Association between hepatocellular carcinoma and tumor necrosis factor alpha polymorphisms in South Korea

AIM: To investigate associations between the tumor necrosis factor alpha(TNF-α)-1031 T>C,-863 C>A,-857 C>T,-308 G>A,and-238 G>A polymorphisms and HCC in ***: Hepatocellular carcinoma(HCC) cases were diagnosed at CHA Bundang Medical Center from June 1996 to August 2008. The association between TNF-α polymorphisms and HCC was analyzed in 157HCC patients and 201 controls using a polymerase chain reaction-restriction fragment length polymorphism assay. We investigated five TNF-α polymorphisms,which are TNF-α-1031 T>C,-863 C>A,-857 C>T,-308 G>A,and-238 G>A. The TNF-α genotype frequencies,genotype combinations and haplotypes were analyzed to disclose the association with ***: None of the TNF-α polymorphisms was significantly associated with HCC. However,nine genotype combinations had associations with increased likelihood of HCC. Among them,TNF-α-1031/-857/-238 TT/CC/GA(AOR = 18.849,95%CI: 2.203-161.246,P = 0.007),TNF-α-1031/-308/-238 TT/GG/GA(AOR = 26.956,95%CI: 3.071-236.584,P = 0.003),and TNF-α-1031/-238 TT/GA(AOR = 21.576,95%CI: 2.581-180.394,P = 0.005) showed marked association with HCC. There were five haplotypes of TNF-α polymorphisms which were significantly associated with HCC. They are TNF-α-1031/-863/-857/-308/-238 T-C-C-G-A(OR = 25.824,95%CI: 1.491-447.223,P = 0.0005),TNF-α-1031/-857/-308/-238 T-C-G-A(OR = 12.059,95%CI: 2.747-52.950,P < 0.0001),TNF-α-1031/-857/-238 T-C-A(OR = 10.696,95%CI: 2.428-47.110,P = 0.0001),TNF-α-1031/-308/-238 T-G-A(OR = 7.556,95%CI: 2.173-26.280,P = 0.0002) and TNF-α-1031/-238 T-A(OR = 10.865,95%CI: 2.473-47.740,P = 0.0001). Moreover,HCC Okuda stage Ⅲ cases with the TNF-α-1031 CC genotype had better survival than those with the TT genotype(AOR = 5.795,95%CI: 1.145-29.323). CONCLUSION: Although no single TNF-α polymorphism is associated with HCC in this study,some TNF-α genotype combinations and haplotypes are associated with HCC. Identification of differentially expressed miRNAs associated with chronic kidney disease-mineral bone disorder

The purpose of this study is to characterize a meta-signature of differentially expressed mRNA in chronic kidney disease （CKD） to predict putative microRNA （miRNA） in CKD-mineral bone disorder （CKD- MBD） and confirm the changes in these genes and miRNA expression under uremic conditions by using a cell culture system. PubMed searches using MeSH terms and keywords related to CKD, uremia, and mRNA arrays were conducted. Through a computational analysis, a meta-signature that characterizes the significant intersection of differentially expressed mRNA and expected miRNAs associated with CKD-MBD was determined. Additionally, changes in gene and miRNA expressions under uremic conditions were confirmed with human Saos-2 osteoblast-like cells. A statistically significant mRNA meta-signature of upregulated and downregulated mRNA levels was identified. Furthermore, miRNA expression profiles were inferred, and computational anaIyses were performed with the imputed mieroRNA regulation based on weighted ranked expression and putative microRNA targets （IMRE） method to identify miRNAs associated with CKD occurrence. TLR4 and miR-146b levels were significantly associated with CKD-MBD. TLR4 levels were significantly downregulated, whereas pri- miR-146b and miR-146b were upregulated in the presence of uremic toxins in human Saos-2 osteoblast-like cells. Differentially expressed miRNAs associated with CKD-MBD were identified through a computational analysis, and changes in gene and miRNA expressions were confirmed with an in vitro cell culture system.

Diel vertical migration of the copepod Calanus sinicus before and during formation of the Yellow Sea Cold Bottom Water in the Yellow Sea

To understand the effects of the Yellow Sea Cold Bottom Water （YSCBW） on the diel vertical migration （D- VM） of the copepod Calanus sinicus, we surveyed vertical distribution of C. sinicus at a fixed station in the Yellow Sea before （spring） and during （summer） formation of the YSCBW. Cold water （〈10℃） was observed in the bottom layer when the water column was thermally stratified in summer, but the water column was thermally well-mixed in spring 2010. Samples were collected from five different layers at 3-h intervals using an opening-closing net. Adult females （1-155 ind./m3） showed a clear normal DVM pattern throughout the entire water column in spring, whereas adult males did not migrate. DVM of copepodite V （CV） individuals was not clear, but the maximum abundance of CI-CIV occurred consistently in the upper 10-20 m layer, where there was a high concentration of chlorophyll-a （Chl-a）（0.49-1.19μg/L）. In summer, weak DVM was limited to cold waters beneath the thermocline for adult females （〈30 ind./m3）, but not for adult males. The maximum abundance of CI-CIV also occurred consistently in the subsurface layer （20-40 m） together with high concentrations of Chl-a （0.81-2.36 μg/L）. CV individuals （1-272 ind./m3） moved slightly upward noc- turnally to the near-surface layer （10-20 m）, where the average temperature was 25.74℃, but they were not found in the surface layer （0-10 m; 28.31℃）. These results indicate that the existence of the YSBCW affected food availability at depth and the vertical temperature distribution, leading to variation in the amplitude and shape of stage-specific vertical distributions （CI to adults） in C. sinicus before and during the formation of cold waters in the Yellow Sea during the study period.

