Trans-10,cis-12, not cis-9,trans-11, conjugated linoleic acid decreases ErbB3 expression in HT-29 human colon cancer cells

AIM： To examine whether trans-10,cis-12 CLA （t10C12） or cis-9,trans-11 CLA （c9t11） inhibits heregulin （HRG）-β- stimulated cell growth and HRG-β-ErbB3 signaling in HT-29 cells. METHODS： We cultured HT-29 cells in the absence or presence of the CLA isomers and/or the ErbB3 ligand HRG-β. MTT assay, [^3H]thymidine incorporation, Annexin V staining, RT-PCR, Western blotting, immunoprecipitation, and in vitro kinase assay were performed. RESULTS： HRG-β increased cell growth, but did not prevent DNA t10c12-induced growth inhibition. T10C12 inhibited DNA synthesis and induced apoptosis of HT-29 cells, whereas c9t11 had no effect. T10c12 decreased the levels of ErbB1, ErbB2, and ErbB3 proteins and transcripts in a dose-dependent manner, whereas c9t11 had no effect. Immunoprecipitation/ Western blot studies revealed that t10c12 inhibited HRG- β-stimulated phosphorylation of ErbB3, recruitment of the p85 subunit of phosphoinositide 3-kinase （PI3K） to ErbB3, ErbB3-associated PI3K activities, and phosphorylation of Akt. However, c9t11 had no effect on phospho Aid： levels. Neither t10c12 nor c9t11 had any effect on HRG-β-induced phosphorylation of ERK-1/2. CONCLUSION： These results indicate that the inhibition of HT-29 cell growth by t10c12 may be induced via its modulation of ErbB3 signaling leading to inhibition of Akt activation.

Is mannitol combined with furosemide a new treatment for refractory lymphedema?A case report

BACKGROUND Mannitol is a hyperosmolar agent and the combination of mannitol and furosemide is a widely used treatment for intracranial pressure *** the hypertonic properties of mannitol to move water out of intracellular spaces,we hypothesized that mannitol combined with furosemide could relieve focal tissue swelling in refractory *** SUMMARY A 90-year-old female had been diagnosed with intracranial hemorrhage and received a combination of mannitol and furosemide for intracranial pressure *** of the intracranial hemorrhage,she had refractory lymphedema of the left lower extremity since ***,after receiving the mannitol and furosemide,the patient’s lower extremity lymphedema improved *** the mannitol and furosemide were discontinued,the lymphedema worsened in spite of complete decongestive therapy(CDT)and intermittent pneumatic compression treatment(IPC).To identify the presumed effect of mannitol and furosemide on the lymphedema,these agents were resumed,and the lymphedema improved *** The present case raises the possibility that a combination of mannitol and furosemide might be considered another effective therapeutic option for refractory lymphedema when CDT and IPC are ineffective.

Superior mesenteric vein thrombosis induced by influenza infection:A case report

BACKGROUND Among the various types and causes of mesenteric ischemia,superior mesenteric vein(SMV)thrombosis is a rare and ambiguous *** a patient presents with SMV thrombosis,past medical history should be reviewed,and the patient should be screened for underlying *** thrombosis may also occur due to systemic *** this report,we describe a case of SMV thrombosis complicated by influenza B *** SUMMARY A 64-year-old male visited the hospital with general weakness,muscle aches,fever,and abdominal *** patient underwent computed tomography(CT)and was diagnosed with SMV *** the patient’s muscle pain and fever could not be explained by the SMV thrombosis,the clinician performed a test for influenza,which produced a positive result for influenza *** patient had a thrombus in the SMV only,with no invasion of the portal or splenic veins,and was clinically *** treatment was prescribed without surgery or other *** follow-up CT scan showed improvement,and the patient was subsequently discharged with continued oral anticoagulant *** This case provides evidence that influenza may be a possible risk factor for SMV *** unexplained abdominal pain is accompanied by an influenza infection,examination of an abdominal CT scan may be necessary to screen for possible SMV thrombosis.

