Background:Increased Galectin 3-binding protein(LGALS3BP)serum levels have been used to assess hepatic fibrosis stages and the severity of hepatocellular carcinoma(HCC).Considering the crucial role of transforming growth factor-β1(TGF-β1)in the emergence of these diseases,the present study tested the hypothesis that LGALS3BP regulates the TGF-β1 signaling ***:The expression levels of LGALS3BP and TGFB1 were analyzed in patients with metabolic dysfunction-associated steatohepatitis(MASH)and *** omics techniques,such as RNA-sequencing,transposaseaccessible chromatin-sequencing assay,and liquid chromatography-tandem mass spectrometry proteomics,were used to identify the regulatory mechanisms for the LGALS3BP-TGF-β1 *** effects of altered TGF-β1 signaling by LGALS3BP were investigated in conditional LGALS3BP-knockin and LGALS3BP-knockout ***:In patients with MASH and HCC,the levels of LGALS3BP and TGFB1 exhibited positive *** of LGALS3BP by the inflammatory cytokine interferonαin HCC cells or ectopic overexpression of LGALS3BP in hepatocytes promoted the expression levels of *** fibrosis was observed in the livers of hepatocyte-specific LGALS3BP-knockin mice,with increased TGFB1 ***3BP directly bound to and assembled integrinαV,an integral mediator required for releasing active TGF-β1 from extracellular latent complex with the rearranged F-actin *** released TGF-β1 activated JunB transcription factor,which in turn promoted the TGF-β1 positive feedback ***3BP deletion in the hepatocytes downregulated TGF-β1 signaling and CCl4 induced ***,LGALS3BP depletion hindered hepatocarcinogenesis by limiting the availability of fibrogenic TGF-β***:LGALS3BP plays a crucial role in hepatic fibrosis and carcinogenesis by controlling the TGF-β1 signaling pathway,making it a promising therapeutic target in TGF-β1-related diseases.

The design and development of cheap,abundant,and efficient electrocatalysts for selective CO_(2)electroreduction is highly desirable yet remains an ongoing ***,we report on our discovery of the use of two-dimensional MgO rich in oxygen vacancies(VO)as an electrocatalyst for the efficient reduction of CO_(2)to yield CO in 1-butyl-3-methylimidazolium hexafluorophosphate dissolved in *** faradaic efficiency toward CO reaches as high as 99.6±0.24%with a current density of up to 40.8 mA cm−*** functional theory calculations illustrate that the introduction of VO in MgO substantially lowers the reaction-free energy for the transformation of ^(*)COO^(−) to ^(*)COOH,the potential determining step,which greatly boosts CO_(2)conversion *** work opens the way to realizing economic and efficient CO_(2)electrolysis of group II metals.

The ex-situ incorporation of the secondary SiC reinforcement,along with the in-situ incorporation of the tertiary and quaternary Mg_(3)N_(2) and Si_(3)N_(4) phases,in the primary matrix of Mg_(2)Si is employed in order to provide ultimate wear resistance based on the laser-irradiation-induced inclusion of N_(2) gas during laser powder bed *** is substantialized based on both the thermal diffusion-and chemical reactionbased metallurgy of the Mg_(2)Si–SiC/nitride hybrid *** study also proposes a functional platform for systematically modulating a functionally graded structure and modeling build-direction-dependent architectonics during additive *** strategy enables the development of a compositional gradient from the center to the edge of each melt pool of the Mg_(2)Si–SiC/nitride hybrid ***,the coefficient of friction of the hybrid composite exhibits a 309.3%decrease to–1.67 compared to–0.54 for the conventional nonreinforced Mg_(2)Si structure,while the tensile strength exhibits a 171.3%increase to 831.5 MPa compared to 485.3 MPa for the conventional *** outstanding mechanical behavior is due to the(1)the complementary and synergistic reinforcement effects of the SiC and nitride compounds,each of which possesses an intrinsically high hardness,and(2)the strong adhesion of these compounds to the Mg_(2)Si matrix despite their small sizes and low concentrations.

