Appropriately controlled gene expression is fundamental for normal growth and survival of all living organisms. In eukaryotes, the tran- scription of protein-coding mRNAs is dependent on RNA polymerase II （Pol II）. The multi-subunit transcription cofactor Mediator complex is proposed to regulate most, if not all, of the Pol II-dependent transcription. Here we focus our discussion on two subunits of the Mediator complex, cyclin-dependent kinase 8 （CDK8） and its regulatory partner Cyclin C （CycC）, because they are either mutated or amplified in a variety of human cancers. CDK8 functions as an oncoprotein in melanoma and colorectal cancers, thus there are considerable interests in developing drugs specifically targeting the CDK8 kinase activity. However, to evaluate the feasibility of targeting CDK8 for cancer therapy and to understand how their dysregulation contributes to tumorigenesis, it is essential to elucidate the in vivo function and regulation of CDKS-CycC, which are still poorly understood in multi-cellular organisms. We summarize the evidence linking their dysregulation to various cancers and present our bioinformatics and computational analyses on the structure and evolution of CDK8. We also discuss the implications of these observations in tumorigenesis. Because most of the Mediator subunits, including CDK8 and CycC, are highly conserved during eukaryotic evolution, we expect that investigations using model organisms such as Drosophila will provide important insights into the function and regulation of CDK8 and CycC in different cellular and developmental contexts.

Objective To investigate the anti-inflammation effect and possible mechanism of Salvianic acid A(SAA)in mouse peritoneal *** Peritoneal macrophages were obtained from BALB/c *** induced nitric oxide(NO),tumor necrosis factor-alpha(TNF-α)and interleukin-6(IL-6)in supernatant,protein expression of inducible nitric oxide synthase(iNOS),matrix metalloproteinase-9(MMP-9)and activation of nuclear factor-kappa B(NF-κB)in the extract were *** SAA strongly inhibited the excessive production of NO,TNF-α and IL-6 in LPS-induced peritoneal macrophages in a concentration-dependent manner and blocked the expression of iNOS and *** with LPS alone increased the translocation of NF-κB(p65)from cytosol to the nucleus,but the SAA inhibited the translocation of NF-κB(p65).Conclusions The results showed that SAA had strong anti-inflammatory effects in LPS-stimulated peritoneal *** important mechanism is due to its inhibition of NF-κB activation.

Fine particle detachment and subsequent migration can lead to severe pore plugging and consequent permeability decline. Therefore, it is crucial to quantify the critical condition when fine particle detachment occurs. The frequently observed deviations or even contradictions between experimental results and theoretical predictions of fines detachment arise from an insufficient understanding of adhesion force that can be highly influenced by salinity and temperature. To clarify the intrinsic influence of salinity and temperature on fines detachment, adhesion forces between carboxyl microspheres and hydrophilic silica substrates in an aqueous medium were measured at various salinities and temperatures using atomic force microscopy(AFM). The AFM-measured adhesion force decreases with increasing salinity or temperature. Trends of mean measured adhesion forces with temperature and salinity were compared with the DLVO and XDLVO theories. DLVO theory captured the trend with temperature via the impact of temperature on electric double layer interactions, whereas XDLVO theory captured the observed trend with salinity via the impact of salinity on the repulsive hydration force. Our results highlight the significance of hydration force in accurately predicting the fate of fines in porous media.

Aerosol hygroscopicity and liquid water content(ALWC)have important influences on the environmental and climate effect of *** this study,we measured the hygroscopic growth factors(GF)of particles with dry diameters of 40,80,150,and 200 nm during the wintertime in *** the GF-derived hygroscopicity parameter(κ_(gf))and ALWC increased with particle size,but displayed differing diurnal variations,withκ_(gf)peaking around the midday,while ALWC peaking in the early ***,ammonium and oxygenated organic aerosols(OOA)were found as the chemical components mostly strongly correlated with ALWC.A closure study suggests that during midday photo-oxidation and nighttime high ALWC periods,theκof organic aerosols(κ_(org))was underestimated when using previous ***,we re-constructed parameterizations forκ_(org)and the oxidation level of organics for these periods,which indicates a higher hygroscopicity of photochemically formed OOA than the aqueous OOA,yet both being much higher than the generally assumed OOA ***,in a typical high ALWC episode,concurrently increased ALWC,nitrate,OOA as well as aerosol surface area and mass concentrations were observed under elevated ambient *** strongly indicates a coupled effect that the hygroscopic secondary aerosols,in particular nitratewith strong hygroscopicity,led to large increase in ALWC,which in turn synergistically boosted nitrate and OOA formation by heterogeneous/aqueous *** interaction may represent an important mechanism contributing to enhanced formation of secondary aerosols and rapid growth of fine particulate matter under relatively high RH conditions.