A method for automatically translating print books into electronic Braille books

In this paper, a method for automatically translating scanned images from print books into electronic Braille books is proposed with the objective of reducing the amount of time and cost required for producing Braille books. The proposed method consists of processes for identifying character and image areas in a scanned image, automatically translating characters and images into Braille and tactile graphics, respectively, and positioning Braille and tactile graphics into an electronic Braille page. Experimental results show that the proposed method drastically reduces the time required to translate a print book into an electronic Braille book. Despite the drastic reduction in translation time, the method proposed in this paper does not compromise the ability to recognize information for the visually impaired compared to manually produced Braille books, demonstrating its feasibility in practical applications. Therefore, the proposed method is expected to significantly reduce the time and cost required for producing Braille books, and provide more reading materials for the visually impaired,making significant contributions to enhancing their knowledge and welfare.

Survival improvement and prognostic factors in recent management of extrahepatic cholangiocarcinoma:A single-center study

Background:Cholangiocarcinoma was considered as a dismal disease with very poor prognosis until *** is increasingly found due to increased life *** surgical and medical management were advanced recently,data on the prognosis,especially extrahepatic cholangio-carcinoma(ECC),were *** study aimed to identify clinicopathologic features and prognosis of patients with ***:Patients followed up and diagnosed with ECC between January 2014 and December 2016 at a tertiary hospital were included,whereas those with intrahepatic cholangiocarcinoma,gallbladder cancer,and ampullary cancer were ***:A total of 83 patients were followed after the treatment(49 men and 34 women;median age 73.3 years).Cancer location was classified as distal common bile duct(25 patients),proximal com-mon bile duct(24 patients),common hepatic duct(20 patients),and hilar(14 patients).About 14.5%of patients had history of another malignant neoplasm,and 24.1%patients had chronic ***-cal resection was performed in 54 patients(65%)and dysplasia was combined in 63%(34/54).Adjuvant chemotherapy was performed in 54%(29/54),but only 7 underwent palliative chemotherapy in 29 non-surgical *** median overall survival in all patients was 30.9 *** analyzing the treatment modality,median survival of adjuvant chemotherapy,surgery only,palliative chemotherapy,and support-ive care groups were 42.9,30.9,12.0,and 8.9 months,respectively(P<0.05).In the Cox regression analy-sis of survival,age,surgical resection,chemotherapy,and comorbidity were significant prognostic factors,and the comorbidity was the only significant prognostic factor in the multivariable analysis(hazard ratio[HR]=2.80;95%CI:1.32–5.95;P=0.007).In a subgroup analysis of surgical patients,the presence of dysplasia was a favorable prognostic factor in the multivariable analysis(HR=0.29;95%CI:0.09–0.91;P=0.033).Conclusions:The overall survival of patients with ECC wa

Reconstruction of Rabbit Corneal Layer Composed of Corneal Fibroblasts and Corneal Epithelium on the Lyophilized Amniotic Membrane

Many researchers have employed the cryopreserved amniotic membrane（CAM） and corneal epithelial cells in the treatment of a severely damaged burned cornea, with corneal epithelial cells cultured on an amniotic membrane （AM）. The lyophilized amniotic membrane（LAM） has a higher graft take and a longer shelf life; it is easier to store and safer because of gamma irradiation. Two Teflon rings（ Ahn＇s supporter） were made for culturing the cells on the LAM, and were then used to support the LAM. To reconstruct a corneal layer composed of corneal fibroblasts and epithelium, the corneal fibroblasts were first cultivated on the stromal side of LAM for five days, foUowed by epithelial cells culture on the epithelial side, by using the air-liquid interface culture. The reconstructed corneal layer composed of corneal fibroblasts and corneal epithelial cells has a much healthier basal layer of corneal epithelium than the reconstructed corneal epithelium, which was got by using only corneal epithelial cells, and resembles the epithelium of normal corneas, without the horny layer. Thus, the reconstruction of the corneal layer by using a LAM is considered to be a good in vitro model, not only for its application in toxicological test kits, but also for transplantation in patients with a severely damaged cornea.

