Evaluation of the gastrointestinal anti-motility effect of Anacardium occidentale stem bark extract:A mechanistic study of antidiarrheal activity

Diarrhea is a prevalent gastrointestinal problem associated with fatal implications.It is a huge public health concern that requires better alternatives to current drugs.This study investigated the mechanisms involved in the antidiarrheal activity of Anacardium occidentale(Ao) stem bark extract,a plant commonly used in the management of diarrhea in Nigeria.Methanolic stem bark extract of the plant was partitioned into three fractions:hexane fraction,ethyl acetate fraction(Ao EF) and methanol fraction.In vitro studies on the effect of these fractions on guinea pig ileum(GPI) strips,as well as the modulatory effect of Ao EF on standard agonists-and antagonists-induced GPI contraction and relaxation,revealed Ao EF as the most active fraction.In vivo studies to assess the effect of Ao EF on the dopaminergic,muscarinic,and serotonergic pathways were carried out using gastric emptying(GE) and gastrointestinal transit(GT) as experimental end points.Ao EF was subjected to GC-MS analysis,while the identified compounds were docked with the muscarinic acetylcholine receptor M3(CHRM3) using Autodock Vina.Results indicated that Ao EF inhibited GE and GT via inhibition of CHRM3.In addition,GC-MS analysis revealed the presence of 24 compounds in Ao EF,while docking indicated that octadecanoic acid 2-(2-hydroxylethoxy)ethyl ester exhibited the highest binding affinity to CHRM3.This study indicated that the antidiarrheal activity of Ao is through its antimotility effect via the inhibition of the muscarinic pathway.And since none of the identified compounds exhibited higher binding affinity to CHRM3 relative to loperamide,the antimotility activity of these phytoconstituents may be via synergism.

Portable dielectrophoresis for biology:ADEPT facilitates cell trapping,separation,and interactions

Dielectrophoresis is a powerful and well-established technique that allows label-free,non-invasive manipulation of cells and particles by leveraging their electrical properties.The practical implementation of the associated electronics and user interface in a biology laboratory,however,requires an engineering background,thus hindering the broader adoption of the technique.In order to address these challenges and to bridge the gap between biologists and the engineering skills required for the implementation of DEP platforms,we report here a custom-built,compact,universal electronic platform termed ADEPT(adaptable dielectrophoresis embedded platform tool)for use with a simple microfluidic chip containing six microelectrodes.The versatility of the open-source platform is ensured by a custom-developed graphical user interface that permits simple reconfiguration of the control signals to address a wide-range of specific applications:(i)precision positioning of the single bacterium/cell/particle in the micrometer range;(ii)viability-based separation by achieving a 94%efficiency in separating live and dead yeast;(iii)phenotype-based separation by achieving a 96%efficiency in separating yeast and Bacillus subtilis;(iv)cell–cell interactions by steering a phagocytosis process where a granulocyte engulfs E.coli RGB-S bacterium.Together,the set of experiments and the platform form a complete basis for a wide range of possible applications addressing various biological questions exploiting the plug-and-play design and the intuitive GUI of ADEPT.

Detection and observation of reactive intermediates is an essential step in investigation of reaction pathways.However,most reactive intermediates are unstable and present at low concentrations;their short lifetimes make them difficult to detect and characterize.Supramolecular containers offer opportunities for the stabilization and characterization of those labile species,through isolation from the media and protection inside the cavity of the host.In this review,we summarize the examples of labile reaction intermediates that are stabilized and characterized with the help of supramolecular containers.The container compounds include carcerands,deep cavitands and amide naphthotubes.We focus on unstable guest species-cyclobutadiene,benzocyclopropenone,o-benzyne,1,2,4,6-cycloheptatetraene,anti-Bredt's olefin,fluorophenoxycarbene,O-acylisoamide,and hemiaminalthat act as intermediates in certain organic reactions.

Background There is a strong,bi-directional link between tuberculosis(TB)and undernutrition:TB often causes undernutrition,and undernourished people are more likely to contract TB and experience worse outcomes.Globally,several TB nutritional support programmes exist;however,evidence on their effectiveness is limited and contested.This study evaluates the effect of a nutritional support programme implemented for people with TB in the Atsimo-Andrefana region,Madagascar in 2022.Within this programme,undernourished people with TB[with a body mass index(BMI)ofkg/m]receive 0.6 L of vegetable oil and 6.0 kg of a soy-wheat blend per month throughout their TB treatment.Methods We analysed secondary non-governmental organisation data collected between January and Novem-ber 2022 in the Atsimo-Andrefana region,Southern Madagascar,including information on an individual's medical conditions(e.g.,type of TB,treatment outcomes)and nutritional status measured prior to,during,and after comple-tion of treatment(e.g.,height,weight,mid-upper arm circumference).We conducted descriptive analyses of patient baseline characteristics and outcomes to assess the impact of the provided nutritional support on the BMI of people With TB.Results A total of 1310 people with TB were included in the study[9.9%(130)children under the age of 5,32.1%(420)children between 5 and 18 years,58.0%(760)adults].55.4% of children under 5,28.1% of children between ages 5 and 18,and 81.3%of adults were undernourished at treatment initiation.42.3%(55/130)of children under 5 experi-enced severe acute malnutrition at treatment uptake.While the average BMl of adults with TB receiving food support increased over time,from 17.1 kg/m^(2)(interquartile range:15.8-18.3,range:10.3-22.5)to 17.9 kg/m^(2)(interquartile range:16.6-19.1,range:11.9-24.1),most adults remained undernourished even after completing TB treatment.Conclusions The current TB nutritional support programme falls short of sufficiently increasing the BMl of people with TB to overcome malnutrition.The

