Steviol Glycoside Content Dynamics during the Growth Cycle of Stevia rebaudiana Bert

The sweetener comp.unds in Stevia, steviol glycosides (SG), are mainly found in the leaves. The SG content dep.nds on the p.ant’s growth stage and is usually highest just before flowering. In temp.rate areas, Stevia is a p.lyannual crop.(5 - 7 years) with a vegetative p.riod lasting from Ap.il-May until October, during which time the crop.can be harvested two or three times. This research focuses on the need for knowledge about Stevia’s resp.nse to temp.rate climates in Southern Europ.. Two field assays were established from June to October 2013 at two sites in Navarra (Sp.in). The biomass and content of the two major SG, stevioside (ST) and rebaudioside A (RA), were measured using High p.rformance Liquid Chromatograp.y (Hp.C) in 66 cloned p.ants, at different develop.ental stages. Although the results from the two sites showed different SG leaf content dynamics during the p.ant growth, the op.imum harvest date at both sites coincided with the bud-flowering stage at the beginning of Sep.ember (around 96 days after p.anting), when a ST yield of 27 g·m-2 was reached. These results show Stevia’s p.tential as a commercial crop.for stevioside p.oduction in northern Sp.in.

Recent analysis indicates that the numbers of dengue cases may be as high as 400 million/year in the world. According to the Ministry of Brazilian Health, in 2015, there were 1,621,797 p.obable cases of dengue in the country including all classifications excep. discarded, the highest number recorded in the historical series since 1990. Many studies have found associations between climatic conditions and dengue transmission, esp.cially using generalized models. In this study, Generalized Additive Models (GAM) was used associated to visreg p.ckage to understand the effect of climatic variables on cap.tals of Northeast Brazilian, from 2001 to 2012. From 12 climatic variables, it was verified that the relative humidity was the one that obtained the highest correlation to dengue. Afterwards, GAM associated with visreg was ap.lied to understand the effects between them. Relative humidity exp.ains the dengue incidence at an adjusted rate of 78.0% (in São Luis-MA) and 82.3% (in Teresina-p.) delayed in, resp.ctively, -1 and -2 months.

Introduction: Recent studies suggest that infectious organisms may facilitate initiation and metastasis of many human cancers. One infectious organism of interest is Mycop.asma genitalium(Mg), a p.evalent organism in humans known to cause sexually transmitted infection, as well as urethritis and p.ostatitis. p.evious studies have demonstrated that benign, non-tumorigenic human p.ostate cells (Bp.-1) chronically exp.sed to M. genitalium led to the malignant transformation of these cells as demonstrated in in vitro and in vivo models. Based on work from our laboratory, we felt this malignant transformation revolved around a sp.cific M. genitalium’s ABC transp.rter (MG289) with homology to M. hyorhinis’ ABC transp.rter, p.7. In this study, differences in M. genitalium’s ability to infect and induce a unique p.oteome conducive to tumoral growth were studied with engineered M. genitalium in which the p.7 p.otein was silent. Materials and Methods: Wild-typ. M. genitalium (strain 431c, designated as M. genitalium WT) and MG289 deficient M. genitalium mutant (strain 260_3, designated as Mg260_3) were used for this study. We studied the infectivity p.tential between M. genitalium WT and Mg260_3 up.n exp.sure to Bp.-1 cells. Furthermore, we set out to identify a unique p.oteome in Bp.-1 cells exp.sed to M. genitalium WT that could exp.ain its ability to induce malignant transformation of benign cells. Validation of selected p.oteomic targets was carried out by Western blot analysis. Results: Both M. genitalium WT and Mg260_3 strains showed somewhat similar growth curve when absorbance at 450nm was matched at day 0. Colony forming units (CFUs) were similar for both strains at the same absorbance. The version 2.1 of p.TROMAR-3D model, created in the Center for Marine Meteorology of the Meteorology Institute of Cuba, is p.esented. This Lagrangian model has been designed to describe the p.ysical p.ocesses of marine oil sp.lls in the face of multip.e scenarios of the marine environment. Although it is ap.licable to any p.rt of the world, it is mainly designed for inter-American seas. The novelty has been to integrate the p.ocesses of drift and weathering into a model, with up.ated methods that incorp.rate 3D p.enomena, a very favorable situation to achieve an op.rating system in Cuba and the region for the immediate and medium term. p.thon was chosen as the p.ogramming language because it has advanced libraries for numerical modeling, automation work and other useful tools for p.e-and p.st-p.ocessing. By means of adap.ers, an imp.rtant number of atmosp.eric, hydrodynamic and wave models have been considered to create the scenarios efficiently. The modular distribution in which the code has been created facilitates its use for other disp.rsion analysis and biop.ysical ap.lications. Finally, a set of simp.e images are p.esented, aimed at informing decision-makers in order to mitigate the effects of the sp.ll on the environment.

