Protective effects of a polysaccharide from Spirulina platensis on dopaminergic neurons in an MPT-Pinduced Parkinson's disease model in C57BL/6J mice

The present study aimed to determine whether a polysaccharide obtained fromSpirulina platensis shows protective effects on dopaminergic neurons. A Parkinson’s disease model was established through the intraperitoneal injection of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyr-idine （MPTP） in C57BL/6J mice. Prior to the MPTP injection, some mice were pretreated with intraperitoneal injections of a polysaccharide derived fromSpirulina platensis once daily for 10 days. The results showed that the immunoreactive staining and mRNA expression of the dopa-mine transporter and tyrosine hydroxylase, the rate-limiting enzyme in dopamine synthesis, in the substantia nigra, were signiifcantly increased in mice pretreated with 800 mg/kg of the poly-saccharide compared with those in MPTP-treated mice. The activities of superoxide dismutase and glutathione peroxidase in the serum and midbrain were also increased signiifcantly in mice injected with MPTP after pretreatment with the polysaccharide fromSpirulina platensis. By con-trast, the activity of monoamine oxidase B in serum and midbrain maintained unchanged. These experimental ifndings indicate that the polysaccharide obtained fromSpirulina platensis plays a protective role against the MPTP-induced loss of dopaminergic neurons in C57BL/6J mice, and that the antioxidative properties of this polysaccharide likely underlie its neuroprotective effect.

Curcumin protects nigral dopaminergic neurons by iron-chelation in the 6-hydroxydopamine rat model of Parkinson's disease

Objective Curcumin is a plant polyphenolic compound and a major component of spice turmeric （Curcuma longa）. It has been reported to possess free radical-scavenging, iron-chelating, and anti-inflammatory properties in dif- ferent tissues. Our previous study showed that curcumin protects MES23.5 dopaminergic cells from 6-hydroxydopamine （6-OHDA）-induced neurotoxicity in vitro. The present study aimed to explore this neuroprotective effect in the 6-OHDA-lesioned rat model of Parkinson＇s disease in vivo. Methods Rats were given intragastric curcumin for 24 days. 6-OHDA lesioning was conducted on day 4 of curcumin treatment. Dopamine content was assessed by high-performance liquid chromatography with electrochemical detection, tyrosine hydroxylase （TH）-containing neurons by immunohistochemistry, and iron-containing cells by Perls＇ iron staining. Results The dopamine content in the striatum and the number of TH-immunoreactive neurons decreased after 6-OHDA treatment. Curcumin pretreatment reversed these changes. Further studies demonstrated that 6-OHDA treatment increased the number of iron-staining cells, which was dramatically decreased by curcumin pretreatment. Conclusion The protective effects of curcumin against 6-OHDA may be attributable to the ironchelating activity of curcumin to suppress the iron-induced degeneration of nigral dopaminergic neurons.

Protection of MES23.5 dopaminergic cells by obestatin is mediated by proliferative rather than anti-apoptotic action

Obestatin is an endogenous peptide sharing a precursor with ghrelin. This study aims to investigate whether and how obestatin protects MES23.5 dopaminergic cells against 1-methyl-4- phenylpyridinium （MPP^＋）-induced neurotoxicity. MES23.5 cells were pretreated with obestatin （10^-13- 10^-6 mol/L） for 20 min prior to incubation with 200 μmol/L MPP^＋ for 12 or 24 h, or treated with obestatin alone （10^-13 to 10^-6 mol/L） for 0, 6, 12, and 24 h. The methyl thiazolyl tetrazolium （MTT） assay was used to measure cell viability. Flow cytometry was used to measure the caspase-3 activity and the mitochondrial transmembrane potential. Proliferating cell nuclear antigen （PCNA） protein levels were determined by Western blotting. Obestatin （10^-13 to 10^-7 mol/L） pretreatment blocked or even reversed the MPP^＋ induced reduction of viability in MES23.5 cells, but had no effect on MPP^＋-induced mitochondrial transmembrane potential collapse and caspase-3 activation. When applied alone, obestatin increased viability. Elevated PCNA levels occurred with 10^-7, 10^-9, 10^-11 and 10^-13 mol/L obestatin treatment for 12 h. The results suggest that the protective effects of obestatin against MPP^＋ in MES23.5 cells are due to its proliferation-promoting rather than anti-apoptotic effects.