Objective:The purpose of the study was to identify the best sequence of therapy beginning with a tyrosine kinase inhibitor(TKI)as the first-line therapy for patients with metastatic renal cell carcinoma(mRCC)in terms of overall survival(OS),progression-free survival(PFS),and rates of discontinuation and adverse effects during the treatment ***:This is a retrospective,nationwide multicenter study of patients with mRCC after diagnosis at 10 different tertiary medical centers in Korea from January 1992 to December *** focused on patients at either“favorable”or“intermediate”risk according to the International mRCC Database Consortium criteria,and they were followed up(median 335 days).Finally,a total of 1409 patients were selected as the study *** generated a Cox proportional hazard model adjusted for covariates,and the different therapy schemes were statistically tested in terms of OS as well as *** addition,frequencies of discontinuation and adverse events were compared among the therapy ***:Of the primary patterns of treatment sequences(24 sequences),“sunitinib epazopanib”and“sunitinibeeverolimuseimmunotherapy”showed the most beneficial results in both OS and PFS with significantly lower hazards than“sunitinib”,which is the most commonly treated agent in *** that the“TKIeTKI”structure showed relatively higher discontinuation rates with higher adverse effects,the overall beneficial sequence would be“sunitinibeeverolimuseimmunotherapy”.Conclusion:Among several sequential therapy starting with TKIs,“sunitinibeeverolimuse immunotherapy”was found to be the best scheme for mRCC patients with“favorable”or“intermediate”risks.

During the last decade,the generation and accumulation of petabase-scale high-throughput sequencing data have resulted in great challenges,including access to human data,as well as transfer,storage,and sharing of enormous amounts of *** promote data-driven biological research,the Korean government announced that all biological data generated from government-funded research projects should be deposited at the Korea BioData Station(K-BDS),which consists of multiple databases for individual data ***,we introduce the Korean Nucleotide Archive(KoNA),a repository of nucleotide sequence *** of July 2022,the Korean Read Archive in KoNA has collected over 477 TB of raw next-generation sequencing data from national genome *** ensure data quality and prepare for international alignment,a standard operating procedure was adopted,which is similar to that of the International Nucleotide Sequence Database *** standard operating procedure includes quality control processes for submitted data and metadata using an automated pipeline,followed by manual *** ensure fast and stable data transfer,a high-speed transmission system called GBox is used in ***,the data uploaded to or downloaded from KoNA through GBox can be readily processed using a cloud computing service called *** seamless coupling of KoNA,GBox,and Bio-Express enhances the data experience,including submission,access,and analysis of raw nucleotide *** not only satisfies the unmet needs for a national sequence repository in Korea but also provides datasets to researchers globally and contributes to advances in *** KoNA is available at https://***/kona/.

Hepatocellular carcinoma(HCC)is the second most common cancer and the third leading cause of cancer-related death worldwide[1].Recently,HCC incidence and mortality rates have further increased due to changes in the global social environment and dietary habits[2].HCC is treated by surgical resection or combination chemotherapy,but the overall survival rate of HCC patients has not improved,and the recurrence rate is still high due to strong invasiveness and resistance to chemotherapy[3].

Development of practical postprocessor for 5-axis machine tool with non-orthogonal rotary axes

In order to develop a practical postprocessor for 5-axis machine tool,the general equations of numerically controlled(NC) data for 5-axis configurations with non-orthogonal rotary axes were exactly expressed by the inverse kinematics,and a windows-based postprocessor written with Visual Basic was developed according to the proposed *** developed postprocessor is a general system suitable for all kinds of 5-axis machines with orthogonal and non-orthogonal rotary *** implementation of the developed postprocessor and verification by a cutting simulation and machining experiment,the effectiveness of the proposed algorithm is *** is improved by allowing exchange of data formats such as rotational total center position(RTCP) controlled NC data,vector post NC data,and program object file(POF) cutter location(CL) data,and convenience is increased by adding the function of work-piece origin ***,a practical post-processor for 5-axis machining is developed.

