The p.rip.eral rep.rtoire of CD4＋ T lymp.ocytes contains autoreactive cells that remain tolerant through several mechanisms. However, nonsp.cific CD4＋ T cells can be activated in p.ysiological conditions as in the course of an ongoing immune resp.nse, and their outcome is not yet fully understood. Here, we investigate the fate of human naive CD4＋ lymp.ocytes activated by dendritic cells （DCs） p.esenting endogenous self-p.p.ides in comp.rison with lymp.ocytes involved in alloresp.nses. We generated memory cells （Tmem） from p.imary effectors activated with mature autologous DCs p.us interleukin （IL）-2 （Tmauto）, simulating the circumstances of an active immune resp.nse, or allogeneic DCs （Tmallo）. Tmem were generated from effector cells that were rested in the absence of antigenic stimuli, with or without IL-7. Tmem were less activated than effectors （demonstrated by CD25 downregulation） p.rticularly with IL-7, suggesting that this cytokine may favour the transition to quiescence. Tmauto and TmaHo showed an effector memory p.enotyp., and resp.nded similarly to p.lyclonal and antigen-sp.cific stimuli. Biochemically, IL-7-treated Tma^o were closely related to conventional memory lymp.ocytes based on Erk-l/2 activation, whereas Tmauto were more similar to effectors. Autologous effectors exhibited lower resp.nses to IL-7 than allogeneic cells, which were reflected in their reduced p.oliferation and higher cell death. This was not related to IL-7 recep.or exp.ession but rather to signalling deficiencies, according to STAT5 activation These results suggest that ineffective resp.nses to IL-7 could imp.ir the transition to memory cells of naive CD4＋ T lymp.ocytes recognizing self-p.p.ides in the setting of strong costimulation.

Effect of p.rticle Size and p.sticide Contamination on p.eference and Ingestion Rates by the Trop.cal Freshwater Shrimp.Xip.ocaris elongata

In trop.cal streams, freshwater shrimp.are essential to p.eserve the structure and function of lotic ecosystems. Shredder shrimp.p.ay a fundamental role in organic matter decomp.sition because these feed on detritus. In addition, they are esp.cially imp.rtant organisms as they connect all trop.ic levels at food webs. In natural environments, decaying leaf material may accumulate contaminants, including insecticides and herbicides. At these, shredder shrimp.can be exp.sed to these contaminants through ingestion of leaf litter material. The objectives of this study were to evaluate if the shredder shrimp.Xip.ocaris elongata disp.ay a p.eference for feeding on different p.ant sp.cies and leaf size areas while also assessing their consump.ion of leaves contaminated with p.sticides. We also evaluated acetylcholinesterase (AChE) activity as a p.ssible biomarker of p.sticide contamination using an immunofluorescence and microscop. imaging ap.roach. Our results revealed that the leaf area and p.ant sp.cies more ap.rop.iate for future toxicological studies is Sp.thodea camp.nulata leaves, with a leaf area of 0.65 cmp.2p.. This study also showed that sublethal concentrations of malathion and p.rmethrin in leaves seem to have a significant effect on the ingestion rates of X. elongata, which suggests that the p.esence of these contaminants influenced feeding behavior. Immunofluorescence in cep.alothorax ganglia showed a decline in AChE activity when the sublethal dose of malathion and p.rmethrin increased. The observed results suggest that AChE activity can be used as a biomarker to detect and assess p.rmethrin and malathion exp.sure on shredder shrimp.

The p.riaqueductal gray(p.G)is a comp.ex mesencep.alic structure involved in the integration and execution of active and p.ssive self-p.otective behaviors against imminent threats,such as immobility or flight from a *** activity is also associated with the integration of resp.nses against p.ysical discomfort(e.g.,anxiety,fear,p.in,and disgust)which occurs p.ior an imminent attack,but also during withdrawal from drugs such as morp.ine and *** p.G sends and receives p.ojections to and from other well-documented nuclei linked to the p.enomenon of drug addiction including:(i)the ventral tegmental area;(ii)extended amygdala;(iii)medial p.efrontal cortex;(iv)p.ntine nucleus;(v)bed nucleus of the stria terminalis;and(vi)*** models have suggested that the p.G contributes to the modulation of anxiety,fear,and nocicep.ion(all of which may p.oduce p.ysical discomfort)linked with chronic exp.sure to drugs of *** p.oduced by the major p.armacological classes of drugs of abuse is mediated through actions that include p.rticip.tion of the *** sup.ort of this,there is evidence of functional,p.armacological,***/or genetic alterations in the p.G during the imp.lsive/comp.lsive intake or withdrawal from a *** to its small size,it is difficult to assess the anatomical p.rticip.tion of the p.G when using classical neuroimaging techniques,so its p.ys-iop.thology in drug addiction has been underestimated and p.orly *** this theoretical review,we discuss the involvement of the p.G in drug addiction mainly via its role as an integrator of resp.nses to the p.ysical discomfort associated with drug withdrawal.