Spatial and Temporal Differences of Climate Suitability of Ice and Snow Sports in Major Ski Tourism Destinations in China

Ice and snow tourism in China has grown significantly since the country successfully hosted the Beijing Winter *** conditions profoundly impact the development of ice and snow tourism;however,most studies have focused on constructing different climate suitability indicators for ice and snow tourism to evaluate individual regions,lacking horizontal comparative studies across multiple *** study aims to enrich the connotation of climate suitability for ice and snow sports,establish an evaluation model based on snowfall amount,temperature,and wind speed,and use daily meteorological data from 1991 to 2021 to horizontally compare the climate suitability for ice and snow sports in major ski tourism destinations in *** study boasts four major findings:1)the average ice and snow sports climate index of each region decreases over time,and the overall suitability of the climate for ice and snow sports is reducing;2)northern Xinjiang exhibits the most evident regional differentiation from‘very suitable’to‘generally suitable’;3)the spatial zoning of climate suitability for ice and snow sports exhibits heterogeneity,as northern Xinjiang is divided into two‘suitable and above’zones with rotating empirical orthogonal function(REOF).Correspondingly,the four provinces of Hebei,Heilongjiang,Jilin,and Liaoning are divided into three‘generally suitable and above’zones;4)snowfall amount is the main factor affecting the climate suitability of ice and snow sports in the major ski tourist destinations in China.

Metallogenesis of the Baidi Au-Sb deposit, southwest Guizhou Province, China: mineralogical and geochemical evidence from sulfur-bearing minerals

The Baidi Au-Sb deposit, which contains 8 t of Au and 10,979 Mt of Sb, is a typical and rare paragenetic deposit located in southwestern Guizhou Province, *** studies have focused on individual ores, but have not combined them to identify their paragenetic mechanism or metallogenic regularity. Therefore, we used field investigations, microscopic observations, and in situ analyses to identify the spatial distribution, mineral paragenesis, compositional evolution, and metallogenic material sources of the ore bodies. We also determined the Au and Sb paragenetic characteristics and the metallogenesis of the deposit. The main Au-bearing minerals in the deposit were early(Apy1–2) and late(Apy3) stage arsenopyrites, as well as pre-mineralization(Py1), mineralization(Py2–5), and late mineralization(Py6–7) stage pyrites. The main Sb-bearing minerals were stibnite(Snt), skinnerite, bournonite,and valentinite. The minerals formed in the order of Py1,Py2–3 + Apy1, Py4–5 + Apy2, Snt, and Py6–7 + *** δ34S values of the arsenopyrites and pyrites ranged from-5 to 5‰, while those of stibnite were mostly less than-5‰ in the later mineralization stages. Sulfur was provided by deep magmatic hydrothermal fluids, but sedimentary sulfur was added in the later stages. Moreover,the trace elemental contents fluctuated and eventually became similar to those of the sedimentary strata. By comprehensively considering the ores along with the geological characteristics of the deposit, we determined that deep magma provided the Au during ore formation. Later tectonic changes provided Sb from the sedimentary strata,which precipitated along fault expansion areas and produced Au and Sb paragenesis.

Classification and spectrum optimization method of grease based on infrared spectrum

The infrared(IR)absorption spectral data of 63 kinds of lubricating greases containing six different types of thickeners were obtained using the IR *** Kohonen neural network algorithm was used to identify the type of the lubricating *** results show that this machine learning method can effectively eliminate the interference fringes in the IR spectrum,and complete the feature selection and dimensionality reduction of the high-dimensional spectral *** 63 kinds of greases exhibit spatial clustering under certain IR spectrum recognition spectral bands,which are linked to characteristic peaks of lubricating greases and improve the recognition accuracy of these *** model achieved recognition accuracy of 100.00%,96.08%,94.87%,100.00%,and 87.50%for polyurea grease,calcium sulfonate composite grease,aluminum(Al)-based grease,bentonite grease,and lithium-based grease,*** on the different IR absorption spectrum bands produced by each kind of lubricating grease,the three-dimensional spatial distribution map of the lubricating grease drawn also verifies the accuracy of classification while recognizing the *** paper demonstrates fast recognition speed and high accuracy,proving that the Kohonen neural network algorithm has an efficient recognition ability for identifying the types of the lubricating grease.

