Objective:Xeroderma pigmentosum(XP)is a rare autosomal recessive dermatosis caused by genetic defects of DNA *** study was performed to detect and analyze the genes of 2 Uygur patients with XP and their families and assess the patients’phenotypes,which may enrich the understanding of the genetic skin disorder spectrum in Xinjiang ***:We collected the clinical data from 2 patients with XP and peripheral blood samples from the patients and their family *** patients’DNA was sequenced and detected by Sanger sequencing,and gene mutations were ***:The proband in family 1 presented with brown maculae at the exposure site and squamous cell carcinoma secondary to a facial *** proband had a homozygous nucleotide variation of XPC c.2251-2A>G(A change from A to G in the penultimate position of the intron before the 2251 position in the coding region),which was a shear *** this family,both parents were heterozygous,and no similar mutation detected in the *** family 2,the proband had scattered black brown spots and papules on the trunk and *** his younger sister was also a *** proband and his younger sister had homonucleotide variation of XPA c.631C>T,which was nonsense mutation,resulting in the codon for Arg No.211 being changed into termination codon(***211X),thus terminating the peptide chain synthesis *** the normal individuals in the two families were heterozygotes,and homozygous mutations occurred in all the patients,which was consistent with the autosomal recessive ***:XP is rare in Uygur *** study expanded the mutation spectrum of XP and provided a basis for early diagnosis,treatment,prognostic prediction,and prenatal genetic consultation.

Rapamycin induces differentiation of glioma stem/progenitor cells by activating autophagy

Glioma stem/progenitor cells(GSPCs) are considered to be responsible for the initiation,propagation,and recurrence of *** factors determining their differentiation remain poorly *** evidences indicate that alterations in autophagy may influence cell fate during mammalian development and ***,we investigated the role of autophagy in GSPC ***-2 cells were treated with rapamycin,3-methyladenine(3-MA) plus rapamycin,E64d plus rapamycin,or untreated as ***-2 cell xenografts in nude mice were treated with rapamycin or 3-MA plus rapamycin,or untreated as *** blotting and immunocytochemistry showed up-regulation of microtubule-associated protein light chain-3(LC3)-II in rapamycin-treated *** neurosphere formation rate and the number of cells in each neurosphere were significantly lower in the rapamycin treatment group than in other ***-time PCR and immunocytochemistry showed down-regulation of stem/progenitor cell markers and up-regulation of differentiation markers in rapamycin-treated *** electron microscopy revealed autophagy activation in rapamycin-treated tumor cells in *** revealed decreased Nestin-positive cells and increased GFAP-positive cells in rapamycin-treated tumor *** results indicate that rapamycin induces differentiation of GSPCs by activating autophagy.

Transient state analysis of a rub-impact rotor system during maneuvering flight

Maneuvering flight substantially affects the dynamic behavior of rotors;particularly,such flight may cause rubbing between a rotor and stator,which is one of the most serious damages in aircraft *** this paper,a nonlinear dynamic model for describing the dynamic characteristics of a rub-impact rotor system during maneuvering flight is established based on the Lagrange ***,numerical simulations employing the Newmark method are performed,delving into the detailed discussion of the influence of parameters such as rotational speed and maneuvering flight on the transient and steady-state responses of the rotor *** effect mechanism of maneuver load and its coupling with rub impact is *** results show that the impact response induced by maneuvering flight is more obvious in the subcritical state than in the supercritical *** additional stiffness and damping are also induced;in particular,the additional damping has a coupling ***,the rub impact imposes an additional constraint on the rotor system,thereby weakening the influence of the maneuver load and becoming the major factor that determines the dynamic behavior of the rotor system at high speeds.

Full waveform inversion based on hybrid gradient

The low-wavenumber components in the gradient of full waveform inversion(FWI)play a vital role in the stability of the ***,when FWI is implemented in some high frequencies and current models are not far away from the real velocity model,an excessive number of low-wavenumber components in the gradient will also reduce the convergence rate and inversion *** solve this problem,this paper firstly derives a formula of scattering angle weighted gradient in FWI,then proposes a hybrid *** hybrid gradient combines the conventional gradient of FWI with the scattering angle weighted gradient in each inversion frequency band based on an empirical formula derived *** weighted hybrid mode,we can retain some low-wavenumber components in the initial lowfrequency inversion to ensure the stability of the inversion,and use more high-wavenumber components in the high-frequency inversion to improve the convergence *** results of synthetic data experiment demonstrate that compared to the conventional FWI,the FWI based on the proposed hybrid gradient can effectively reduce the low-wavenumber components in the gradient under the premise of ensuring inversion *** also greatly enhances the convergence rate and inversion accuracy,especially in the deep part of the *** the field marine seismic data experiment also illustrates that the FWI based on hybrid gradient(HGFWI)has good stability and adaptability.

