The addition of packing material for high performance hydrophobic interaction chromatograghy （HPHIC） into the denaturant solution to prevent, or depress protein aggregation in the denatuaration process is presented. The renaturation of α-chymotrypsin （α-Chy） denatured with guanidine hydrochloride （GuHCl） solution indicated that renaturation efficiency can be enhanced from 36.1% to 59.0% by this new method. The structure of the ligand linking of HPHIC packings is also important for the protein renaturation.

To analyze the differences in the transport and distribution of different types of proppants and to address issues such as the short effective support of proppant and poor placement in hydraulically intersecting fractures,this study considered the combined impact of geological-engineering factors on *** reservoir production parameters and the discrete elementmethod,multispherical proppants were ***,a 3D fracture model,based on the specified conditions of the L block,employed coupled(Computational Fluid Dynamics)CFD-DEM(Discrete ElementMethod)for joint simulations to quantitatively analyze the transport and placement patterns of multispherical proppants in intersecting *** indicate that turbulent kinetic energy is an intrinsic factor affecting proppant ***,the efficiency of placement and migration distance of low-sphericity quartz sand constructed by the DEM in the main fracture are significantly reduced compared to spherical ceramic proppants,with a 27.7%decrease in the volume fraction of the fracture surface,subsequently affecting the placement concentration and damaging fracture *** to small-angle fractures,controlling artificial and natural fractures to expand at angles of 45°to 60°increases the effective support length by approximately 20.6%.During hydraulic fracturing of gas wells,ensuring the fracture support area and post-closure conductivity can be achieved by controlling the sphericity of proppants and adjusting the perforation direction to control the direction of artificial fractures.

Suitable temperature and light intensity play important roles in the formation of harmful algae blooms(HABs),which can pose serious threats to aquatic ecosystems and human *** this study,we measured the growth,physiological function,and paralytic shellfish toxins(PSTs)production of Alexandrium pacificum(CCMA-272),a strain isolated from East China Sea,at different temperatures(15,20,and 25℃)and light intensities(30,60,and 90μmol photons/(m^(2)·s)).Results indicate that temperature and light intensity significantly affected the growth,physiology,and toxigenic potentials of *** optimal conditions for the growth of *** were observed at 20℃ under60μmol photons/(m^(2)·s).Regarding the production of PSTs,this strain of *** produced 12 PSTs,including carbamate toxins:saxitoxin(STX),neosaxitoxin(NEO),and gonyautoxin 1–4(GTX1,GTX2,GTX3,GTX4);dicarbamoyl toxins:dicarbamoylsaxitoxin(dcSTX),dicarbamoylgonyautoxin 2,3(dcGTX2,dcGTX3);and N-sulfocarbamoyl toxins:N-sulfocarbamoylgonyautoxin 1,2(C1,C2),and gonyautoxin 5(GTX5).Among all the PSTs,C2 was the most *** temperature(15℃)and high light intensity(90μmol photons/(m^(2)·s))were beneficial for the production of PSTs in *** cultured at 20 and 25℃,*** generated comparable total quantities of PSTs,yet the toxicity levels were lower at 25℃.Intra-cellular PSTs contents were greater than extra-cellular PSTs contents,except those under the condition of 25℃ with 30μmol photons/(m^(2)·s).However,as the increase of temperature,*** released more amounts of analogues with higher toxicity levels(e.g.,STX and dcGTX_(2))into the environment than *** findings emphasize the significant sensitivity of *** to temperature and light intensity,highlighting the importance of evaluating both intra-cellular and extra-cellular PSTs for assessing its toxicity and aiding in the prediction and management of HABs.

Control of N-nitrosodimethylamine(NDMA)in drinking water could be achieved by removing its precursors as one practical ***,superfine powdered activated carbons with a diameter of about 1μm(SPACs)were successfully prepared by grinding powdered activated carbon(PAC,D50=24.3μm)and applied to remove model NDMA precursors,***(RAN)and nizatidine(NIZ).Results fromgrain diameter experiments demonstrated that the absorption velocity increased dramatically with decreasing particle size,and the maximum increase in k2 was 26.8-folds for RAN and 33.4-folds for ***,kinetic experiments explained that rapid absorption could be attributed to the acceleration of intraparticle diffusion due to the shortening of the diffusion ***,performance comparison experiments suggested that the removal of RAN and NIZ(C_(0)=0.5 mg/L)could reach 61.3%and 60%,respectively,within 5 min,when the dosage of SAPC-1.1(D_(50)=1.1μm)was merely 5 mg/L,while PAC-24.3 could only eliminate 17.5%and 18.6%.The adsorption isotherm was well defined by Langmuir isotherm model,indicating that the adsorption of RAN/NIZ was a monolayer coverage *** adsorption of RAN or NIZ by SAPC-1.1 and PAC-24.3 was strongly pH dependent,and high adsorption capacity could be observed under the condition of pH>pk_(a)+*** coexistence of humic acid(HA)had no significant effect on the adsorption performance because RAN/NIZ may be coupled with HA and removed *** coexistence of anions had little effect on the adsorption *** study is expected to provide an alternative strategy for drinking water safety triggered by NDMA.

