Deep learning-based virtual staining,segmentation,and classification in label-free photoacoustic histology of human specimens

In pathological diagnostics,histological images highlight the oncological features of excised specimens,but they require laborious and costly staining *** recent innovations in label-free microscopy that simplify complex staining procedures,technical limitations and inadequate histological visualization are still problems in clinical ***,we demonstrate an interconnected deep learning(DL)-based framework for performing automated virtual staining,segmentation,and classification in label-free photoacoustic histology(PAH)of human *** framework comprises three components:(1)an explainable contrastive unpaired translation(E-CUT)method for virtual H&E(VHE)staining,(2)an U-net architecture for feature segmentation,and(3)a DL-based stepwise feature fusion method(StepFF)for *** framework demonstrates promising performance at each step of its application to human liver *** virtual staining,the E-CUT preserves the morphological aspects of the cell nucleus and cytoplasm,making VHE images highly similar to real H&E *** segmentation,various features(e.g.,the cell area,number of cells,and the distance between cell nuclei)have been successfully segmented in VHE ***,by using deep feature vectors from PAH,VHE,and segmented images,StepFF has achieved a 98.00%classification accuracy,compared to the 94.80%accuracy of conventional PAH *** particular,StepFF’s classification reached a sensitivity of 100%based on the evaluation of three pathologists,demonstrating its applicability in real clinical *** series of DL methods for label-free PAH has great potential as a practical clinical strategy for digital pathology.

Pancreatic pseudocystocolonic fistula treated without surgical or endoscopic intervention

We report here a case of pancreatic pseudocystocolic fistula that was treated without surgical or endoscopic intervention.A 76-year-old woman,presenting with a fever and epigastric pain,was referred to our *** months prior to this admission,the patient had been admitted to the hospital for acute *** computerized tomography(CT)revealed a 9 cm pseudocyst containing air,and a fistular opening was observed via *** colonoscopy,the abdominal pain was slightly improved,the fever subsided and laboratory results showed decreased C-reactive protein *** observed improvement was likely due to the cleansing of the bowel,which induced spontaneous drainage from the pseudocyst into the *** therapy was administered and daily bowel cleansing was performed using a polyethylene glycol *** three weeks,a follow-up CT revealed that the size of the pseudocyst had decreased significantly from 9 to 5.3 *** addition,laboratory tests were *** patient was able to resume a normal diet and was discharged in good overall health from the hospital,without aggravation of the symptoms.A colonoscopy performed 3 mo later and a follow-up CT performed 6 mo later confirmed that both the fistula and pseudocyst had completely disappeared.

Expression of micro RNAs in fibroblast of pterygium

AIM：To screen microRNAs（miRNAs） and set up target miRNAs in ***：Primary fibroblasts were isolated from pterygium and Tenon＇s capsule and *** analysis and Western blotting were performed to confirm the culture of *** all,1733 miRNAs were screened in the first step by using GeneChip miRNA3.0 *** miRNAs involved in the pathogenesis of pterygium were subsequently determined using the following criteria：1） high reproducibility in a repetitive test;2） base log value of 〉7.0 for both control and pterygial fibroblasts;and 3） log ratio of 〉1.0 between pterygial fibroblasts and control ***：Primary screening showed that 887/1733 miRNAs were up-regulated and 846/1733 miRNAs were down-regulated in pterygial fibroblasts compared with those in control *** the 1733 miRNAs screened,4 miRNAs,namely,miRNA-143a-3p,miRNA-181a-2-3p,miRNA-377-5p and miRNA-411a-5p,met the above-mentioned *** screening showed that these 4 miRNAs were up-regulated in pterygial fibroblasts compared with control fibroblasts and that miRNA-143a-3p had the highest mean ratio compared with the miRNAs in control ***：miRNA-143a-3p,miRNA-181a-2-3p,miRNA-377-5p and miRNA-411a-5p are up-regulated in pterygial fibroblasts compared with control fibroblasts,suggesting their involvement in the pathogenesis of pterygium.