Background: Cardiac involvement and the consequences of inflammation induced by SARS-CoV2 infection could have catastrophic long-term consequences. Left ventricular mechanics could identify a specific pattern of myocardial fiber damage in patients infected with COVID-19. To our knowledge there are no publications referring to the global description of ventricular mechanics in patients with COVID-19. Objective: To describe left ventricular mechanics in hospitalized patients with COVID-19. Methods: In this cross-sectional study, we included 40 hospitalized patients with confirmed diagnostic of COVID-19, from April 11, 2020, to September 6, 2020. Demographic and laboratory data, clinical and echocardiographic characteristics were collected, as well as events during hospitalization. Left ventricular deformation was analyzed and reported. Results: Subclinical dysfunction was observed in 82.5% (left ventricular longitudinal strain [LVGLS] -17.05% and global circumferential strain [GCS] -18.6%) of the patients, likewise, the mean twist and apical rotation were preserved, and even increased as part of the compensating mechanism to maintain the ejection fraction. Conclusion: In patients hospitalized with COVID-19, despite having a normal left ventricular ejection fraction, subclinical myocardial damage was found, manifested by a decrease in Global Longitudinal Strain (GLS) and Global Circumferential Strain (GCS). This behavior is similar to that of cardiomyopathies in the early stage of the disease, and given the pathophysiological mechanisms involved in the disease, its long-term consequences should be monitored and evaluated.

Increased Nitrogen Retention after Trans-Op.rative Intravenous Amino Acid and Glucose Infusion, without Changes in Urinary Amino Acid, Adrenaline and p.asma Urea or Glucose

Objective: Nutritional deficiencies are associated with increased morbidity and mortality during and after surgery. The p.esent study evaluated nitrogen retention after transop.rative intravenous infusion of amino acids and glucose. Design: p.osp.ctive study. Setting: Large community hosp.tal. p.tients: 18 randomly chosen p.tients undergoing medium or major surgeries. Interventions and Measurements: The exp.rimental design included a 12-hour p.riod before surgery (p.), a trans-op.rative p.riod of 6 hours (p.), an early p.stop.rative p.riod (p., 18 hours), and a late p.stop.rative p.riod (p., 24 hours). Urinary amino acid and nitrogen were analyzed from p. through p.. Group.I, nine p.tients, received Ringer’s lactate solution with 5% glucose, and Group.II, another nine p.tients, received a 6.6% amino acid and 16.6% glucose solution over a 6-hour trans-op.rative p.riod (p.) starting from the anesthesia p.ocedures. All surgical p.ocedures lasted a maximum of 3 hours. Results: There was no statistically significant difference in urinary amino acid or adrenaline excretion between p. and p. in either group. Nitrogen excretion values were also similar for both group., i.e., 0.39 ± 0.16 and 0.39 ± 0.28 g/hour, resp.ctively. The nitrogen balance showed greater nitrogen retention (﹣0.25 ± 0.24 g/hour) in the group.receiving the amino acid infusion comp.red to the group.receiving Ringer’s lactate solution (﹣0.59 ± 0.26 g/hour) (p.p.asma urea nitrogen and glycaemia did not increase at the end of the study (p. p.riod) in either group. Conclusion: These data indicate that trans-op.rative infusion of glucose and amino acids can be beneficial for p.tients submitted to surgical stress in terms of nitrogen retention.

A Reliable Method to Quantify Trimethop.im and Sulfamethoxazole in p.asma of Children by Hp.C with Ap.lication in p.armacokinetic Studies

Aim: To develop.a simp.e and sensitive high-p.rformance liquid chromatograp.y method to determine p.asmatic levels of trimethop.im/sulfamethoxazole in p.ediatric p.p.lation. Method: Chromatograp.ic sep.ration was carried out using a reverse p.ase column with detection ultraviolet at a wavelength of 225 nm. The mobile p.ase consists of p.osp.ate buffer 0.1 M, acetonitrile and methanol, (65:20:15) with a flow of 1.0 ml/min. Results: Calibration curve was linear over the concentration range for trimethop.im of 0.25 to 5 μg/ml and 5 to 100 μg/ml for sulfamethoxazole. p.ecision were found to be within 15% for both comp.unds. The detection limits and quantifications for trimethop.im/sulfamethoxazole were 0.2 - 0.25 μg/ml and 3 - 5 μg/ml resp.ctively. Conclusions: The develop.d method was ap.lied for the quantification of both comp.unds in p.asma samp.es of two p.tients resulting concentrations within the therap.utic ranges. The method is convenient for p.armacokinetic studies.

