not show toxicity against Tenon＇s tissue, but inhibited conjunctival fibroblast proliferation by reducing collagen synthesis. EP2 receptor activation enhanced the c AMP cascade, which might be an important mechanism underlying this ***： Butaprost effectively reduces the subconjunctival scarring response. Given the significanceof wound healing modulation in blebs, butaprost＇s inhibitory effect on subconjunctival Tenon＇s fibroblasts may be beneficial in managing postoperative scarring in glaucoma surgery.

ASL Recognition by the Layered Learning Model Using Clustered Groups

American Sign Language(ASL)images can be used as a communication tool by determining numbers and letters using the shape of the ***,ASL can have an key role in communication for hearing-impaired persons and conveying information to other persons,because sign language is their only channel of *** ASL recognition methods primarily adopt images,sensors,and pose-based recognition techniques,and employ various gestures together with *** study briefly reviews these attempts at ASL recognition and provides an improved ASL classification model that attempts to develop a deep learning method with *** the proposed model,the collected ASL images were clustered based on similarities in shape,and clustered group classification was first performed,followed by reclassification within the *** experiments were conducted with various groups using different learning layers to improve the accuracy of individual image *** selecting the optimized group,we proposed a meta-layered learning model with the highest recognition rate using a deep learning method of image *** proposed model exhibited an improved performance compared with the general classification model.

Cyberattacks on the Industrial Control System(ICS)have recently been increasing,made more intelligent by advancing *** such,cybersecurity for such systems is attracting *** a core element of control devices,the Programmable Logic Controller(PLC)in an ICS carries out on-site control over the ICS.A cyberattack on the PLC will cause damages on the overall ICS,with Stuxnet and Duqu as the most representative ***,cybersecurity for PLCs is considered essential,and many researchers carry out a variety of analyses on the vulnerabilities of PLCs as part of preemptive efforts against *** this study,a vulnerability analysis was conducted on the XGB *** vulnerabilities were identified by analyzing the network protocols and memory structure of PLCs and were utilized to launch replay attack,memory modulation attack,and FTP/Web service account theft for the verification of the *** on the results,the attacks were proven to be able to cause the PLC to malfunction and disable it,and the identified vulnerabilities were defined.

BACKGROUND： Thyroid hormones modulate proliferation of astrocytes and microglia depending on maturation stage and localization. Studies have demonstrated that triiodothyronine treatment or thyroidectomy during developmental stages results in morphological alterations and changes in the number of astrocytes and microglia. Little is known about the effects of hypothyroidism on astrocytes and microglia in adults. OBJECTIVE： To investigate the effects of hypothyroidism on morphology and number of astrocytes and microglia in the adult rat hippocampus. DESIGN, TIME AND SETTING： A randomized, controlled, neuroendocrinological, animal study was performed at the College of Medicine, Hallym University, South Korea between May 2008 and April 2009. MATERIALS： Methimazole, rabbit anti-glial fibrillary acidic protein （GFAP） antiserum, and rabbit anti-lba-1 antiserum were purchased from Sigma, USA. Rabbit anti-GFAP polyclonal antibody was provided by Chemicon, USA. Rabbit anti-lba-1 polyclonal antibody was purchased from Wako, Japan. Terminal deoxynucleotidyl transferase dUTP-biotin nick-end-labeling （TUNEL） kit was provided by Roche Molecular Biochemicals, Mannheim, Germany. METhoDS： Hypothyroidism was induced in Wistar rats via methimazole administration （0.025%） in drinking water for 5 weeks, starting at 6 months of age. MAIN OUTCOME MEASURES： Following methimazole treatment, hippocampai neuronal death was determined using TUNEL staining. The morphology and number of GFAP and lba-1 immunoreactive cells were detected by immunohistochemistry. Hippocampal GFAP and lba-1 protein levels were detected by Western blot analysis. Serum-free triiodothyronine and thyroxine levels were quantified. RESULTS： TUNEL-positive neurons were not observed in the hippocampus of euthyroid and hypothyroid rats. Reasons for Calling Doctor： Visualizing Callers＇ Multi-unit Turns and Filming Television Call-ins as Institutional Interaction