Refinement Mechanism of Microstructure of Undercooled Nickel Based Alloys

Through the use of purification and recirculation superheating techniques on molten glass,the Ni65Cu33Co2 alloy was successfully undercooled to a maximum temperature of 292 k.High-speed photography was employed to capture the process of interface migration of the alloy liquid,allowing for an analysis of the relationship between the morphological characteristics of the alloy liquid solidification front and the degree of undercooling.Additionally,the microstructure of the alloy was examined using metallographic microscopy,leading to a systematic study of the microscopic morphological characteristics and evolution laws of the refined structure during rapid solidification.The research reveals that the grain refining mechanism of the Ni-Cu-Co ternary alloy is consistent with that of the binary alloy(Ni-Cu).Specifically,under low undercooling conditions,intense dendritic remelting was found to cause grain refinement,while under high undercooling conditions,recrystallization driven by accumulated stress and plastic strain resulting from the interaction between the liquid flow and the primary dendrites caused by rapid solidification was identified as the main factor contributing to grain refinement.Furthermore,the study highlights the significant role of the Co element in influencing the solidification rate and reheat effect of the alloy.The addition of Co was also found to facilitate the formation of non-segregated solidification structure,indicating its importance in the overall solidification process.

Highly efficient and stable organic solar cells with SnO_(2)electron transport layer enabled by UV-curing acrylate oligomers

The interfaces between the inorganic metal oxide and organic photoactive layer are of outmost importance for efficiency and stability in organic solar cells(OSCs).Tin oxide(SnO_(2))is one of the promising candidates for the electron transport layer(ETL)in high-performance inverted OSCs.When a solution-processed SnO_(2)ETL is employed,however,the presence of interfacial defects and suboptimal interfacial contact can lower the power conversion efficiency(PCE)and operational stability of OSCs.Herein,highly efficient and stable inverted OSCs by modification of the SnO_(2)surface with ultraviolet(UV)-curable acrylate oligomers(SAR and OCS)are demonstrated.The highest PCEs of 16.6%and 17.0%are achieved in PM6:Y6-BO OSCs with the SAR and OCS,respectively,outperforming a device with a bare SnO_(2)ETL(PCE 13.8%).The remarkable enhancement of PCEs is attributed to the optimized interfacial contact,leading to mitigated surface defects.More strikingly,improved light-soaking and thermal stability strongly correlated with the interfacial defects are demonstrated for OSCs based on SnO_(2)/UV cross-linked resins compared to OSCs utilizing bare SnO_(2).We believe that UV cross-linking oligomers will play a key role as interfacial modifiers in the future fabrication of large-area and flexible OSCs with high efficiency and stability.

Prevalence of Work-Aggravated Asthma among Bakery Workers in Abidjan (Cote d’Ivoire)

Introduction: Work-aggravated asthma is pre-existing or concomitant asthma whose symptoms are aggravated by the work environment. The aim of this study was to determine the prevalence of this pathology and its associated factors among bakery workers in Abidjan. Materials and Methods: This descriptive and analytical cross-sectional study was conducted over a period of four (4) months from 18 December 2019 to 18 April 2020. Two questionnaires were used, one on employees and the other on the indoor environment of bakeries. In addition, a spirometry test was performed on all bakery workers. Statistical analysis was performed using stata 15.1 software. Results: A total of 599 bakery employees, including bakers (59.73%), sales assistants/ cashiers (23.52%), cleaners (6.34%) and administrative staff (10.18%), were investigated. The mean age was 30.8 ± 8 years, with a sex ratio (M/F) = 2.2. Asthma symptoms were found in 95 (15.86%) employees, of whom 74 (77.9%) had work-related asthma and 11 (14.9%) had asthma aggravated by work. The factors associated with work-aggravated asthma were personal or family history of allergy or atopy [ORa = 3.75;CI95%: 1.56 - 8.93;p = 0.003], exposure to dust [ORa = 5.01;CI95%: 1.43 - 7.50;p = 0.011] and humidity level (60% - 70%) [ORa = 1.80;CI 95%: 0.99 - 3.28;p = 0.05]. Conclusion: Work-aggravated asthma is a reality in bakeries in Abidjan, with an estimated prevalence of 14.9%. Two of the three factors associated with this condition suggest a link with indoor air pollution. Combating air pollution in these establishments must therefore be a priority for the relevant authorities, in order to provide employees with a working environment that protects their health.