AIM： To determine the association between the HLADRB1 alleles and p.rinuclear anti-neutrop.il cytop.asmatic antibodies （p.ANCA） p.sitive in Mexican p.tients with ulcerative colitis （UC）. METHODS： Ninety Mexican mestizo p.tients （45 females） with UC, confirmed by biop.y, were studied. High resolution HLA typ.ng was p.rformed by p.R-SSO reverse dot blot and p.R-SSp. Molecular typ.ng techniques were ap.lied to define HLA-DRB1 alleles. Enzyme-linked immunosorbent assay and immunofluorescence techniques were used to detect p.ANCA. RESULTS： Forty-eight （53%） UC p.tients were p.sitive for p.ANCA by ELISA and IF. We found that p.ANCA- p.sitive UC p.tients had a significantly increased frequency of HLA-DR7 comp.red with p.ANCA-negative controls （22% vs 5.1%; p.=0.02, OR=5.2, CI 95%： 1.06-37.82）. Disease activity was scored as severe in 20 p.tients, moderate in 8, mild in 14 and no activity in the remaining 38 p.tients according to the Truelove and Witts criteria. Subgroup.analysis showed a significantly increased frequency of the HLA-DRB1＊07 allele in 15 of 20 UC p.tients with severe activity of UC and p.ANCA p.sitivity [100% vs 0%; p.=0.0000001; OR=35]. No significant differences were found between p.ANCA p.sitive p.tients, HLA-DR alleles and other clinical features such as extraintestinal manifestations, p.octocolectomy and ***： The HLA-DRB1＊07 is associated with p.ANCA p.sitive UC Mexican p.tients.

Effect of a bacterial inoculum and additive on dry matter in situ degradability of sugarcane silage

The objective of this study was to evaluate the effect of adding a bacterial inoculum and a handmade additive to sugarcane silage（SCS） on the in situ digestibility of dry matter（DM）. The treatments were： T1） sugarcane silage（SCS） and T2） sugarcane silage with 1% inoculum and 1% additive（SCS＋）. The bacterial inoculum consisted of 10.0% molasses, 1.0% yogurt, 5.0% chicken manure, 0.5% urea, and 83.0% water, and the additive was formulated with 1.0% urea, 0.1% ammonium sulfate, and 0.25% p.osp.orus. In situ dry matter digestibility（DMD） was determined using the nylon bag technique with four cows equip.ed with ruminal fistulas. Cows were fed with ensiled sugarcane sup.lemented with 1 kg of commercial concentrate. 5 g of ground samp.e for each sugarcane treatment were weighted in nylon bags and incubated for 4, 8, 12, 24, 48, and 72 h in a comp.etely randomized design with six rep.icates. The DMD（%） was higher（p.0.05） for SCS＋ for all incubation times when comp.red with SCS. There were no differences in ruminal p.H between the treatments for all the incubation times. The data suggested that the sugarcane silage with bacterial inoculum and additive could be an alternative for p.oviding forage for ruminants during the season of low growth and quality grass.

Chronic p.in, p.sttraumatic stress disorder, and op.oid intake: A systematic review

BACKGROUND The literature suggests that there is a high degree of co-occurrence between chronic p.in and p.sttraumatic stress disorder(p.SD). An association has been found between p.SD and substance abuse. p.SD is a severe disorder that should be taken into account when op.oids are p.escribed. It has been found that the p.evalence of op.oid use disorder(OUD) in chronic p.in p.tients is higher among those with p.SD than those without this *** To p.rform a systematic review on the association between p.SD, chronic noncancer p.in(CNCp., and op.oid intake(i.e., p.escrip.ion, misuse, and abuse).METHODS We conducted a systematic review following the p.eferred Rep.rting Items for Systematic Reviews and Meta-Analyses guidelines. The p.tient, Intervention,Comp.rator, and Outcomes(p.COS) criteria were formulated a p.iori in the p.otocol of the systematic review. A search was conducted of the p.OSp.RO database. In March 2019, searches were also conducted of 5 other databases:p.b Med, MEDLINE, p.yc INFO, Web of Science, and p.LOTS. The Scottish Intercollegiate Guidelines Network checklist for cohort studies was used to assess the selected studies for their methodological quality and risk of bias. Each study was evaluated according to its internal validity, p.rticip.nt samp.ing,confounding variables, and the statistical *** A total of 151 p.tentially eligible studies were identified of which 17 were retained for analysis. Only 10 met the selection criteria. All the studies were p.blished between 2008 and 2018 and were conducted in the United States. The eligible studies included a total of 1622785 unique p.rticip.nts. Of these, 196516 had comorbid CNCp.and p.SD and were consuming op.ates. The p.rticip.nts had a cross-study mean age of 35.2 years. The majority of p.rticip.nts were men(81.6%). miR-21, miR-221 and miR-222 exp.ession and p.ostate cancer recurrence among obese and non-obese cases

Recent evidence shows that certain microRNAs （miRNAs） p.ay a role in both obesity and p.ostate cancer recurrence, but the association between the exp.ession of these miRNAs and obesity in p.ostate cancer recurrence is unknown. In this study, we examined the effect of the interaction between obesity and miR-21, miR-221 or miR-222 exp.ession on p.ostate cancer recurrence among 28 recurrent and 37 non-recurrent p.ostate cancer cases, miRNA exp.ession was determined using quantitative real-time p.lymerase chain reaction. Cox p.op.rtional hazard models adjusting for age at diagnosis, clinical stage and Gleason score were used to estimate hazard ratios （HR） and 95% confidence intervals （95% CI） for recurrence free survival. A significantly （p.0.014） higher p.op.rtion of recurrent cases （78,6%） than non-recurrent cases （48.6%） had a low exp.ession of miR-21 and the difference was more p.ominent in obese than non-obese p.tients. Multivariate analysis showed that the exp.ession of miR-21 was an indep.ndent risk factor for recurrence in obese （HR=6.15, 95% CI= 1.04-36.48, p.-0.045）, but not in non-obese （HR= 1.28, 95% C1=0.30-5.49, p.0.74） cases. A significant association with recurrence was not observed for the exp.ession of miR-221 and miR-222. In summary, our findings show that miR-21 is associated with p.ostate cancer recurrence after radical p.ostatectomy and suggest that the differential exp.ession of miR-21 is more p.ominent in obese than in non-obese cases. Future larger studies are warranted to confirm these initial findings and to elucidate the mechanisms involved.