This study describes discursive and visual features of telephone openings to a live broadcast television call-in program based on the analysis of the ＂multi-unit reason-for-the-call turn＂（Couper-Kuhlen, 2001）. The data were based on video-recordings of the call-in program broadcast daily on Hakka channel in Taiwan, in which callers call to seek advice on health problems. The discourse analysis shows that callers＇＂multi-unit reason-for-the-call turns＂ can be classified into three forms including request, epistemic frame and descriptions of symptoms, and telling of lifestyle. The visual analysis demonstrates that the visual shots of ＂multi-unit reason-for-the-call turns＂ orient viewers to the studio scene in which the invited guest, usually the medical doctor, can be seen listening to callers＇ upcoming questions. Based on the analyses and an interview with the camera director of this television program, the discussion sheds light on how television production practices use callers＇ multi-unit turns as a resource for switching to the studio shot, which helps to build an image of the expert and his/her role and function in the advice-seeking program. In this sense, filming and television production practices rely on linguistic resources and the visualization of callers＇ multi-unit turns has meanings in the organization of studio interaction as ＂institutional interaction＂（Drew ＆ Heritage, 1992）, being shaped by interactants＇ identities, roles, and tasks relevant for the specific institutional needs or purposes.

Induction of vascular endothelial growth factor receptor expression in human umbilical vein endothelial cells after repeated bevacizumab treatment in vitro

AIM： To investigate the mechanism underlying the loss of responsiveness to anti-vascular endothelial growth factor（VEGF） treatment after repeated injections for choroidal neovascularization, VEGF and VEGF receptor（VEGFR） expressions were evaluated following repeated bevacizumab treatments in hypoxic human umbilical vein endothelial cells（HUVECs） in ***： HUVECs were incubated under hypoxic conditions in two media of different bevacizumab concentrations（1.0 or 2.5 mg/m L） for 17 h, and then in a new medium without bevacizumab for 7h. This procedure was repeated twice more. A culture with an identical volume of excipients served as the control. Cytotoxicity and cell proliferation were assessed using 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide and Ki-67 assays, respectively. Levels of VEGF and VEGFR were assessed using enzyme-linked immunosorbent assay and Western blot ***： Cytotoxic effects were not reported for either bevacizumab concentration. Cell proliferation was not reduced after anti-VEGF treatments. VEGF level after single treatment was significantly higher than that of the control and after repeated treatments. Phosphorylated VEGFR-2 expression increased significantly after singleand repeated bevacizumab treatments compared with the control. The 1.0 mg/m L bevacizumab induced significantly higher expressions of VEGFR-2 than the 2.5 mg/m L in single and repeated treatment ***： Bevacizumab treatment of HUVECs elevated VEGFR expression in both single and repeated treatments, indicating a mechanism for the reduced efficacy of anti-VEGF therapy in ocular neovascular disorders.

Programmable Logic Controller Block Monitoring System for Memory Attack Defense in Industrial Control Systems

Cyberattacks targeting industrial control systems(ICS)are becoming more sophisticated and advanced than in the past.A programmable logic controller(PLC),a core component of ICS,controls and monitors sensors and actuators in the ***,PLC has memory attack threats such as program injection and manipulation,which has long been a major target for attackers,and it is important to detect these attacks for ICS *** detect PLC memory attacks,a security system is required to acquire and monitor PLC memory *** addition,the performance impact of the security system on the PLC makes it difficult to apply to the *** address these challenges,this paper proposes a system to detect PLC memory attacks by continuously acquiring and monitoring PLC *** proposed system detects PLC memory attacks by acquiring the program blocks and block information directly from the same layer as the PLC and then comparing them in bytes with previous *** with Siemens S7-300 and S7-400 PLC were conducted to evaluate the PLC memory detection performance and performance impact on *** experimental results demonstrate that the proposed system detects all malicious organization block(OB)injection and data block(DB)manipulation,and the increment of PLC cycle time,the impact on PLC performance,was less than 1 *** proposed system detects PLC memory attacks with a simpler detection method than earlier ***,the proposed system can be applied to ICS with a small performance impact on PLC.