Growth, Development and Yield of kenaf as Affected by Planting Dates and N Fertilization

kenaf (Hibiscus cannabinus L) consists of various beneficial components like stalks, seeds, leaves, fibers, oils, proteins, allelopathic chemicals, and fiber strands, among other things. Despite the numerous uses of the crop, there is little or no information on optimum agronomic practices such as planting date and N fertilization of the crop in the Upper East Region (UER) of Ghana where the crop is widely cultivated by smallholder farmers. Field experiments were therefore carried out in 2020 and repeated during the 2021 cropping season in the study area. The objective of the study was to determine appropriate planting date and N fertilization for increased kenaf productivity. In each year, the treatments consisted of 3 × 5 factorial combinations of three planting dates (1st July, 7th July and 14th July) and five levels of N (0, 20, 40, 60 and 80 kg/ha) replicated three times. The design of the experiment was a split-plot with the N fertilizer as the main plot and the planting date assigned to sub plot. The results showed that, planting kenaf in early (1st) July or N fertilization at the rate of 60 kg/ha increased plant density, stem height, stem diameter, dry bast and core yields in both cropping seasons.

Background: Since 2021, high-risk Human Papilloma Virus (HR-HPV) testing has been the recommended screening test for cervical cancer for all settings;either used alone in a “test and treat” strategy, or with a triage test, with or without biopsy, before treatment. Cameroon has rolled out immunization against HPV 16 and 18, but studies show a higher prevalence of non-16/18 HR-HPV types. Objectives: Determine the prevalence of precancerous lesions, in women with HR-HPV infection and evaluate association of digital cervicography (DC) VIA/VILI positivity with HPV serotype, as a measure of their contribution to precancer and cancer incidence. Methodology: The study was cross-sectional, descriptive, and analytic. It took place at the Etoug-Ebe and Ekoudoum Baptist Hospitals in Yaoundé, during the period April-September 2022. We reviewed the records of women screened for cervical cancer between February 2020 and December 2021 and evaluated the prevalence of lesions on digital cervicography (DC) with VIA/VILI for women positive for HR-HPV serotypes. The data were analyzed using SPSS version 20.0 for Windows. P values Results: We identified 315 cases with a positive HR-HPV deoxyribonucleic acid (DNA) test, 224 (71.1%) had a DC VIA/VILI triage test done. Of these, 30 (13.4%) women had a positive DC VIA/VILI, with five women (2.2%) having lesions suggestive of cancer. Out of 11 cases positive for HPV 16 alone, 05 (45.5%) had a positive DC VIA/VILI test. Of the 14 cases positive for HPV 18 alone, 03 (21.4%) had a positive VIA/VILI, meanwhile only 19 (10.7%) of the 177 cases positive for non-16/18 HPV had a positive VIA/VILI test. Conclusion: A high proportion of women (13.4%) with HR HPV had a positive DC VIA/VILI, with a significant proportion (2.2%) having lesions suggestive of invasive cervical cancer HR-HPV serotype was associated with DC VIA/VILI positivity;HPV 16 had the strongest association (45.5%), followed by HPV 18 (21.4%), and non-16/18 HR-HPV (10.7%), suggesting a decreasing order of oncogenicity.

DNA Barcoding of Insects and Its Direct Application for Plant Protection

The introduction of invasive insect pests across national borders has become a major concern in crop production. Accordingly, national plant protection organizations are challenge to reinforce their monitoring strategies, which are hampered by the weight and size of inspection equipment, as well as the taxonomic extensiveness of interrupted species. Moreover, some insect pests that impede farmer productivity and profitability are difficult for researchers to address on time due to a lack of appropriate plant protection measures. Farmers’ reliance on synthetic pesticides and biocontrol agents has resulted in major economic and environmental ramifications. DNA barcoding is a novel technology that has the potential to improve Integrated Pest Management decision-making, which is dependent on the ability to correctly identify pest and beneficial organisms. This is due to some natural traits such as phenology or pesticide susceptibility browbeaten by IPM strategies to avert pest establishment. Specifically, Deoxyribonucleic acid (DNA) sequence information was applied effectively for the identification of some micro-organisms. This technology, DNA barcoding, allows for the identification of insect species by using short, standardized gene sequences. DNA barcoding is basically based on repeatable and accessible technique that allows for the mechanisation or automation of species discrimination. This technique bridges the taxonomic bio-security gap and meets the International Plant Protection Convention diagnostic standards for insect identification. This review therefore discusses DNA barcoding as a technique for insect pests’ identification and its potential application for crop protection.