Spatial control of lithium deposition is the most important issue in lithium-metal batteries because of the considerable control of lithium dendrite suppression via the uniform distribution of Li^(+)*** seed materials are crucial for the behavior of lithium deposition,in-depth studies on their physical and chemical control have not been ***,we describe a new design of seed structure comprising a wrinkled Cu/graphene substrate surrounded by copper(Ⅰ)oxide(Cu_(2)O)on a graphene grain boundary over a large area,which is fabricated by the oxidation of the Cu surface via graphene boundary defects by using chemical vapor deposition(CVD).Scanning and transmission electron microscopy results reveal that Cu_(2)O on the graphene boundary can render a preferential reaction with lithium during the first deposition and assist in the uniform deposition of lithium by preventing the agglomeration of lithium clusters during the second *** two-step process is attributed to the degree of selectivity due to the difference in lithium affinity,which allows long-term electrochemical stability and a high rate capability via boundary *** study highlights the significance of the boundary effect,which can open new avenues for the formation of a large family of seed structures in lithium-metal batteries.

Anti-MMP-2 and MMP-9 activity of Salsola komarovii Iljin extract and its solvent fractions

Objective: To investigate matrix metalloproteinases(MMP)-2 and MMP-9 inhibitory effect of Salsola komarovii Iljin, an edible halophyte with health beneficial ***: Salsola komarovii crude extracts(SKI), and solvent(n-hexane, 85% aq. Meoh, n-Buoh, and H2O) fractionated extracts of SKI were prepared. Gelatin zymography was carried out to observe MMP enzymatic activity. The release of the MMP enzymes was measured by enzyme-linked immunosorbent assay. Expression of MMPs in m RNA and protein level were investigated by polymerase chain reaction analysis and immunoblotting, ***: SKI and SKI fractions inhibited active MMP-2 and MMP-9 amount in the treated cell culture medium. Also, SKI suppressed the release of MMP-2 and MMP-9 from stimulated HT1080 human fibrosarcoma cells. Furthermore, SKI suppressed the m RNA and protein expression of MMP-2 and MMP-9. SKI fractions showed parallel effects except for H2O fraction which did not yield any significant MMP inhibitory effect. Among fractions, 85% aq. Me oh was the most active fraction to inhibit both the enzymatic effect and expression of MMP-2 and ***: SKI may contain potential MMP release inhibitory compounds. Salsola komarovii is a promising source of compounds against MMP and could be utilized in the development of antitumor agents.

Herein, we report a simple and effective preparation of ultrafine CNFs （u-CNFs） with high surface area via electrospinning of two immiscible polymers [polyacrylonitrile （PAN） and poly（methyl methacry- late）（PMMA）] followed by calcination at high temperature in an inert atmosphere. Various electrospinning conditions were optimized in detail. Four different kinds of PAN/PMMA ratios （10/0, 7：3, 5：5 and 3：7） were chosen and found that the PAN/PMMA ratio of 3：7 （PAN/PMMA-3：7） is the optimum one. BET anal- ysis showed the specific surface area of the u-CNFs-3：7 was 46Z57 m2/g with an excellent pore volume （1.15 cms g-l） and an average pore size （9.48 nm）： it is about 25 times higher than the conventional CNFs （c-CNFs）. TEM and FE-SEM images confirmed the ultrafine structure of the CNFs with a thinner fiber di- ameter of-50 nm. The graphitic nature and atomic arrangement of the u-CNFs were investigated by Raman and XPS analyses. For the supercapacitor application, unlike the common electrode preparation methods, the u-CNFs-3：7 was used without any activation, chemical or mechanical modifications. The u-CNFs- 3：7 showed a better specific capacitance of 86 Fig in 1 mol/L 1-12S04 when compared to pure CNFs. The excellent physicochemical properties make the u-CNFs-3：7 an alternative choice to the existing CNFs for the supercapacitors.

Influence of glass-based dental ceramic type and thickness with identical shade on the light transmittance and the degree of conversion of resin cement

The purpose of this study was to assess the influence of the types and thicknesses of glass ceramic plates on light transmittance and compare the degrees of conversion(DC) of resin cement under the ceramic materials. Three ceramic plates with thicknesses of 0.5, 1.0, 2.0, and 4.0 mm were fabricated from each of five commercial ceramic blocks in shade A2: high-translucency and low-translucency IPS Empress CAD(EmpHT and EmpLT); high-translucency and low-translucency IPS *** CAD(EmxHT and EmxLT); and Vita Mark Ⅱ(Vita). The translucency parameter was obtained using a colorimeter. The light transmittance rate was measured using a photodetector attached to an optical power meter. The DC of a resin cement(Variolink N) underneath the ceramic plates was examined by Fourier transform infrared spectroscopy. The translucency parameter, light transmittance rate, and DC showed significant differences by ceramic type and thickness(P 0.05). Both type and thickness of the glass ceramics significantly influenced the light transmittance and DC of the light-cured resin cement beneath the ceramic of the same shade.
