Predictability of polygenic risk score for progression to dementia and its interaction with APOEε4 in mild cognitive impairment

Background:The combinatorial efect of multiple genetic factors calculated as a polygenic risk score(PRS)has been studied to predict disease progression to Alzheimer’s disease(AD)from mild cognitive impairment(MCI).Previous studies have investigated the performance of PRS in the prediction of disease progression to AD by including and excluding single nucleotide polymorphisms within the region surrounding the APOE *** studies may have missed the APOE genotype-specifc predictability of PRS for disease progression to ***:We analyzed 732 MCI from the Alzheimer’s Disease Neuroimaging Initiative cohort,including those who progressed to AD within 5 years post-baseline(n=270)and remained stable as MCI(n=462).The predictability of PRS including and excluding the APOE region(PRS_(+APOE) and PRS_(−APOE))on the conversion to AD and its interaction with the APOEε4 carrier status were assessed using Cox regression ***:PRS_(+APOE)(hazard ratio[HR]1.468,95%CI 1.335-1.615)and PRS_(−APOE)(HR 1.293,95%CI 1.157-1.445)were both associated with a signifcantly increased risk of MCI progression to *** interaction between PRS_(+APOE) and APOEε4 carrier status was signifcant with a P-value of *** association of PRSs with the progression risk was stronger in APOEε4 non-carriers(PRS_(+APOE):HR 1.710,95%CI 1.244-2.351;PRS_(−APOE):HR 1.429,95%CI 1.182-1.728)than in APOEε4 carriers(PRS_(+APOE):HR 1.167,95%CI 1.005-1.355;PRS_(−APOE):HR 1.172,95%CI 1.020-1.346).Conclusions:PRS could predict the conversion of MCI to dementia with a stronger association in APOEε4 noncarriers than APOEε4 *** indicates PRS as a potential genetic predictor particularly for MCI with no APOEε4 alleles.

CORE:Common Region Extension Based Multiple Protein Structure Alignment for Producing Multiple Solution

Over the past several decades, biologists have conducted numerous studies examining both general and specific functions of proteins. Generally, if similarities in either the structure or sequence of amino acids exist for two proteins, then a common biological function is expected. Protein function is determined primarily based on the structure rather than the sequence of amino acids. The algorithm for protein structure alignment is an essential tool for the research. The quality of the algorithm depends on the quality of the similarity measure that is used, and the similarity measure is an objective function used to determine the best alignment because of their individual strength and weakness however, none of existing similarity measures became golden standard They require excessive filtering to find a single alignment. In this paper, we introduce a new strategy that finds not a single alignment, but multiple alignments with different lengths. This method has obvious benefits of high quality alignment. however, this novel method leads to a new problem that the running time for this method is considerably longer than that for methods that find only a single alignment. To address this problem~ we propose algorithms that can locate a common region （CORE） of multiple alignment candidates, and can then extend the CORE into multiple alignments. Because the CORE can be defined from a final alignment, we introduce CORE＊ that is similar to CORE and propose an algorithm to identify the CORE＊. By adopting CORE＊ and dynamic programming, our proposed method produces multiple alignments of various lengths with higher accuracy than previous methods. In the experiments, the alignments identified by our algorithm are longer than those obtained by TM-align by 17% and 15.48%, on average, when the comparison is conducted at the level of super-family and fold, respectively.

Checkpoint Management with Double Modular Redundancy Based on the Probability of Task Completion

This paper proposes a checkpoint rollback strategy for real-time systems with double modular *** built-in fault-detection and spare processors,our scheme is able to recover from both transient and permanent *** comparisons are conducted at each ***,the states stored in two consecutive checkpoints of one processor are compared for checking integrity of the *** states of two processors are also compared for detecting faults and the system rolls back to the previous checkpoint whenever required by logic of the proposed scheme.A Markov model is induced by the fault recovery scheme and analyzed to provide the probability of task completion within its *** optimal number of checkpoints is selected so as to maximize the probability of task completion.

Induction of vascular endothelial growth factor receptor expression in human umbilical vein endothelial cells after repeated bevacizumab treatment in vitro

AIM： To investigate the mechanism underlying the loss of responsiveness to anti-vascular endothelial growth factor（VEGF） treatment after repeated injections for choroidal neovascularization, VEGF and VEGF receptor（VEGFR） expressions were evaluated following repeated bevacizumab treatments in hypoxic human umbilical vein endothelial cells（HUVECs） in ***： HUVECs were incubated under hypoxic conditions in two media of different bevacizumab concentrations（1.0 or 2.5 mg/m L） for 17 h, and then in a new medium without bevacizumab for 7h. This procedure was repeated twice more. A culture with an identical volume of excipients served as the control. Cytotoxicity and cell proliferation were assessed using 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide and Ki-67 assays, respectively. Levels of VEGF and VEGFR were assessed using enzyme-linked immunosorbent assay and Western blot ***： Cytotoxic effects were not reported for either bevacizumab concentration. Cell proliferation was not reduced after anti-VEGF treatments. VEGF level after single treatment was significantly higher than that of the control and after repeated treatments. Phosphorylated VEGFR-2 expression increased significantly after singleand repeated bevacizumab treatments compared with the control. The 1.0 mg/m L bevacizumab induced significantly higher expressions of VEGFR-2 than the 2.5 mg/m L in single and repeated treatment ***： Bevacizumab treatment of HUVECs elevated VEGFR expression in both single and repeated treatments, indicating a mechanism for the reduced efficacy of anti-VEGF therapy in ocular neovascular disorders.

