Stomatal movements are critical in regulating gas exchange for photosynthesis and water balance between plant tissues and the atmosphere. The plant hormone abscisic acid （ABA） plays key roles in regulating stomatal closure under various abiotic stresses. In this study, we revealed a novel role of BAKI in guard cell ABA signaling. We found that the brassinosteroid （BR） signaling mutant bakl lost more water than wild- type plants and showed ABA insensitivity in stomatal closure. ABA-induced OST1 expression and reactive oxygen species （ROS） production were also impaired in bakl. Unlike direct treatment with H202, overex-pression of OST1 did not completely rescue the insensitivity of bakl to ABA. We demonstrated that BAK1 forms a complex with OST1 near the plasma membrane and that the BAKI/OST1 complex is increased in response to ABA in planta. Brassinolide, the most active BR, exerted a negative effect on ABA-induced formation of the BAK1/OST1 complex and OST1 expression. Moreover, we found that BAK1 and ABI1 oppositely regulate OST1 phosphorylation in vitro, and that ABI1 interacts with BAK1 and inhibits the interaction of BAK1 and OST1. Taken together, our results suggest that BAK1 regulates ABA-induced stomatal closure in guard cells.

BACKGROUND Inflammatory bowel disease(IBD)is a chronic inflammatory condition of the gastrointestinal tract,with tumor necrosis factor(TNF)-αplaying a key role in its ***,a decoy receptor for TNF,is used to treat inflammatory *** secretome derived from adipose-derived stem cells(ASCs)has anti-inflammatory effects,making it a promising therapeutic option for *** To investigate the anti-inflammatory effects of the secretome obtained from ASCs synthesizing etanercept on colon cells and in a dextran sulfate sodium(DSS)-induced IBD mouse *** ASCs were transfected with etanercept-encoding mini-circle plasmids to create etanercept-producing *** secretory material from these cells was then tested for anti-inflammatory effects both in vitro and in a DSS-induced IBD mouse *** This study revealed promising results indicating that the group treated with the secretome derived from etanercept-synthesizing ASCs[Etanercept-Secretome(Et-Sec)group]had significantly lower expression levels of inflammatory mediators,such as interleukin-6,Monocyte Chemoattractant Protein-1,and TNF-α,when compared to the control secretome(Ct-Sec).Moreover,the Et-Sec group exhibited a marked therapeutic effect in terms of preserving the architecture of intestinal tissue compared to the *** These results suggest that the secretome derived from ASCs that synthesize etanercept has potential as a therapeutic agent for the treatment of IBD,potentially enhancing treatment efficacy by merging the anti-inflam-matory qualities of the ASC secretome with etanercept's targeted approach to better address the multifaceted pathophysiology of IBD.

Two hundred endoscopic extraperitoneal inguinal hernioplasties: cost containment by reusable instruments

Objective To report our experience of 200 endoscopic totally extraperitoneal inguinal hernioplasties utilizing reusable *** Between August 1999 and june 2000, 200 endoscopic totally extraperitoneal hernioplasties were performed on 163 patients. The mean age of the study population was 63 years with a male to female ratio of 157∶6. Perioperative details and postoperative outcomes were prospectively evaluated and *** A total of 196 (98%) endoscopic extraperitoneal inguinal hernioplasties were successfully performed. Conversion rates to transabdominal preperitoneal and open repairs were 1.5% (n=3) and 0.5% (n=1), respectively. There were no other intraoperative complications. Postoperative morbidity included retention of urine (n=7), wound bruising (n=2), atelectasis (n=2) and gouty arthritis (n=1). The mean visual analogue pain scores at rest were 2.3, 1.6 and 1.9 on postoperative days 0, 1 and 2, respectively. The mean length of hospital stay was 1.9 days. 113 patients (69%) returned to normal activities within one week. Of the 35 patients who experienced both open and laparoscopic repair, 80% expressed preference for endoscopic hernioplasty in the event of future *** Endoscopic extraperitoneal inguinal hernioplasty can be safely performed utilizing reusable trocars. Substantial reduction of operative cost could be achieved by the elimination of disposable instruments. Deficiencies of the reusable metallic trocar, namely peri-cannula air-leak and sliding movements of the trocar, can be overcome by purse-string suture of the fascial opening.

We present a new variant of penalty method, which is different from the existing penalty methods, for solving the weak linear bilevel programming problems. We then transform it into a single-level optimization problem using Kuhn-Tucker optimality condition and discuss the relations between them. Finally, two examples are used to illustrate the feasibility of the proposed penalty method.