Beta-nerve growth factor gene therapy alleviates pyridoxine-induced neuropathic damage by increasing doublecortin and tyrosine kinase A in the dorsal root ganglion

Beta-nerve growth factor(β-NGF) is known to be a major leading cause of neuronal plasticity. To identify the possible action mechanisms of β-NGF gene therapy for sciatic nerve recovery, experimental dogs were randomly divided into control, pyridoxine, and pyridoxine + β-NGF groups. We observed chronological changes of morphology in the dorsal root ganglia in response to pyridoxine toxicity based on cresyl violet staining. The number of large neurons positive for cresyl violet was dramatically decreased after pyridoxine intoxication for 7 days in the dorsal root ganglia and the neuron number was gradually increased after pyridoxine withdrawal. In addition, we also investigated the effects of β-NGF gene therapy on neuronal plasticity in pyridoxine-induced neuropathic dogs. To accomplish this, tyrosine kinase receptor A(TrkA), βIII-tubulin and doublecortin(DCX) immunohistochemical staining was performed at 3 days after the last pyridoxine treatment. TrkA-immunoreactive neurons were dramatically decreased in the pyridoxine group compared to the control group, but strong TrkA immunoreactivity was observed in the small-sized dorsal root ganglia in this group. TrkA immunoreactivity in the dorsal root ganglia was similar between β-NGF and control groups. The numbers of βIII-tubulin-and DCX-immunoreactive cells decreased significantly in the pyridoxine group compared to the control group. However, the reduction of βIII-tubulin-and DCX-immunoreactive cells in the dorsal root ganglia in the β-NGF group was significantly ameliorated than that in the pyridoxine group. These results indicate that β-NGF gene therapy is a powerful treatment of pyridoxine-induced neuropathic damage by increasing the TrkA and DCX levels in the dorsal root ganglia. The experimental protocol was approved by the Institutional Animal Care and Use Committee(IACUC) of Seoul National University, South Korea(approval No. SNU-060623-1, SNU-091009-1) on June 23, 2006 and October 9, 2009, respectively.

A feasible modified biopsy method for tissue diagnosis of gastric subepithelial tumors

AIM: To evaluate the diagnostic yield and safety of a modified technique for the histological diagnosis of subepithelial tumors (SETs). METHODS: A retrospective review of patients who underwent a modified technique for the histological diagnosis of gastric SETs, consisting of a mucosal incision with a fixed flexible snare (MIF) and deep-tissue biopsy under conventional endoscopic view, from January 2012 to January 2013 was performed. Eleven patients with gastric SETs 10-30 mm in diameter and originating from the third or fourth layer on endoscopic ultrasonography were included. RESULTS: The mean age was 59.8 (range, 45-76) years, and 5 patients were male. The mean size of the SETs was 21.8 (range, 11-30) mm. The number of biopsy specimens was 6.3 (range 5-8). The mean procedure time was 9.0 min (range, 4-17 min). The diagnostic yield of MIF biopsies was 90.9% (10/11). The histological diagnoses were leiomyoma (4/11, 36.4%), aberrant pancreas (3/11, 27.3%), gastrointestinal stromal tumors (2/11, 18.2%), an inflammatory fibrinoid tumor (1/11, 9.1%); one result was non-diagnostic (1/11, 9.1%). There were six mesenchymal tumors; the specimens obtained in each case were sufficient for an immunohistochemical diagnosis. There was no major bleeding, but one perforation occurred that was successfully controlled by endoscopic clipping. CONCLUSION: The MIF biopsy was simple to perform, safe, and required a shorter procedure time, with a high diagnostic yield for small SETs.