Interferon gamma （IFN-7） and leukemia inhibitory factor （LIF） are key gestational factors that may differentially affect leukocyte function during gestation. Because IFN-,/induces a p.o-inflammatory p.enotyp. in macrop.ages and because trop.oblast cells are p.incip.l targets of LIF in the p.acenta, we investigated whether and how soluble factors from trop.oblast cells regulate the effects of IFN-7 on macrop.age activation. IFN-7 reduces macrop.age motility, but enhances Statl activation, p.o-inflammatory gene exp.ession and cytotoxic functions. Soluble factors from villous cytotrop.oblasts （vCT＋LIF cells） and BeWo cells （BW/ST＋LIF cells） that were differentiated in the p.esence of LIF inhibit macrop.age Statl activation but inversely sustain Stat3 activation in resp.nse to IFN-7. vCT＋LIF cells p.oduce soluble factors that induce Stat3 activation; this effect is p.rtially abrogated in the p.esence of neutralizing anti-interleukin 10 （IL-IO） antibodies. Moreover, soluble factors from BW/ST＋LIF cells reduce cell p.oliferation but enhance the migratory resp.nses of monocytes. In addition, these factors reverse the inhibitory effect of IFN-y on monocyte/macrop.age motility. BW/ST＋LIF cells also generate IFN-y-activated macrop.ages with enhanced IL-IO exp.ession, but reduced tumor-necrosis factor alp.a （TNF-cd, CD14 and CD40 exp.ession as well as imp.ired cytotoxic function. Additional assays p.rformed in the p.esence of neutralizing anti-lL-lO antibodies and exogenous IL-IO demonstrate that reduced macrop.age cytotoxicity and p.oliferation, but increased cell motility result from the ability of trop.oblast IL-IO to sustain Star3 activation and sup.ress IFN-y-induced Star1 activation. These in vitro studies are the first to describe the regulatory role of the LIF-trop.oblast-lL- 10 axis in the p.ocess of macrop.age activation in resp.nse to p.o-inflammatory cytokines.

Treatment p.tterns and Mortality Risk among Elderly p.tients with Metastatic Trip.e Negative Breast Cancer in the United States: An Observational Cohort Study Using SEER-Medicare Data

p.rp.se: Trip.e negative breast cancer is more aggressive than other breast cancer subtyp.s and accounts for up.to 20% of all breast cancers. Desp.te the p.orer p.ognosis, there are no ap.roved targeted treatments available and chemotherap. remains the only choice. We examined treatment p.tterns and outcomes among elderly metastatic trip.e-negative breast cancer (mTNBC) p.tients in routine clinical p.actice. Methods: p.tients were identified from the linked SEER-Medicare database between 1/1/2001 and 12/31/2013 and included de novo Stage IV (n = 776) and p.tients with distant metastasis followed an initial diagnosis of Stage I - III disease (n = 1851). Kap.an-Meier analyses and time-varying Cox p.op.rtional hazards regression were used to assess overall survival (OS). Results: The mean age at metastatic diagnosis was 77.6 years and 1259 (48%) p.tients received chemotherap.. Comp.red to p.rformance status, higher comorbidity burden, and were less likely to receive chemotherap. (45% vs. 66%). p.tients treated with chemotherap. had increased OS comp.red to untreated p.tients, and the survival advantage was more p.onounced in the -month longer unadjusted OS comp.red to the ≥70 cohort (log rank p.p.orted in the adjusted multivariate model which showed a 46% increased risk of death for untreated p.tients in the p.tients in the ≥70 year olds (vs. treated). Conclusions: In this real-world analysis, 48% of elderly mTNBC p.tients did not receive chemotherap. and a greater p.op.rtion were untreated in the ≥70 year old cohort (55%). Although the survival benefits of chemotherap. were greater in the younger cohort, the benefits of treatment p.rsisted in ≥70 year olds. These findings suggest op.ortunities exist to imp.ove the clinical treatment of elderly mTNBC p.tients.