Phonon-blockade-based multiple-photon bundle emission in a quadratically coupled optomechanical system

We propose a scheme to realize antibunched multiple-photon bundles based on phonon blockade in a quadratically coupled optomechanical *** adjusting the detunings to match the conditions of phonon blockade in the photon sidebands,we establish super-Rabi oscillation between zero-photon state and multiple-photon states with adjustable super-Rabi frequencies under appropriate single-phonon resonant *** the system dissipation into account,we numerically calculate the standard and generalized second-order functions of the cavity mode as well as the quantum trajectories of the state populations with Monte Carlo simulation to confirm that the emitted photons form antibunched multiple-photon ***,the desirable n-photon states are reconstructed after a direct phonon emission based on phonon blockade,and thus the single-phonon emission heralds the cascade emission of n-photon *** proposal shows that the optomechanical system can simultaneously behave as antibunched multiple-photon emitter and single-phonon *** a nonclassical source could have potential applications in quantum information science.

Copper (Cu^(2+)) induces degeneration of dopaminergic neurons in the nigrostriatal system of rats

Objective To study the effects of intranigral injection of different doses of CuSO45H2O on dopaminergic neuron in the nigrostriatal system of rats. Methods Wistar rats were divided into four groups, including control group, 10 nmol, 50 nmol and 200 nmol copper injected into left substantia nigra （SN） groups. Seven days after the intranigral injection of copper, dopamine （DA） contents in the striatum （Str） were measured by high performance lipid chromotophotography （HPLC）; the density of tyrosine hydroxylase （TH） positive axons in the Str was measured by TH staining method; TH and Caspase-3 mRNA expression in the SN were measured by semi-quantitative RT-PCR. We detected the activity of superoxide dismutase （SOD） in the lesioned midbrain of rats using biochemical methods. Results DA and its metabolites contents had no significant difference between control group and low dose （10 nmol） copper group. But from 50 nmol copper group, DA contents in the lesioned sides were reduced with the increase in the copper doses injected, showing a significant linear correlation （F = 34.16, P 〈 0.01）. In the 50 nmol copper group, TH positive axons in the Str decreased compared with those of the control and unlesioned sides （F = 121.9, P 〈 0.01）. In the 50 nmol copper group, TH mRNA expression decreased （t =3.12, P 〈 0.01） while Caspase-3 mRNA expression increased （t =8.96, P 〈 0.01） in the SN compared with the control. SOD activity decreased in the midbmin of rots treated with 50 nmol copper compared with that of the control （t = 2.33, P〈0.01）. Conclusion Copper could induce damage of dopaminergic neurons in the SN of rats through destroying antioxidant defenses and promoting apoptosis.

As ethnic medicine,the whole grass of plants in Cirsium was used as *** review focuses on the antimicrobial activity of plants in Cirsium,including antimicrobial components,against different types of microbes and bacteriostatic *** results showed that the main antimicrobial activity components in Cirsium plants were flavonoids,triterpenoids and phenolic acids,and the antimicrobial ability varied according to the species and the content of *** them,phenolic acids showed a strong antibacterial ability against Acinetobacter baumannii,Pseudomonas aeruginosa,and Enterococcus *** antibacterial mechanisms include:(1)damaging the cell membrane,cell walls,mitochondria and nucleus of bacteria;(2)inhibiting the synthesis of proteins and nucleic acids;(3)suppressing the synthesis of enzymes for tricarboxylic acid cycle pathways and glycolysis,and then killing the bacteria via inhibition of energy ***,most research results on antimicrobial activity of Cirsium plants are reported based on in vitro *** evidence from clinical data and comprehensive evaluation are needed.