Dynamic interwell connectivity analysis of multi-layer waterflooding reservoirs based on an improved graph neural network

The analysis of interwell connectivity plays an important role in the formulation of oilfield development plans and the description of residual oil distribution. In fact, sandstone reservoirs in China's onshore oilfields generally have the characteristics of thin and many layers, so multi-layer joint production is usually adopted. It remains a challenge to ensure the accuracy of splitting and dynamic connectivity in each layer of the injection-production wells with limited field data. The three-dimensional well pattern of multi-layer reservoir and the relationship between injection-production wells can be equivalent to a directional heterogeneous graph. In this paper, an improved graph neural network is proposed to construct an interacting process mimics the real interwell flow regularity. In detail, this method is used to split injection and production rates by combining permeability, porosity and effective thickness, and to invert the dynamic connectivity in each layer of the injection-production wells by attention *** on the material balance and physical information, the overall connectivity from the injection wells,through the water injection layers to the production layers and the output of final production wells is established. Meanwhile, the change of well pattern caused by perforation, plugging and switching of wells at different times is achieved by updated graph structure in spatial and temporal ways. The effectiveness of the method is verified by a combination of reservoir numerical simulation examples and field example. The method corresponds to the actual situation of the reservoir, has wide adaptability and low cost, has good practical value, and provides a reference for adjusting the injection-production relationship of the reservoir and the development of the remaining oil.

Learning in games:a systematic review

Game theory studies the mathematical models for self-interested individuals. Nash equilibrium is arguably the most central solution in game theory. While finding the Nash equilibrium in general is known as polynomial parity arguments on directed graphs(PPAD)-complete, learning in games provides an alternative to approximate Nash equilibrium, which iteratively updates the player's strategy through interactions with other players. Rules and models have been developed for learning in games, such as fictitious play and no-regret learning. Particularly, with recent advances in online learning and deep reinforcement learning,techniques from these fields greatly boost the breakthroughs in learning in games from theory to *** a result, we have witnessed many superhuman game AI systems. The techniques used in these systems evolve from conventional search and learning to purely reinforcement learning(RL)-style learning methods,gradually getting rid of the domain knowledge. In this article, we systematically review the above techniques,discuss the trend of basic learning rules towards a unified framework, and recap applications in large ***, we discuss some future directions and make the prospect of future game AI systems. We hope this article will give some insights into designing novel approaches.

Monotropein Induced Apoptosis and Suppressed Cell Cycle Progression in Colorectal Cancer Cells

Objective: To determine whether monotropein has an anticancer effect and explore its potential mechanisms against colorectal cancer(CRC) through network pharmacology and molecular docking combined with experimental verification. Methods: Network pharmacology and molecular docking were used to predict potential targets of monotropein against CRC. Cell counting kit assay, plate monoclonal assay and microscopic observation were used to investigate the antiproliferative effects of monotropein on CRC cells HCT116, HT29 and LoVo. Flow cytometry and scratch assay were used to analyze apoptosis and cell cycle, as well as cell migration, respectively in HCT116, HT29, and LoVo cells. Western blotting was used to detect the expression of proteins related to apoptosis, cell cycle, and cell migration, and the expression of proteins key to the Akt pathway. Results: The Gene Ontology and Reactome enrichment analyses indicated that the anticancer potential of monotropein against CRC might be involved in multiple cancer-related signaling pathways. Among these pathways, RAC-beta serine/threonine-protein kinase(Akt1, Akt2), cyclin-dependent kinase 6(CDK6), matrix metalloproteinase-9(MMP9), epidermal growth factor receptor(EGFR), cell division control protein 42 homolog(CDC42) were shown as the potential anticancer targets of monotropein against CRC. Molecular docking suggested that monotropein may interact with the 6 targets(Akt1, Akt2, CDK6, MMP9, EGFR, CDC42). Subsequently, cell activity of HCT116, HT29 and LoVo cell lines were significantly suppressed by monotropein(P<0.05). Furthermore, our research revealed that monotropein induced cell apoptosis by inhibiting Bcl-2 and increasing Bax, induced G_1–S cycle arrest in colorectal cancer by decreasing the expressions of CyclinD1, CDK4 and CDK6, inhibited cell migration by suppressing the expressions of CDC42 and MMP9(P<0.05), and might play an anticancer role through Akt signaling pathway. Evolution and Determinants of Population Agglomeration in Less Developed Metropolitan Areas:A Case Study of the Taiyuan Metropolitan Area,China

It is of importance to enhance the urban areas'capacity for population aggregation in underdeveloped regions,aiming to rectify the imbalanced and insufficient pattern of economic development in *** the Taiyuan Metropolitan Area(TMA)in central China as a case study,this paper examines the evolutionary process and characteristics of population agglomeration from 2000 to 2020,and identifies factors associated with agglomeration and their spatial *** findings indicated that:1)against the background of sustained population shrinkage in the provincial area,the TMA showed a demographic trend of steady increase,albeit with a decelerated growth *** the metropolitan area,urban population size continued to grow rapidly,whereas the rural areas endured sustained *** in city size continued to widen,and the polarization of concentrated population in the core cities kept increasing.2)Agglomerations in both secondary and service industries had significant positive effects on local population agglomeration,with the former effect being *** economic development,government fiscal expenditure,and financial advancement all contributed to facilitating local population *** a spatial spillover perspective,service agglomeration and financial development promoted population agglomeration in surrounding ***,fiscal expenditure inhibited such *** for industrial agglomeration and regional economic development,their spatial spillover effects were *** results obtained reveal several policy implications aimed at enhancing the population agglomeration capacity of the metropolitan area in underdeveloped regions during the new era.