Development of a skill assessment tool for the Korea operational oceanographic system

A standard skill assessment （SA） tool was developed and implemented to evaluate the performance of op- erational forecast models in the Korea operational oceanographic system. The SA tool provided a robust way to assess model skill in the system by comparing predictions and observations, and involved the com- putation of multiple skill metrics including correlation and error skills. User- and system-based acceptance criteria of skill metrics were applied to determine whether predictions were acceptable for the system. To achieve this, the tool produced a time series comparison plot, a skill score table, and an advanced sum- marized diagram to effectively demonstrate the multiple skill scores. Moreover, the SA was conducted to evaluate both atmospheric and hydrodynamic forecast variables. For the atmospheric variables, acceptable error criteria were preferable to acceptable correlation criteria over short timescales, since the mean square error overwhelmed the observation variance. Conversely, for the hydrodynamic variables, acceptable root mean square percentage error （e.g., perms） criteria were preferable to acceptable error （e.g., erms） criteria owing to the spatially variable tidal intensity around the Korean Peninsula. Furthermore, the SA indicated that predetermined acceptance error criteria were appropriate to satisfy a target central frequency （fc） for which errors fell within the specified limits （i.e., the .fc equals 70%）.

In multicellular organisms, controlling the timing and extent of asymmetric cell divisions （ACDs） is crucial for correct patterning. During post-embryonic root development in Arabidopsis thaliana, ground tissue （GT） maturation involves an additional ACD of the endodermis, which generates two different tissues： the endo- dermis （inner） and the middle cortex （outer）. It has been reported that the abscisic acid （ABA） and gibberellin （GA） pathways are involved in middle cortex （MC） formation. However, the molecular mechanisms under- lying the interaction between ABA and GA during GT maturation remain largely unknown. Through transcriptome analyses, we identified a previously uncharacterized C2H2-type zinc finger gene, whose expression is regulated by GA and ABA, thus named GAZ （GA- AND ABA-RESPONSIVE ZINC FINGER）. Seedlings ectopically overexpressing GAZ （GAZ-OX） were sensitive to ABA and GA during MC formation, whereas GAZ-SRDX and RNAi seedlings displayed opposite phenotypes. In addition, our results indicated that GAZ was involved in the transcriptional regulation of ABA and GA homeostasis. In agreement with pre- vious studies that ABA and GA coordinate to control the timing of MC formation, we also confirmed the unique interplay between ABA and GA and identified factors and regulatory networks bridging the two hor- mone pathways during GT maturation of the Arabidopsis root.

A cobalt-silica hybrid nanocatalyst bearing small cobalt particles of diameter -5 nm was prepared through a hydrothermal reaction and hydrogen reduction. The resulting material showed very high CO conversion （〉82%） and high hydrocarbon productivity （-1.0gHC·gcat^-1·h^-1 ） with high activity （-8.5 × 10^-5 ***^-1.s-1） in the Fischer-Tropsch synthesis reaction.

Effect of the herbal medicine Danggui Sini plus Wuzhuyu Shengjiang Tang on erythrocyte deformability in normal subjects: a cross-over trial

OBJECTIVE: To show whether Danggui Sini plus Wuzhuyu Shengjiang Tang(DSWST) has any transient effect on erythrocyte deformability in normal ***: A total of 25 subjects [mean age(27.8± 1.8) years] was enrolled in this study. The study was designed as a cross-over trial in which the subjects took part for 2 d. On the first day, blood samples were collected at baseline and 1-2 h after administration of water, whereas, on the second day, instead of water, the subjects were administered DSWST after the baseline blood sampling. The blood samples collected at baseline and after the administration water or DSWST, were examined for erythrocyte ***: The elongation index increased significantly after 2 h(P = 0.009) compared to the baseline after DSWST intake. However, after water intake, there was no significant differenceobserved. When comparing the percent change of erythrocyte deformability between DSWST and water, we found that after 2 h of administration,DSWST improved erythrocyte deformability significantly compared to water(P < 0.001).CONCLUSION: DSWST has a transient effect on erythrocyte deformability in normal subjects.