Schottky mass spectrometry utilizing heavy-ion storage rings is a powerful technique for the precise mass and decay half-life measurements of highly charged *** to the nondestructive ion detection features of Schottky noise detectors,the number of stored ions in the ring is determined by the peak area in the measured revolution frequency *** of their intrinsic amplitude-frequency characteristic(AFC),Schottky detector systems exhibit varying sensitivities at different *** low-energy electron-cooled stored ions,a new method is developed to calibrate the AFC curve of the Schottky detector system of the Experimental Cooler Storage Ring(CSRe)storage ring located in Lanzhou,*** the amplitude-calibrated frequency spectrum,a notable refinement was observed in the precision of both the peak position and peak *** a result,the storage lifetimes of the electron-cooled fully ionized^(56)Fe^(26+)ions were determined with high precision at beam energies of 13.7 and 116.4 MeV/u,despite of frequency drifts during the *** electron cooling was turned off,the effective vacuum condition experienced by the 116.4 MeV/u^(56)Fe^(26+)ions was determined using amplitude-calibrated spectra,revealing a value of 2×10^(−10)mbar,which is consistent with vacuum gauge readings along the CSRe *** method reported herein will be adapted for the next-generation storage ring of the HIAF facility under construction in Huizhou,*** can also be adapted to other storage ring facilities worldwide to improve precision and enhance lifetime measurements using many ions in the ring.

Measurements from a hyperspectral infrared (HIR) sounder onboard a satellite in geostationary orbit not only provide atmospheric thermodynamic information, but also can be used to infer dynamic information with high temporal resolution. Radiance measurements from the Geostationary Interferometric Infrared Sounder (GIIRS), obtained with 15-min temporal resolution during Typhoon Maria (2018) and 30-min temporal resolution during Typhoon Lekima (2019), were used to derive three-dimensional (3D) horizontal winds by tracking the motion of atmospheric moisture. This work focused on the impact of assimilation of 3D winds on typhoon analyses and forecasts using the operational NWP model of the China Meteorological Administration (CMA-MESO), and improved understanding of the potential benefits of assimilating dynamic information from geostationary sounder data with higher temporal resolution. The standard deviation of the observations minus simulations revealed that the accuracy of the derived 3D winds with 15-min resolution was higher than that of derived winds with 30-min resolution. Experiments showed that the assimilation system can effectively absorb the information of the derived 3D winds, and that dynamic information from clear-sky areas can be transferred to typhoon areas. In typhoon prediction, assimilation of the derived 3D winds had greatest influence on the typhoon track, and less influence on the maximum wind speed. Assimilation of the derived 3D winds reduced the average track error by 17.4% for Typhoon Maria (2018) and by 3.5% for Typhoon Lekima (2019) during their entire 36-h forecasts initiated at different times. Assimilation of GIIRS dynamic information can substantially improve forecasts of heavy precipitation by CMA-MESO. Results indicate that the assimilation of dynamic information from high-temporal-resolution geostationary HIR sounder data adds value for improved numerical weather prediction. Tanreqing Injection Inhibits Activation of NLRP3 Inflammasome in Macrophages Infected with Influenza A Virus by Promoting Mitophagy

Objective: To investigate the inhibitory effect of Tanreqing Injection(TRQ) on the activation of nucleotide-binding oligomerization domain-like receptor pyrin domain containing 3(NLRP3) inflammasome in macrophages infected with influenza A virus and the underlying mechanism based on mitophagy pathway. Methods:The inflammatory model of murine macrophage J774A.1 induced by influenza A virus [strain A/Puerto Rico/8/1934(H1N1), PR8] was constructed and treated by TRQ, while the mitochondria-targeted antioxidant Mito-TEMPO and autophagy specific inhibitor 3-methyladenine(3-MA) were used as controls to intensively study the anti-inflammatory mechanism of TRQ based on mitophagy-mitochondrial reactive oxygen species(mt ROS)-NLRP3 inflammasome pathway. The levels of NLRP3, Caspase-1 p20, microtubule-associated protein 1 light chain 3 Ⅱ(LC3Ⅱ) and P62 proteins were measured by Western blot. The release of interleukin-1β(IL-1β) was tested by enzyme linked immunosorbent assay, the mt ROS level was detected by flow cytometry, and the immunofluorescence and co-localization of LC3 and mitochondria were observed under confocal laser scanning microscopy. Results: Similar to the effect of Mito-TEMPO and contrary to the results of 3-MA treatment, TRQ could significantly reduce the expressions of NLRP3, Caspase-1 p20, and autophagy adaptor P62, promote the expression of autophagy marker LC3Ⅱ, enhance the mitochondrial fluorescence intensity, and inhibit the release of mt ROS and IL-1β(all P<0.01).Moreover, LC3 was co-localized with mitochondria, confirming the type of mitophagy. Conclusion: TRQ could reduce the level of mt ROS by promoting mitophagy in macrophages infected with influenza A virus, thus inhibiting the activation of NLRP3 inflammasome and the release of IL-1β, and attenuating the inflammatory response.