Intelligent electronic devices(IEDs)are interconnected via communication networks and play pivotal roles in transmitting grid-related operational data and executing control *** the context of the heightened security challenges within smart grids,IEDs pose significant risks due to inherent hardware and software vulner-abilities,as well as the openness and vulnerability of communication *** grid security,distinct from traditional internet security,mainly relies on monitoring network security events at the platform layer,lacking an effective assessment mechanism for ***,we incorporate considerations for both cyber-attacks and physical faults,presenting security assessment indicators and methods specifically tailored for ***,we outline the security monitoring technology for IEDs,considering the necessary data sources for their security ***,we classify IEDs and establish a comprehensive security monitoring index system,incorporating factors such as running states,network traffic,and abnormal *** index system contains 18 indicators in 3 ***,we elucidate quantitative methods for various indicators and propose a hybrid security assessment method known as GRCW-hybrid,combining grey relational analysis(GRA),analytic hierarchy process(AHP),and entropy weight method(EWM).According to the proposed assessment method,the security risk level of IEDs can be graded into 6 levels,namely 0,1,2,3,4,and *** higher the level,the greater the security ***,we assess and simulate 15 scenarios in 3 categories,which are based on monitoring indicators and real-world situations encountered by *** results show that calculated security risk level based on the proposed assessment method are consistent with actual ***,the reasonableness and effectiveness of the proposed index system and assessment method are validated.

Nanocrystalline 90W-7Ni-3Fe (wt pct) composite powder was prepared by mechanical alloying and mixed with binder to form a feedstock. Its rheological and sintering behaviors were compared with those of the feedstock from the original powder. It is found that milling can increase the maximum powder loading of feedstock and enhance the sintering densification process.

A comparative study on 3-D solar wind structure observed by Ulysses and MHD simulation

During Ulysses’ first rapid pole-to-pole transit from September 1994 to june 1995, its observations showed that middle- or high-speed solar winds covered all latitudes except those between ?20° and +20° near the ecliptic plane, where the velocity was 300―450 km/s. At poleward 40°, however, only fast solar winds at the speed of 700―870 km/s were observed. In addition, the transitions from low-speed wind to high-speed wind or vice versa were abrupt. In this paper, the large-scale structure of solar wind observed by Ulysses near solar minimum is simulated by using the three-dimensional numerical MHD model. The model com- bines TVD Lax-Friedrich scheme and MacCormack II scheme and decomposes the calculation region into two re- gions: one from 1 to 22 Rs and the other from 18 Rs to 1 AU. Based on the observations of the solar photospheric magnetic field and an addition of the volumetric heating to MHD equations, the large-scale solar wind structure mentioned above is reproduced by using the three-dimensional MHD model and the numerical results are roughly consistent with Ulysses’ observations. Our simulation shows that the initial magnetic field topology and the addition of volume heating may govern the bimodal structure of solar wind observed by Ulysses and also demonstrates that the three-dimensional MHD numerical model used here is efficient in modeling the large-scale solar wind structure.

Novel T cells with improved in vivo anti-tumor activity generated by RNA electroporation

The generation of T cells with maximal anti-tumor activities will significantly impact the field of T-cell- based adoptive immunotherapy. In this report, we found that OKT3/IL-2-stimulated T cells were phenotypically more heterogeneous, with enhanced anti-tumor activity in vitro and when locally administered in a solid tumor mouse model. To further improve the OKT3/IL-2-based T cell manufacturing procedure, we developed a novel T cell stimulation and expansion method in which peripheral blood mononuclear cells were electroporated with mRNA encoding a chimeric membrane protein consisting of a single-chain variable fragment against CD3 and the intracellular domains of CD28 and 4-1BB （OKT3-28BB）. The expanded T cells were phenotypically and functionally similar to T cells expanded by OKT3/IL-2. Moreover, co-electroporation of CD86 and 4-1BBL could further change the phenotype and enhance the in vivo anti-tumor activity. Although T cells expanded by the co- electroporation of OKT3-28BB with CD86 and 4-1BBL showed an increased central memory phenotype, the T cells still maintained tumor lytic activities as potent as those of OKT3/IL-2 or OKT3-28BB-stimulated T cells. In different tumor mouse models, T cells expanded by OKT3-28BB RNA electroporation showed anti-tumor activities superior to those of OKT3/IL-2 T cells. Hence, T cells with both a less differentiated phenotype and potent tumor killing ability can be generated by RNA electroporation, and this T cell manufacturing procedure can be further optimized by simply co-delivering other splices of RNA, thus providing a simple and cost-effec- tive method for generating high-quality T cells for adop- tive immunotherapy.