The effects of anti-vascular endothelial growth factor agents on human retinal pigment epithelial cells under high glucose conditions

AIM： To investigate the effects of high glucose levels and anti-vascular endothelial growth factor（VEGF） agents（bevacizumab,ranibizumab and aflibercept） on retinal pigment epithelium（RPE） ***： ARPE-19 cells were cultured at different glucose levels（5.5 mmol/L,25 mmol/L,and 75 mmol/L）.Cell viability was evaluated by MTT assay at 3d after treatment with *** migration ability was measured by wound healing assay at 3d.A cell death detection kit was used to assess apoptosis at 3 and 14 *** proliferation was assessed by EdU assay at *** culture medium was treated with anti-VEGF agents at clinically relevant *** experiment was then repeated at a different glucose ***： The viability and migration of ARPE-19 cells were significantly decreased in the presence of 75 mmol/L as compared to 5.5 mmol/L *** percentage of TUNEL-positive cells was significantly increased and the proliferative potential was decreased with 75 mmol/L compared to 5.5 mmol/L *** were no significant differences in the results between 25 mmol/L and 5.5 mmol/L *** the presence of 75 mmol/L glucose,the groups treated with anti-VEGF showed decreased cell viability and proliferation and increased ***,there were no significant differences between the anti-VEGF ***： High glucose level decreases the viability,wound healing ability,and proliferation of RPE cells,while increasing ***,anti-VEGF agents interfered with the physiological functions of RPE cells under high-glucose conditions,accompanied by decreases in cell viability and proliferation.

Combination of Korean Red Ginseng Extract and Hydrogen-Rich Water Improves Spermatogenesis and Sperm Motility in Male Mice

Objective: To investigate the effect of hydrogen-rich Korean Red Ginseng(KRG) water(HRGW) mixture on the spermatogenesis and sperm motility of mice of different ages. Methods: Eighty young(3 month-old) and aged(12 month-old) male mice were randomly assigned to 4 groups(n=10 per group) including control group, hydrogen-rich water(HRW) group(10 m L/kg daily), KRG group(50 mg/kg daily) and HRGW group(10 m L/kg HRW and 50 mg/kg KRG daily) by an oral zoned needle for 4 weeks. Sperm count and motility were measured using sperm suspension released from cauda epididymis. Serum follicle stimulating hormone(FSH), luteinizing hormone(LH), testosterone, and reactive oxygen species(ROS) in serum have also been estimated. Tubular changes were examined through histological hematoxylin and eosin staining. Expression of antioxidation(PPx3, PPx4, GSTm5 and GPx4), spermatogenesis(inhibin-α, neptin-2 and CREM), antiaging(SIRT1 and SIRT2), and angiogenesis [visfatin and vascular endothelial growth factor(VEGF)] related genes were examined through realtime polymerase chain reaction. Results: HRW and KRG treatment stimulated spermatogenesis followed by increasing sperm production and sperm motility(Phancing antioxidation and stimulating spermatogenesis and sex hormone production, particularly in aged mice.

Surface effects of hybrid vibration-assisted femtosecond laser system for micro-hole drilling of copper substrate

The ultrafast laser based hybrid machining system was studied and a novel approach was demonstrated to improve laser machining quality on metals by vibrating the optical objective lens with a low frequency (500 Hz) and various displacements (0-16.5 μm) during a femtosecond laser machining *** laser used in this experiment is an amplified Ti:sapphire femtosecond (10-15 s) laser system that generates 100 femtosecond pulses having an energy of 3.5 mJ/pulse with a 5 kHz repetition rate at a central wavelength of 790 *** is found that both the wall surface finish of the machined structures and the aspect ratio obtained using the frequency vibration assisted laser machining are improved compared with those derived via laser machining without vibration assistance.