Dynamic enhanced computed tomography imaging findings of an inflammatory fibroid polyp with massive fibrosis in the stomach

Inflammatory fibroid polyp(IFP) is a rare benign lesion of the gastrointestinal tract. We report a case of computed tomography(CT) imaging finding of a gastric IFP with massive fibrosis. CT scans showed thickening of submucosal layer with overlying mucosal hyperenhancement in the gastric antrum. The submucosal layer showed increased enhancement on delayed phase imaging. An antrectomy with gastroduodenostomy was performed because gastric cancer was suspected, particularly signet ring cell carcinoma. The histopathological diagnosis was an IFP with massive fibrosis. The authors suggest that when the submucosal layer of the gastric wall is markedly thickened with delayed enhancement and preservation of the mucosal layer, an IFP with massive fibrosis should be considered in the differential diagnosis.

Korean freshwater snails of the genus Semisulcospira are widely distributed across East *** has been a very popular nutritional food in Korea,and is an ecologically important water quality indicator because it lives only in clean ***,no microsatellite markers have been generated to study the population genetic diversity of this *** the present study,we developed and characterized 18 novel microsatellite loci from Semisulcospira coreana genomic *** microsatellites were isolated using 454 GS-FLX titanium sequencing and 18 markers were used for genotyping in *** addition,we also tested the cross-species transferability of the microsatellite markers in four additional Semisulcospira *** identified 18 polymorphic loci and the number of alleles per loci,and their polymorphism information content values ranged from 2 to 17 and 0.203 to 0.902,*** observed and expected heterozygosities of the loci ranged from 0.063 to 0.924 and 0.226 to 0.924,*** to the analysis of the cross-species transferability of these markers,four species,***,***,***,and ***,showed a very high transferability(80%–85%).These results show that this set of nuclear markers could be useful for population genetics studies of this species and closely related species.

Spatial Ecology of Translocated and Resident Amur Ratsnakes(Elaphe schrenckii) in Two Mountain Valleys of South Korea

The translocation of snakes has been viewed as a useful tool to augment declining populations and to mitigate human-wildlife conflicts, even though released snakes often exhibit relatively high mortality. We radio-tracked 13 Amur Ratsnakes （Elaphe schrenckii） in the Woraksan National park in South Korea from July 2008 to May 2009. Two of these snakes were residents, and 11 had been illegally captured in areas remote from the study site and were donated by the park office. During the study period, six of the translocated snakes were lost： two were killed by predators, one died of unknown causes, and the radio signals of three of the snakes were lost. In the field, the ratsnakes laid eggs in early August, moved into hibemacula in late November, and moved away from the hibemacula in mid-April. Compared to the resident snakes, five of the translocated snakes traveled approximately 1.3 times farther per week, and the home ranges of the translocated snakes were three to six times larger than those of the resident snakes. In addition, the translocated snakes were found underground more frequently than the resident snakes. The management recommendations resulting from this study will guide biologists and land use managers in making appropriate decisions regarding release sites and the use of gravid females in the translocation of this endangered ratsnake.

Surface-roughened current collectors for anode-free all-solid-state batteries

Anode-free all-solid-state batteries(AFASSBs), composed of a fully lithiated cathode and a bare current collector(CC) that eliminates excess lithium, can maximize the energy density(because of a compact cell configuration) and improve the safety of solid-state systems. Although significant progress has been made by modifying CCs in liquid-based anode-free batteries, the role of CCs and the mechanism of Li formation on CCs in AFASSBs are still unexplored. Here, we systematically investigate the effect of the surface roughness of the CCs on the Li plating/stripping behavior in AFASSBs. The results show that the moderately roughened CC substantially improves the Coulombic efficiency and cycle stability of AFASSBs owing to the increased contact points between the solid electrolyte and the roughened CC. In contrast, the excessively roughened CC deteriorates the performance owing to the contact ***, an ex situ interface analysis reveals that the roughened surface of the CC could suppress the interfacial degradation during the Li ion extraction from a sulfide solid electrolyte to a CC. This provides an indication to the origin that hinders the electrochemical performance of AFASSBs. These findings show the potential for the application of surface-engineered CCs in AFASSBs and provide guidelines for designing advanced CCs.

Synthesis and properties of Cr-Al-Si-N films deposited by hybrid coating system with high power impulse magnetron sputtering (HIPIMS) and DC pulse sputtering

The CrN and Cr-Al-Si-N films were deposited on Si wafer and SUS 304 substrates by a hybrid coating system with high power impulse magnetron sputtering (HIPIMS) and a DC pulse sputtering using Cr and AlSi targets under N2/Ar *** varying the sputtering current of the AlSi target in the range of 0-2.5 A,both the Al and Si contents in the films increased gradually from 0 to 19.1% and 11.1% (mole fraction),*** influences of the AlSi cathode DC pulse current on the microstructure,phase constituents,mechanical properties,and oxidation behaviors of the Cr-Al-Si-N films were investigated *** results indicate that the as-deposited Cr-Al-Si-N films possess the typical nanocomposite structure,namely the face centered cubic (Cr,Al)N nano-crystallites are embedded in the amorphous Si3N4 *** increasing the Al and Si contents,the hardness of the film first increases from 20.8 GPa for the CrN film to the peak value of 29.4 GPa for the Cr0.23Al0.14Si0.07 N film,and then decreases *** the meanwhile,the Cr0.23Al0.14Si0.07N film also possesses excellent high-temperature oxidation resistance that is much better than that of the CrN film at 900 or 1000 °C.