Rigidity(or stiffness)of materials and extracellular matrix has proven to be one of the most significant extracellular physicochemical cues that can control diverse cell behaviors,such as contractility,motility,and spreading,and the resultant pathophysiological *** 2D materials engineered with tunable rigidity have enabled researchers to elucidate the roles of matrix biophysical cues in diverse cellular events,including migration,lineage specification,and mechanical ***,the recent findings accumulated under 3D environments with viscoelastic and remodeling properties pointed to the importance of dynamically changing rigidity in cell fate control,tissue repair,and disease ***,here we aim to highlight the works related with material/matrix-rigidity-mediated cell and tissue behaviors,with a brief outlook into the studies on the effects of material/matrix rigidity on cell behaviors in 2D systems,further discussion of the events and considerations in tissue-mimicking 3D conditions,and then examination of the in vivo findings that concern material/matrix *** current discussion will help understand the material/matrix-rigidity-mediated biological phenomena and further leverage the concepts to find therapeutic targets and to design implantable materials for the treatment of damaged and diseased tissues.

Effect of monovalent charge compensators on the photoluminescence properties of Ca_(3)(PO_(4))_(2):Tb^(3+),A^(+)(A=Li,Na,K)phosphors

Ca_(3-x)(PO_(4))_(2):xTb^(3+)(0.2≤x≤0.4),Ca_(2.3)(PO_(4))_(2):0.35 Tb^(3+),0.35 A^(+)(A=Li,Na,K),and Ca_(2.3)(PO_(4))_(2):0.35 Tb^(3+),yLi^(+)(0.35≤y≤0.455)phosphors were prepared by solid-state *** the prepared phosphors formed a rhombohedral unit cell with the R3c space *** improve the photoluminescence(PL)properties of the Ca_(2.65)(PO_(4))_(2):0.35 Tb^(3+)phosphor,monovalent charge compensators such as Li^(+),NA^(+),and K^(+)were added to the Ca_(2.65)(PO_(4))_(2):0.35 Tb^(3+)*** charge compensators acted as fluxes,so they improved the *** excitation and emission properties were significantly improved through the incorporation of charge *** particular,among the charge compensators,Li^(+)ion substantially enhanced the emission intensity and color ***,considering the evaporation of Li_(2)CO_(3)during the annealing process,we optimized the concentration of Li^(+)charge compensator to enhance its PL ***,the green emission intensity of the Ca_(2.3)(PO_(4))_(2):0.35 Tb^(3+),0.385 Li^(+)phosphor was 260%higher than that of the Ca_(2.65)(PO_(4))_(2):0.35 Tb^(3+)*** believe that the effect of charge compensators on the PL properties and the optimum concentration of Li^(+)cha rge compensator are useful for the design of phosphors in light-emitting diodes.

Nitric oxide(NO)has been shown to promote revascularization and nerve regeneration after peripheral nerve ***,in vivo application of NO remains challenging due to the lack of stable carrier materials capable of storing large amounts of NO molecules and releasing them on a clinically meaningful time ***,a silica nanoparticle system capable of reversible NO storage and release at a controlled and sustained rate was *** this study,NO-releasing silica nanoparticles(NO-SNs)were delivered to the peripheral nerves in rats after acute crush injury,mixed with natural hydrogel,to ensure the effective application of NO to the *** using a polymer dye and immunohistochemical staining for the detection of CD34(a marker for revascularization)results showed that NO-releasing silica nanoparticles increased revascularization at the crush site of the sciatic *** sciatic functional index revealed that there was a significant improvement in sciatic nerve function in NO-treated *** and anatomical analyses showed that the number of myelinated axons in the crushed sciatic nerve and wet muscle weight excised from NO-treated rats were ***,muscle function recovery was improved in rats treated with *** together,our results suggest that NO delivered to the injured sciatic nerve triggers enhanced revascularization at the lesion in the early phase after crushing injury,thereby promoting axonal regeneration and improving functional recovery.
